Automatic and label-free identification of blood vessels in gliomas using the combination of multiphoton microscopy and image analysis.

Currently, the targeted treatment of tumor based on the tumor microenvironment is newly developed. Blood vessels are the key parts in the tumor microenvironment, which is taken as a new visible target for tumor therapy. Multiphoton microscopy (MPM), based on the second harmonic generation and two-photon excited fluorescence, is available to make the label-free analysis on the blood vessels in human gliomas. MPM can reveal the vascular morphological characteristics in gliomas, including vascular malformation, intense vascular proliferation, perivascular collagen deposition, perivascular lymphoc

Serelaxin improves cardiac and renal function in DOCA-salt hypertensive rats.

Serelaxin, a recombinant form of the naturally occurring peptide hormone relaxin-2, is a pleiotropic vasodilating hormone that has been studied in patients with acute heart failure. In this study, the effects of serelaxin on cardiac and renal function, fibrosis, inflammation and lipid accumulation were studied in DOCA-salt treated rats. Uninephrectomized rats were assigned to two groups: controls provided with normal drinking water and DOCA provided with DOCA pellets and sodium chloride drinking water. After 4 weeks, the DOCA-salt rats were randomly selected and implanted with osmotic minipump

Second harmonic generation imaging as a potential tool for staging pregnancy and predicting preterm birth.

We use second harmonic generation (SHG) microscopy to assess changes in collagen structure of murine cervix during cervical remodeling of normal pregnancy and in a preterm birth model. Visual inspection of SHG images revealed substantial changes in collagen morphology throughout normal gestation. SHG images collected in both the forward and backward directions were analyzed quantitatively for changes in overall mean intensity, forward to backward intensity ratio, collagen fiber size, and porosity. Changes in mean SHG intensity and intensity ratio take place in early pregnancy, suggesting that

Substrate-Directed Underlayer Growth of Bilayer MoS2 Revealed by Mo Isotope Labeling.

Direct control over the vertical formation sequence and stacking registry in van der Waals (vdW) bilayers is essential for device performance and moiré engineering yet difficult to resolve unambiguously with conventional probes. We use Mo isotope labeling in a two-step chemical vapor deposition process to synthesize bilayer MoS2 and trace its vertical formation on common substrates. By combining site-selective laser thinning, Raman spectroscopy, time-of-flight secondary ion mass spectrometry, and atomic-resolution scanning transimission electron microscopy (STEM), we find a clear substrate dep

Endogenous optical biomarkers of ovarian cancer evaluated with multiphoton microscopy.

PURPOSE: Among gynecologic cancers, ovarian cancer is the second most common and has the highest mortality. Currently, there is no accurate early diagnostic technique for ovarian cancer. Furthermore, little is understood regarding the early progression of this disease. We have imaged multiphoton interactions of endogenous tissue constituents from normal and abnormal ovarian biopsies that were kept viable during transport from the operating room and microscopy. EXPERIMENTAL DESIGN: The ovarian surface and underlying stroma were assessed with two-photon excited fluorescence (2PEF) and second ha

Light and metabolism: label-free optical imaging of metabolic activities in biological systems [Invited].

Metabolic imaging is critical for understanding cellular functions beyond morphology, offering significant insights into various biological processes and disease states. Label-free optical imaging techniques stand out by providing high-resolution, molecularly specific, and/or non-invasive assessments of metabolic activity without relying on exogenous contrast agents. This review discusses the key photon-tissue interactions-absorption, emission, and scattering-that underpin label-free optical imaging modalities for interrogating tissue's metabolic activities at various scales. Specifically, pho

Comparison of confocal microscopy and two-photon microscopy in mouse cornea in vivo.

High-resolution imaging of the cornea is important for studying corneal diseases at cellular levels. Confocal microscopy (CM) has been widely used in the clinic, and two-photon microscopy (TPM) has recently been introduced in various pre-clinical studies. We compared the performance of CM and TPM in normal mouse corneas and neovascularized mouse corneas induced by suturing. Balb/C mice and C57BL/6 mice expressing green fluorescent protein (GFP) were used to compare modalities based on intrinsic contrast and extrinsic fluorescence contrast. CM based on reflection (CMR), CM based on fluorescence

Label-free 3D characterization of cardiac fibrosis in muscular dystrophy using SHG imaging of cleared tissue.

