Exploring Senolytic and Senomorphic Properties of Medicinal Plants for Anti-Aging Therapies.

Senolytic and senomorphic therapies have gained more and more attention in the last decade. This kind of therapy is based on the killing of cellular senescent cells without harming the "normal" cells. Aging is not a disease. Clinical studies on healthy people will be difficult to conduct. Therefore, one possibility is to draw on the large repertoire of medicinal plants and use their senolytic properties to provide mild anti-aging therapies. Chamomile, goldenrod, reishi, and green tea were tested for their ability to trigger senolysis. Quercetin was used as control substance. Cellular senescenc

Endometrial receptivity in women of advanced age: an underrated factor in infertility.

BACKGROUND: Modern lifestyle has led to an increase in the age at conception. Advanced age is one of the critical risk factors for female-related infertility. It is well known that maternal age positively correlates with the deterioration of oocyte quality and chromosomal abnormalities in oocytes and embryos. The effect of age on endometrial function may be an equally important factor influencing implantation rate, pregnancy rate, and overall female fertility. However, there are only a few published studies on this topic, suggesting that this area has been under-explored. Improving our knowled

Senescent immune cells, inflammaging, and bone loss: therapeutic potential and translational challenges of senolytics in osteoporosis.

Osteoporosis is a common age-related skeletal disorder characterized by low bone mass, deterioration of bone microarchitecture, and increased susceptibility to fragility fractures. Although antiresorptive and anabolic therapies have substantially improved fracture prevention, current treatments do not fully address the aging-related biological processes that disrupt skeletal homeostasis. Increasing evidence from osteoimmunology, immunosenescence, and cellular senescence research suggests that inflammaging contributes to age-related bone loss by reshaping the bone marrow immune microenvironment

Therapeutic Targeting of Inflammaging and Emerging Strategies for Managing Age-Related Multimorbidity: A Narrative Review.

The global aging population faces an increasing burden of chronic multimorbidity, much of which is driven by inflammaging. Inflammaging is defined as a persistent, low-grade inflammatory state recognized as a hallmark of biological aging. This narrative review examines therapeutic strategies targeting key mechanisms of inflammaging, with particular focus on cellular senescence and the senescence-associated secretory phenotype (SASP), nicotinamide adenine dinucleotide (NAD+) decline, and inflammasome activation. Senolytic therapies designed to eliminate senescent cells have progressed into earl

Evaluation of exploratory fluid biomarkers from a phase 1 senolytic trial in mild Alzheimer's disease.

Senescent cell accumulation contributes to the progression of age-related disorders including Alzheimer's disease (AD). Clinical trials focused on cellular senescence are in early stages and have yet to establish reliable outcome measures reflecting senescent cell burden or response to senolytics, therapeutics that clear senescent cells. Results from the first open-label trial of senolytics, dasatinib plus quercetin (D + Q), in older adults (N = 5) with early AD demonstrated central nervous system penetration of dasatinib and favorable safety and tolerability. Herein, we present explorator

Reverse aging to treat lupus.

Systemic lupus erythematosus (SLE) is a complex autoimmune disease with multifaceted pathogenetic processes, including abnormalities of T-cell subset distribution and function. Accumulation of senescent CD4+ T cells has been found to contribute to the development of the disease. In this issue, Jiang et al. provide compelling evidence that links an expanded pool of CD4+CD57+ senescent T cells in patients with SLE to disease activity favored by interleukin-15. Importantly, treatment of lupus-prone mice with a senolytic drug resulted in decreased autoimmune pathology. The findings of this study s

Anti-inflammatory phytochemicals from medicinal herbs based on senoinflammation schema for healthy aging.

