Impact of overexpression of wild-type CFTR and elexacaftor-tezacaftor-ivacaftor on oxylipin production by the CFBE41o- bronchial epithelial cell line.

A hallmark of cystic fibrosis (CF) is dysregulated lipid metabolism marked by an imbalance of pro-inflammatory to pro-resolving metabolites. Despite the breadth of evidence associating mutation of the cystic fibrosis conductance regulator (CFTR) with dysregulation of the production of oxylipins, oxidized lipid mediators with specialized functions generated during inflammation, few studies have directly measured whether overexpression of wild-type (WT) CFTR is sufficient to equilibrate oxylipin levels in CF models. In this study, targeted lipidomics was used to compare the oxylipin profiles of

Green synthesis of silver nanoparticles using Padina tetrastromatica against malaria and deciphering the mechanism through machine learning-driven metabolomics and network pharmacology.

Malaria remains a major public health challenge due to drug and insecticide resistance, underscoring the need for novel therapies with distinct mechanisms of action. In this study, silver nanoparticles were green-synthesized using the brown marine algae Padina tetrastromatica (Ag-PT) and evaluated through integrated in vitro, in vivo, metabolomics, network pharmacology, and in silico approaches. Ag-PT showed potent antiplasmodial activity, with significantly lower IC50 values and superior parasite suppression compared to chemically synthesized silver nanoparticles. Untargeted metabolomics reve

Maresin-1 ameliorates renal oxidative stress and inflammation in diabetic kidney disease mice partially dependent on apolipoprotein D upregulation.

Diabetic kidney disease (DKD) is a prevalent diabetic microvascular complication driven by persistent, unresolved renal inflammation, while specialized pro-resolving mediators (SPMs) represented by Maresin-1 (MaR1) have emerged as promising candidates against DKD. This study investigated whether apolipoprotein D (APOD) acts as a critical downstream mediator of MaR1. A mouse model of type 2 diabetic kidney disease was established using a high-fat diet combined with streptozotocin. In vivo pharmacologic intervention and in vitro high glucose-induced HK2 cell culture combined with proteomic scree

DHA-Derived Lipid Mediators Attenuate Osteoarthritis by Resolving Inflammation and Protecting Cartilage in Association with the SIRT1 Signaling Pathway.

Osteoarthritis (OA) is a chronic degenerative joint disease characterized by persistent low-grade inflammation and progressive cartilage destruction. Macrophage-driven inflammatory responses contribute to extracellular matrix (ECM) degradation and accelerate disease progression. Here, we investigated the therapeutic potential of a DHA-derived lipid mediator mixture (LM), generated via soybean lipoxygenase and composed of 17S-hydroxydocosahexaenoic acid, resolvin D5, and protectin DX (3:47:50), in regulating macrophage-chondrocyte crosstalk and OA progression. LM significantly reduced IL-6, IL-

Drug Therapy for Abdominal Aortic Aneurysms Utilizing Omega-3 Unsaturated Fatty Acids and Their Derivatives.

BACKGROUND: Abdominal aortic aneurysms (AAA) are life-threatening because of the potential for rupture, resulting in death. The current standard treatment for AAA is surgery, comprising laparotomic graft replacement and endovascular repair. However, because surgery carries the risk of major complications and re-intervention, drug therapies are desirable because they may reduce the occurrence of enlargement and rupture. OBJECTIVE: Recent research shows that the progression of AAA is related to inflammatory reactions, especially those in the NF-κB pathway. Omega-3 polyunsaturated fatty acids (P

A cautionary tale: Non-steroidal anti-inflammatory drug use and localized provoked vulvodynia.

Localized provoked vulvodynia (LPV) is characterized by chronic vulvar pain upon light touch to the vulvar vestibule, a specialized ring of tissue immediately surrounding the vaginal opening. LPV affects ∼14 million people in the US. Current treatments for LPV include nonsteroidal anti-inflammatory drugs (NSAIDs), but clinical evidence has demonstrated that they are ineffective for vulvar pain. Here, the effects of NSAID treatment on inflammation and resolution in an in vitro model of LPV were explored using enzyme-linked immunosorbent assays (ELISAs), calcium imaging, and metabololipidomics.

