Methods were developed that allow invasion of sporozoites from simian malaria parasite species (Plasmodium cynomolgi, P. knowlesi, P. coatneyi, P. inui, P. gonderi, P. fragile) and development to schizont stages in rhesus and Saimiri monkey hepatocytes. The P. cynomolgi-rhesus monkey model was used to study inhibition of schizont development using monoclonal antibodies (MAbs) produced against the circumsporozoite (CS) protein of various strains and species of malaria parasites. Immunoelectron microscopy, using gold-labelled MAbs and cultured parasites, demonstrated that the CS protein persists

Merozoites are the invasive stage of the malaria parasite, which are released from infected erythrocytes to invade other erythrocytes. Antibody to surface antigens on merozoites may prevent invasion by agglutinating merozoites as they are released from infected erythrocytes or by blocking receptors before contact of merozoites with the host erythrocyte. Monoclonal antibodies were produced to a 140,000-m.w. protein on the merozoite surface. The protein was synthesized by the mature intraerythrocytic parasite, the schizont, as a 143,000-m.w. protein and had a m.w. of 140,000 on the surface of fr

Theileria infections were induced in cattle by feeding ticks on them from 3 sources: (a) adult rhipicephalid ticks obtained from the vegetation in a paddock containing an eland EAO at the Animal Orphanage, Nairobi National Park, Kenya, (b) Rhipicephalus appendiculatus adults fed as nymphs on the same eland, (c) R. pulchellus adults fed as nymphs on an eland W 68 captured in the Machakos district of Kenya. Both eland were harbouring Theileria parasites at the time nymphal ticks were fed. Mild infections were produced when adult ticks from these 3 batches were applied to cattle associated with l

Cystoisospora (Isospora) belli is a coccidian parasite of humans. It can cause serious digestive disorders involving infection of intestines, biliary tract and gallbladder, especially in those with depressed immunity. It has a direct fecal-oral transmission cycle. After ingestion of sporulated oocysts, the parasite multiplies asexually and sexually within host epithelial cells, resulting in unsporulated oocysts that are excreted in feces. The details of asexual and sexual stages are not known and certain inclusions in epithelial cells in biopsy samples have been erroneously identified recently

Of the many tick-borne haemoparasites of sheep and goats, three may be controlled by vaccination. A live virulent blood vaccine for Cowdria ruminantium infection (heartwater) is used widely in southern Africa in an infection and treatment procedure. A live vaccine for Babesia ovis attenuated by passage in splenectomised sheep has been used extensively in Bulgaria with good results. A live vaccine for Theileria hirci, consisting of schizonts propagated in a lymphoid cell culture and attenuated by passage, has been used successfully in the Middle East.

A 40-year-old man was admitted to hospital with a 5 day history of fever, restlessness and altered sensorium. Peripheral blood smears showed a Plasmodium vivax and Plasmodium falciparum mixed infection as revealed by the presence of rings, schizonts and gametocyte forms of the parasites. The patient soon became unconscious due to subdural haematoma (SDH) associated with disseminated intravascular coagulation and thrombocytopenia. Immediate intervention with a right fronto-parieto temporal craniectomy, evacuation of the SDH and intravenous quinine administration resulted in the patient's comple

Malaria is a haemato-protozoan disease which causes thousands of deaths every year. Due to the alarming increase of drug resistant strains of P. falciparum, malaria is now becoming more deadly. Helicases are the most important components of the cellular machinery without which cells are unable to survive. The importance of helicases has been proven in variety of organisms. In this study we have reported detailed biochemical characterization of human homologue of DDX3X from Plasmodium falciparum (PfDDX3X). Our study revealed that PfDDX3X is ATP- dependent DNA helicase whereas in human host it i

Plasmodium knowlesi poses a health threat throughout Southeast Asian communities and currently causes most cases of malaria in Malaysia. This zoonotic parasite species has been studied in Macaca mulatta (rhesus monkeys) as a model for severe malarial infections, chronicity, and antigenic variation. The phenomenon of Plasmodium antigenic variation was first recognized during rhesus monkey infections. Plasmodium-encoded variant proteins were first discovered in this species and found to be expressed at the surface of infected erythrocytes, and then named the Schizont-Infected Cell Agglutination

One of the major barriers in the prevention and control of malaria programs worldwide is the growing emergence of multidrug resistance in Plasmodium parasites, and this necessitates continued efforts to discover and develop effective drug molecules targeting novel proteins essential for parasite survival. In recent years, epigenetic regulators have evolved as an attractive drug target option owing to their crucial role in survival and development of Plasmodium at different stages of its life cycle. PfMYST, a histone acetyltransferase protein, is known to regulate key cellular processes, such a

Artemisinin (ART) resistance has spread through Southeast Asia, posing a serious threat to the control and elimination of malaria. ART resistance has been associated with mutations in the Plasmodium falciparum kelch-13 (Pfk13) propeller domain. Phenotypically, ART resistance is defined as delayed parasite clearance in patients due to the reduced susceptibility of early ring-stage parasites to the active metabolite of ART dihydroartemisinin (DHA). Early rings can enter a state of quiescence upon DHA exposure and resume growth in its absence. These quiescent rings are referred to as dormant ring

