Bioinformatics analysis of CYP1B1 mutation hotspots in Chinese primary congenital glaucoma patients.

Primary congenital glaucoma (PCG) is an inherited blinding eye disease. The CYP1B1 gene was identified as a causal gene for PCG, and many mutations have been found, but no studies have focussed on the molecular epidemiology of CYP1B1 in Chinese populations. We aimed to explore the CYP1B1 mutation hotspots in Chinese PCG patients and the possible impact of these mutations on the protein structure and function. First, we performed a meta-analysis on seven datasets of Chinese populations and found L107V and R390H to be the most common CYP1B1 mutations with allele frequencies of 3.19% and 3.09%, r

In silico screening and molecular dynamics analysis of natural DHPS enzyme inhibitors targeting Acinetobacter baumannii.

Over time, antimicrobial agents are losing their credibility in curbing infections due to the development of resistant pathogen strains. The resistant strains have proven to invade living beings and cause various diseases, leading to deaths at an alarming rate. Acinetobacter baumannii is one such pathogen, and to target it through enzyme inhibition, Dihydropteroate synthase enzyme's active site is virtually screened for antimicrobial agents against in-house libraries of natural molecules from medicinally important plants and Agaricus spp. fungus. Two ligands (MSID_000725 and CID_291096) are fo

Computational evaluation of carnosic acid as a 4EY7 inhibitor for potential anti-Alzheimer therapy.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized in part by cholinergic dysfunction resulting from excessive acetylcholinesterase (AChE) activity. Although currently approved AChE inhibitors provide symptomatic relief, their clinical benefits remain modest and are often associated with adverse effects. Natural products represent an important source of structurally diverse bioactive compounds with potential therapeutic relevance. In this study, we computationally evaluated carnosic acid, a diterpene from Rosmarinus officinalis, as a potential AChE inhibitor usi

Non-fluorescent benzothiadiazole probe with protein-triggered turn-on fluorescence for imaging SARS-CoV-2 spike protein in biological systems.

Herein, we report a non-fluorescent 2,1,3-benzothiadiazole (BTD) derivative, BTD-PhCOOH, as a protein-triggered fluorogenic platform for SARS-CoV-2 spike protein detection. Direct coupling under mild refrigerated conditions afforded the BTD-Spike conjugate, producing an intense green fluorescence signal and demonstrating efficient fluorescence turn-on after protein ligation. Solvent-accessible surface area analysis (SASA) identified Lys529 as the most accessible lysine residue, providing a structural rationale for conjugate formation. In MCF-7 cells, BTD-Spike enabled time-dependent visualizat

Aortogastric Fistula Caused by a Foreign Body in a Hiatal Hernia.

Foreign body ingestion is more common in children than in adults, and sharp foreign body ingestion is extremely rare. We report a 93-year-old woman who was unaware of foreign body ingestion and who presented with sudden hematemesis and circulatory collapse. Computed tomography showed a hiatus hernia and part of the stomach herniating in the posterior mediastinum. Additionally, a needle-shaped object was seen penetrating the aorta through the herniated gastric wall. Therefore, immediate endovascular repair of the aorta was performed. Subsequent endoscopy identified a bamboo stick. Therefore, em

Mutation studies on degradation of 3-phenoxybenzoic acid by Lacticaseibacillus paracasei - human gut microbiome in controlling risk for Parkinson's disease using molecular simulation dynamics.

Parkinson's disease (PD) is a debilitating neurodegenerative disease affecting millions worldwide, especially the elderly. Pesticides, particularly pyrethroids like Cypermethrin, have been linked to the development of PD. Cypermethrin, when ingested, is broken down into 3-phenoxybenzoic acid (3PBA), which can lead to the dysfunction or death of dopaminergic neurons. Catechol-2,3-dioxygenase is an enzyme that breaks down 3PBA into catechol, that can further be processed and excreted by the human body. This enzyme is produced by the bacteria Lacticaseibacillus paracasei, a naturally present huma

In vitro digestion and molecular dynamics reveal mechanisms of digestion resistance in type V resistant starch: Binary versus ternary complexes with lipids and/or proteins.

To elucidate the resistance of type V resistant starch to enzymatic digestion, this study investigated the relationship between the crystalline structure and digestibility of complexes formed with lipids or proteins. The incorporation of octadecanoic acid (OA) and rice protein (RP) facilitated the formation of type V crystalline structures in native rice starch (NRS), increasing its crystallinity in the early stages of digestion. During in vitro digestion, the short-range ordered structure and type V crystal content of the NRS-RP and NRS-OA-RP complexes progressively increased. In contrast, th