Lipid mediators in glaucoma: Unraveling their diverse roles and untapped therapeutic potential.

Glaucoma is a complex neurodegenerative disease characterized by optic nerve damage and visual field loss, and remains a leading cause of irreversible blindness. Elevated intraocular pressure (IOP) is a critical risk factor that requires effective management. Emerging research underscores dual roles of bioactive lipid mediators in both IOP regulation, and the modulation of neurodegeneration and neuroinflammation in glaucoma. Bioactive lipids, encompassing eicosanoids, specialized pro-resolving mediators (SPMs), sphingolipids, and endocannabinoids, have emerged as crucial players in these proce

Sleep disturbance affects inflammatory resolution in Long COVID.

BACKGROUND: Sleep disturbance, which is a common symptom in Long COVID, promotes a pro-inflammatory state and dysregulates lipid-derived specialized pro-resolving mediators (SPMs), presumably contributing to chronic unresolved inflammation. This study aimed to investigate the role of sleep disturbance in inflammatory resolution in Long COVID. METHODS: We studied 39 participants (30F/9M, age range 22-68 years), including 31 individuals with Long COVID and 8 SARS-CoV-2-infected controls, who did not develop Long COVID. The study consisted of a 14-day at-home phase followed by a 1-day (24-h) in-

B10 Adoptive Transfer Ameliorates Inflammation and Bone Loss in Experimental Periodontitis Through Promoting Macrophage-Mediated Pro-Resolving Functions.

Immune cell-cell interaction plays a critical role in the regulation of inflammatory responses. IL-10-expressing regulatory B cells (B10) alleviate the periodontal inflammation and bone loss by secreting IL-10, which can stimulate pro-resolving macrophage differentiation. However, little is known about the dynamic B10-macrophage interaction and the specific B10-induced pro-resolving functions in vivo in periodontitis. This study aimed to determine the dynamic local infiltration of B10 and macrophages, and the B10-induced macrophage differentiation and functions in experimental periodontitis. B

Inflammation pro-resolving mechanisms associated with puerperal metritis in dairy cows.

In healthy dairy cows, a puerperal transient inflammation of the endometrium is necessary for the clearance of placental debris and bacteria and to achieve complete uterine involution. However, metabolic, immune and inflammatory disturbances leading to uterine dysbiosis may disturb the control of this physiological inflammation, and allow the establishment of metritis. There is a gap in knowledge, regarding the pro-resolving mechanisms controlling the puerperal endometrial physiologic inflammation, and their putative disturbance behind the establishment of metritis. This observational retrospe

Aspirin and blood pressure: Effects when used alone or in combination with antihypertensive drugs.

Arterial hypertension is a major risk factor for cardiovascular and renal events. Lowering blood pressure is thus an important strategy for reducing morbidity and mortality. Since low-dose aspirin is a cornerstone in the prevention of adverse cardiovascular outcomes, combined treatment with aspirin and antihypertensive drugs is very common. However, the impact of aspirin therapy on blood pressure control remains a subject of intense debate. Recent data suggest that the cardioprotective action of aspirin extends beyond its well-known antithrombotic effect. Aspirin has been shown to trigger the

Signaling lipids: An overview of emerging physiological functions.

Lipids are now recognized as central regulators of cellular signaling, extending well beyond their traditional roles in membrane structure and energy storage. As spatially confined and rapidly inducible messengers, signaling lipids integrate membrane dynamics, metabolism, and signal transduction to control processes like inflammation, immune responses, vesicular trafficking, cytoskeletal organization, and cell fate decisions. This review highlights key mechanistic principles underlying lipid signaling specificity, including localized biosynthesis, enzymatic turnover, and receptor engagement. W

Pro-resolving lipid mediators in diseases: exploring the molecular basis and clinical implication.

Inflammation resolution is now understood as an active and highly coordinated biological process rather than a passive decline in inflammatory signals. When this program fails, inflammation may become persistent and gradually shift to a chronic pathological state. Such unresolved inflammation is increasingly recognized as a core driver of cardiovascular disease, metabolic disorders, autoimmune pathologies, neurodegeneration, and tumor progression. Although conventional anti-inflammatory drugs can suppress inflammatory mediators, they do not restore immune balance or actively promote resolution

Fatty Acid Oxidation and Pro-Resolving Lipid Mediators Are Related to Male Infertility.

