Effects of exogenous lactoferrin on characteristics and functions of bovine epididymal, ejaculated and frozen-thawed sperm.

The purpose of this study was to investigate effects of addition of lactoferrin on characteristics and functions of bovine epididymal, ejaculated, and frozen-thawed sperm. The addition of lactoferrin was significantly (p < .05) effective on increasing values of progressive motility, straightness, and linearity in caput epididymal sperm and values of motility in cauda epididymal sperm. When ejaculated sperm were incubated in capacitation medium, percentages of motile and progressively motile sperm decreased largely within the first period of 30 min, followed by only minor changes. However, the

Dysregulation of sphingolipid and cholesterol homeostasis imposes oxidative stress in human spermatozoa.

Infertility is a significant public health concern, affecting one in six couples globally, with male-factor infertility representing half of all cases. Obesity is an important health problem and is increasingly linked to poor reproductive outcomes as it induces metabolic disturbances and dyslipidemia, both of which impair male fertility. Dyslipidemia, characterized by abnormal lipid profiles, is associated with declining sperm quality and alters cholesterol and sphingolipid abundances in the seminal plasma. Sperm capacitation enables spermatozoa to acquire fertilizing competence by promoting c

Development of a biomolecular approach to identify sperm functions and fertility using sperm RNAs.

Infertility affects a significant percentage of couples worldwide, and male factors contribute significantly to this problem. Traditional assessments of male fertility rely primarily on parameters such as sperm motility, morphology, viability, and concentration. However, these metrics often do not provide a comprehensive understanding of sperm function, which is critical not only for fertilization but also for successful embryo development. Herein, we used porcine spermatozoa as a model to investigate the potential of sperm RNA markers in assessing various aspects of sperm function from motili

Bicarbonate and its role in mammalian sperm function.

This paper deals with the effects of bicarbonate induced signaling pathways on plasma membrane lipid organization and downstream protein signaling, and their role in sperm-egg interactions. It also provides an overview of results that indicate that bicarbonate responses are not related to cell death or apoptosis. The information presented shows that only those sperm cells that have functionally completed maturation in the epididymis are sensitive to bicarbonate whereas immature sperm fail to respond to this physiological challenge. Therefore, it is important to selectively analyze the responsi

Caput Ligation Renders Immature Mouse Sperm Motile and Capable to Undergo cAMP-Dependent Phosphorylation.

Mammalian sperm must undergo two post-testicular processes to become fertilization-competent: maturation in the male epididymis and capacitation in the female reproductive tract. While caput epididymal sperm are unable to move and have not yet acquired fertilization potential, sperm in the cauda epididymis have completed their maturation, can move actively, and have gained the ability to undergo capacitation in the female tract or in vitro. Due to the impossibility of mimicking sperm maturation in vitro, the molecular pathways underlying this process remain largely unknown. We aimed to investi

Sperm Adhesion Molecule 1 (SPAM1) Distribution in Selected Human Sperm by Hyaluronic Acid Test.

The failures of binding to the oocyte zona pellucida are commonly attributed to defects in the sperm recognition, adhesion, and fusion molecules. SPAM1 (sperm adhesion molecule 1) is a hyaluronidase implicated in the dispersion of the cumulus-oocyte matrix. Therefore, the aim of this study was to characterize the SPAM1 distribution in the different physiological conditions of human sperm. Specifically, we evaluated the location of the SPAM1 protein in human sperm before capacitation, at one and four hours of capacitation and after hyaluronic acid (HA) selection test by fluorescence microscopy.

Oviductal high concentration of K+ suppresses hyperpolarization but does not prevent hyperactivation, acrosome reaction and in vitro fertilization in hamsters.

Mammalian sperm have to undergo capacitation to be fertilization competent. Capacitated sperm in vitro show hyperpolarization of the membrane potential. It has been reported that in mouse membrane hyperpolarization is necessary for the acrosome reaction. We recently found that the fluid of the hamster oviduct, where fertilization occurs, contained a high potassium (K+) concentration (~20 mEq/l). This high K+ concentration could depolarize the membrane potential and prevent the acrosome reaction/fertilization. Conversely, some beneficial effects on capacitation of high K+ concentration or a hig

Nitric oxide regulates the phosphorylation of the threonine-glutamine-tyrosine motif in proteins of human spermatozoa during capacitation.

