Glucagon-like peptide-1 receptor (GLP-1R) agonists such as semaglutide are highly effective treatments for obesity, yet the mechanisms by which they reduce food intake remain incompletely understood. Because taste plays a critical role in guiding food intake, several clinical studies have investigated whether GLP-1R agonists alter taste function, but these reports have yielded conflicting results. Here, we systematically tested the effects of chronic semaglutide treatment on taste responsivity in diet-induced obese mice. Mice were evaluated using brief-access gustometer tests to assess respons

AIMS: Semaglutide, a glucagon-like peptide-1 receptor agonist, has demonstrated cardiovascular benefit in patients with type 2 diabetes mellitus (T2DM) and established atherosclerotic disease. Its role in the immediate postacute coronary syndrome (ACS) setting, however, remains unexplored. This study evaluated the early real-world use of semaglutide prescribed at hospital discharge after ACS.
METHODS: We conducted a retrospective, multicenter observational study of adults with T2DM hospitalized for ACS between January 2022 and December 2024 and discharged with a prescription for semaglutide.

Glucagon-like peptide-1 receptor agonists are widely used for the management of obesity and dysglycaemia, and current evidence does not support a causal association with pancreatic cancer. We report a 55-year old woman with obesity and prediabetes who started once-weekly semaglutide 0.25 mg for weight management and glycaemic control. Baseline fasting plasma glucose was 107 mg/dL and glycated haemoglobin was 6.0%. After one month, despite a 4 kg weight loss and no significant gastrointestinal symptoms, fasting plasma glucose increased to 169 mg/dL and glycated haemoglobin to 6.6%. Latent autoi

We report a paradoxical case of a 40-year-old man with class 2 obesity (body mass index 39.8 kg/m2) and insulin resistance (homeostatic model assessment of insulin resistance [HOMA-IR] 5.0; reference <2.0) who achieved minimal weight loss (<5%) despite tirzepatide dose escalation to 15 mg weekly over 6 months. Following switch to semaglutide 2.4 mg weekly, the patient achieved 20% weight loss over 6 months, with normalization of fasting glucose, resolution of insulin resistance, and marked improvement in appetite control. This case challenges the assumption that dual glucose-dependent insulino

AIMS: Neuropsychiatric safety concerns, including depression and suicidal ideation, have emerged during the expanding clinical use of incretin-based therapies for type 2 diabetes mellitus (T2DM) and obesity. However, whether risks differ across successive generations of incretin-based therapies remains uncertain.
MATERIALS AND METHODS: This retrospective cohort study used the TriNetX Global Federated Network (> 192 million patients). Adults with T2DM, obesity, or both initiating tirzepatide, semaglutide, or other glucagon-like peptide-1 receptor agonists (GLP-1 RAs) between July 2022 and June

This systematic review examines the potential association between semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, and the development of non-arteritic anterior ischemic optic neuropathy (NAION). Nine studies were included, consisting of retrospective cohort analyses, case series, and pharmacovigilance reports. Findings across the literature were inconsistent, with some studies reporting an increased risk while others found no significant association. Variations in study design, diagnostic criteria, and population characteristics limited comparability and prevented meta-analysi

BACKGROUND: The cardiovascular safety of oral semaglutide, a glucagon-like peptide 1 receptor agonist, has been established in persons with type 2 diabetes and high cardiovascular risk. An assessment of the cardiovascular efficacy of oral semaglutide in persons with type 2 diabetes and atherosclerotic cardiovascular disease, chronic kidney disease, or both is needed.
METHODS: In this double-blind, placebo-controlled, event-driven, superiority trial, we randomly assigned participants who were 50 years of age or older, had type 2 diabetes with a glycated hemoglobin level of 6.5 to 10.0%, and ha

BACKGROUND: Semaglutide is a medication for weight loss in obese patients. Recently, many plastic surgeons have recommended semaglutide following bariatric surgery to increase weight loss. However, postoperative complications such as wound dehiscence, delayed healing, and infection pose significant risks.
OBJECTIVES: In this study we aimed to examine the relationship between preoperative semaglutide and postoperative complications in nondiabetic obese patients undergoing body contouring surgery. We hypothesized that semaglutide would increase the risk of postoperative complications.
METHODS:

BACKGROUND AND PURPOSE: Myocardial fibrosis (MF), a hallmark of structural cardiac remodelling, drives disease progression across most forms of heart failure and plays a central role in heart failure with preserved ejection fraction (HFpEF). Despite its clinical relevance, effective treatments remain scarce. In preclinical models, current methods for quantifying MF fail to capture its regional heterogeneity, limiting reliable assessment of novel anti-fibrotic compounds. This study aimed to develop a whole-heart imaging and deep learning (DL)-based quantification pipeline for MF, and to validat

OBJECTIVES: Some concerns persist regarding the safety of semaglutide. The objective of this updated meta-analysis is to assess the risk of acute pancreatitis with the use of semaglutide, assessing the results according to the different administration regimens.
METHODS: We performed an updated meta-analysis of randomised, placebo-controlled studies of semaglutide therapy that report acute pancreatitis. This meta-analysis was performed in line with PRISMA guidelines. A global and stratified analysis according to the therapeutic scheme used was performed using the fixed-effects model.

BACKGROUND: Diabetes mellitus (DM) and obesity are associated with total knee arthroplasty (TKA) complications. Semaglutide, a medication for DM and weight loss, can potentially affect TKA outcomes. This study investigated whether semaglutide use during TKA demonstrates fewer: (1) medical complications; (2) implant-related complications; (3) readmissions; and (4) costs.
METHODS: A retrospective query was performed using a National database to 2021. Patients undergoing TKA for osteoarthritis with DM and semaglutide use were successfully propensity score-matched to controls semaglutide = 7,051;

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) reduce food intake and body weight in humans, partly via neural circuits governing reward and energy homeostasis. Using Caenorhabditis elegans as a tractable model, we examined neurobehavioral and physiological effects of semaglutide, a long-acting GLP-1RA. Wild-type N2 animals exposed to semaglutide (1 μM and 10 μM) showed significantly reduced chemotaxis toward the volatile odorant diacetyl. Semaglutide also reduced egg-laying and delayed larval development at both concentrations tested. These phenotypes demonstrate that semaglutide alters
