Murine typhus: a re-emerging rickettsial zoonotic disease.

Murine typhus, caused by Rickettsia typhi, is re-emerging in many parts of the world. The disease is also called endemic typhus to differentiate from epidemic typhus (caused by Rickettsia prowazekii), and sometimes also named flea-borne typhus. Occasionally, literature sources will include Rickettsia felis as a causative agent of flea-borne typhus, but illnesses caused by R. felis are actually flea-borne spotted fever. Murine typhus occurs in warm, coastal areas worldwide. In the United States, most cases are reported from California, Texas, and Hawaii. Murine typhus is usually a self-limited

Population survey of Egyptian arthropods for rickettsial agents.

Between June 2002 and July 2003, 987 fleas, representing four species, and 1019 ticks, representing one argasid and eight ixodid species, were collected from Egyptian animals. These arthropods were tested for rickettsial agents using polymerase chain reaction. DNAs from Anaplasma and Ehrlichia spp. were detected in 13 ticks. Previously undescribed Bartonella spp. were detected in 21 fleas. Coxiella burnetii was detected in two fleas and 20 ticks. Rickettsia typhi was detected in 27 fleas from 10 cities. Spotted fever group rickettsiae were detected in both fleas and ticks and included Ricketts

Sennetsu Neorickettsiosis, Spotted Fever Group, and Typhus Group Rickettsioses in Three Provinces in Thailand.

We estimated the seroprevalence and determined the frequency of acute infections with Neorickettsia sennetsu, spotted fever group rickettsiae, Rickettsia typhi, and Orientia tsutsugamushi among 2,225 febrile patients presenting to community hospitals in three rural Thailand provinces during 2002-2005. The seroprevalence was 0.2% for sennetsu neorickettsiosis (SN), 0.8% for spotted fever group (SFG) rickettsiae, 4.2% for murine typhus (MT), and 4.2% for scrub typhus (ST). The frequency of acute infections was 0.1% for SN, 0.6% for SFG, 2.2% for MT, and 1.5% for ST. Additional studies to confirm

Development and characterization of high-titered, group-specific fluorescent-antibody reagents for direct identification of rickettsiae in clinical specimens.

Rabbits were inoculated with purified antigen preparations of Coxiella burnetii and representative species of the spotted fever and typhus groups of rickettsiae. Their antibody responses were monitored by complement fixation tests; high-titered antisera were fractionated with ammonium sulfate and then labeled with fluorescein isothiocyanate by the dialysis method. The conjugates had homologous 3+ staining titers of 1:256 to 1:2,048 and did not exhibit nonspecific staining. The Rickettsia rickettsii, R. conorii, and R. akari conjugates reacted only with rickettsiae of the spotted fever group; t

Rickettsioses in Colombia during the 20th century: A historical review.

Rickettsioses are vector-borne zoonotic diseases whose etiological agents are bacteria of the genera Rickettsia and Orientia. In Colombia, literature published on rickettsioses during the 20th century is limited and disperse; to date, there are no detailed publications that compile the affected regions or the involved Rickettsia species in each outbreak. Since early works of Luis Patiño-Camargo, the study of rickettsioses gained major importance with the subsequent description of significant outbreaks in some departments of the country. During the 1940s, many articles were published reporting

Prevalence of Rickettsia and Bartonella species in Spanish cats and their fleas.

The aim of this study was to determine the prevalence of Bartonella henselae, Rickettsia felis, and Rickettsia typhi in fleas and companion cats (serum and claws) and to assess their presence as a function of host, host habitat, and level of parasitism. Eighty-nine serum and claw samples and 90 flea pools were collected. Cat sera were assayed by IFA for Bartonella henselae and Rickettssia species IgG antibodies. Conventional PCRs were performed on DNA extracted from nails and fleas collected from cats. A large portion (55.8%) of the feline population sampled was exposed to at least one of the

Spotted fever group and typhus group rickettsioses in humans, South Korea.

