Sample size and classification error for Bayesian change-point models with unlabelled sub-groups and incomplete follow-up.

Many medical (and ecological) processes involve the change of shape, whereby one trajectory changes into another trajectory at a specific time point. There has been little investigation into the study design needed to investigate these models. We consider the class of fixed effect change-point models with an underlying shape comprised two joined linear segments, also known as broken-stick models. We extend this model to include two sub-groups with different trajectories at the change-point, a change and no change class, and also include a missingness model to account for individuals with incom

Endogenous testosterone concentration, mental rotation, and size of the corpus callosum in a sample of young Hungarian women.

In the present study brain laterality, hemispheric communication, and mental rotation performance were examined. A sample of 33 women were tested for a possible linear relationship of testosterone level and mental rotation with structural background of the brain. Subjects with a smaller splenial area of corpus callosum tended to have lower levels of testosterone (r =.37, p<.05). However, there were no significant differences in mean scores of mental rotation of object and hand between groups with high and low levels of testosterone. There was a significant difference in relative size of the 6t

Planning genetic studies on primary adult-onset dystonia: sample size estimates based on examination of first-degree relatives.

Primary adult-onset dystonia is thought to be partly genetic, but families large enough for a genome wide search are difficult to find. We examined the first-degree relatives of 76 primary adult-onset dystonia patients to assess the feasibility of model-free nonparametric methods that allow either screening of candidate loci (case-control design, transmission disequilibrium test [TDT], and sibling-TDT [S-TDT]) or identification of novel genes (affected sib-pair [ASP] method). Among the examined relatives, 1/34 parents, 13/149 siblings and 10/125 offspring were affected by adult-onset dystonia.

Bayesian sample size determination for prevalence and diagnostic test studies in the absence of a gold standard test.

Planning studies involving diagnostic tests is complicated by the fact that virtually no test provides perfectly accurate results. The misclassification induced by imperfect sensitivities and specificities of diagnostic tests must be taken into account, whether the primary goal of the study is to estimate the prevalence of a disease in a population or to investigate the properties of a new diagnostic test. Previous work on sample size requirements for estimating the prevalence of disease in the case of a single imperfect test showed very large discrepancies in size when compared to methods tha

Sample size estimation for a flexible phase II oncology trial design using two dependent endpoints with nested response metrics.

Phase II oncology trials are often designed using a single primary efficacy endpoint, even when multiple clinically relevant response metrics are available. When two endpoints are based on nested response metrics, such as objective response rate and disease control rate or progression-free survival at two ordered time points, success on the more stringent endpoint implies success on the broader endpoint. This structure induces a known dependence relationship that can be incorporated into sample size estimation and decision-rule construction. We develop a flexible phase II design framework for

Metabolic mechanism analysis of abdominal aortic aneurysm tissues by internal extractive electrospray ionization mass spectrometry with minimal sample size.

Abdominal aortic aneurysm (AAA) is a major clinical challenge, with a mortality rate of over 80% if rupture. Metabolic imbalance is a critical factor in the progression of AAA. Therefore, using modern analytical techniques to study the tissue metabolic characteristics of AAA is crucial for elucidating the pathological mechanisms and identifying novel drug targets. However, existing methodologies often require large sample sizes and involve complex experimental procedures, which limit their applicability in studies with limited sample availability, especially when rodent AAA models are used. Composite endpoint for liver and colon simultaneous surgery: a proposed approach to reduce sample size of future clinical trials.

BACKGROUND: With expanding indications for resection of colorectal liver metastasis (CRLM), simultaneous resections for primary colon cancer (CC) with synchronous colorectal liver metastases (sCRLM) has increased. sCRLM remains debated, however, and only a handful, often underpowered, trials have evaluated this approach. We sought to develop a composite postoperative endpoint (composite endpoint for liver and colorectal simultaneous surgery [CELCSS]) combining colon- and liver-specific complications to reduce sample size requirements in future prospective randomized clinical trials. METHODS:

A digital twin methodology using real patient data for sample size reduction in Alzheimer's disease randomized controlled clinical trials.

INTRODUCTION: Recruitment for Alzheimer's disease randomized controlled trials (RCTs) is difficult and expensive. To reduce RCT sample sizes, our Digital Twin Trial (DTT) methodology combines an interpretable cognitive decline prediction model with prediction-powered inference. METHODS: For DTT participants, our model identifies similar individuals ("Digital Twins") from a retrospective database and uses their cognitive scores to predict decline. Predictions adjust observed scores, reducing variance within treatment groups. We simulated 18-month DTTs and standard RCTs using mixed effects mode

How to use learning curves to evaluate the sample size for malaria prediction models developed using machine learning algorithms.

BACKGROUND: Machine learning algorithms have been used to predict malaria risk and severity, identify immunity biomarkers for malaria vaccine candidates, and determine molecular biomarkers of antimalarial drug resistance. Developing these prediction models requires large training datasets to ensure prediction accuracy when applied to new individuals in the target population. Learning curves can be used to assess the sample size required for the training dataset by evaluating the predictive performance of a model trained using different dataset sizes. These curves are agnostic to the specific p

Assessing new treatments for central sleep Apnoea in heart failure: sample size considerations for reliable detection of safety signals.

