Enterotoxin production by strains of Staphylococcus aureus isolated from clinical specimens of human and animal origin and from healthy human carriers was investigated. All nine patients admitted to hospital with symptoms of toxic shock syndrome (TSS) yielded enterotoxin-producing strains of S. aureus. Eight of these produced staphylococcal enterotoxin F (SEF). A significantly smaller proportion of strains (42% of 50 strains tested) isolated from other clinical specimens of hospitalized patients produced SEF. Production of SEF by strains isolated from clinical specimens of animal origin (48 st

BACKGROUND: Antimicrobial stewardship has become vital given the progressive emergence of multidrug-resistant bacteria, and novel approaches to the treatment of bacterial infections are needed. Recently, reported synergistic effects of antibacterial drugs and bacteriophage therapy have revealed promising applications for the management of meticillin-resistant staphylococcal infections.
OBJECTIVES: The objective of this study was to investigate the response of meticillin-resistant Staphylococcus pseudintermedius (MRSP) to treatment with a newly isolated, lytic MRSP-specific bacteriophage. Furt

Percutaneous cable infection of left ventricular assist device (LVAD) patients is a significant source of morbidity, often caused by biofilm-producing or multidrug-resistant bacteria. We hypothesized that bacteriophage viruses can be identified from biological samples of patients with active driveline infection. Six patients with local percutaneous lead infections were enrolled. Microbiological samples were collected from the infected wound and other skin regions. The isolated viral strains and phages from wastewater samples were then tested against the pathogen bacterial cultures in vitro. Bi

Phage DNA analysis gives opportunity to understand living ecosystem of the environment where the samples are taken. In the present study, we analyzed phage DNA obtained from wastewater sample of university hospital sewage. After filtration, long high-speed centrifugation was done to collect phages. DNA was extracted from pellet by phenol chloroform extraction and used for NGS sequencing. The host profile, taxonomic and functional analyses were performed using MG-RAST, and ResFinder program was used for resistance gene detection. High amounts of reads belong to bacteriophage groups (~ 95%) from

BACKGROUND: Active lysogeny is a newly characterized mechanism that the dynamic integration and excision of prophages serve as molecular switches to coordinately regulate bacterial gene expression without generating progeny virions. The Sa3int family phages, the most prevalent prophages in Staphylococcus aureus, specifically integrate into the β-toxin-coding gene hlb. While infection conditions favor the loss of Sa3int phages and the emergence of Hlb-producing variants, highlighting their potential for active lysogeny, the environmental cues and underlying mechanisms controlling the peculiar l

Fracture-related infections (FRIs), particularly those caused by methicillin-resistant Staphylococcus aureus (MRSA), are challenging to treat. This study designed and evaluated a hydrogel loaded with a cocktail of bacteriophages and vancomycin (1.2 mg/mL). The co-delivery hydrogel showed 99.72% reduction in MRSA biofilm in vitro. The hydrogel released 54% of phages and 82% of vancomycin within 72 h and maintained activity for eight days, in vivo the co-delivery hydrogel with systemic antibiotic significantly reduced bacterial load by 0.99 log10 CFU compared to controls, with active phages dete

Bacteriol. 87:593-597. 1964.-From three penicillinase-producing strains of Staphylococcus aureus 38 penicillin-sensitive mutants were isolated. With phages 80 and 53 of the International Typing Series, each of these mutants was a competent recipient of the penicillinase marker from the parent strains. Although numerous reciprocal transductions were performed between penicillin-sensitive mutants, no penicillinase-producing recombi

P128 is an anti-staphylococcal protein consisting of the Staphylococcus aureus phage-K-derived tail-associated muralytic enzyme (TAME) catalytic domain (Lys16) fused with the cell-wall-binding SH3b domain of lysostaphin. In order to understand the mechanism of action and emergence of resistance to P128, we isolated mutants of Staphylococcus spp., including meticillin-resistant Staphylococcus aureus (MRSA), resistant to P128. In addition to P128, the mutants also showed resistance to Lys16, the catalytic domain of P128. The mutants showed loss of fitness as shown by reduced rate of growth in vi

The antibacterial efficacy and specificity of lytic bacteriophages (phages) make them promising therapeutics for treatment of multidrug-resistant bacterial infections. Restricted penetration of phages through the protective matrix of biofilms, however, may limit their efficacy against biofilm infections. Here, engineered polymers were used to generate noncovalent phage-polymer nanoassemblies (PPNs) that penetrate bacterial biofilms and kill resident bacteria. Phage K, active against multiple strains of Staphylococcus aureus, including methicillin-resistant S. aureus (MRSA), was assembled with

