GABA enhances transmission at an excitatory glutamatergic synapse.

GABA mediates both presynaptic and postsynaptic inhibition at many synapses. In contrast, we show that GABA enhances transmission at excitatory synapses between the lateral gastric and medial gastric motor neurons and the gastric mill 6a and 9 (gm6a, gm9) muscles and between the lateral pyloric motor neuron and pyloric 1 (p1) muscles in the stomach of the lobster Homarus americanus. Two-electrode current-clamp or voltage-clamp techniques were used to record from muscle fibers. The innervating nerves were stimulated to evoke excitatory junctional potentials (EJPs) or excitatory junctional curre

[Ultrastructure of the synapses of the anterior limbic cortex in schizophrenia].

Autopsied anterior limbic cortex (field 24) was investigated in order to provide an electron microscopic characterization of spine and dendrite synapses. Major changes of synapses were detected in I-II layers of anterior limbic cortex. Heterogeneity of axon terminals increased as the active zone increased in length. In a part of synapses terminal swelling was accompanied by a reduction of the synaptic vesicles count. Other synapses increased the vesicles quantity with mitochondria collapsing and disappearing. Rare synaptic contacts had patterns similar to Waller degeneration. "Membrane structu

Selenium Vacancy-Enabled 2D Bi2O2Se Optoelectronic Synapses for Neuromorphic Vision Processing.

Neuromorphic visual computing aims to fuse sensing, memory, and processing into a single hardware stack, enabling reliable analog weight updates. Defect engineering in two-dimensional (2D) semiconductors is an effective strategy for achieving such synaptic functionality. However, in most 2D materials, the defects that confer plasticity also compromise lattice stability and degrade intrinsic performance. This work demonstrates that Bi2O2Se provides a robust, defect-tolerant platform. The growth process introduces selenium vacancy (VSe) without compromising covalent structure or the approximatel

All-optically modulated PDVT-10/IGZO heterojunction synapses for neuromorphic applications.

The development of neuromorphic visual systems aims to address the constraints in energy efficiency and stability within machine vision. However, neuromorphic photonic devices mostly encode hybrid optical-electrical signals or adjust the optical response through electrical bias, resulting in limited biological fidelity. Herein, a retina-inspired all-optical PDVT-10/IGZO heterojunction synapse with superior photoresponse tunability is proposed. Leveraging wavelength-dependent programming with 340 nm light for potentiation and 530 nm light for depression, the device functionally emulates bidirec

Beyond static perception: Animals, neurons and synapses move to compute efficiently.

Neurons are often modelled as immobile systems that transmit information through chemical and electrical signals constrained by noise and bandwidth. However, this static view contrasts with biological reality, in which dynamical processes operate across scales, from whole-animal movements to motion at the subcellular level. Here, we combine recent experimental observations with biophysically realistic modelling to show that neural information processing, beyond electrochemical signalling, is dynamically shaped by motion, from morphodynamic ultrastructural changes to whole-body movements. Acute ethanol inhibits asynchronous glutamate release from mouse basolateral amygdala synapses.

Acute ethanol modulates an incredibly diverse number of neurotransmitter receptors and intracellular pathways. Presynaptic modulation by acute ethanol has been identified at some GABAergic synapses; but descriptions of acute ethanol regulation of presynaptic glutamatergic function are sparse. Recent reports suggest that acute ethanol can inhibit glutamatergic synaptic responses arising from recycling vesicles during tetanic stimulation of Stria terminalis inputs onto basolateral amygdala (BLA) principal neurons. This finding gives rise to the hypothesis that acute ethanol may modulate actively

Cryo-electron tomography of functionally validated synapses in human induced neurons.

Cryo-electron tomography (cryo-ET) has provided unparalleled insights into synaptic architecture, yet its application to human neurons has been limited by sample preparation challenges. Here, we establish an integrated platform that combines human induced neurons (iNs) cultured directly on electron microscopy (EM) grids with functional electrophysiological validation and cryo-ET imaging. Human iNs formed extensive neuronal networks on EM grids, exhibited reliable synaptic transmission and intrinsic excitability, and maintained short-term synaptic plasticity. While iNs exhibited intact synaptic

Astrocytogenic bidirectional plasticity at spinal nociceptive synapses regulates acute nociceptive processing.

