Penetration in 3D tumor spheroids and explants: Adding a further dimension to the structure-activity relationship of cell-penetrating peptides.

Drug delivery into tumors and metastases is a major challenge in the eradication of cancers such as epithelial ovarian carcinoma. Cationic cell-penetrating peptides (CPPs) are a promising group of delivery vehicles to mediate cellular entry of molecules that otherwise poorly enter cells. However, little is known about their penetration behavior in tissues. Here, we investigated penetration of cationic CPPs in 3D ovarian cancer spheroids and patient-derived 3D tumor explants. Penetration kinetics and distribution after long-term incubation were imaged by confocal microscopy. In addition, sphero

Mitochondrial-targeted nano-enhancer driving a self-amplifying NAD+ feedback loop to reprogram MSC spheroids metabolism for accelerated bone regeneration.

Mitochondrial oxidative phosphorylation (OXPHOS) plays a crucial role in determining the functional fate of mesenchymal stem cells (MSCs). However, effective strategies to precisely modulate mitochondrial bioenergetics within three-dimensional (3D) MSC spheroids to improve bone repair remain limited. In this study, we have developed a mitochondria-targeted nano-enhancer (BP@PDA-TPP/Q) to enhance the therapeutic potential of MSC spheroids by reprogramming their energy metabolism for bone regeneration. The BP@PDA-TPP/Q nano-enhancer was constructed with black phosphorus (BP) nanosheets as a biod

Tuning the Testicular Microenvironment for Enhancing Human Sertoli Cells Maturation and Functionality In Vitro.

Spermatogenesis relies on the highly specialized interaction between germ cells and a supportive somatic niche, yet replicating this environment in vitro remains a major challenge. Primary human Sertoli cells (hSCs), key architects of this niche, often lose their phenotype under conventional culture conditions, limiting the establishment of a stable blood-testis barrier (BTB)-associated phenotype and their ability to support germ cells. Here, we present a structurally organized, human ECM-derived connective tissue equivalent (CTE) designed to support long-term maintenance and organization of h

The role of Piezo1 mechanotransduction in high-grade serous ovarian cancer: Insights from an in vitro model of collective detachment.

Slowing peritoneal spread in high-grade serous ovarian cancer (HGSOC) would improve patient prognosis and quality of life. HGSOC spreads when single cells and spheroids detach, float through the peritoneal fluid and take over new sites, with spheroids thought to be more aggressive than single cells. Using our in vitro model of spheroid collective detachment, we determine that increased substrate stiffness led to the detachment of more spheroids. We identified a mechanism where Piezo1 activity increased MMP-1/MMP-10, decreased collagen I and fibronectin, and increased spheroid detachment. Piezo

Intra-spheroidal BMP-2 delivery in co-cultured spheroids enables concurrent vasculogenesis and osteogenesis in 3D engineered bone constructs.

Critical-sized bone defects demand rapid vascularization for graft success, yet conventional pre-vascularization strategies have failed to coordinate vessel formation with osteoinduction. Here, we developed a GelMA-based 3D construct encapsulating co-cultured spheroids composed of human adipose-derived stem cells and human umbilical vein endothelial cells, incorporating BMP-2-immobilized nanofibers that directly deliver BMP-2 within the spheroid microenvironment. This spatially confined delivery system allowed BMP-2 to concurrently enhance hADSC osteogenesis, HUVEC functionality, and pericyte-

Spatially compartmentalized fibrotic tumor microtissues via embedded coaxial 3D bioprinting for enhanced barrier function and drug resistance.

Fibrosis is a prominent feature of the tumor microenvironment (TME) that contributes to tumor progression and therapeutic resistance. Here, we developed an embedded coaxial 3D bioprinting strategy to fabricate spatially compartmentalized fibrotic tumor microtissues with controlled tumor-stromal organization. Compared with mixed spheroids, coaxial spheroids exhibited enhanced hypoxia, fibroblast activation, and ECM deposition, together with reduced drug penetration and increased therapeutic resistance. These responses were supported by molecular and functional analyses. Collectively, our findin

Patient-derived 3D nasal spheroids reveal epithelial changes following Dupilumab therapy in CRSwNP: a preliminary report.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a type 2 inflammatory disease associated with epithelial dysfunction and impaired mucosal barrier integrity. Dupilumab, an IL-4 receptor alpha antagonist, has shown clinical efficacy, but its cellular effects on nasal epithelium remain poorly understood. Advanced in vitro models such as 3D spheroid cultures may provide insight into epithelial organization under treatment. We conducted a preliminary study using nasal epithelial cells obtained from three patient groups: CRSwNP treated with Dupilumab for 16 weeks (n = 3), untreated CRSwNP (n =

Impact of understudied PFAS on Lipid Metabolism in Hepatocyte 2D and 3D in vitro model.

