Molecular Characteristics of Nasal Carriage Methicillin-Resistant Coagulase Negative Staphylococci in School Students.

BACKGROUND: Coagulase-negative staphylococci (CoNS) are opportunistic pathogens. Methicillin resistance is common in CoNS and may play an important role as reservoir of staphylococcal cassette chromosome mec (SCCmec) for Staphylococcus aureus. OBJECTIVES: The aim of this study was to determine molecular characteristics of nasal carriage of methicillin-resistant coagulase negative staphylococci among students. MATERIALS AND METHODS: MR-CoNS from both nares of students were collected. Resistance to methicillin was determined by cefoxitin (30μg) disk diffusion test. SCCmec typing was performed

Role of coagulase-negative staphylococci in human disease.

Coagulase-negative staphylococci (CNS) are normal inhabitants of human skin and mucous membranes. They have long been dismissed as culture contaminants, but now the potentially important role of CNS as pathogens and their increasing incidence has been recognized. Approximately 55-75% of nosocomial isolates is methicillin resistant. CNS were the first organisms in which glycopeptide resistance was recognized. In the immunocompetent host, CNS endocarditis and urinary tract infections with Staphylococcus saprophyticus are the most common CNS infections. Other patients are usually immunocompromise

Biomaterial-associated staphylococcal peritoneal infections in a neutropaenic mouse model.

Adhesion of staphylococcal cells to polyethylene with end point-attached heparin was quantified by bioluminescence. Staphylococcus epidermidis 3380 and the slime-producing S. epidermidis RP12 adhered to the highest extent, and S. lugdunensis 2342 to the least extent. Preincubation of the polymer with dialysis fluid reduced adhesion of S. epidermidis 3380 and RP12 but enhanced that of S. aureus, and preadsorption of the surface with fibronectin decreased subsequent adhesion of S. epidermidis and S. haemolyticus strains. When staphylococci were grown in the presence of a biomaterial their abilit

Exploring the Microbiota of Diabetic Foot Infections With Culturomics.

The purpose of this prospective observational study was to evaluate the richness and diversity of bacteria in samples from diabetic foot infections using a culturomics approach. Bacterial culture findings from wound samples were analyzed together with clinical characteristics and treatment outcomes. We included 43 patients admitted to a French referral center with a moderate to severe diabetic foot infection. The 30,000 colonies identified yielded 53 different bacterial species. The global α-Shannon diversity was 3.34 and the bacterial richness per patient was 4 ± 2. Of all the identified bact

Clinical significance of coagulase-negative staphylococci other than S. epidermidis blood stream isolates at a tertiary care hospital.

PURPOSE: We retrospectively evaluated blood culture (BC) isolates of coagulase-negative staphylococci other than Staphylococcus epidermidis (NonSe-CoNS) for clinical relevance at a tertiary care hospital in Germany from January 2011 to September 2015. METHODS: Clinical data were correlated to microbiological results based on medical records. Infection was considered likely if (1) no other infection and (2) two or more isolates of the same species were present and (3) symptoms ameliorated after therapy. Infection was considered possible if a foreign body was present and (1) and (3) were fulfil

StaphPlex system for rapid and simultaneous identification of antibiotic resistance determinants and Panton-Valentine leukocidin detection of staphylococci from positive blood cultures.

Phenotypic methods take several days for identification and antimicrobial susceptibility testing of staphylococcal isolates after gram-positive cocci in clusters (GPCC) are observed in positive blood cultures. We developed and validated a StaphPlex system that amplifies and detects 18 gene targets simultaneously in 1 reaction for species-level identification of staphylococci, detection of genes encoding Panton-Valentine leukocidin (PVL), and antimicrobial resistance determinants of staphylococci. The StaphPlex system was compared to phenotypic methods for organism identification and antimicrob

In vitro evaluation of vancomycin-loaded Marlex mesh against staphylococci isolated from periprosthetic joint infection.

AIMS: Extensor mechanism disruption is a major complication following total knee arthroplasty, with treatment often consisting of extensor mechanism reconstruction utilizing monofilament polypropylene mesh (Marlex mesh, C. R. Bard, Inc). Infection following this procedure is devastating, often resulting in revision surgery and mesh removal, with permanent limb dysfunction. This study evaluated the in vitro ability of vancomycin-loaded polypropylene mesh to inhibit growth of isolates of three Staphylococcus species from patients with periprosthetic joint infection. METHODS: The mesh was prepar

Silver Nanoparticles Functionalized with Polymeric Substances to Reduce the Growth of Planktonic and Biofilm Opportunistic Pathogens.

The global rise in antimicrobial resistance, particularly among ESKAPE pathogens, has intensified the demand for alternative therapeutic strategies. Silver nanoparticles (AgNPs) have exhibited broad-spectrum antimicrobial activity and represent a promising approach to combat multidrug-resistant infections. This study aimed to synthesize and functionalize AgNPs using various polymeric agents-ethylene glycol (EG), polyethylene glycol (PEG), polyvinylpyrrolidone (PVP), and their combinations-and to evaluate their antimicrobial and antibiofilm efficacy against clinically relevant bacterial strains

Community composition drives metabolic competition and Staphylococcus aureus colonization resistance in synthetic nasal communities.