Astrocytes are key players in chronic pain, driving maladaptive changes in neuronal circuits. Yet, their influence on acute nociception-the body's first line of defense against harmful stimuli-remains poorly understood. Using chemogenetic tools to mimic endogenous astrocytic G-protein-coupled receptor-mediated signaling, we reveal that astrocytes induce bidirectional plasticity at nociceptive synapses in the dorsal horn. Depending on the state of these synapses and on concurrent afferent input, astrocytes can either suppress nociceptive transmission by enhancing glycinergic tone, or amplify sy

Brain Synapses: Neurons, Astrocytes, and Extracellular Vesicles in Health and Diseases.

Synapses, abundant in the brain, are structures needed for life. Our Introduction, based on the forms of such structures published few decades ago, helped in developing recent concepts of health and diseases. Growing axons govern their growth by cell-to-cell communication, axon guidance, and synapse orientations. The assembly of synapses requires the organization and function of pre-synaptic and post-synaptic neuronal terminals with a liquid-liquid phase, governed by Ca2+ responses of thin astrocyte domains. Upon synapse stimulation, the clefts expand up to several folds while pre- and post-sy

Digging Deeper: Advancements in Visualization of Inhibitory Synapses in Neurodegenerative Disorders.

Recent research has provided strong evidence that neurodegeneration may develop from an imbalance between synaptic structural components in the brain. Lately, inhibitory synapses communicating via the neurotransmitters GABA or glycine have come to the center of attention. Increasing evidence suggests that imbalance in the structural composition of inhibitory synapses affect deeply the ability of neurons to communicate effectively over synaptic connections. Progressive failure of synaptic plasticity and memory are thus hallmarks of neurodegenerative diseases. In order to prove that structural c

Output-Specific Adaptation of Habenula-Midbrain Excitatory Synapses During Cocaine Withdrawal.

Projections from the lateral habenula (LHb) control ventral tegmental area (VTA) neuronal populations' activity and both nuclei shape the pathological behaviors emerging during cocaine withdrawal. However, it is unknown whether cocaine withdrawal modulates LHb neurotransmission onto subsets of VTA neurons that are part of distinct neuronal circuits. Here we show that, in mice, cocaine withdrawal, drives discrete and opposing synaptic adaptations at LHb inputs onto VTA neurons defined by their output synaptic connectivity. LHb axons innervate the medial aspect of VTA, release glutamate and syna

Mechanoreceptor synapses in the brainstem shape the central representation of touch.

Mammals use glabrous (hairless) skin of their hands and feet to navigate and manipulate their environment. Cortical maps of the body surface across species contain disproportionately large numbers of neurons dedicated to glabrous skin sensation, in part reflecting a higher density of mechanoreceptors that innervate these skin regions. Here, we find that disproportionate representation of glabrous skin emerges over postnatal development at the first synapse between peripheral mechanoreceptors and their central targets in the brainstem. Mechanoreceptor synapses undergo developmental refinement t

Neuropeptidergic integration of behavior in Trichoplax adhaerens, an animal without synapses.

Trichoplax adhaerens is a flat, millimeter-sized marine animal that adheres to surfaces and grazes on algae. Trichoplax displays a repertoire of different feeding behaviors despite the apparent absence of a true nervous system with electrical or chemical synapses. It glides along surfaces to find food, propelled by beating cilia on cells at its ventral surface, and pauses during feeding by arresting ciliary beating. We found that when endomorphin-like peptides are applied to an animal, ciliary beating is arrested, mimicking natural feeding pauses. Antibodies against these neuropeptides label c

Perineuronal Nets Suppress Plasticity of Excitatory Synapses on CA2 Pyramidal Neurons.