Felids are the unique definitive host of Toxoplasma gondii. The intestine of felid is the only site for initiating Toxoplasma gondii sexual reproduction. T. gondii excretes millions of infectious oocysts from the intestine, which are the primary source of infection. There are many difficulties in developing vaccines and drugs to control oocyst excretion due to the lack of an appropriate experimental model. Here, we established an in vitro feline intestinal epithelial cell (IEC) infection system and an efficient animal model of T. gondii Chinese 1 genotype, Wh6 strain (TgCtwh6). The Kunming mic

Thirty calves 12 weeks of age raised under essentially parasite-free conditions were used to determine the effects of Cooperia punctata and Eimeria bovis (in single and combination infections) on mucosal alkaline phosphatase activities at 8 locations in the small intestine. In experiment 1, 5 calves infected with C punctata 3 weeks previously had highly significant (P less than 0.01) reductions in mucosal alkaline phosphatase activities compared with those in 5 noninfected control calves. These reductions were greatest in the 2 locations of the intestine closest to the pylorus. Infected calves

This study presents a photonic crystal fibre based optical sensor with a simple and practical architecture for malaria detection using refractive index variations in red blood cells. The proposed sensor consists of a hollow central core surrounded by five concentric layers of heptagonal cladding, enabling efficient sample infiltration and enhanced light matter interaction. This configuration provides high sensitivity to subtle refractive index changes while maintaining structural simplicity suitable for real world diagnostic deployment. Refractive index variations corresponding to different Pl

The present study investigates the potential effect of externally added unsaturated fatty acids on P. falciparum growth. Our results indicate that polyunsaturated fatty acids (PUFAs) inhibit the growth of Plasmodium in proportional to their degree of unsaturation. At higher concentration the PUFA Docosahexaenoic acid (DHA) induces pyknotic nuclei in infected erythrocytes. When Plasmodium stages were treated transiently with DHA, the ring stage culture recovered from the drug effect and parasitemia was increased post DHA removal with delayed growth of 12 h, compared to untreated control. OBJECTIVE: Glucose-6-phosphate dehydrogenase (G6PD) deficiency offers some protection against malaria; however, the degree of protection is poorly described and likely to vary with G6PD genotype and Plasmodium species. We present a novel approach to quantify the differential invasion rates of P. falciparum between G6PD deficient and normal red blood cells (RBCs) in an ex vivo model. A flow-cytometry based assay was developed to distinguish G6PD deficient and normal, parasitized and non-parasitized RBCs within the same sample. Venous blood collected from a G6PD heterozygous female was infected

Hepato-biliary coccidiosis is reported in a diary goat that died of liver failure. Two types of lesions were seen in the liver. In the biliary type, coccidial oocysts, schizonts, gametes and gametocytes were seen in the epithelium of the bile duct, the wall of which was highly fibrosed. The hepatic type was in the form of granuloma, composed of oocysts and macrophages encapsulated in a fibrous capsule. Hepatic lymph nodes were diffusely scattered with oocysts and macrophages. Liver coccidiosis was found to be unresponsive to oral sulphamethazine treatment.

There is an emerging concern that snakes are definitive hosts of certain species of Sarcocystis that cause muscular sarcocystosis in human and non-human primates. Other species of Sarcocystis are known to cycle among snakes and rodents, but have been poorly characterized in the USA and elsewhere. Although neurological sequalae are known for certain species of Sarcocystis, no such neurological symptoms are known to typify parasites that naturally cycle in rodents. Here, sporocysts of a species of Sarcocystis were found in the intestinal contents of a rat snake (Pantherophis alleghaniensis) from

Sexual commitment in Plasmodium parasites is essential for malaria transmission, yet much remains unknown about the underlying signaling events initiating sexual conversion in a subpopulation of parasites. We discovered a single valine (V2163) to leucine (L2163) mutation in an Apetala 2 (AP2) transcription factor required for P. falciparum gametocytogenesis, ap2-g that abrogates sexual differentiation and confirmed this with forward and reverse mutation editing. Mutated AP2-G.L2163 does not bind the ap2-g consensus motif, GnGTAC, or stimulate AP2-G-dependent gene transcription including autore

Tropical theileriosis is an important protozoan tick-borne disease in cattle. Vaccination using attenuated schizont-infected cell lines is one of the methods used for controlling the disease. This study describes the production of attenuated schizont-infected cell lines from Egypt and an evaluation of its use as a vaccine to protect calves against clinical disease upon field challenge. Two groups of exotic and crossbred male calves were divided into vaccinated and control groups. The vaccinated groups were inoculated with 4 ml (1 × 106 cells/ml) of the attenuated cell line. Three weeks after v

Eukaryotic Armadillo (ARM) repeat proteins are multifaceted with prominent roles in cell-cell adhesion, cytoskeletal regulation and intracellular signaling among many others. One such ARM repeat containing protein, ARM Repeats Only (ARO), has recently been demonstrated in both Toxoplasma (TgARO) and Plasmodium (PfARO) parasites to be targeted to the rhoptries during the late asexual stages. TgARO has been implicated to play an important role in rhoptry positioning i.e. directing the rhoptry towards the apical end of the parasite. Here, we report for the first time that PfARO exhibits a DNA bin
