Forebrain Control of Behaviorally Driven Social Orienting in Zebrafish.

Deficits in social engagement are diagnostic of multiple neurodevelopmental disorders, including autism and schizophrenia [1]. Genetically tractable animal models like zebrafish (Danio rerio) could provide valuable insight into developmental factors underlying these social impairments, but this approach is predicated on the ability to accurately and reliably quantify subtle behavioral changes. Similarly, characterizing local molecular and morphological phenotypes requires knowledge of the neuroanatomical correlates of social behavior. We leveraged behavioral and genetic tools in zebrafish to b

Directional Gaussian Mixture Models of the Gut Microbiome Elucidate Microbial Spatial Structure.

The gut microbiome is spatially heterogeneous, with environmental niches contributing to the distribution and composition of microbial populations. A recently developed mapping technology, MaPS-seq, aims to characterize the spatial organization of the gut microbiome by providing data about local microbial populations. However, information about the global arrangement of these populations is lost by MaPS-seq. To address this, we propose a class of Gaussian mixture models (GMM) with spatial dependencies between mixture components in order to computationally recover the relative spatial arrangeme

A Human Open Field Test Reveals Thigmotaxis Related to Agoraphobic Fear.

BACKGROUND: Thigmotaxis refers to a specific behavior of animals (i.e., to stay close to walls when exploring an open space). Such behavior can be assessed with the open field test (OFT), which is a well-established indicator of animal fear. The detection of similar open field behavior in humans may verify the translational validity of this paradigm. Enhanced thigmotaxis related to anxiety may suggest the relevance of such behavior for anxiety disorders, especially agoraphobia. METHODS: A global positioning system was used to analyze the behavior of 16 patients with agoraphobia and 18 healthy

Analysis of learning deficits in aged rats on the W-track continuous spatial alternation task.

Young and aged animals were tested on a spatial alternation task that consisted of two interleaved components: (1) an "outbound" or alternation component (working memory) and (2) an "inbound" component, requiring the animal to remember to return to a central location in space (spatial memory). In the present study, aged rats made more outbound errors throughout testing, resulting in significantly more days to reach learning criterion, as compared to young rats. Furthermore, while all animals were able to learn the hippocampus-dependent inbound component of the task, most aged animals remained

SciGeneX: enhancing transcriptional analysis through gene module detection in single-cell and spatial transcriptomics data.

The standard pipeline to analyze single-cell RNA-seq or spatial transcriptomics data focuses on a gene-centric approach that overlooks the collective behavior of genes. However, understanding cell populations necessitates recognizing intricate combinations of activated and repressed pathways. Therefore, a broader view of gene behavior offers more accurate insights into cellular heterogeneity in single-cell or spatial transcriptomics data. Here, we describe SciGeneX (Single-cell informative Gene eXplorer), a R package implementing a neighborhood analysis and a graph partitioning method to gener

Interactions of pain and opioids on conditioned place preference in rodents.

RATIONALE: Opioid analgesics are the most effective medications used for the treatment of pain, however there are significant risks associated with repeated opioid use including opioid misuse and opioid use disorder development. Chronic pain affects millions of adults in the United States, and opioid misuse is often comorbid with pain conditions in individuals who are repeatedly treated with opioids. In addition to providing pain relief, opioids produce rewarding effects, but in chronic pain states, reward processing can become dysregulated. The conditioned place preference task is commonly us

A proteomic signature for CNS adaptations to the valence of environmental stimulation.

Environmental Enrichment leads to a significant improvement in long-term performance across a range of cognitive functions in mammals and it has been shown to produce an increased synaptic density and neurogenesis. Nevertheless it is still an open question as to whether some key aspects of spatial learning & memory and procedural learning might be embodied by different molecular pathways to those of social cognition. Associated with synaptic changes and potentially underlying conditions, the Ras-ERK pathway has been proposed to be the primary mediator of in vivo adaptations to environmental en

The medial frontal cortex contributes to but does not organize rat exploratory behavior.

Animals use multiple strategies to maintain spatial orientation. Dead reckoning is a form of spatial navigation that depends on self-movement cue processing. During dead reckoning, the generation of self-movement cues from a starting position to an animal's current position allow for the estimation of direction and distance to the position movement originated. A network of brain structures has been implicated in dead reckoning. Recent work has provided evidence that the medial frontal cortex may contribute to dead reckoning in this network of brain structures. The current study investigated th

Functional dissociation between fornix and hippocampus in spatial conditional learning.

Do lesions of the fornix or the hippocampus impair the performance of spatial conditional associative learning tasks, and to what extent does damage to these brain structures result in comparable deficits in this type of spatial behavior? The available evidence is not clear. In the present study, rats with lesions of the fornix, hippocampus, and normal control animals were trained on two spatial-visual conditional learning tasks in which they had to form arbitrary associations between visual stimuli and the context in which these stimuli were embedded. In one condition, rats were required to c

Mucosal inherent activity patterns in the rat: evidence from voltage-sensitive dyes.

1. Fluorescence changes in the dye di-4-ANEPPS were monitored on the rat's nasal septum and medial surface of the turbinates in response to odorant stimuli. For each mucosal surface a 6.0 x 6.0-mm area was sampled at 100 contiguous sites with a 10 x 10 photodiode array. The odorants were propyl acetate, 2-propanol, citral, L-carvone and ethylacetoacetate, each presented at a low and high concentration. 2. Like previous work using optical recording techniques and potential-sensitive dyes on the amphibian epithelium, the fluorescence signals elicited by odorant stimuli in the rat preparation wer

Somatostatin and (D-Trp8, D-Cys14)-somatostatin delay extinction and reverse electroconvulsive shock induced amnesia in rats.