Specialized pro-resolving lipid mediators regulate the resolution of acute inflammation. They are formed by enzymatic oxygenation of polyunsaturated fatty acids and are divided into families including lipoxins, resolvins, protectins, and maresins. Resolvin D1 (RvD1), produced by docosahexaenoic acid, exerts anti-inflammatory and pro-resolving activities. This research aimed to investigate the implication of seminal RvD1 in human infertility. Infertile patients (n° 67) were grouped based on pathological reproductive conditions as idiopathic infertility, varicocele, and leukocytospermia; the fou

Targeted Modulation of Lipid Mediator-Biosynthetic Enzymes as Strategy for Inflammation Resolution Pharmacology.

Lipid mediators (LMs) comprise a large variety of signaling molecules that are enzymatically produced from polyunsaturated fatty acids (PUFA) in a wide biosynthetic network. These pivotal mediators regulate all stages of inflammation, from initiation, progression, and maintenance to resolution and regain of tissue homeostasis. While leukotrienes and prostaglandins, produced from arachidonic acid (AA) by the 5-lipoxygenase (LOX) and cyclooxygenase (COX) pathways, respectively, mediate/promote inflammatory reactions and related symptoms, the specialized pro-resolving mediators (SPMs) terminate a

Polyunsaturated fatty acids in kidney diseases: Navigating the fine line between healing and damage.

Polyunsaturated fatty acids (PUFAs) regulate renal inflammation through metabolites generated by COX, LOX, and CYP pathways. While prostaglandins, leukotrienes, and 20-hydroxyeicosatetraenoic acid (20-HETE) exacerbate kidney injury, epoxyeicosatrienoic acids (EETs), lipoxins, and other specialized pro-resolving mediators (SPMs) counteract inflammation and promote tissue repair. These lipid mediators also modulate nuclear receptors such as peroxisome proliferator-activated receptors (PPARs) and fibrotic pathways like TGF-β signaling. Disease-specific imbalances in PUFA metabolism have been impl

The Role of Endogenous Eicosapentaenoic Acid and Docosahexaenoic Acid-Derived Resolvins in Systemic Sclerosis.

In the acute inflammation, these lipid mediators limit polymorphonuclear cells infiltration, proinflammatory cytokine production; promote efferocytosis, and regulate several cell types being important roles in innate and adaptive immunity. Any dysregulation or defect of the resolution phase result in prolonged, persistent inflammation and eventually fibrosis. Resolvins are implicate

RvD1/FPR2 attenuates cerebral ischemic injury through inhibiting inflammation and oxidative stress.

Oxidative stress and inflammation are two closely related pathogenesis processes during ischemic injury. The current study investigated the neuroprotective effects of resolvin D1 (RvD1, a pro-resolving mediator) and its corresponding receptor formyl peptide receptor 2 (FPR2) in a rat model of middle cerebral artery occlusion (MCAO). Dynamic expressions of endogenous RvD1 and FPR2 in ischemic hemisphere were firstly examined by ELISA and western blot respectively. To explore the therapeutic effects of RvD1/FPR2 on ischemic injury, exogenous RvD1 were given at 0.4 µg/kg after MCAO (i.p.). Boc-2

Resolvin D5 Inhibits CXCL8 Expression in Colonic Epithelial Cells Through Activating GPR101 to Impede Neutrophil Recruitment and Consequently Alleviate Ulcerative Colitis.

In the colonic mucosal tissues of patients with ulcerative colitis (UC), the levels of resolvin D5 (RvD5), a specialized pro-resolving mediator, are significantly reduced and negatively correlate with disease severity. Whether RvD5 can ameliorate UC remain unclear. Here, we show that exogenous supplement of RvD5 remarkably reduces neutrophil infiltration within the colonic mucosal epithelial layers and alleviates dextran sulfate sodium-induced colitis in mice. Adoptive transfer of neutrophils significantly attenuates the alleviation of RvD5 against colitis, indicating that inhibition of neutro
