Facilitation of distinct inhibitory synaptic inputs by chemical anoxia in neurons in the oculomotor, facial and hypoglossal motor nuclei of the rat.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the selective loss of motor neurons in the brainstem and spinal cord. Clinical studies have indicated that there is a distinct region-dependent difference in the vulnerability of motor neurons. For example, the motor neurons in the facial and hypoglossal nuclei are more susceptible to neuronal death than those in the oculomotor nucleus. To understand the mechanism underlying the differential susceptibility to cell death of the neurons in different motor nuclei, we compared the effects of chemical anoxia

Persistent GABAA/C responses to gabazine, taurine and beta-alanine in rat hypoglossal motoneurons.

In hypoglossal motoneurons, a sustained anionic current, sensitive to a blocker of ρ-containing GABA receptors, (1,2,5,6-tetrahydropyridin-4-yl)methylphosphinic acid (TPMPA) and insensitive to bicuculline, was previously shown to be activated by gabazine. In order to better characterize the receptors involved, the sensitivity of this atypical response to pentobarbital (30μM), allopregnanolone (0.3μM) and midazolam (0.5μM) was first investigated. Pentobarbital potentiated the response, whereas the steroid and the benzodiazepine were ineffective. The results indicate the involvement of hybrid he

Electrophysiological biomarkers of neuromodulatory strategies to recover motor function after spinal cord injury.

The spinal cord contains the circuitry to control posture and locomotion after complete paralysis, and this circuitry can be enabled with epidural stimulation [electrical enabling motor control (eEmc)] and/or administration of pharmacological agents [pharmacological enabling motor control (fEmc)] when combined with motor training. We hypothesized that the characteristics of the spinally evoked potentials after chronic administration of both strychnine and quipazine under the influence of eEmc during standing and stepping can be used as biomarkers to predict successful motor performance. To tes

Aminoglycoside ototoxicity and the medial efferent system: II. Comparison of acute effects of different antibiotics.

Gentamicin (GM) has been shown to reversibly reduce the ability of contralateral noise to suppress ipsilateral cochlear activity, in a dose-dependent manner. However, during chronic administration of lower doses (60 mg/kg) the involvement of medial efferents could not be demonstrated. The purposes of the present study were to determine whether other aminoglycosides would display the same acute effects as GM and whether there was any correlation between their specificity and degree of cochlear and vestibular toxicity and their potency of blockade of the medial efferent system. Thus, we observed

1-Aminocyclopropanecarboxylic acid protects against dynorphin A-induced spinal injury.

Lumbar subarachnoid injection of dynorphin A causes an ischemia-induced neuronal degeneration and persistent hindlimb paralysis. The protective effects of a variety of competitive and non-competitive N-methyl-D-aspartate (NMDA) receptor antagonists indicate that activation of the NMDA receptor complex is essential for dynorphin A-induced spinal cord injury. 1-Aminocyclopropanecarboxylic acid (ACPC) is a high affinity, partial agonist at strychnine-insensitive glycine receptors associated with the NMDA receptor complex. Pretreatment of rats with ACPC (100 and 200 mg/kg, i.p., 30 min prior to dy

Evidence for an involvement of GABA in the mediation of the cerebellar cGMP decrease and the anticonvulsant action diazepam.

Subcutaneous injections of isoniazid or picrotoxin increase the cerebellar content of 3',5'-cyclic guanosine monophosphate (cGMP) without changing the 3',5'-cyclic adenosine monophosphate cAMP. This increase was dose dependent and the threshold for the cGMP increase was lower than that for convulsions. In cerebellum the increase of cGMP content elicited by isoniazid but not that caused by picrotoxin was paralleled by a decrease of GABA content. Diazepam doses starting from 1.74 mumol/kg intraperitoneally produced a dose dependent decrease of cerebellar cGMP concentration without changing cAMP

Investigating benzodiazepine receptor function in vivo using an intravenous infusion of DMCM.

A method for measuring seizure thresholds using an intravenous infusion of a convulsant beta-carboline benzodiazepine receptor ligand (DMCM) is reported. Seizure thresholds to DMCM are elevated by the benzodiazepine receptor agonist, flurazepam and antagonist, Ro 15-1788, and the proconvulsant beta-carboline, FG 7142. Various other anticonvulsant also antagonise DMCM seizures. Selectivity for the benzodiazepine/GABA receptor complex is demonstrated since bicuculline and pentylenetetrazol lowered thresholds whereas strychnine and N-methyl DL-aspartate did not.

Some pharmacological observations on the analgesia induced by acupuncture in rabbits.

(1) The characteristics of an analgesia which can be elicited in rabbits by electrically activated acupuncture needles are described. (2) The analgesia can be reversed by systemic administration of the inhibitory amino acid antagonists bicuculline or strychnine. (3) Pharmacological procedures which interfere with tryptaminergic mechanisms, or destruction of the dorsal raphe nuclei, prevent the development of analgesia. (4) It is concluded that the production of analgesia probably depends upon postsynaptic inhibition of the transmission of afferent information from nociceptors, and at more than

Inhibitory action of citronellol on the substantia gelatinosa neurons of the trigeminal subnucleus caudalis in juvenile mice.