Aging is characterized by chronic inflammation involving a continuous, multifarious, low-grade inflammatory response that contributes to age-related chronic diseases. Recently, researchers have focused on age-related chronic inflammation and anti-inflammatory phytochemicals to ameliorate age-related degenerative conditions. As proposed earlier, senoinflammation is a broader view of age-related inflammation with an insightful perspective, which differs from the traditional phenomenal view. Recent advances have identified the senescence-associated secretory phenotype (SASP) as a central mediator

Senolytic Therapy as a Preventive Strategy for Spine Degeneration and Pain.

Cellular senescence drives inflammation and tissue breakdown and is a key hallmark of aging. The accumulation of senescent cells is strongly linked to the degeneration of spinal tissues and back pain. Here, we show that administration of the senolytic agents, o-vanillin and RG-7112, prevents the development of pain-related behavior in sparc-/- mice. Treated mice exhibit a reduced expression of senescence markers in the intervertebral discs, vertebral endplates, vertebral bone, and spinal cord, alongside a dampening of pro-inflammatory senescence-associated secretory factors in these tissues. Pseudo-senescence induced by palbociclib does not sensitise pleural mesothelioma cells to combinations with senolytics.

Pleural Mesothelioma (PM) is an aggressive neoplasm of the lung pleura with poor survival rates, highlighting the urgent need for novel therapeutic options. The CDK4/6 inhibitors abemaciclib and palbociclib have demonstrated promising results in patient-derived xenograft models of PM. In this study, we observed that palbociclib reduced proliferation, leading to increased cell size, enhanced SA-β-galactosidase activity, and elevated secretion of IL-6 and IL-8 (SASP), all of which are hallmarks of senescence. However, upon drug removal, the cells regrew. To enhance therapeutic efficacy, we attem

A Review of Senescence-Associated Secretory Phenotype-Mediated Remodeling of the Tumor Microenvironment: Implications for Cancer Progression and Therapy.

BACKGROUND: Cellular senescence influences cancer progression through irreversible cell-cycle arrest, immune surveillance, and paracrine signaling mediated by the senescence-associated secretory phenotype (SASP). SASP comprises inflammatory cytokines, chemokines, growth factors, and matrix-remodeling enzymes that regulate the tumor microenvironment (TME). OBJECTIVE: This review describes the current evidence on the mechanisms of SASP induction, its condition-dependent roles in cancer, TME remodeling, and emerging strategies targeting senescent cells and SASP signaling. METHODS: Relevant lite

Protocol for a pilot clinical trial of the senolytic drug combination Dasatinib Plus Quercetin to mitigate age-related health and cognitive decline in mental disorders.

BACKGROUND: Major depressive disorder (MDD) and schizophrenia are linked to accelerated aging leading to reduced lifespan, health span and cognitive decline. Cellular senescence, in which cells lose proliferative capacity and develop a senescence-associated secretory phenotype (SASP), plays a role in this process. Emerging research suggests that the senolytic regimen of dasatinib+quercetin (D+Q) reduces senescent cells, potentially mitigating age-related health and cognitive decline. This pilot study aims to assess the feasibility and safety of D+Q in older adults with schizophrenia, schizoaff

Senescent cell heterogeneity and responses to senolytic treatment are related to cell cycle status during senescence induction.

Cellular senescence has been strongly linked to aging and age-related diseases. It is well established that the phenotype of senescent cells is highly heterogeneous and influenced by their cell type and senescence-inducing stimulus. Recent single-cell RNA-sequencing studies identified heterogeneity within senescent cell populations. However, proof of functional differences between such subpopulations is lacking. To identify functionally distinct senescent cell subpopulations, we employed high-content image analysis to measure senescence marker expression in primary human endothelial cells and

Senescence Targeting Methods Impact Alzheimer's Disease Features in 3xTg Mice.

BACKGROUND: Cellular senescence has been associated with neurodegenerative disease and clearance of senescent cells using genetic or pharmaceutical strategies (senolytics) has demonstrated beneficial effects in mouse models investigating individual disease etiologies of Alzheimer's disease (AD). However, it has remained unclear if senescent cell clearance in a mouse model exhibiting both plaque and tau pathologies modifies the disease state (3xTg). OBJECTIVE: To investigate the effects of senescent cell clearance in the 3xTg mouse model. METHODS: 3xTg mice were treated with senolytics (ABT26

Senolytics cocktail dasatinib and quercetin alleviate chondrocyte senescence and facet joint osteoarthritis in mice.

