The present investigation was conducted to document a role of L-threo-3,4-dihydroxyphenylserine (L-DOPS), precursor of L-norepinephrine (NE), in the functional recovery from beam-walking performance deficits in rats after unilateral sensorimotor cortex ablation. L-DOPS was administered simultaneously with benserazide (BSZ; a peripheral aromatic amino acid decarboxylase inhibitor), and the regional contents of NE in the cerebral cortex, hippocampus, and cerebellum were assayed. Behavioral recovery was demonstrated by the rats treated with L-DOPS and BSZ, and the rate of recovery was significant

A better understanding of the neural and muscular mechanisms underlying motor responses is essential for advancing neurorehabilitation protocols, brain-computer interfaces (BCI), feature engineering for biosignal classification algorithms, and identifying biomarkers of disease and performance enhancement strategies. In this study, we examined the neuromuscular dynamics of healthy individuals during a sequential finger-pinching task, focusing on the relationships between cortical oscillations and muscle activity in simultaneous electroencephalography (EEG) and electromyography (EMG) recordings.

Previous studies have reported changes in cortical responses to stimulation of the plegic or nonplegic hand after unilateral neuronal injury. We aimed to investigate the relationship between these responsiveness changes in central sensorimotor systems and functional recovery. Five macaque monkeys underwent lower cervical cord subhemisection surgery. Their hand movements were observed, and longitudinal functional MRI (fMRI) was performed under anaesthesia to assess responses to tactile stimulation of each hand and resting-state connectivity. Eigenvector centrality was computed from resting-stat

We studied cortical connections of functionally distinct movement zones of the posterior parietal cortex (PPC) in galagos identified by intracortical microstimulation with long stimulus trains ( approximately 500 msec). All these zones were in the anterior half of PPC, and each of them had a different pattern of connections with premotor (PM) and motor (M1) areas of the frontal lobe and with other areas of parietal and occipital cortex. The most rostral PPC zone has major connections with motor and visuomotor areas of frontal cortex as well as with somatosensory areas 3a and 1-2 and higher ord

The purpose of this study was to compare the location and intensity of activation of the sensorimotor cortex between finger tapping and palm scratching paradigms in normal volunteers and to consider using passive task instead of motor task in patients who are unable to perform motor task. Multishot echo-planar T2*-weighted imaging sequences at the level of the sensorimotor cortex were performed in axial plane during finger tapping and palm scratching paradigms in 13 normal volunteers. The authors found that the location of activation was slightly posterior only in bilateral passive task compar

The integrity of the structural cerebral cortex is disrupted after stroke either at the macroscopic or microscopic levels. Therefore, many attempts have been gathered to circumvent stroke-associated problems in the brain tissue. The current study was aimed to design an animal model of photochemical stroke using rose bengal (RB) plus laser irradiation (L) after 10, 15, and 20 min (´) and evaluate its effect on the cerebral tissue using unbiased stereological quantitative methods and morphometric histological analysis. Photochemical stroke was induced by intraperitoneal injection of RB dye and f

Tourette syndrome (TS) is a neurological disorder characterized by vocal and motor tics and is associated with cortical-striatal-thalamic-cortical circuit (CSTC) dysfunction and hyperexcitability of cortical limbic and motor regions, which are thought to lead to the occurrence of tics. Importantly, individuals with TS often report that their tics are preceded by 'premonitory sensory phenomena' (PSP) that are described as uncomfortable cognitive or bodily sensations that precede the execution of a tic, and are experienced as a strong urge for motor discharge. While the precise role played by PS

RATIONALE: Aspects of schizophrenia, including deficits in sensorimotor gating, have been linked to glutamate dysfunction and/or oxidative stress in the prefrontal cortex. System xc(-), a cystine-glutamate antiporter, is a poorly understood mechanism that contributes to both cellular antioxidant capacity and glutamate homeostasis.
OBJECTIVES: Our goal was to determine whether increased system xc(-) activity within the prefrontal cortex would normalize a rodent measure of sensorimotor gating.
METHODS: In situ hybridization was used to map messenger RNA (mRNA) expression of xCT, the active sub

BACKGROUND: Knee osteoarthritis (KOA) is characterized not only by peripheral joint pathology but also by mechanisms related to central sensitization. Previous studies have reported reduced activation of sensorimotor cortex during isolated joint movements in patients with KOA. However, cortical activation during walking remains unclear. This study aimed to investigate sensorimotor cortex activation during different walking tasks in patients with KOA to identify potential targets for central interventions.
METHODS: Nineteen patients with KOA and 18 demographically matched healthy controls (HCs

BACKGROUND: Acquired brain injury, whether the result of trauma, stroke, or iatrogenic complications, may cause persistent motor and cognitive deficits with limited effective treatments. Recent studies suggest that deep brain stimulation (DBS) of the lateral cerebellar nucleus (LCN) promotes long-term motor recovery after ischemic stroke and traumatic brain injury. Here, we examined the effect of LCN-DBS on motor recovery, and associated mechanisms, in an aspiration lesion model of surgical brain injury (SBI) in female rats.
METHODS: Ten female Long-Evans rats underwent unilateral, catheter-a