Reactive oxygen species (superoxide anion, hydrogen peroxide, and nitric oxide) are involved in human sperm capacitation and associated tyrosine (Tyr) phosphorylation through a cAMP- and protein kinase A-mediated pathway. Recently, we evidenced the double phosphorylation of the threonine-glutamine-Tyr motif (P-Thr-Glu-Tyr-P) in human sperm proteins of 80 and 105 kDa during capacitation. The objective of the present study was to investigate the role of reactive oxygen species in the regulation of this process and to immunolocalize the P-Thr-Glu-Tyr-P motif in human spermatozoa. The sodium-proton exchangers sNHE and NHE1 control plasma membrane hyperpolarization in mouse sperm.

Sperm capacitation is a complex process that takes place in the female reproductive tract and empowers mammalian sperm with the competence to fertilize an egg. It consists of an intricate cascade of events that can be mimicked in vitro through incubation in a medium containing essential components, such as bicarbonate, albumin, Ca2+, and energy substrates, among others. Genetic and pharmacological studies have underscored the unique significance of the K+ channel SLO3 in membrane potential hyperpolarization, as evidenced by the infertility of mice lacking its expression. Notably, two key molec

Src Kinase Is the Connecting Player between Protein Kinase A (PKA) Activation and Hyperpolarization through SLO3 Potassium Channel Regulation in Mouse Sperm.

Plasma membrane hyperpolarization is crucial for mammalian sperm to acquire acrosomal responsiveness during capacitation. Among the signaling events leading to mammalian sperm capacitation, the immediate activation of protein kinase A plays a pivotal role, promoting the subsequent stimulation of protein tyrosine phosphorylation that associates with fertilizing capacity. We have shown previously that mice deficient in the tyrosine kinase cSrc are infertile and exhibit improper cauda epididymis development. It is therefore not clear whether lack of sperm functionality is due to problems in epidi

Exogenous neurotensin modulates sperm function in Japanese Black cattle.

Recently, the conception rates after artificial insemination have been pointed out to decline continuously. To overcome this problem, the control of frozen and thawed sperm quality is required. However, the mechanism of bovine sperm functional regulation is still largely unknown. In mammals, the ejaculated sperm are capable of showing fertilizing ability during migration in the female reproductive organs. It is well known that these female organs secrete several factors contributing to sperm capacitation. We previously reported that neurotensin (NT) secreted from the oviduct and cumulus cells

ACRBP (Sp32) is involved in priming sperm for the acrosome reaction and the binding of sperm to the zona pellucida in a porcine model.

In boar sperm, we have previously shown that capacitation is associated with the appearance of the p32 tyrosine phosphoprotein complex. The principal tyrosine phosphoprotein involved in this complex is the acrosin-binding protein (ACRBP), which regulates the autoconversion of proacrosin to intermediate forms of acrosin in both boar and mouse sperm. However, the complete biological role of ACRBP has not yet been elucidated. In this study, we tested the hypothesis that tyrosine phophorylation and the presence of the ACRBP in the sperm head are largely necessary to induce capacitation, the acroso

Intracellular calcium and protein tyrosine phosphorylation during the release of bovine sperm adhering to the fallopian tube epithelium in vitro.

In mammals, sperm adhesion to the epithelial cells lining the oviductal isthmus plays a key role in the maintenance of motility and in the selection of superior quality subpopulations. In the bovine species, heparin and other sulfated glycoconjugates powerfully induce the synchronous release of sperm adhering to tubal epithelium in vitro and may represent the signal which triggers release at ovulation in vivo. Sperm detachment may be due either to surface remodeling or to hyperactivation brought about by capacitation. In this paper, the dynamics of intracellular free Ca2+concentration ([Ca2+]i

Prokaryotic expression, purification, and functional characterization of recombinant buffalo (Bubalus bubalis) cysteine-rich secretory protein 1 on sperm physiology.