BACKGROUND CONTEXT: Low back pain (LBP) is a pervasive issue, causing substantial economic burden and physical distress worldwide. Facet joint osteoarthritis (FJ OA) is believed to be a significant contributor to this problem. However, the precise role of chondrocyte senescence in FJ OA remains unclear, as does whether the clearance of chondrocyte senescence can alleviate the progression of FJ OA. PURPOSE: The goal of this study was to understand the potential of Dasatinib (D) and Quercetin (Q) as a treatment to clear chondrocyte senescence during the progression of FJ OA. STUDY DESIGN: We u

Selective Vulnerability of Senescent Glioblastoma Cells to BCL-XL Inhibition.

UNLABELLED: Glioblastoma (GBM) is a rapidly fatal malignancy typically treated with radiation and temozolomide (TMZ), an alkylating chemotherapeutic. These cytotoxic therapies cause oxidative stress and DNA damage, yielding a senescent-like state of replicative arrest in surviving tumor cells. Unfortunately, recurrence is inevitable and may be driven by surviving tumor cells eventually escaping senescence. A growing number of so-called "senolytic" drugs have been recently identified that are defined by their ability to selectively eliminate senescent cells. A growing inventory of senolytic dru

A Senolytic-Eluting Coronary Stent for the Prevention of In-Stent Restenosis.

The vast majority of drug-eluting stents (DES) elute either sirolimus or one of its analogues. While limus drugs stymie vascular smooth muscle cell (VSMC) proliferation to prevent in-stent restenosis, their antiproliferative nature is indiscriminate and limits healing of the endothelium in stented vessels, increasing the risk of late-stent thrombosis. Oxidative stress, which is associated with vascular injury from stent implantation, can induce VSMCs to undergo senescence, and senescent VSMCs can produce pro-inflammatory cytokines capable of inducing proliferation of neighboring nonsenescent V

The Respiratory Epithelial Barrier as an Immunopharmacological Rheostat: From Homeostatic Sentinel to Therapeutic Target in Chronic Lung Disease.

The respiratory epithelium is a dynamic immunopharmacological interface that transcends its traditional role as a passive barrier to become a central coordinator of pulmonary immunity. This review introduces and elaborates on the paradigm of the "Epithelial Immunopharmacological Rheostat," a conceptual framework that posits the epithelium as a dynamic, multidimensional signal processor that continuously calibrates the threshold of immune activation. Distinct from static models of barrier dysfunction or isolated alarmin biology, this rheostat operates through four hierarchically interacting axe

p16 Beyond Senescence: Cancer Biology Lessons Applied to Endometriosis.

Endometriosis is an estrogen-dependent disease characterized by chronic inflammation and fibrosis and is a major cause of pelvic pain and infertility. Recent studies have reported dysregulated expression of p16INK4a, a canonical cellular senescence marker, in endometriotic lesions. Although p16 is a proliferation-suppressive factor, its altered expression has paradoxically been associated with disease persistence, and its pathophysiological significance remains unclear. This review aims to clarify the context-dependent roles of p16 in endometriosis in relation to cellular senescence, the senes

Uremic Toxins, CKD, and Cognitive Dysfunction.

Cognitive impairment involves alterations to one's cognitive status that affects everyday life. Individuals with CKD, and particularly kidney failure, experience higher rates of cognitive impairment (20%-70%) compared with the general population. The highest prevalence is described in kidney failure such that dialysis-dependent patients have twice the prevalence of age-matched controls. In the past 5 years, the number of investigations examining the "kidney-brain axis," mechanisms of CKD-related cognitive impairment, and potential therapeutics have exponentially increased.