Neural stem cells are characterized by the ability to differentiate and stably express exogenous ge-nes. Vascular endothelial growth factor plays a role in protecting local blood vessels and neurons of newborn rats with hypoxic-ischemic encephalopathy. Transplantation of vascular endothelial growth factor-transfected neural stem cells may be neuroprotective in rats with cerebral palsy. In this study, 7-day-old Sprague-Dawley rats were divided into five groups: (1) sham operation (control), (2) cerebral palsy model alone or with (3) phosphate-buffered saline, (4) vascular endothelial growth fac

Cortical and subcortical lesions to the motor system, as often occur with stroke, typically lead to transitions through a stereotyped upper limb recovery sequence. After initial weakness and loss of dexterity, spasticity and fixed muscle coactivation patterns (synergies) appear. Early work suggested that different features arise from distinct primary motor cortex (M1) subdivisions. Here we investigated this with modern methods, using ischemic lesions of various cortical areas and electrocoagulation lesions of magnocellular red nucleus (RNm) in rhesus monkeys. Nine animals were trained on a rea

Transcranial alternating current stimulation (tACS) can modulate endogenous brain oscillations in a frequency-specific manner. Previous studies have reported that beta tACS modulates the excitability of primary motor cortex and improves task performance. Tactile and proprioceptive stimuli also elicit event-related synchronization of the beta rhythm in contralateral sensorimotor cortex, termed beta rebound, and a strong correlation was reported between proprioception-induced rebound strength and clinical recovery in stroke patients. We investigated the effects of tACS matched to the dominant be

During the hypotensive phase of electronocuous shock in rabbits, injection of nalorphine to the hypothalamus provoked elevation in the blood pressure (BP), recovery of the evoked potential (EP) of the sensorimotor brain cortex in response to electrodermal stimulation (EDS) of the hind limb. Injection of the drug to the central gray of the midbrain resulted in an insignificant short-term elevation of the BP followed by its drop. It is concluded that the endogenous opioid system of the hypothalamus is of importance in the genesis of the hypotensive response and suppression of the EP of the rabbi

Two female rhesus monkeys experienced 6 months of forced-use upper-extremity rehabilitation following localized damage to the sensorimotor cortex. One monkey received rehabilitation of the severely affected (contralesional) hand to model traditional poststroke therapeutic intervention. The second monkey received the same rehabilitation paradigm, but of the less affected (ipsilesional) hand, to model patients who preferentially use their ipsilesional hand or develop learned nonuse of the contralesional hand. Changes in the terminal distribution of the corticospinal projection (CSP) at C5-T1 fro

BACKGROUND AND HYPOTHESIS: Although the 5-hydroxytr-yptamine 2C receptor (5-HT2C) receptor agonists have demonstrated efficacy in mitigating sensorimotor gating deficits in the schizophrenic animal model of glutamatergic dysfunction, the precise neural mechanisms underlying these effects remain incompletely understood. This study investigates how 5-HT2C receptor activation improves sensorimotor gating and cognitive function by modulating Excitation/Inhibition balance in the medial prefrontal cortex (mPFC).

We used a dual anterograde-tracing paradigm to characterize the organization of corticocortical projections from primary somatosensory (SI) barrel cortex. In one group of rats, biotinylated dextran amine (BDA) and Fluoro-Ruby (FR) were injected into separate barrel columns that occupied the same row of barrel cortex; in the other group, the tracers were deposited into barrel columns residing in different rows. The labeled corticocortical terminals in the primary motor (MI) and secondary somatosensory (SII) cortices were plotted, and digital reconstructions of these plots were quantitatively an

INTRODUCTION: Why some people develop chronic pain following an acute episode of low back pain is unknown. Recent cross-sectional studies have suggested a relationship between aberrant sensorimotor cortex activity and pain persistence. The UPWaRD (Understanding persistent Pain Where it ResiDes) cohort study is the first prospective, longitudinal investigation of sensorimotor cortex activity in low back pain. This paper describes the development of a causal model and statistical analysis plan for investigating the causal effect of sensorimotor cortex activity on the development of chronic low b

Although treatment with N-methyl-D-aspartate (NMDA) receptor antagonists reduce neuronal loss after cerebral infarction and brain trauma in laboratory animals, there is little data concerning the effects of these drugs on behavioral recovery. Because NMDA receptor antagonists impede certain kinds of learning, and because motor recovery after sensorimotor cortex injury in the rat is dependent on post-lesion experience, we hypothesized that treatment with MK-801 after focal brain injury would be detrimental. Groups of rats were first trained to traverse a narrow elevated beam and then subjected

BACKGROUND: The Rolandic ∼20-Hz beta rhythm of the sensorimotor cortex is associated with motor function and perception. However, the modulation of this rhythm by different emotional stimuli is an innovative area of research.
AIMS: This study aims at investigating the impact of affective pictures (positive, negative, and neutral) on the proprioceptive modulation of the Rolandic ∼20 Hz beta rhythm in typically developing children and children with neurodevelopmental disorders (i.e. cerebral palsy and autism).
METHODS AND PROCEDURES: EEG was recorded while participants experienced passive wris