Perfluoroalkyl substances (PFAS) are industrial chemicals with several applications, including heat and water resistance. These chemicals are ubiquitous in our environment, and human exposure to them has been linked to liver injury and disruption of lipid metabolism. This study investigated the effects of PFAS (fluorotelomer sulfonic acids 6:2 FTSA and 8:2 FTSA, and perfluorooctane sulfonamide (PFOSA)) on hepatic lipid metabolism using in vitro human cell models. Initially, we assessed the effects of 10 μM PFOSA, 100 μM 6:2 FTSA, and 100 μM 8:2 FTSA in HepG2 monolayer (2D) cultures, focusing o

New Knowledge About Tissue Engineering Under Microgravity Conditions in Space and on Earth.

Microgravity (µg)-generated three-dimensional (3D) multicellular aggregates can serve as models of tissue and disease development. They are relevant in the fields of cancer and in vitro metastasis or regenerative medicine (tissue engineering). Driven by the 3R concept-replacement, reduction, and refinement of animal testing-µg-exposure of human cells represents a new alternative method that avoids animal experiments entirely. New Approach Methodologies (NAMs) are used in biomedical research, pharmacology, toxicology, cancer research, radiotherapy, and translational regenerative medicine. Vario

Evaluating nanoparticle localisation in glioblastoma multicellular tumour spheroids by surface enhanced Raman scattering.

Glioblastoma multiforme (GBM) is a particularly aggressive and high-grade brain cancer, with poor prognosis and life expectancy, in urgent need of novel therapies. These severe outcomes are compounded by the difficulty in distinguishing between cancerous and non-cancerous tissues using conventional imaging techniques. Metallic nanoparticles (NPs) are advantageous due to their diverse optical and physical properties, such as their targeting and imaging potential. In this work, the uptake, distribution, and location of silica coated gold nanoparticles (AuNP-SHINs) within multicellular tumour sph

Squaramide-Based Supramolecular Materials Drive HepG2 Spheroid Differentiation.

A major challenge in the use of HepG2 cell culture models for drug toxicity screening is their lack of maturity in 2D culture. 3D culture in Matrigel promotes the formation of spheroids that express liver-relevant markers, yet they still lack various primary hepatocyte functions. Therefore, alternative matrices where chemical composition and materials properties are controlled to steer maturation of HepG2 spheroids remain desired. Herein, a modular approach is taken based on a fully synthetic and minimalistic supramolecular matrix based on squaramide synthons outfitted with a cell-adhesive pep

Hyperbolastic growth models: theory and application.

BACKGROUND: Mathematical models describing growth kinetics are very important for predicting many biological phenomena such as tumor volume, speed of disease progression, and determination of an optimal radiation and/or chemotherapy schedule. Growth models such as logistic, Gompertz, Richards, and Weibull have been extensively studied and applied to a wide range of medical and biological studies. We introduce a class of three and four parameter models called "hyperbolastic models" for accurately predicting and analyzing self-limited growth behavior that occurs e.g. in tumors. To illustrate the

Reversible Intracellular Gelation of MCF10A Cells Enables Programmable Control Over 3D Spheroid Growth.

In nature, some organisms survive extreme environments by inducing a biostatic state wherein cellular contents are effectively vitrified. Recently, a synthetic biostatic state in mammalian cells is achieved via intracellular network formation using bio-orthogonal strain-promoted azide-alkyne cycloaddition (SPAAC) reactions between functionalized poly(ethylene glycol) (PEG) macromers. In this work, the effects of intracellular network formation on a 3D epithelial MCF10A spheroid model are explored. Macromer-transfected cells are encapsulated in Matrigel, and spheroid area is reduced by ≈50% com

Application and Comparison of Supervised Learning Strategies to Classify Polarity of Epithelial Cell Spheroids in 3D Culture.

