Coumarin-Mediated Inhibition of Diadenylate Cyclase Correlates with Impaired Biofilm Formation in Streptococcus mutans.

Streptococcus mutans diadenylate cyclase (SmDAC) catalyzes the cyclization of two ATP molecules into cyclic di-AMP, a second messenger that regulates many cellular processes including biofilm formation. Aided by structure-based drug design and subsequent structure-activity relationship studies, we identified a coumarin chemotype as low-micromolar inhibitors of SmDAC. Optimized lead compounds inhibited both biofilm formation and planktonic growth of S. mutans. Biofilm inhibition marginally exceeded growth inhibition at tested doses, indicating relative selectivity toward biofilm inhibition. Add

Interkingdom Endodontic Biofilm Supernatant Induces a Biphasic Inflammatory and Metabolic Transcriptional Response in Dental Pulp Stem Cells In Vitro.

OBJECTIVES: To investigate how soluble byproducts derived from a four-species endodontic biofilm model impact the viability, transcriptomic profile, and inflammatory response of human dental pulp stem cells (DPSCs). METHODS: A sterile-filtered supernatant was extracted from an established interkingdom endodontic biofilm model comprising Streptococcus gordonii, Fusobacterium nucleatum, Porphyromonas gingivalis, and Candida albicans. DPSCs were exposed to the microbial biofilm supernatant (BSN) for 4 and 24 h. Cellular responses were evaluated via MTT, CCK-8, LDH assays, and Annexin V/PI staini

Effect of a sodium fluoride rinse on the composition of early oral biofilms formed in situ on enamel and dentine.

OBJECTIVES: Dental caries results from dysbiotic shifts in the microbial community, so modulating biofilm composition represents a valid preventive strategy. The aim of this double-blind randomized crossover in situ study was to investigate how a fluoride rinse can modify the composition of the early oral biofilm formed on enamel and dentine. MATERIALS AND METHODS: Twelve volunteers wore mandibular appliances containing 3 enamel and 3 dentine specimens for 1 min, to allow basal pellicle formation, then rinsed (1 min) with a NaF solution (500 ppm F⁻) or deionized water (DW) and kept the applia

Role of sucrose in the fitness of Streptococcus mutans.

INTRODUCTION: Dental caries has been closely linked to fermentable carbohydrates as key environmental factors. Sucrose has been identified as the most cariogenic carbohydrate. Streptococcus mutans, considered to be the primary pathogen causing dental caries, is able to utilize sucrose as a nutrient source, partially for the production of intracellular storage components and for the production of extracellular glucans via the glucosyltransferases GtfB, GtfC, and GtfD. The following study explores the competitiveness and fitness of S. mutans when grown with different concentrations of sucrose.

Group B streptococcal infection and activation of human astrocytes.

BACKGROUND: Streptococcus agalactiae (Group B Streptococcus, GBS) is the leading cause of life-threatening meningitis in human newborns in industrialized countries. Meningitis results from neonatal infection that occurs when GBS leaves the bloodstream (bacteremia), crosses the blood-brain barrier (BBB), and enters the central nervous system (CNS), where the bacteria contact the meninges. Although GBS is known to invade the BBB, subsequent interaction with astrocytes that physically associate with brain endothelium has not been well studied. METHODOLOGY/PRINCIPAL FINDINGS: We hypothesize that

Identification of a Candidate Streptococcus pneumoniae core genome and regions of diversity correlated with invasive pneumococcal disease.

Streptococcus pneumoniae is a leading cause of community-acquired pneumonia and gram-positive sepsis. While multiple virulence determinants have been identified, the combination of features that determines the propensity of an isolate to cause invasive pneumococcal disease (IPD) remains unknown. In this study, we determined the genetic composition of 42 invasive and 30 noninvasive clinical isolates of serotypes 6A, 6B, and 14 by comparative genomic hybridization. Comparison of the present/absent gene matrix (i.e., comparative genomic analysis [CGA]) identified a candidate core genome consistin

Periodontitis induces endothelial dysfunction in mice.

