In Parkinson's disease (PD), aggregation of alpha-synuclein (α-syn) contributes to neuronal dysfunction and death, particularly in dopaminergic neurons, driving disease progression. Several pathogenic point mutations have been identified in human α-syn such as A30P, E46K, H50Q, G51D, A53T and A53E that have been associated with PD. In this study, a sandwich ELISA assay was developed to quantify total α-syn levels in various Drosophila melanogaster genotypes expressing A30P, E46K, G51D and A53T. Using this approach, we compared α-syn levels across wild-type (WT) and mutant forms of the protein.

The pathological aggregation of α-synuclein (α-syn) is a hallmark of Parkinson's disease, where extracellular α-syn aggregates can disrupt intracellular α-synuclein conformation in neurons. In this study, we investigated the impact of 2',3'-dihydroxy-4',6'-dimethoxychalcone (DDC), isolated from green perilla, on α-syn fibrillization and subsequently on intracellular α-synuclein conformation. DDC concentration-dependently inhibited α-syn fibrillization in vitro, as determined by thioflavin S fluorescence and blue native polyacrylamide gel electrophoresis. DDC also disaggregated preformed α-syn

The autophagy-lysosomal pathway is crucial for maintaining homeostasis and survival of neurons, hence defects in this system have been associated with neurodegeneration, including Parkinson's disease (PD). The cysteine proteases cathepsin B (CTSB) and cathepsin L (CTSL) are involved in the clearance of various neurodegenerative disease-related proteins such as amyloid-[Formula: see text], huntingtin and the prion protein. While there are studies implicating CTSB and CTSL as mediators of α-synuclein/SNCA clearance, their exact roles remain unclear. We previously demonstrated that recombinant pr

Selective loss of discrete neuronal populations is a prominent feature of many neurodegenerative conditions, but the molecular basis of this is poorly understood. A central role of α-synuclein in the selective neurodegeneration of Parkinson's disease has been speculated, as its level of expression critically determines the propensity of this protein to misfold. To investigate whether the propensity of neuronal cell loss is associated with the level of endogenous α-synuclein expression, non-transgenic rats were given a single intravenous administration of α-synuclein pre-formed fibrils (PFFs) r

In most subjects with Parkinson's disease and dementia with Lewy bodies, alpha-synuclein (alphaS) immunoreactive pathology is found not only in the brain but also in the autonomic nuclei of the spinal cord. However, neither has the temporal course of alphaS pathology in the spinal cord in relation to the brain progression been established, nor has the extent of alphaS pathology in the spinal cord been analyzed in population-based studies. Using immunohistochemistry, the frequency and distribution of alphaS pathology were assessed semiquantitatively in the brains and spinal cord nuclei of 304 s

Abnormal α-synuclein (α-syn) accumulation and mitochondrial dysfunction are central features of Parkinson's disease (PD), and increasing evidence suggests that these processes are closely connected. Using yeast models to manipulate mitochondrial metabolic states, we investigated how mitochondrial activity may affect α-syn accumulation and toxicity. Under the fermentative growth condition, we found that α-syn localized predominantly to the plasma membrane and was non-toxic. Respiratory growth induced cytoplasmic α-syn accumulation, mitochondrial abnormalities, and marked toxicity dependent on α

The structure of α-synuclein is predominantly α-helical when bound to cellular membranes. However, under pathological or destabilizing conditions, this α-helical structure transitions into β-sheet-rich conformations, promoting protein aggregation and formation of Lewy bodies (LBs) in neurodegenerative disorders such as Parkinson's disease dementia (PDD), dementia with Lewy bodies (DLB), multiple system atrophy (MSA), and Alzheimer's disease (AD). DJ-1 is a multifunctional brain protein acting as a chaperone or protease under oxidative stress and associating with abnormal protein aggregates. Al

The synuclein family comprises three presynaptic proteins-alpha-synuclein (α-Syn), beta-synuclein (β-Syn), and gamma-synuclein (γ-Syn)-which are crucial for synaptic transmission. Our previous studies have detailed the precise cellular and subcellular localization of α- and β-Syn in the adult mouse retina, revealing distinct expression patterns in excitatory and inhibitory synaptic elements. The balance between excitatory and inhibitory neurotransmitters in the inner plexiform layer (IPL) is critical for neural development in the mouse retina. Here, we employed light-microscopic immunocytochem

Parkinson's disease (PD), the second most prevalent neurodegenerative disorder, is associated with α-synuclein (α-syn) overexpression or mutation, leading to harmful aggregates and neuronal apoptosis. Effective drugs that inhibit or reduce α-syn accumulation remain challenging. Targeted protein degradation (TPD) technology offers a novel solution by utilizing the ubiquitin-proteasome pathway to target specific proteins for destruction. Here, we have developed Proteolysis Targeting Chimera (PROTAC) to target α-syn for degradation. Specifically, our PROTACs employ the amino acid arginine (Arg) a

