Hexachlorocyclohexane impairs human sperm motility by affecting lysine glutarylation and mitochondrial functions.

Decreased sperm motility is a leading cause of male infertility and persistent organic pollutants are known to contribute significantly to the development of this disease. The effects of organochlorine pesticides such as hexachlorocyclohexane (HCH) on human sperm function and their mechanisms of action have received much attention, but are still not fully understood. Herein, we discovered that HCH has a concentration- and time-dependent inhibitory effect on human sperm motility in vitro. Moreover, HCH could reduce the levels of lysine glutarylation (Kglu) and glucose-6-phosphate dehydrogenase

Multistage Optimization Using a Modified Gaussian Mixture Model in Sperm Motility Tracking.

Infertility is a condition whereby pregnancy does not occur despite having unprotected sexual intercourse for at least one year. The main reason could originate from either the male or the female, and sometimes, both contribute to the fertility disorder. For the male, sperm disorder was found to be the most common reason for infertility. In this paper, we proposed male infertility analysis based on automated sperm motility tracking. The proposed method worked in multistages, where the first stage focused on the sperm detection process using an improved Gaussian Mixture Model. A new optimizatio

Prohibitin (PHB) interacts with AKT in mitochondria to coordinately modulate sperm motility.

Prohibitin (PHB), an evolutionarily conserved mitochondrial inner membrane protein, is highly expressed in cells that require strong mitochondrial function. Recently, we demonstrated that the deletion of Phb in spermatocytes results in impaired mitochondrial function. In addition, PHB expression in the mitochondrial sheath of human sperm has a significantly negative correlation with mitochondrial reactive oxygen species levels, but a positive one with mitochondrial membrane potential and sperm motility. These results suggest that mitochondrial PHB expression plays a role in sperm motility. How

Effect of low-frequency electric field screening on motility of human sperm.

INTRODUCTION: The human body is constantly exposed to an extremely low electromagnetic field (ELF-EMF), in particular at 50 Hz, emitted by power lines, domestic distribution lines, electrical appliances, etc. It is assumed that the increase in electromagnetic exposure may cause adverse effects upon human health, as well as raising concerns regarding the impact on human fertility. OBJECTIVE: The aim of this in vitro study was to investigate the influence of ELF-EMF with a frequency of 50 Hz on the motility of human sperm. At the same time, the effectiveness of the dielectric screen constructed

Abnormal arachidonic acid metabolic network may reduce sperm motility via P38 MAPK.

Asthenozoospermia is a common cause of male infertility, the aetiology of which remains unclear in 50-60% of cases. The current study aimed to characterize metabolic alterations in asthenozoospermic seminal plasma and to explore the signalling pathways involved in sperm motility regulation. At first, high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry was used to detect the targeted metabolic network of arachidonic acid (AA). Metabolomic multivariate data analysis showed significant distinction of AA metabolites between asthenozoospermic and healthy seminal

Measuring Sperm Guidance and Motility within the Caenorhabditis elegans Hermaphrodite Reproductive Tract.

Successful fertilization is fundamental to sexual reproduction, yet little is known about the mechanisms that guide sperm to oocytes within the female reproductive tract. While in vitro studies suggest that sperm of internally fertilizing animals can respond to various cues from their surroundings, the inability to visualize their behavior inside the female reproductive tract creates a challenge for understanding sperm migration and mobility in its native environment. Here, we describe a method using C. elegans that overcomes this limitation and takes advantage of their transparent epidermis.

Ubiquitin Carboxy-Terminal HydrolaseL3 Correlates with Human Sperm Count, Motility and Fertilization.

Ubiquitin C-terminal hydrolase L3 (UCHL3) belongs to the group of deubiquitinating enzymes and plays a part in apoptosis of germ cells and the differentiation of spermatocytes into spermatids. However, the exact role of UCHL3 in human spermatogenesis and sperm function remains unknown. Here we examined the level and activity of UCHL3 in spermatozoa from men with asthenozoospermia (A), oligoasthenozoospermia (OA) or normozoospermia (N). Immunofluorescence indicated that UCHL3 was mainly localized in the acrosome and throughout the flagella, and western blotting revealed a lower level in A or OA

Optimum calcium concentration: a crucial factor in regulating sperm motility in vitro.

Sperm motility can be maintained in vitro by incubation in a defined medium under specific conditions. In most studies, the exact role of various constituents of epididymal fluid, including calcium, has remained obscure. Most of the culture media have included millimolar concentrations of calcium, but previous reports have indicated that millimolar calcium inhibits sperm motility. In this present study, we sought the optimum concentration of extracellular calcium required for optimum sperm motility. This study showed that extracellular calcium has a concentration-dependent biphasic role in mot

Transient Ca2+ mobilization caused by osmotic shock initiates salmonid fish sperm motility.

Salmonid fish sperm motility is known to be suppressed in millimolar concentrations of extracellular K(+), and dilution of K(+) upon spawning triggers cAMP-dependent signaling for motility initiation. In a previous study, however, we demonstrated that suspending sperm in a 10% glycerol solution and subsequent dilution into a low-osmotic solution induced motility independently of extracellular K(+) and cAMP. In the present study, we further investigated the glycerol-induced motility mechanism. We found that treatment with solutions consisting of organic or inorganic ions, as well as glycerol, i

In vitro initiated sperm forward motility in caput spermatozoa: weak and transient.