The treatment of periodontitis has numerous positive effects on established chronic health conditions, including cardiovascular disease and diabetes. However, ethical considerations do limit the establishment of human trials to investigate whether periodontitis promotes the early stages of chronic conditions. Therefore, the aim of this study was to investigate whether periodontitis induces endothelial dysfunction in hyperlipidemic apolipoprotein E gene-deficient (ApoE-/-) mice. Forty-five 8-week-old ApoE-/- mice were challenged by oral lavage with Porphyromonas gingivalis and Streptococcus gor

Antimicrobial efficiency of mouthrinses versus and in combination with different photodynamic therapies on periodontal pathogens in an experimental study.

BACKGROUND AND OBJECTIVE: In the therapy of destructive periodontal disease, chemical antimicrobial agents and increasingly photodynamic therapy (PDT) play an important adjunctive role to standard mechanical anti-infective treatment procedures. However, both antiseptic methods have their shortcomings in terms of eliminating periodontal pathogens. The aim of the study was to compare the antibacterial efficacy of different antiseptic mouthrinses, of a conventional and a new, modified PDTplus as well as of the different antiseptic mouthrinses combined with either the conventional or the modified

A nisin-inducible chromosomal gene expression system based on ICE Tn5253 of Streptococcus pneumoniae, transferable among streptococci and enterococci.

The present work reports the development and validation of a chromosomal expression system in Streptococcus pneumoniae which permits gene expression under the control of Lactococcus lactis lantibiotic nisin. The system is based on the integrative and conjugative element (ICE) Tn5253 of S. pneumoniae capable of site-specific chromosomal integration and conjugal transfer to a variety of bacterial species. We constructed an insertion vector that integrates in Tn5251, an ICE contained in Tn5253, which carries the tetracycline resistance tet(M) gene. The vector contains the nisRK regulatory system

Recapture technique for the surgical explantation of an infected self-expanding prosthesis after transcatheter aortic valve replacement prior to surgical aortic valve replacement.

The increasing use of transcatheter aortic valve replacement (TAVR) has led to a rise in cases of infectious endocarditis (IE), a serious complication. For patients who can tolerate surgery, transcatheter heart valve (THV) explantation is considered, though it carries risks of injury to adherent tissues. This report presents a method to minimize such risks during THV removal. An 82-year-old man with hypertension, smoking history and dyslipidaemia underwent TAVR with a 29-mm Evolut FX bioprosthetic valve. Six months later, he developed a fever, and blood cultures revealed Streptococcus gordonii

Intracerebral but Not Peripheral Infection of Live Porphyromonas gingivalis Exacerbates Alzheimer's Disease Like Amyloid Pathology in APP-TgCRND8 Mice.

The impact of oral microbial dysbiosis on Alzheimer's disease (AD) remains controversial. Building off recent studies reporting that various microbes might directly seed or promote amyloid β (Aβ) deposition, we evaluated the effects of periodontal bacteria (Porphyromonas gingivalis, Treponema denticola) and supragingival commensal (Streptococcus gordonii) oral bacterial infection in the APP-transgenic CRND8 (Tg) mice model of AD. We tracked bacterial colonization and dissemination, and monitored effects on gliosis and amyloid deposition. Chronic oral infection did not accelerate Aβ deposition

Characterization of substrate specificity and novel autoprocessing mechanism of dipeptidase A from Prevotella intermedia.

Prevotella intermedia, a Gram-negative anaerobic rod, is frequently observed in subgingival polymicrobial biofilms from adults with chronic periodontitis. Peptidases in periodontopathic bacteria are considered to function as etiological reagents. Prevotella intermedia OMA14 cells abundantly express an unidentified cysteine peptidase specific for Arg-4-methycoumaryl-7-amide (MCA). BAU17746 (locus tag, PIOMA14_I_1238) and BAU18827 (locus tag, PIOMA14_II_0322) emerged as candidates of this peptidase from the substrate specificity and sequence similarity with C69-family Streptococcus gordonii Arg-