Parkinson's disease (PD) is a progressive neurodegenerative condition that affects the brain's ability to control movement. The disease occurs when nerve cells of the brain become impaired. Alpha-synuclein protein (α-Syn) aggregation followed by fibrillation is a hallmark of PD. Recent studies showed that the liquid-liquid phase separation (LLPS) and droplet formation of α-Syn protein play a crucial role in the pathology of PD. We synthesized novel zwitterionic compounds (3), which inhibited α-Syn phase separation and subsequent formation of fibrillary aggregates in a concentration-dependent m

Deposition of fibrillar aggregates of α-synuclein (αS) in the brain is a hallmark of αS-associated neurodegenerative diseases. Heparin, a structural analog of cell-surface heparan sulfate, modulates the kinetics of αS fibrillation and the morphology of the resulting fibrils. In this study, we investigated the kinetic mechanism of heparin-induced αS fibrillation using physicochemical techniques. Thioflavin T fluorescence and fibril-pelleting assays demonstrated that heparin greatly induces αS fibril formation and increases the mass of fibrillar αS in a concentration-dependent manner. Clinical and epidemiological studies suggest similarities in dysregulation of pathways in type 2 diabetes (T2DM) and Parkinson's disease (PD). Efficacy of several antidiabetic drugs has been tested in PD. Exenatide, a synthetic version of exendin-4, an incretin-mimetic drug, is an agonist of glucagon-like peptide 1 receptor (GLP1R) and is approved for the treatment of T2DM. Exenatide can cross the blood-brain barrier and exerts neuroprotective and neurorestorative effects via GLP1R at doses similar to those used in T2DM, resulting in improved motor performance, behaviour, learning and memory i

Interaction with lipid membranes is important in the physiological and pathological functioning of α-synuclein (αS) in brain neuronal cells. In this study, we investigated the effect of lipid composition on the membrane-binding behavior of αS using multiple biophysical techniques. Circular dichroism measurement revealed that, although negatively charged phospholipids are necessary for αS to bind to small unilamellar vesicles, the presence of phosphatidylethanolamine (PE) significantly enhances α-helical structure formation, specifically within the first 35 αS residues. To obtain residue-level

The accumulation of alpha-synuclein (Asyn) fibrils is the defining pathologic feature in Parkinson's disease, Lewy body dementia, and multiple system atrophy. As such, the process of Asyn fibril formation has been an important research area and fibrils themselves have become attractive targets for disease diagnosis and therapeutic intervention. Due to the presence of mixed populations of fibrillar proteins associated with neurodegenerative diseases in brain tissue, high-resolution structures of Asyn fibrils are essential for the design of high-specificity imaging and therapeutic agents. Alpha-synuclein is a 140 aa neuronal protein pathologically and genetically linked to Parkinson's disease (PD). In PD, the major protein modification of alpha-synuclein is phosphorylation at serine 129. Alpha-synuclein phosphorylation also occurs at low levels in normal brains and cultured CNS neurons, but its regulation and role are only beginning to be investigated. The enteric nervous system is now recognized as a second brain in its own right, and therefore, we set out to examine the regulation of alpha-synuclein phosphorylation in enteric neurons. To this end, primary cultures of rat ente

Chaperones may retard the aggregation of other proteins and increase their solubility. An important goal is a thermodynamic understanding of such an action. Here, the chaperone DNAJB6b (JB6) is found to suppress amyloid formation of the protein α-synuclein (α-syn) leading to a reduced rate of fibril formation and an increase in apparent solubility of α-syn. These findings were reached at mildly acidic pH and with light seeding under conditions where the effect on secondary nucleation is visible. Cryo-transmission electron microscopy (cryo-TEM) imaging reveals that coaggregates of α-syn and JB6

α-Synuclein (α-syn), a pathological hallmark of PD, is emerging as a bridging element at the crossroads between neuro/immune-inflammatory responses and neurodegeneration in PD. Several evidence show that pathological α-syn accumulates in neuronal and non-neuronal cells (i.e., neurons, microglia, macrophages, skin cells, and intestinal cells) in central and peripheral tissues since the prodromal phase of the disease, contributing to brain pathology. Indeed, pathological α-syn deposition can promote neurogenic/immune-inflammatory responses that contribute to systemic and central neuroinflammatio

Progressive aggregation of α-synuclein (α-Syn), a small cytosolic protein involved in cell vesicle trafficking, in the midbrain, hypothalamus, and thalamus is linked to Parkinson's disease (PD). Amyloid oligomers and fibrils formed as a result of such aggregation are highly toxic to neurons. However, it remains unclear whether amyloid-induced toxicity of neurons is the primary mechanism of the progressive neurodegeneration observed upon PD. In the current study, we investigated cytotoxicity exerted by α-Syn fibrils formed in the lipid-free environment, as well as in the presence of two phospho