UNLABELLED: Long-term potentiation of excitatory synapses on pyramidal neurons in the stratum radiatum rarely occurs in hippocampal area CA2. Here, we present evidence that perineuronal nets (PNNs), a specialized extracellular matrix typically localized around inhibitory neurons, also surround mouse CA2 pyramidal neurons and envelop their excitatory synapses. CA2 pyramidal neurons express mRNA transcripts for the major PNN component aggrecan, identifying these neurons as a novel source for PNNs in the hippocampus. We also found that disruption of PNNs allows synaptic potentiation of normally p

Activity-dependent degeneration of axotomized neuromuscular synapses in Wld S mice.

Activity and disuse of synapses are thought to influence progression of several neurodegenerative diseases in which synaptic degeneration is an early sign. Here we tested whether stimulation or disuse renders neuromuscular synapses more or less vulnerable to degeneration, using axotomy as a robust trigger. We took advantage of the slow synaptic degeneration phenotype of axotomized neuromuscular junctions in flexor digitorum brevis (FDB) and deep lumbrical (DL) muscles of Wallerian degeneration-Slow (Wld(S)) mutant mice. First, we maintained ex vivo FDB and DL nerve-muscle explants at 32°C for

Localization of kainate receptors in inner and outer hair cell synapses.

Glutamate plays a role in hair cell afferent transmission, but the receptors that mediate neurotransmission between outer hair cells (OHCs) and type II ganglion neurons are not well defined. A previous study using in situ hybridization showed that several kainate-type glutamate receptor (KAR) subunits are expressed in cochlear ganglion neurons. To determine whether KARs are expressed in hair cell synapses, we performed X-gal staining on mice expressing lacZ driven by the GluK5 promoter, and immunolabeling of glutamate receptors in whole-mount mammalian cochleae. X-gal staining revealed GluK5 e

Palmitoylation of gephyrin controls receptor clustering and plasticity of GABAergic synapses.

Postsynaptic scaffolding proteins regulate coordinated neurotransmission by anchoring and clustering receptors and adhesion molecules. Gephyrin is the major instructive molecule at inhibitory synapses, where it clusters glycine as well as major subsets of GABA type A receptors (GABAARs). Here, we identified palmitoylation of gephyrin as an important mechanism of strengthening GABAergic synaptic transmission, which is regulated by GABAAR activity. We mapped palmitoylation to Cys212 and Cys284, which are critical for both association of gephyrin with the postsynaptic membrane and gephyrin cluste

Precocious synapses in 13.5-week fetal holoprosencephalic cortex and cyclopean retina.

BACKGROUND: Genetic programming of cerebral development involves tissue morphogenesis and also timing of developmental processes. Precocious synaptogenesis in the neocortical plate was previously demonstrated in 5 of 6 fetuses of 20-31 weeks gestation. MATERIALS AND METHODS: Neuropathological examination was performed of a 13-week-5-day fetus with trisomy-13, a lobar holoprosencephaly, hydrocephalus, cyclopia and absence of ears. Immunocytochemical demonstration of the synaptic vesicle protein synaptophysin was performed in the brain and retina, along with other neuronal markers. RESULTS: Sy

Metabotropic glutamate receptors and dendrodendritic synapses in the main olfactory bulb.

The main olfactory bulb (MOB) is the first site of synaptic processing in the central nervous system for odor information that is relayed from olfactory receptor neurons in the nasal cavity via the olfactory nerve (ON). Glutamate and ionotropic glutamate receptors (iGluRs) play a dominant role at ON synapses. Similarly, glutamate and iGluRs mediate dendrodendritic transmission between several populations of neurons within the MOB network. Neuroanatomical studies demonstrate that metabotropic glutamate receptors (mGluRs) are densely expressed through the MOB network, and they are particularly a

Bidirectional Hebbian plasticity at hippocampal mossy fiber synapses on CA3 interneurons.

Hippocampal area CA3 is critically involved in the formation of nonoverlapping neuronal subpopulations ("pattern separation") to store memory representations as distinct events. Efficient pattern separation relies on the strong and sparse excitatory input from the mossy fibers (MFs) to pyramidal cells and feedforward inhibitory interneurons. However, MF synapses on CA3 pyramidal cells undergo long-term potentiation (LTP), which, if unopposed, will degrade pattern separation because MF activation will now recruit additional CA3 pyramidal cells. Here, we demonstrate MF LTP in stratum lacunosum-m