In the present study the effects of somatostatin and its analogs on active avoidance behavior, electroconvulsive shock (ECS)-induced retrograde amnesia, and spatial-discrimination learning were compared in rats. (D-Trp8, D-Cys14)-somatostatin (as did the somatostatin molecule itself) delayed the extinction of active avoidance behavior, antagonized ECS-induced amnesia, and did not modify spatial-discrimination learning. Des-Asn5-(D-Trp8, D-Ser13) somatostatin and des-AA1,2,4,5,12,13,-(D-Trp8) somatostatin did not influence these behaviors. The data suggest that certain parts of the somatostatin

Functional Connectivity Pattern Using Resting-state fMRI as an Assessment Tool for Spatial Neglect during the Recovery Stage of Stroke: A Pilot Study.

PURPOSE: To determine if functional connectivity measured with resting-state functional MRI could be used as a tool to assess unilateral spatial neglect during stroke recovery. METHODS: Resting-state functional MRI was performed on 13 stroke patients with lesions in the right cerebral hemisphere and 31 healthy subjects. The functional connectivity score was defined as a correlation of a target region with the right inferior parietal lobule. Spatial neglect was measured with a behavioral inattention test. RESULTS: First, the functional connectivity scores between the right inferior parietal l

Visual scanning behavior in patients with homonymous hemianopia.

This study examined oculomotor scanning behavior in 60 patients suffering from homonymous hemianopia due to postgeniculate damage. Eye movements were recorded using an infra-red recording technique during performance of a visual searching task. In 24 patients (40%) scanning behavior was found to be normal; the remaining 60% showed significantly increased search times. Detailed analysis of patients' eye movements revealed that the pronounced slowing of visual scanning was mainly due to the disordered spatial organization of scanning not only in the affected, but also, to a lesser degree, in the

GABAergic Signaling Underlying REM Sleep Deprivation-Induced Spatial Working Memory Deficits.

INTRODUCTION: Declining spatial working memory (WM) is an early hallmark of Alzheimer's disease (AD). Sleep disturbance exacerbates spatial WM and increases AD risk. The GABAergic system, crucial for sleep regulation, may mediate this link. We thus investigate the relationship between spatial WM and hippocampal GABAergic signaling during rapid eye movement sleep deprivation (REM-SD) in AD model mice. METHODS: We assessed spatial and non-spatial WM, locomotor activity, and anxiety-like behavior in 6-month-old triple transgenic (3xTg) AD mice and wild-type (WT) controls, with and without REM-SD

A single dose of ethanol in adult mice transiently alters the organization of spontaneous exploratory behaviors in a dark open field.

Acute exposure to ethanol is known to produce significant alterations to the functions of the hippocampus leading to "blackouts" which are defined as temporary disruptions to the encoding and retention of memories. The hippocampus has a prominent role in generating representations of spatial location and contributes to spatial navigation. In open environments, spatial exploratory behaviors are organized such that animals spontaneously alternate between long duration stops (absence of movement) at a specific home base location and exploratory trips or progressions (locomotion between locations)

Intelligent judgements over health risks in a spatial agent-based model.

BACKGROUND: Millions of people worldwide are exposed to deadly infectious diseases on a regular basis. Breaking news of the Zika outbreak for instance, made it to the main media titles internationally. Perceiving disease risks motivate people to adapt their behavior toward a safer and more protective lifestyle. Computational science is instrumental in exploring patterns of disease spread emerging from many individual decisions and interactions among agents and their environment by means of agent-based models. Yet, current disease models rarely consider simulating dynamics in risk perception an

Pathfinder: open source software for analyzing spatial navigation search strategies.

Spatial navigation is a universal behavior that varies depending on goals, experience and available sensory stimuli. Spatial navigational tasks are routinely used to study learning, memory and goal-directed behavior, in both animals and humans. One popular paradigm for testing spatial memory is the Morris water maze, where subjects learn the location of a hidden platform that offers escape from a pool of water. Researchers typically express learning as a function of the latency to escape, though this reveals little about the underlying navigational strategies. Recently, a number of studies hav

Spatiotemporal encoding of search strategies by prefrontal neurons.

Working memory is capacity-limited. In everyday life we rarely notice this limitation, in part because we develop behavioral strategies that help mitigate the capacity limitation. How behavioral strategies are mediated at the neural level is unclear, but a likely locus is lateral prefrontal cortex (LPFC). Neurons in LPFC play a prominent role in working memory and have been shown to encode behavioral strategies. To examine the role of LPFC in overcoming working-memory limitations, we recorded the activity of LPFC neurons in animals trained to perform a serial self-ordered search task. This tas

Overview of the Role of Spatial Factors in Indoor SARS-CoV-2 Transmission: A Space-Based Framework for Assessing the Multi-Route Infection Risk.

The COVID-19 pandemic has lasted from 2019 to 2022, severely disrupting human health and daily life. The combined effects of spatial, environmental, and behavioral factors on indoor COVID-19 spread and their interactions are usually ignored. Especially, there is a lack of discussion on the role of spatial factors in reducing the risk of virus transmission in complex and diverse indoor environments. This paper endeavours to summarize the spatial factors and their effects involved in indoor virus transmission. The process of release, transport, and intake of SARS-CoV-2 was reviewed, and six tran