Three-dimensional culture systems that allow generation of monolayered epithelial cell spheroids are widely used to study epithelial function in vitro. Epithelial spheroid formation is applied to address cellular consequences of (mono)-genetic disorders, that is, ciliopathies, in toxicity testing, or to develop treatment options aimed to restore proper epithelial cell characteristics and function. With the potential of a high-throughput method, the main obstacle to efficient application of the spheroid formation assay so far is the laborious, time-consuming, and bias-prone analysis of spheroid

NAD(P)H Quinone Oxidoreductase-1 Expression Promotes Self-Renewal and Therapeutic Resistance in Non-Small Cell Lung Cancer.

Identifying cellular drivers responsible for enhancing cancer cell resistance to therapeutics provides critical information for designing more effective drugs. Populations of slowly growing, self-renewing, chemo-resistant cells purportedly contribute to the development of therapeutic resistance in many solid tumors. In the current study, we implemented a tumor spheroid model to determine whether NAD(P)H quinone oxidoreductase-1 (NQO1) was requisite for self-renewal and promotion of the drug-resistant phenotype in non-small cell lung cancer (NSCLC). We found that stable depletion of NQO1 in A54

Noninvasive Viability Profiling of 3D Spheroids via Flow-through Electrical Impedance Spectroscopy.

Three-dimensional (3D) cell models, such as spheroids and organoids, are powerful tools to enhance the predictive power of preclinical in vitro studies and support clinical translation of anti-cancer therapies. However, their integration into drug discovery pipelines remains limited, particularly for high-throughput screening, where rapid and reliable viability assessment is essential. Conventional viability assays, often dye-based or destructive, remain labor-intensive and poorly suited for high-throughput applications and downstream re-use of the models. In this study, we demonstrate, for th

Acoustofluidic separation of oblate spheroids from spheres using acoustic radiation torque and force.

Cells, bacteria, and other bioparticles exist in diverse shapes, and their morphology plays a pivotal role in biological functions and clinical significance. Various microfluidic approaches have been developed for shape-based separation of bioparticles with the same or similar volume; however, most of them were limited to separation of prolate spheroids from spheres or required a priori labeling and consequent detection. Here, we propose a vertical-type acoustofluidic method for the first separation of oblate spheroids from spheres in a label-free manner. The acoustic radiation torque suppress

Engineered Deoxyribonucleic Acid-Tunable Spheroid Platform for Analysis of Tumor-Macrophage Interactions.

Tumor architecture strongly influences immune cell behavior, yet the individual contributions of cell-cell spacing and hypoxia remain difficult to isolate in vivo. Here, we use a DNA-programmable 3D tumor spheroid platform to generate 4T1 spheroids with tunable intercellular spacing and predictable hypoxia gradients, enabling structural and biochemical cues to be decoupled. Using this system, we compared infiltration and polarization dynamics of immortalized RAW 264.7 macrophages and primary BMDMs. Macrophages readily infiltrated preformed spheroids across all DNA lengths, indicating that the

A multi-well axon array device for evaluation of stimulus responses on human sensory neural spheroid-derived axon fascicles.

Understanding the response of sensory neurons involved in pain perception to external stimuli is crucial for developing chronic pain treatments and determining pain thresholds. In the field of drug discovery, cultured neurons derived from human induced pluripotent stem cells (hiPSCs) are used to evaluate pain perception. Recently, three-dimensional axonal tissues derived from hiPSC-derived sensory neural spheroids have been constructed. These three-dimensional axonal tissues can mimic axon fascicles (AFs) found in the peripheral nervous system, making these tissues promising for cellular exper

A 3D In Vitro Model of the Human Hepatobiliary Junction.

Bile flow is an essential feature of the liver, and disruption of this process contributes to a range of liver pathologies. Efficient bile transport requires coordinated organization between hepatocytes and cholangiocytes at the hepatobiliary junction, a structure that remains poorly captured in existing in vitro models of liver disease. Here, we present a 3D multicellular spheroid-based model of the human hepatobiliary junction. Building on advances in organoid and spheroid engineering, we co-aggregate human hepatocytes and intrahepatic cholangiocytes, supported by murine fibroblasts, into ad