Testicular spermatozoa during journey through epididymis acquire forward motility, which is essential for fertility. To understand the biochemistry of sperm motility initiation, various initiation media have been developed that permitted high level of motility induction (55-60%) in the immature caput-spermatozoa in presence of activating principles: theophylline, bicarbonate and epididymal plasma (EP) when analysed microscopically. Here, we show for the first time using caprine model that stability and quality of in vitro-induced motility in the caput spermatozoa is insignificant in contrast t

Effects of Shao-Fu-Zhu-Yu-Tang on motility of human sperm.

Shao-Fu-Zhu-Yu-Tang (SFZYT) is reportedly beneficial to sperm. In this study, we examined sperm acrosomal activity and serum free radical changes to evaluate the possible mechanism of SFZYT. A clinical study evaluated the sperm count and motility in 36 patients with chronic prostatitis before and after treatment for 60 days. The results revealed a significant increase in sperm motility after treatment as evaluated by computer-assisted semen analysis (17.27 +/- 9.00 versus 28.29 +/- 10.00, p < 0.01). An increase in sperm quantity and quality was observed by count and morphology with a high-powe

Purification and characterization of reversible sperm motility inhibitors from porcine seminal plasma.

Two sperm motility inhibitors (SMI1 and SMI2) were purified from porcine seminal plasma with high performance liquid chromatography (HPLC). Their molecular weights are about 15,000 as estimated by SDS-polyacrylamide gel electrophoresis (SDS-PAGE). Both of them decreased the percentage of motile spermatozoa in a dose-dependent manner. The inhibitory effect can be abolished by addition of the porcine follicular fluid. Both SMI1 and SMI2 have similar amino acid composition, suggesting that they may be structurally related. They also have inhibitory effect on chymotrypsin.

Alloplastic spermatocele: poor sperm motility in intraoperative epididymal fluid contraindicates prosthesis implantation.

After vasectomy reversal by vasovasostomy or vasoepididymostomy motile sperm appear commonly in the semen even when only nonmotile sperm are present in the intraoperative vasal or epididymal fluid. We studied patients with bilateral congenitally absent vasa deferentia to see if relief of obstruction by implantation of an alloplastic spermatocele also benefits sperm motility in such patients. A total of 130 alloplastic spermatoceles was implanted in 91 patients. Of 21 patients with only nonmotile sperm in the epididymal fluid intraoperatively only 1 had motile sperm in the postoperative aspirat

LCAT acts as a molecular switch and regulates rooster sperm motility in vitro.

Sperm motility is a critical determinant of semen quality, yet the molecular mechanisms regulating motility remain unclear. In this study, we identified Yufen 1 roosters with significant differences in sperm motility among highly motile sperm samples stored under different conditions. By integrating the seminal plasma proteome and seminal plasma metabolome data, 181 proteins and 112 metabolites were identified as differentially expressed between the high motility group (HMG) and low motility group (LMG). Lecithin cholesterol acyltransferase (LCAT) was upregulated in the LMG. Targeted LCAT acti

The structure of mammalian sperm axoneme provides insights into motility and disease mechanisms.

Mammalian sperm are highly specialized cells consisting of a head containing paternal genome and a flagellar tail responsible for propulsion. At the core of the flagellum lies the axoneme-an evolutionarily conserved microtubule-based organelle essential for generating motility. Since the first visualization of the canonical "9 + 2" axonemal architecture by transmission electron microscopy (TEM) in the 1950s, advancements in cryo-electron microscopy (cryo-EM) have revealed unprecedented details of its structural organization and molecular composition. These breakthroughs have illuminated how ax

Sperm Motility Is Modulated by F4-Neuroprostane via the Involvement of Ryanodine Receptors.

F4-Neuroprostanes (F4-NeuroPs), oxidative metabolites of docosahexaenoic acid, act as bioactive lipid mediators enhancing sperm motility and induce capacitation-like changes in vitro. Their biological action is proposed to involve sperm ion channels, in particular ryanodine receptors (RyRs), which regulate intracellular calcium homeostasis. We evaluated the effects of dantrolene, a RyR inhibitor, on motility and vitality of a selected spermatozoa at different concentrations (10, 30, 50, 100 μM). Then sperm motility, acrosome integrity, and RyR localization following co-incubation with dantrole

Receptor expression is essential for forward motility in the course of sperm cell maturation.

Forward motility stimulating factor (FMSF) is a glycoprotein previously purified from buffalo blood serum that promotes progressive motility of caprine caudal spermatozoa. We have prepared a functionally active covalent conjugate of this factor with horseradish-peroxidase (HRP) to obtain an idea of its binding efficacy on maturing spermatozoa. Receptor-assay was performed using FMSF-HRP conjugate in saturating conditions to bind with spermatozoa isolated from different epididymal segments. Activity and binding profile of the motility stimulating factor coincided, suggesting both these paramete
