Molecular diagnostics of disorders of sexual development: an Indian survey and systems biology perspective.

We aimed to survey the monogenic causes of disorders of sex development (DSD) and thereby its prevalence in India. This study revealed mutations resulting in androgen insensitivity syndrome, 5α-reductase type 2 deficiency, and gonadal dysgenesis were commonly reported. Intriguingly, AR deficits were the most prevalent (32 mutations) and of 11/26 missense mutations were in exons 4-8 (encoding ligand binding domain). The unique features of SRD5A2 defects were p.R246Q (most prevalent) and p.G196S could be mutational hotspots, dual gene defects (p.A596T in AR and p.G196S in SRD5A2) in a patient wi

Systems biology in the central nervous system: a brief perspective on essential recent advancements.

As recent advances in human genetics have begun to more rapidly identify the individual genes contributing to risk of psychiatric disease, the spotlight now turns to understanding how disruption of these genes alters the brain, and thus behavior. Compared to other tissues, cellular complexity in the brain provides both a substantial challenge and a significant opportunity for systems biology approaches. Current methods are maturing that will allow for finally defining the 'parts list' for the functioning mouse and human brains, enabling new approaches to defining how the system goes awry in di

A systems biology approach identifies a regulatory network in parotid acinar cell terminal differentiation.

OBJECTIVE: The transcription factor networks that drive parotid salivary gland progenitor cells to terminally differentiate, remain largely unknown and are vital to understanding the regeneration process. METHODOLOGY: A systems biology approach was taken to measure mRNA and microRNA expression in vivo across acinar cell terminal differentiation in the rat parotid salivary gland. Laser capture microdissection (LCM) was used to specifically isolate acinar cell RNA at times spanning the month-long period of parotid differentiation. RESULTS: Clustering of microarray measurements suggests that ex

Anti-infectious drug repurposing using an integrated chemical genomics and structural systems biology approach.

The emergence of multi-drug and extensive drug resistance of microbes to antibiotics poses a great threat to human health. Although drug repurposing is a promising solution for accelerating the drug development process, its application to anti-infectious drug discovery is limited by the scope of existing phenotype-, ligand-, or target-based methods. In this paper we introduce a new computational strategy to determine the genome-wide molecular targets of bioactive compounds in both human and bacterial genomes. Our method is based on the use of a novel algorithm, ligand Enrichment of Network Top

Nutritional regulation of division of labor in honey bees: toward a systems biology perspective.

Organisms adapt their behavior and physiology to environmental conditions through processes of phenotypic plasticity. In one well-studied example, the division of labor among worker honey bees involves a stereotyped yet plastic pattern of behavioral and physiological maturation. Early in life, workers perform brood care and other in-hive tasks and have large internal nutrient stores; later in life, they forage for nectar and pollen outside the hive and have small nutrient stores. The pace of maturation depends on colony conditions, and the environmental, physiological, and genomic mechanisms b

The miRNA-virus axis in RNA virus infections: Mechanisms, therapeutic targeting, and systems biology approaches.

RNA viruses are a diverse and rapidly evolving group of pathogens that significantly contribute to worldwide morbidity and mortality, propelled by elevated mutation rates and effective use of host cellular mechanisms. MicroRNAs (miRNAs) have emerged as an essential post-transcriptional regulator of host-virus interactions, influencing viral replication, immunological responses and disease progression. This review offers a comprehensive overview of the miRNA-virus axis in RNA virus infections, highlighting the dynamic and context-dependent roles of host miRNAs. We investigate the mechanistic ba

Immune-Metabolic Interactions in the Degenerative Retina: A Systems Biology Approach Based on Diabetic Retinopathy.

Diabetic retinopathy (DR) is increasingly recognized as a complex neurovascular degenerative disorder driven by intertwined immune and metabolic disturbances within the retinal microenvironment. Chronic hyperglycemia induces metabolic stress, mitochondrial dysfunction, and oxidative imbalance, which, in turn, activate innate and adaptive immune pathways. Key mechanisms-including complement dysregulation, microglial activation, leukostasis, cytokine and chemokine signaling, and advanced glycation end-product-mediated inflammation-contribute to endothelial injury, barrier breakdown, and progress

A co-evolutionary perspective on humans and Mycobacterium tuberculosis in the era of systems biology.