The presence of the nucleic acid of the spotted fever group (SPG) and typhus group (TG) rickettsiae was investigated in 200 serum specimens seropositive for SFG rickettsiae by multiplex-nested polymerase chain reaction with primers derived from the rickettsial outer membrane protein B gene. The DNA of SFG, TG, or both rickettsiae was amplified in the 24 serum specimens, and sequence analysis showed Rickettsia conorii, R. japonica, and R. felis in the specimens. R. conorii and R. typhi were found in 7 serum specimens, which indicated the possibility of dual infection in these patients. These fi

First description of a clinical case of murine typhus in Campeche, Mexico.

Murine typhus is a flea-borne disease caused by Rickettsia typhi, which was first detected in Mexico in 1927. It was not until 1996 that the first systematized study involving this pathogen was conducted in two coastal states of Mexico. We now report the first confirmed case of murine typhus in the state of Campeche, which occurred in a male patient who exhibited fever, thrombocytopenia, hyperbilirubinemia, and a rash. Furthermore, the patient reported having had previous contact with Rickettsia reservoirs.

Postinfectious optic neuropathy in endemic typhus.

Endemic typhus (Rickettsia typhi), also known as murine typhus, is a flea-borne bacterial disease rarely found in most of the developed world. Known ocular manifestations linked to endemic typhus include mild vitritis, retinal lesions, and retinal vascular leakage. Optic neuropathy, however, is rarely associated with R. typhi, and postinfectious optic neuropathy is even less common. To highlight this unusual complication, we report a patient who developed postinfectious optic neuropathy a few weeks after he was successfully treated for endemic typhus.

Flea-borne rickettsioses: ecologic considerations.

Ecologic and economic factors, as well as changes in human behavior, have resulted in the emergence of new and the reemergence of existing but forgotten infectious diseases during the past 20 years. Flea-borne disease organisms (e.g., Yersinia pestis, Rickettsia typhi, R. felis, and Bartonella henselae) are widely distributed throughout the world in endemic-disease foci, where components of the enzootic cycle are present. However, flea-borne diseases could reemerge in epidemic form because of changes in vector-host ecology due to environmental and human behavior modification. The changing ecol

Murine typhus: endemic Rickettsia in southwest Texas.

Murine Typhus is a zoonosis caused by the organism Rickettsia typhi and is transmitted to humans by fleas. It is endemic in several areas of Texas, California and Hawaii where the vector is supported predominantly by rodents in addition to opossums, domestic and feral cats and domestic dogs. We present a typical case in an adult from Corpus Christi, located in one of the four endemic areas in Texas. Included is an overview of the organism's pathogenicity and our host responses, both influencing the milder clinical course seen with this species of Rickettsia.

Sensorineural hearing loss complicating severe rickettsial diseases: report of two cases.

Sub-acute focal neurological manifestations are reported rarely in systemic rickettsial diseases and are considered to be secondary to immune-mediated mechanisms. We present two cases of transient sensorineural hearing loss complicating the course of severe rickettsial diseases caused by Rickettsia typhi and Rickettsia conorii, respectively. The diagnosis was based on the presence of high IgM antibodies and the prompt response to doxycycline treatment. In both cases, hearing loss presented during convalescence and resolved automatically without administration of specific treatment.

SFG and TG seropositivity in Humans suspected of TBD in Yucatan, Mexico.

Since 1996, the incidence of rickettsiosis has been increasing in Yucatán, Mexico, but recent prevalence data are lacking. This study aimed to determine exposure to the Spotted Fever Group (SFG) and Typhus Group (TG) in human serum samples suspected of tick-borne diseases (TBD) between 2015 and 2022. A total of 620 samples were analysed using indirect immunofluorescence assay (IFA) to detect IgG antibodies against SFG (Rickettsia rickettsii) and TG (Rickettsia typhi), considering a titer of ≥64 as positive. Results showed that 103 samples (17%) were positive for R. rickettsii and 145 (24%) for

Murine Typhus: A Life-Threatening Presentation of a Case in Galveston, Texas.