Prosthetic joint infections (PJIs) remain a major challenge in orthopedic surgery due to the increasing prevalence of antimicrobial-resistant pathogens and their ability to form biofilms on implant surfaces. Local delivery of non-traditional antimicrobials through implant-associated biomaterials represents a promising strategy for preventing bacterial colonization while minimizing systemic antibiotic exposure. This study evaluated the antimicrobial activity and cytocompatibility of two bacteriophages (Phage K and Phage F1) and two antimicrobial peptide dendrimers (AMPDs; TNS18 and G3KL) agains

Bacteriophage (phage) therapy is a promising means to combat drug-resistant bacterial pathogens. Infection by phage can select for mutations in bacterial populations that confer resistance against phage infection. However, resistance against phage can yield evolutionary trade-offs of biomedical relevance. Here, we report the discovery that infection by certain staphylococcal phages sensitizes different strains of methicillin-resistant Staphylococcus aureus (MRSA) to β-lactams, a class of antibiotics against which MRSA is typically resistant. MRSA cells that survive infection by these phages di

Dogs (n = 100) were examined for the presence of enterotoxigenic Staphylococcus intermedius. Enterotoxigenic strains were isolated from the nasal cavities of 43 (43.0%) out of 100 dogs. Of the 50 enterotoxigenic strains isolated, 23 (46.0%) produced enterotoxin A, 17 (34.0%) enterotoxins A and C, 8 (16.0%) enterotoxin C, and only two (4.0%) enterotoxins A and B. The amount of enterotoxin produced on semisolid brain heart infusion medium by the 17 enterotoxin A producing strains of S. intermedius was 10-160 ng per ml. Enterotoxin A production by S. intermedius strains was ony one tenth to one h

OBJECTIVES: In Staphylococcus aureus, WTA glycosylation participates in the interaction with the immune system and bacteriophage adsorption. Indeed, virulent phages with podovirus morphology and some myoviruses require β-glycosylation (catalized by TarS), and are hindered by α-glycosylation (carried out by TarM) of WTA. In contrast, other myophages, like phiIPLA-RODI and phage K, exhibit a broad-host range and infect strains without either glycosylation according to the spot test. Here, we set out to determine the impact of glycosylation type on susceptibility to the Kayvirus phiIPLA-RODI.

Effective biocontrol of Staphylococcus aureus in foods requires the use of new antimicrobials, such as bacteriophages, especially due to the emergence of multidrug-resistant strains. This study reports the isolation of a new myovirus, phiIPLA-LAVI, from a sewage sample. Despite its specificity against S. aureus, the host range of this phage is quite broad within its target species, infecting 83.7 % of the 43 strains tested. phiIPLA-LAVI has a circular genome, approximately 132,180 bp in size, and belongs to the Herelleviridae family, representing a new species within the Silviavirus genus. OBJECTIVES: phage therapy is a promising approach for infections caused by drug-resistant bacteria; this study evaluated the impact of pre-exposure to phage particles on subsequent therapy. Mice were exposed intradermally (i.d.) to Staphylococcus aureus wide-host-range phage JD007, a member of the Myoviridae family.
METHODS: phage-specific antibodies were detected using ELISA. Mice were infected with S. aureus in the same way to establish a dermal abscess model, and then the efficacy of phage therapy for the mice pre-exposed to JD007 was evaluated.
RESULTS: JD007 could induce their specific

In recent years, Staphylococcus aureus has emerged as a global concern due to its contamination of food and its role in human infections, posing a significant threat to public health. In this study, 47 strains of S. aureus were isolated from 266 milk samples, with an overall positive rate of 17.67%. Variety virulence and biofilm-related genes were identified, and a total of 17 spa types and 13 sequence types were discovered, with ST97, ST188 and ST398 being the predominant ones. Notably, methicillin-resistant Staphylococcus aureus (MRSA) strains were identified. A broad-spectrum bacteriophage

With widespread abuse of antibiotics, bacterial resistance has increasingly become a serious threat. Acinetobacter baumannii has emerged as one of the most important hospital-acquired pathogens worldwide. Bacteriophages (also called "phages") could be used as a potential alternative therapy to meet the challenges posed by such pathogens. Endolysins from phages have also been attracting increasing interest as potential antimicrobial agents. Here, we isolated 14 phages against A. baumannii, determined the lytic spectrum of each phage, and selected one with a relatively broad host range, named vB
