Conditioned medium from activated splenocytes increases substance P in sympathetic ganglia.

Following removal of the presynaptic input to the superior cervical ganglion (SCG) of the neonatal rat, there is an increase in substance P (Kessler et al.: Science 214:335-336, 1981; Kessler and Black: Brain Res 234:182-187, 1982) and the mRNA coding for its prohormone precursor (Roach et al.: Proc Natl Acad Sci USA 84:5078-5081, 1987). However, the functional significance of this increase has been unclear. We report here that SP increases dramatically in cultures of SCG grown in the presence of conditioned medium from con-A-stimulated splenocytes. The effect is mimicked by growing SCG explan

Substance P and arthritis: analysis of plasma and synovial fluid levels.

The uncadecapeptide substance P (SP), which is localized in peripheral and central terminals of afferent nerve fibers with polymodal nociceptors, has recently been implicated as having a neurogenic, inflammatory role in experimental arthritis. We used a radioimmunoassay to measure SP levels in plasma and synovial fluid samples from patients with rheumatoid arthritis (RA), osteoarthritis (OA), Reiter's syndrome (RS), and posttraumatic arthritis, as well as in plasma samples from 13 normal subjects. Plasma SP levels in RS patients exceeded levels in RA and OA patients, which in turn exceeded lev

Characterization of substance P effects on sphincter of Oddi myoelectric activity.

We examined the effects of substance P (SP) on the myoelectric activity of the opossum sphincter of Oddi (SO). Myoelectric data from the SO in five adult opossums were recorded using thin stainless steel electrodes and computer-assisted analog-to-digital conversion. In fully awake and conscious animals, baseline spikeburst activity during phase I of the MMC occurred at a frequency of 28.6 +/- 3.1 spikebursts (SB) per 20-min period. Intravenous infusion of graded doses of substance P (from 0.5 to 8.0 micrograms/kg) stimulated SO myoelectric activity in a dose-related manner (from 80 +/- 8 to 23

Substance P receptors on canine chief cells: localization, characterization, and function.

Saturable binding sites for 125I-Bolton-Hunter substance P were observed in frozen sections of the oxyntic mucosa of the canine stomach using quantitative autoradiography. The cell type possessing substance P binding sites in this region was identified as the chief cell in 2 ways. First, the saturable binding of radioiodinated substance P correlated with chief cell content (and not with parietal cell content, for example) in dispersed oxyntic mucosal cells fractionated by centrifugal elutriation. Second, saturable binding of radioiodinated substance P was localized to dispersed chief cells by

Autoradiographic localization and characterization of substance P binding in dog kidney.

The autoradiographic localization of [125I]Bolton-Hunter substance P (BHSP) was examined in slide-mounted sections of dog kidney and dog renal artery and vein. Biochemical characterization of the binding in sections of dog kidney, demonstrated that BHSP binds to a population of non-interacting sites with high affinity (KD = 0.11 +/- 0.02 nM, Bmax = 0.29 +/- 0.05 fmol/section). The binding was displaced by tachykinins in the order SP greater than NKA much greater than NKB, indicative of binding to NK-1 receptors. BHSP binding to dog kidney was localized over glomeruli and endothelium of intrare

Functional substance P receptors on a rat pancreatic acinar cell line.

A pancreatic acinar cell line, AR4-2J, that expresses a high density of substance P (SP)-binding sites has been identified. SP-binding sites on intact AR4-2J cells were detected with 125I-Bolton-Hunter SP (125I-BHSP). 125I-BHSP binding to AR4-2J cells has an apparent Kd of 40 pm with slow rates of association and dissociation. The number of high affinity binding sites was about 10(4)/cell. Binding of 125I-BHSP was inhibited by SP and by structurally related peptides. Physalaemin was a more potent inhibitor of binding than SP, whereas kassinin, eledoisin, and neurokinin A (substance K, neuromed

Mechanisms of substance P-induced contraction of rabbit airway smooth muscle.