BACKGROUND INFORMATION: Duchenne muscular dystrophy (DMD) is a neuromuscular disease caused by mutations in the gene encoding dystrophin. It leads to repeated cycles of muscle fiber necrosis and regeneration and progressive replacement of fibers by fibrotic and adipose tissue, with consequent muscle weakness and premature death. Fibrosis and, in particular, collagen accumulation are important pathological features of dystrophic muscle. A better understanding of the development of fibrosis is crucial to enable better management of DMD. Three-dimensional (3D) characterization of collagen organiz

In-vivo nonlinear optical microscopy (NLOM) of epithelial-connective tissue interface (ECTI) reveals quantitative measures of neoplasia in hamster oral mucosa.

The epithelial-connective tissue interface (ECTI) plays an integral role in epithelial neoplasia, including oral squamous cell carcinoma (OSCC). This interface undergoes significant alterations due to hyperproliferating epithelium that supports the transformation of normal epithelium to precancers and cancer. We present a method based on nonlinear optical microscopy to directly assess the ECTI and quantify dysplastic alterations using a hamster model for oral carcinogenesis. Neoplastic and non-neoplastic normal mucosa were imaged in-vivo by both multiphoton autofluorescence microscopy (MPAM) a

Digital quantitation of bridging fibrosis and septa reveals changes in natural history and treatment not seen with conventional histology.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) with bridging fibrosis is a critical stage in the evolution of fatty liver disease. Second harmonic generation/two-photon excitation fluorescence (SHG/TPEF) microscopy with artificial intelligence (AI) provides sensitive and reproducible quantitation of liver fibrosis. This methodology was applied to gain an in-depth understanding of intra-stage fibrosis changes and septa analyses in a homogenous, well-characterised group with MASH F3 fibrosis. METHODS: Paired liver biopsies (baseline [BL] and end of treatment [EOT])

Multi-contrast optical coherence tomography for in vivo visualization and quantification of vascular features and collagen in mouse ovaries in aging.

Multi-contrast optical coherence tomography (MC-OCT), incorporating optical coherence tomography angiography (OCTA) and polarization-sensitive OCT (PS-OCT), offers a powerful non-invasive approach for interrogating ovarian microvascular and extracellular matrix (ECM) remodeling in vivo. However, its application to ovarian aging has remained largely unexplored. Here, we establish and validate MC-OCT as a robust imaging platform for visualizing and quantifying age-related changes in vascular architecture and collagen organization in mouse ovaries. Using in vivo OCTA, we demonstrate a progressive

Modelling Atherosclerotic Plaque Cap Mechanics: Microcalcifications Reduce Mechanical Properties in Mesenchymal Stromal Cell-Based Model.

Rupture of atherosclerotic plaque caps is the cause of many disabling or lethal cardiovascular events, such as stroke and myocardial infarction. Microcalcifications (<50 µm) have been shown, in computational models, to affect the biomechanical stability of the cap. The current study aims to develop a tissue-engineered model of the atherosclerotic fibrous cap with microcalcifications produced by mesenchymal stromal cells (MSCs). Human MSCs are seeded in fibrin gels and cultured for 2 weeks in medium supplemented with TGF-β1 to induce smooth muscle cell differentiation and collagenous matrix for

Exploring thoracic aorta ECM alterations in Marfan syndrome: insights into aorta wall structure.

Marfan syndrome is a connective tissue disorder caused by FBN1 mutations, leading to aortic wall fragility and increased susceptibility to aneurysm and dissection. This study investigated microstructural and molecular alterations in the thoracic aorta of Fbn1mgΔlpn mice, with a focus on the tunica intima and media. Histological and ultrastructural analyses demonstrated elastic fiber fragmentation and reduced fibrillin-1 expression. In the intima, endothelial cells showed partial detachment and decreased levels of fibrillin-1, perlecan, collagen IV, and α5β1 integrins, suggesting compromised ad

Stiffness-Dependent Lysyl Oxidase Regulation through Hypoxia-Inducing Factor 1 Drives Extracellular Matrix Modifications in Psoriasis.