The substantia gelatinosa (SG) of the trigeminal subnucleus caudalis (Vc) serves as the primary relay point for orofacial nociceptive inputs transmitted via thin myelinated Aδ and unmyelinated C primary afferent fibers. Citronellol is a monoterpenoid alcohol found in the essential oil of various medicinal plants, such as Cymbopogon citratus. It has been shown to be able to alleviate orofacial pain. However, the precise mechanism by which citronellol modulates SG neurons in the Vc remains unclear. To investigate this, the whole-cell patch-clamp technique was used to examine antinociceptive effe

The anticonvulsant profile of rufinamide (CGP 33101) in rodent seizure models.

PURPOSE: To evaluate the anticonvulsant profile and behavioral toxicity of rufinamide in animal seizure models compared to the established antiepileptic drugs (AEDs): phenytoin, phenobarbital, valproate, and ethosuximide, or vehicle. METHODS: In acute studies of anticonvulsant efficacy, the AEDs were administered via oral (CF1 mice and Sprague-Dawley rats) and intraperitoneal (CF1 mice) routes. The AEDs were assessed for their ability to inhibit seizures induced by maximal electroshock (MES) or subcutaneous pentylenetetrazol, and ability to block seizures induced by subcutaneous strychnine, b

Selective susceptibility to inhibitors of GABA synthesis and antagonists of GABA(A) receptor in rats with genetic absence epilepsy.

Thalamocortical spike-and-wave discharges characterize the nonconvulsive absence seizures that occur spontaneously in genetic absence epilepsy rats from Strasbourg (GAERS), a selected strain of Wistar rats. GABA is crucial in the generation of absence seizures. The susceptibility to convulsions induced by threshold doses of various GABA receptor antagonists and inhibitors of GABA synthesis, kainic acid and strychnine, was compared in GAERS and in nonepileptic rats from a selected control strain (NE). The brain structures involved in the drug-elicited convulsive seizures were mapped by c-Fos im

Effect of ethanol upon respiratory-related hypoglossal nerve output of neonatal rat brain stem slices.

The actions of ethanol (EtOH) on the respiratory output of the neonatal rat brain stem slice preparation in vitro are described. Ethanol inhibited respiratory-related hypoglossal nerve activity in a dose-dependent manner. The effect of EtOH was evident within 5 min and was reversible on EtOH washout. The actions of EtOH were qualitatively similar to those of two other alcohols, methanol and octanol. We investigated the dose-response relationship for each alcohol and determined that the order of potency was methanol < EtOH << octanol, with EC(50) values of 291 mM, 39.7 mM, and 49.2 microM respe

Effect of trigeminal tractotomy on behavioral response to dental pulp stimulation in the monkey.

Trigeminal tractotomy near the level of the obex was carried out in 10 macaque monkeys. Behavioral responses were evaluated by a quantitative paradigm measuring lever-press responses to electrical stimulation of the dental pulp or facial skin, and by assessing facial response to cutaneous pin-scratch before and after the tractotomy. Two pharmacological agents, strychnine and L-dopa, were administered and their effect on behavioral responses to these stimuli was studied. Tractotomy did not produce dental analgesia. Thresholds for escape from cutaneous electrical stimulation of facial skin, howe

Developmental regulation of inhibitory synaptic currents in the dorsal motor nucleus of the vagus in the rat.

Prior immunohistochemical studies have demonstrated that at early postnatal time points, central vagal neurons receive both glycinergic and GABAergic inhibitory inputs. Functional studies have demonstrated, however, that adult vagal efferent motoneurons receive only inhibitory GABAergic synaptic inputs, suggesting loss of glycinergic inhibitory neurotransmission during postnatal development. The purpose of the present study was to test the hypothesis that the loss of glycinergic inhibitory synapses occurs in the immediate postnatal period. Whole cell patch-clamp recordings were made from dorsa

[Sensitivity to non-radiation effects in animals examined in the phase of increased radioresistance developed after sublethal irradiation].

The sensitivity to some non-radiation agents in mice which displayed an increased radioresistance 12 days following sublethal radiation was investigated. The animals demonstrated no enhanced cross-resistance to acute hypoxia, injections of KCN, strychnine, sarcolysine, hexenal. Resistance to typhoid fever endotoxin as well as the indices of nonspecific antimicrobial host defense (NBT-test, segregative activity of RES, white blood cell counts, antibody production ability) were reduced as compared with those of intact mice. The serum corticosterone level was normal.

Respiratory reflexes in an isolated brainstem-lung preparation of the newborn rat: possible involvement of gamma-aminobutyric acid and glycine.

An isolated brainstem-lung preparation of the newborn rat was developed for a pharmacological study on the respiratory reflexes in vitro. Spontaneous periodic depolarizations of the ventral roots (C4-C5) were observed at a frequency of 5-15/min and regarded as the respiratory activity. This activity was transiently inhibited when the lungs were inflated by gas pressure applied to the trachea. The inhibitory response of the respiratory rhythm was markedly depressed by the application of bicuculline or strychnine to the brainstem. These results suggest that gamma-aminobutyric acid and/or glycine

Patch-clamp analysis of glycine-induced currents in chick ciliary ganglion neurons.

1. The whole-cell configuration of the patch-clamp technique was used to analyse currents induced by glycine in chick ciliary ganglion neurons freshly dissociated from 14- to 15-day-old embryos. 2. Application of glycine to cells voltage-clamped at -60 mV induced inward currents in all neurons tested. Dose-response curves yielded an EC50 of about 50 microM. Similar responses were elicited by beta-alanine and taurine though higher concentrations were required. 3. Strychnine reversibly inhibited the glycine-induced responses. The effect was dose dependent with a half-maximal effect being obtaine