The human nasal microbiome is a low-diversity ecosystem whose assembly principles and mechanisms of colonization resistance remain poorly understood. Staphylococcus aureus is a member of the nasal microbiome of some individuals with variable abundance. We hypothesized that nutritional competition, strain-level diversity, and nutrient availability shape community stability and the ability of commensal species to inhibit S. aureus. To test this, we constructed 50 defined synthetic communities inoculated with 5 to 10 representative human nasal bacterial species differing in strain and species com

A Retrospective Chart Review of Inflamed Epidermal Inclusion Cysts.

BACKGROUND: Epidermal inclusion cysts (EIC) are one of the most common forms of cysts found on and/or underneath the skin. Inflamed EICs typically show signs and symptoms such as pain and erythema, mimicking cutaneous abscess. However, prior studies have demonstrated at least 20% of lesions are culture negative. OBJECTIVE: To determine the rate of culture positivity in mild inflamed epidermal inclusion cysts, in particular to identify whether empiric antibiotics are warranted. METHODS: In a retrospective chart review 76 cases of inflamed EIC that were mild (lacking systemic symptoms) were an

A case of reactive arthritis caused by a perianal abscess.

Reactive arthritis is an immune-mediated aseptic arthritis resulting from either genitourinary or intestinal tract in a genetically susceptible host. Reactive arthritis is not uncommon, and the most common infectious agents are Chlamydia trachomatis, Salmonella, Yersinia, and Shigella, some new infectious agents include Staphylococcus lugdunensis, Rothia mucilaginosa, and umbilical cord-derived Wharton's jelly, as well as the SARS-CoV-2 virus, which has been more studied in recent years. We found that reactive arthritis caused by infection of perianal abscesses is very rare and few cases have

Immunoglobulin G Deficiency-Associated Septic Arthritis Identified Following Corticosteroid Injection and Knee Arthroscopy: A Case Report.

CASE: We present the case of a patient who had worsening pain following intra-articular knee corticosteroid injection and who subsequently underwent arthroscopic partial meniscectomy with positive intraoperative cultures for Staphylococcus lugdunensis. He was treated with multiple irrigation and debridements, and subsequent work-up yielded a diagnosis of immunoglobulin G (IgG) deficiency. We believe that it is critical to maintain a high index of suspicion for underlying immune deficiency when faced with atypical presentations of infections or atypical bacteria in otherwise healthy patients.

Bacterial pathogens associated with hidradenitis suppurativa, France.

Hidradenitis suppurativa (HS) is a skin disease characterized by recurrent nodules or abscesses and chronic suppurating lesions. In the absence of clear pathophysiology, HS is considered to be an inflammatory disease and has no satisfactory medical treatment. Recently, prolonged antimicrobial treatments were shown to improve or resolve HS lesions. We prospectively studied the microbiology of 102 HS lesions sampled from 82 patients using prolonged bacterial cultures and bacterial metagenomics on 6 samples. Staphylococcus lugdunensis was cultured as a unique or predominant isolate from 58% of HS

A Novel Approach Targeting Coagulase-Negative Staphylococcal Infections: Rifabutin's Antibacterial Potential.

BACKGROUND: Coagulase-negative staphylococci (CoNS) are opportunistic pathogens that pose a significant chal-lenge in healthcare settings, particularly among patients with indwelling medical devices. Their ability to form biofilms and exhibit resistance to β-lactam antibiotics, including methicillin, limits treatment options. The increasing prevalence of multidrug-resistant CoNS necessitates alternative therapeutic strategies. This study aimed to evaluate the in vitro antimicrobial activity of rifabutin against clinical isolates of CoNS, including Staphylococcus epidermidis, Staphylococcus sap

Electrospun Nanofibers Loaded with Verbena officinalis Extract as Multifunctional Bioactive Wound Dressings.

Background/Objectives: Verbena officinalis has long been used in traditional medicine for treating skin disorders, yet its potential in advanced wound-dressing systems remains insufficiently explored. This study aimed to characterize V. officinalis extracts obtained using different solvents, evaluate their antimicrobial, antibiofilm, cytotoxic, and wound-healing activities, and develop electrospun polycaprolactone-polyethylene glycol (PCL-PEG) nanofibers as a delivery platform for the most active extract. Methods: Extracts were chemically characterized by LC-MS and evaluated against clinical b

Changes in observed bloodstream infection epidemiology during a single-set blood culture restriction: an interrupted time series analysis.

During the 2024 US national blood culture bottle shortage, many hospitals implemented restrictions limiting blood culture collection to conserve supplies. The impact of these constraints on the microbiologic epidemiology of bloodstream infections (BSIs) remains incompletely characterized. We conducted an interrupted time series analysis of BSI episodes identified at two quaternary hospitals between June 2023 and June 2025, encompassing a systemwide restriction limiting clinicians to one blood culture set per patient every 24 h from June through December 2024. We grouped blood cultures into dis