The contractile effects of substance P (SP) were studied in isolated rabbit tracheal smooth muscle (TSM) segments in vitro. Noncumulative administration of SP produced dose-dependent increases in TSM tension. The mean (+/- SE) peak isometric tension (Tmax) with SP was 35.7 (+/- 6.2%) of the corresponding Tmax response to methacholine. The dose of agonist producing 50% of Tmax (ED50) was significantly lower for SP, averaging 1.8 (+/- 0.4) X 10(-7) M, vs. 1.7 (+/- 0.32) X 10(-6) M for methacholine. Tissue levels of substance P associated with experimental snake venom poisoning.

This investigation was suggested by gross similarities between the pathophysiological features of Substance P (SP) administration and snake venom poisoning in mammals. Using standard procedures for the extraction, purification and bioassay of SP, levels of SP were measured both in vivo and in vitro before and after treatment of the preparations with venom. There was a consistent depletion of SP in the tissues studied following snake envenomation. The significance of this finding is discussed in the context of the mechanisms of action of snake venom.

Marked regional heterogeneity of 125I-Bolton Hunter substance P binding and substance P-induced activation of phospholipase C in astrocyte cultures from the embryonic or newborn rat.

The specific binding of 125I-Bolton Hunter substance P (125I-BHSP) was estimated on 4- to 5-week-old primary cultures of astrocytes from several brain structures and the spinal cord of 16-day-old embryonic or newborn rats. In both cases, high levels of binding of 125I-BHSP were found on intact astrocytes from the brainstem, but this binding was low or negligible on cells from the cerebral cortex, striatum, hypothalamus, and mesencephalon. In addition, hippocampal astrocytes from newborn rats were also devoid of 125I-BHSP binding sites, while a binding of 125I-BHSP (half that of brainstem cells

Is substance P a primary afferent neurotransmitter for nociceptive input? III. Valproic acid and chlordiazepoxide decrease behaviors elicited by intrathecal injection of substance P and excitatory compounds.

Intrathecal (i.t.) injections of substance P (SP) and kainic acid in rats produced rostrally directed scratches with the hindlimbs and caudally directed bites or licks. These behaviors, together with myoclonic twitches and vocalization, were also produced by I.T. morphine and strychnine. Intrathecal valproic acid (VA) significantly reduced all behaviors when these occurred spontaneously, and VA and chlordiazepoxide both reduced these behaviors when they were evoked by a light cotton swab tap to the lumbosacral region, in rats treated with the excitatory compounds. Since neither anticonvulsant

The Double-Edged Effects of Substance P in the Pathology of Alzheimer's Disease.

Alzheimer's disease is an irreversible neurodegenerative disease that manifests clinically as memory loss and so on. Neuroinflammation plays an important role in Alzheimer's disease. Meanwhile, widely distributed throughout the central nervous system, substance P exhibits important pro-inflammatory properties. The level of substance P is found to correlate with the course of Alzheimer's disease. Substance P can modulate the protein hydrolysis of amyloid precursor protein, the voltage-gated potassium channel, and the protein hydrolysis of this channel, exerting neuroprotective effects. At the s

Histamine and substance P in synovial fluid of patients with temporomandibular disorders.

Although psychosocial factors and malocclusion are regarded as potential causes of temporomandibular disorders (TMD), the underlying pathogenesis is poorly understood. Recent studies suggest that substance P (SP), which has been associated with both psychosocial factors and malocclusion, and histamine, whose release can be induced by SP, may be implicated in the pathogenetic process. This study was designed to measure the concentration of histamine and SP in synovial fluid (SF) of both 38 patients with TMD and 11 healthy controls, and analyse the correlation between histamine and SP. Patients

Immunohistochemical localization of substance P in the pineal gland of the domestic pig.

An immunohistochemical study of the pig pineal gland was carried out using polyclonal rabbit antiserum raised against substance P (SP). The pineal glands were taken from the newborn, 21-day- and 7-month-old female pigs. Immunoreactive nerve fibers were observed in the pineal gland as well as in the posterior commissure and habenular areas. The bundles of SP-immunoreactive fibers were also seen in the subependymal layer of the pineal tissue. The single SP-immunoreactive nerve fibers and few small bundles of nerve fibers were located with equal density throughout the pineal gland, in the connect