PURPOSE: Central sleep apnoea (CSA) and Cheyne-Stokes respiration (CSR) are prevalent in patients with heart failure with reduced ejection fraction (HFrEF) and are associated with increased mortality. Previous trials evaluating adaptive servo-ventilation (ASV) for CSA/CSR in HFrEF have yielded conflicting safety outcomes. This analysis aims to determine the sample size and number of events required to reliably detect safety signals, specifically all-cause mortality, in future trials assessing new treatments for CSA/CSR in HFrEF patients. METHODS: Utilizing data from the SERVE-HF trial, we con

Determining an Appropriate Sample Size for Qualitative Interviews to Achieve True and Near Code Saturation: Secondary Analysis of Data.

BACKGROUND: In-depth interviews are a common method of qualitative data collection, providing rich data on individuals' perceptions and behaviors that would be challenging to collect with quantitative methods. Researchers typically need to decide on sample size a priori. Although studies have assessed when saturation has been achieved, there is no agreement on the minimum number of interviews needed to achieve saturation. To date, most research on saturation has been based on in-person data collection. During the COVID-19 pandemic, web-based data collection became increasingly common, as tradi

Relationship between the inclusion/exclusion criteria and sample size in randomized controlled trials for SARS-CoV-2 entry inhibitors.

The coronavirus disease 2019 (COVID-19) pandemic that has been ongoing since 2019 is still ongoing and how to control it is one of the international issues to be addressed. Antiviral drugs that reduce the viral load in terms of reducing the risk of secondary infection are important. For the general control of emerging infectious diseases, establishing an efficient method to evaluate candidate therapeutic agents will lead to a rapid response. We evaluated clinical trial designs for viral entry inhibitors that have the potential to be effective pre-exposure prophylactic drugs in addition to redu

Impact of metabolic syndrome on short-term outcome of carotid revascularization: a large sample size study in Chinese population.

BACKGROUND: Metabolic syndrome (MetS) is relatively common worldwide and an important risk factor for cardiovascular diseases. It is closely linked to arterial stiffness of the carotid artery. However, the association of MetS with the safety of carotid revascularization has been rarely studied. The aim of this study was to observe the current status of MetS and its components in Chinese carotid revascularized patients, and investigate the impact on major adverse clinical events (MACEs) after carotid endarterectomy (CEA) or carotid artery stenting (CAS). METHODS: From January 2013 to December

Effects of the sample size of reference population on determining BMD reference curve and peak BMD and diagnosing osteoporosis.

UNLABELLED: Establishing reference databases generally requires a large sample size to achieve reliable results. Our study revealed that the varying sample size from hundreds to thousands of individuals has no decisive effect on the bone mineral density (BMD) reference curve, peak BMD, and diagnosing osteoporosis. It provides a reference point for determining the sample size while establishing local BMD reference databases. INTRODUCTION: This study attempts to determine a suitable sample size for establishing bone mineral density (BMD) reference databases in a local laboratory. METHODS: The

Implications for cisgender female underrepresentation, small sample sizes, and misgendering in sport and exercise science research.

A sex-data gap, from testing primarily males, results in a lack of scientific knowledge for other groups (females, transgender individuals). It is unknown whether typical recruitment and participant characterization causes incorrect statistical decisions, and three factors were evaluated: 1) underrepresenting cisgender females, 2) recruiting small sample sizes, 3) misgendering. Data from the National Health and Nutrition Examination Survey (2003-2004) were evaluated for sex differences after removing missing values (N = 3,645; F = 1,763). Disparities were determined by utilizing sample sizes c

DLX6 and MSX1 from saliva samples as potential predictors of mandibular size: A cross-sectional study.

INTRODUCTION: Morphologic features of the mandible are influenced by the genes of each individual. Mandible size is important to orthodontists because the mandible is the mechanism by which the lower face influences facial esthetics and dental function. To date, no biological marker has been identified that indicates eventual mandible size. This study aimed to correlate the expression of DLX5, DLX6, EDN1, HAND2, PRRX1, and MSX1 to mandible size. METHODS: Fifty-nine orthodontic patients aged >6 years who had available cephalometric radiographs were studied. Patients were classified on the basi

Statistical Impact of Sample Size and Imbalance on Multivariate Analysis in silico and A Case Study in the UK Biobank.

Large-scale biobank cohorts coupled with electronic health records offer unprecedented opportunities to study genotype-phenotype relationships. Genome-wide association studies uncovered disease-associated loci through univariate methods, with the focus on one trait at a time. With genetic variants being identifiedfor thousands of traits, researchers found that 90% of human genetic loci are associated with more than one trait, highlighting the ubiquity of pleiotropy. Recently, multivariate methods have been proposed to effectively identify pleiotropy. However, the statistical performance in nat

Gonadectomy and Blood Sampling Procedures in the Small Size Teleost Model Japanese Medaka (Oryzias latipes).

Sex steroids, produced by the gonads, play an essential role in brain and pituitary tissue plasticity and in the neuroendocrine control of reproduction in all vertebrates by providing feedback to the brain and pituitary. Teleost fishes possess a higher degree of tissue plasticity and variation in reproductive strategies compared to mammals and appear to be useful models to investigate the role of sex steroids and the mechanisms by which they act. The removal of the main source of sex steroid production using gonadectomy together with blood sampling to measure steroid levels has been well-estab