BACKGROUND Murine typhus is a rare bacterial infection caused by Rickettsia typhi, which is transmitted from rodents to humans through the infected Xenopsylla cheopis flea. The disease presentation is often non-specific, leading to unnecessary tests, and a delay in diagnosis and treatment. CASE REPORT A report is presented of a 22-year-old, previously healthy man, who presented with several symptoms and signs that increased in severity, requiring admission to the medical intensive care unit (MICU). After an extensive bacterial and viral laboratory workup, IgM and IgG titers confirmed the diagn

Rickettsial infection in murine models activates an early anti-rickettsial effect mediated by NK cells and associated with production of gamma interferon.

Natural killer (NK) cell activity was significantly increased on days 2-6 of infection in the Rickettsia conorii-infected C3H/HeN mice and on day 2 in the Rickettsia typhi-infected C57BL/6 mice. Depletion of NK cell activity utilizing anti-NK1.1 monoclonal antibody enhanced the susceptibility of normally resistant C57BL/6 mice to infection with R. typhi, and depletion of NK cell activity with antibody to asialo GM1 enhanced the susceptibility of C3H/HeN mice to infection with R. conorii. Serum gamma interferon was increased in R. conorii-infected C3H/HeN mice compared with NK cell-depleted, in

Rickettsiae and hosts.

Protective immunity to Rickettsia tsutsugamushi in a murine model is dependent upon the development of cell mediated immunity, as demonstrated by lymphocyte transfer, production of lymphokines and -interferon by thymus-derived lymphocytes, activation of macrophages by lymphokines and demonstration of delayed-type hypersensitivity response following exposure to these organisms. Infection of mice with small numbers of Rickettsia typhi inoculated by a peripheral route leads to a more complex pattern of immune development, with a distinction between resistance to local and systemic infection. rRNA intergenic spacer regions for phylogenetic analysis of Rickettsia species.

Species of the genus Rickettsia are responsible for several human diseases, namely epidemic typhus, Rocky Mountain spotted fever, and tick-borne typhus transmitted by arthropod vectors. The rrl-rrf intergenic spacer region (rrl-rrf ITS) was sequenced for 12 Rickettsia strains, including R. typhi, 6 untested species, R. aeschlimannii, Bar29, R. helvetica, R. honei, R. massilae, and R. slovaca as well as 5 Portuguese isolates. Phylogenetic trees inferred from rrl-rrf spacer sequences using maximum-parsimony and distance methods provided largely congruent tree topologies, supported by significant

Short report: surveillance of rickettsial infections in Indonesian military personnel during peace keeping operations in Cambodia.

Indonesian peacekeepers in Cambodia provided a unique study population to estimate the threat of rickettsial exposure to Rickettsia typhi (murine typhus), Orientia tsutsugamushi, (scrub typhus), and R. conorii (spotted fever) for the region. Prescreening prevalence measure showed a large proportion (36%) of soldiers with antibodies to R. typhi. Predeployment prevalence for antibodies to O. tsutsugamushi was 8%, with no evidence of background R. conorii infections. Actual seroconversions of R. typhi (3) and O. tsutsugamushi (1), attributed to exposure(s) in Cambodia, translated into annualized

Rickettsia Lipid A Biosynthesis Utilizes the Late Acyltransferase LpxJ for Secondary Fatty Acid Addition.

Members of the Rickettsia genus are obligate intracellular, Gram-negative coccobacilli that infect mammalian and arthropod hosts. Several rickettsial species are human pathogens and are transmitted by blood-feeding arthropods. In Gram-negative parasites, the outer membrane (OM) sits at the nexus of the host-pathogen interaction and is rich in lipopolysaccharide (LPS). The lipid A component of LPS anchors the molecule to the bacterial surface and is an endotoxic agonist of Toll-like receptor 4 (TLR4). Despite the apparent importance of lipid A in maintaining OM integrity, as well as its inflamm