Psoriasis is a common chronic inflammatory skin disease characterized by a thickened epidermis with elongated rete ridges and massive immune cell infiltration. It is currently unclear what impact mechanoregulatory aspects may have on disease progression. Using multiphoton second harmonic generation microscopy, we found that the extracellular matrix was profoundly reorganized within psoriatic dermis. Collagen fibers were highly aligned and assembled into thick, long collagen bundles, whereas the overall fiber density was reduced. This was particularly pronounced within dermal papillae extending

Metal Ion Mediation of Interfacial Chiral Supramolecular Formation of Amphiphilic Schiff Bases Studied by In Situ Second Harmonic Generation.

The coordination effects between chiral Schiff bases and metal ions can provide an effective strategy for fabricating chiral supramolecular self-assemblies. We studied the supramolecular self-assembly of the amphiphilic Schiff base enantiomer, 2-hydroxy-1-naphthylmethylamino-N,N'-bis(octadecyl)-l/d-glutamic diamide (l/dGJ), at the air/water interfaces by using in situ second harmonic generation linear dichroism (SHG-LD) technique combined with UV-vis spectroscopy and atom force microscopy. lGJ and dGJ monolayers can form mirror-image structures of each other at different interfaces, which can

Distinct reorganization of collagen architecture in lipopolysaccharide-mediated premature cervical remodeling.

Previous work has identified divergent mechanisms by which cervical remodeling is achieved in preterm birth (PTB) induced by hormone withdrawal (mifepristone) or lipopolysaccharide (LPS). Our current study aims to document how collagen architecture is modified to achieve premature cervical remodeling in mice treated with LPS as a model of infection-induced inflammation. Cervices were collected on gestation day (d) 15 from mice with premature cervical ripening induced by LPS and compared to d15 and d18 controls as well as a hormone withdrawal PTB model. Nonlinear optical microscopy and ultrasound imaging of human cervical structure.

The cervix softens and shortens as its collagen microstructure rearranges in preparation for birth, but premature change may lead to premature birth. The global preterm birth rate has not decreased despite decades of research, likely because cervical microstructure is poorly understood. Our group has developed a multilevel approach to evaluating the human cervix. We are developing quantitative ultrasound (QUS) techniques for noninvasive interrogation of cervical microstructure and corroborating those results with high-resolution images of microstructure from second harmonic generation imaging

Dual Twisted Interfaces Enable Tunable Second Harmonic Generation in Chemical Vapor Deposition Grown Trilayer MoS2.

Twisted bilayer transition metal dichalcogenides, governed by a single twisted interface, offer a versatile platform for manipulating interlayer coupling and emergent optical behavior. However, despite extensive studies on single twisted interface systems, how multi-interface twisted stacking influences nonlinear optical behavior remains unclear. Here, we address this gap by systematically investigating the nonlinear optical performance of chemical vapor deposition (CVD) grown twisted trilayer MoS2 (TT-MoS2) with multi-interface twisted stacking. We fabricate all nine distinct trilayer MoS2 co

Electrochemically Enabled Self-Intercalation for Symmetry Breaking and Metallic Ferroelectricity in Nb1+xS2.

Two-dimensional (2D) metallic materials that simultaneously exhibit inversion symmetry breaking are highly desirable for nonlinear optics, unconventional ferroelectrics, and symmetry-driven electronic functionalities, yet controllably inducing structural asymmetry without compromising metallicity remains a central challenge. Here, we report a self-intercalation strategy to engineer symmetry breaking in metallic NbS2 by exploiting the intrinsic oxidation behavior of niobium. Electrochemical anodization precisely tailors the native oxide layer on Nb foil, which serves as a tunable Nb reservoir d

Spatial Proteomics of the Normal Breast Collagen Stroma: Links to Density and Body Mass Index.

Collagen breast stroma can become a breast cancer risk factor, yet proteomic regulation of normal breast stroma remains poorly defined. This study evaluates the spatial regulation of the collagen proteome from normal breast tissue. Normal breast tissue sections from the Susan G. Komen tissue bank were used (n = 40), with data including genetic ancestry (n = 20 African ancestry; n = 20 European ancestry), body-mass-index (BMI), age, and mammogram density by the Breast Imaging Reporting and Data System (BI-RADS). 10-plex cell marker staining showed CD44 and COL1A1 markers modulated with BMI.