Tuberculosis is once again the most fatal global infectious disease and has killed many more humans than any other pathogen. Despite the identification of Mycobacterium tuberculosis (Mtb) over 140 years ago, we have yet to control the epidemic. A central issue is the complexity of the host-pathogen interaction, with multiple underlying pathways leading to tuberculosis disease. This intricate relationship stems from the prolonged co-evolution of the pathogen with humans, resulting in diverse immunological processes leading to tuberculosis disease. Conversely, Mtb exposure may give a survival ad

A molecular renaissance in Alzheimer's disease research: The rise of systems biology and spatial omics.

Recent advances in multi-omics and spatial proteomics are reshaping our understanding of Alzheimer's disease. Guo et al.1 applied integrative multi-omics to stratify mild cognitive impairment into biologically distinct subtypes with divergent progression trajectories: one metabolically impaired and slow-progressing, the other immune-activated and rapidly declining. Current techniques such as STC-DESI, LCM-MS, and machine learning enhance regional proteomic resolution, supporting biomarker discovery and spatially targeted interventions. This work exemplifies a broader shift toward precision med

Systems biology modelling based evaluation of omega-3 formulation in managing cardiovascular and cerebrovascular risk.

Natural therapies for cardiovascular and cerebrovascular disease management often use nutraceuticals and herbal supplements to improve lipid profiles. This in-silico study examined the effects of Omega-3 fatty acids, specifically Eicosapentaenoic acid (EPA) and Docosahexaenoic acid (DHA), on lipid metabolism using systems biology-based models. The pharmacokinetics and pharmacodynamics of EPA (1 g) and DHA (1 g) were analysed to understand their lipid-lowering mechanisms. The study also assessed the impact of omega-3 fatty acid-based formulation, (EPA 180 mg and DHA 120 mg), on lipid biomarkers

Organokine-Mediated Crosstalk: A Systems Biology Perspective on the Pathogenesis of MASLD-A Narrative Review.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent chronic condition with a complex pathophysiology involving multiple organs. Organokines, including hepatokines, myokines, cardiokines, renokines, osteokines, and adipokines, play central roles in lipid metabolism, glucose homeostasis, inflammation, and fibrosis. Dysregulation of these signaling molecules contributes to the progression of MASLD and its systemic complications. This review examines the role of organokine-mediated crosstalk between the liver and peripheral organs (e.g., muscle, heart, kidneys, bone, an

A Systems Biology Perspective on Childhood ADHD: Neurochemical Dysregulation, Brain-Behavior Interactions, and Emerging Therapeutics.

Attention deficit hyperactivity disorder (ADHD) is a pervasive neurodevelopmental condition that typically manifests during childhood. Its symptoms manifest much earlier-often before the age of seven. With the disorder's continuous patterns of inattention, hyperactivity, and impulsiveness, it gets in the way of academic, social, and occupational performance. Individuals affected by ADHD are found among all age groups, though its presentation may change over time. The mechanisms of ADHD seem to be very complex and multifactorial and include disturbances within neuroanatomical structures, neuroc

Evolutionary Systems Biology Identifies Genetic Trade-offs in Rice Defense against Aboveground and Belowground Attackers.

Like other plants, wild and domesticated rice species (Oryza nivara, O. rufipogon, and O. sativa) evolve in environments with various biotic and abiotic stresses that fluctuate in intensity through space and time. Microbial pathogens and invertebrate herbivores such as plant-parasitic nematodes and caterpillars show geographical and temporal variation in activity patterns and may respond differently to certain plant-defensive mechanisms. As such, plant interactions with multiple community members may result in conflicting selection pressures on genetic polymorphisms. Here, through assays with

From CFTR to a CF signalling network: a systems biology approach to study Cystic Fibrosis.

BACKGROUND: Cystic Fibrosis (CF) is a monogenic disease caused by mutations in the gene coding the Cystic Fibrosis Transmembrane Regulator (CFTR) protein, but its overall physio-pathology cannot be solely explained by the loss of the CFTR chloride channel function. Indeed, CFTR belongs to a yet not fully deciphered network of proteins participating in various signalling pathways. METHODS: We propose a systems biology approach to study how the absence of the CFTR protein at the membrane leads to perturbation of these pathways, resulting in a panel of deleterious CF cellular phenotypes.