Cysteine-rich secretory protein 1 (CRISP-1), an epididymis-origin glycoprotein, is recognized as a sperm decapacitation factor. Incorporation of CRISP-1 in semen prior to preservation may extend sperm fertility by preventing premature capacitation. This study aimed to produce bioactive recombinant buffalo CRISP-1 in Escherichia coli and evaluate its effects on sperm motility, capacitation, and associated signalling mechanisms. The expression of a 27.6 kDa fragment of buffalo CRISP-1 (C15-C242) protein was optimized by using E. coli BL21(DE3) and BL21(DE3)-CodonPlus strains under different IPTG

Estrous Sheep Serum-Derived Extracellular Vesicles Deliver Proteins That Modulate Sperm Motility in Sheep.

Sperm capacitation requires signals from the female reproductive environment, although the molecular mechanisms governing this process remain poorly understood. Estrous sheep serum (ESS) is widely used in in vitro fertilization of small ruminants, contributing to mimic the female reproductive fluids. Among ESS components, extracellular vesicles (ESS-EVs) may mediate the transfer of molecular signals to spermatozoa and support the modifications observed when sperm are incubated with ESS. To explore this possibility, ESS-EVs were isolated and validated according to MISEV2023 guidelines, and thei

Dysregulation of intracellular pH is a cause of impaired capacitation in Slc22a14-deficient mice.

Solute carrier 22a member 14 (SLC22A14) plays a critical role in male infertility in mice. We previously revealed that one of the causes of infertility is impaired capacitation. However, the molecular mechanism remained unclear. Here, we show that the influx of HCO3-, a trigger of capacitation, is impaired and intracellular pH (pHi) is decreased in the sperm of Slc22a14 knockout (KO) mice. While intracellular cAMP concentration did not increase during capacitation in Slc22a14 KO spermatozoa, HCO3--dependent soluble adenylate cyclase activity was normal, and the addition of 8-bromo cAMP rescued

Mouse sperm energy restriction and recovery (SER) revealed novel metabolic pathways.

Mammalian sperm must undergo capacitation to become fertilization-competent. While working on mice, we recently developed a new methodology for treating sperm in vitro, which results in higher rates of fertilization and embryo development after in vitro fertilization. Sperm incubated in media devoid of nutrients lose motility, although they remain viable. Upon re-adding energy substrates, sperm resume motility and become capacitated with improved functionality. Here, we explore how sperm energy restriction and recovery (SER) treatment affects sperm metabolism and capacitation-associated signal

Expression of gp20, a human sperm antigen of epididymal origin, is reduced in spermatozoa from subfertile men.

gp20, a sialylglycoprotein of human sperm homologous to CD52, is present everywhere on the surface of the freshly ejaculated sperm but is prevalently localized in the equatorial region of the head of capacitated sperm. In the present study, we confirmed this feature on large scale and correlated equatorial exposure of the antigen to the presence of serum albumin (SA) in the capacitation medium. Furthermore, we analyzed the relationship between the presence of the antigen and its equatorial exposure after capacitation and fertility, by comparing immunostaining for gp20 in the motile fraction of

Progesterone receptor availability in mouse spermatozoa during epididymal transit and capacitation: ligand blot detection of progesterone-binding protein.

The goals of the present study were to determine the availability of progesterone (P4) receptor (P4r) in mouse sperm during maturation and capacitation and to make the first steps toward a characterization of P4r. It has been proposed that P4 is able to induce an acrosomal reaction (AR) by using a membrane P4r. This induction was verified in sperm isolated from the cauda epididymis (fully mature) when incubated in specific conditions that capacitate sperm. First, we set up the conditions in our laboratory to induce an AR in mature and capacitated sperm triggered by P4 that was detected by a ch

The secretion and metabolism of cumulus cells support fertilization in the bovine model.

The cumulus oophorus is a structure that surrounds the mammalian egg and plays a key role in fertilization. However, very little is known with regards to how secretions from the cumulus cells can specifically promote fertilization. We hypothesized that secretions from bovine cumulus cells, and the reduction of oxygen stress by metabolic change, would enhance the fertilization capacity of sperm during in vitro fertilization (IVF) procedures. To prove our hypothesis, sperm were pre-incubated in chemically defined capacitation media containing methyl-beta-cyclodextrin and used to inseminate cumul
