UNLABELLED: Post-translational modifications orchestrate virus-host interactions, yet their roles in the hepatitis B virus (HBV) life cycle remain incompletely defined. The small hepatitis B surface antigen (SHBs)-the most abundant HBV protein-drives immune evasion, pathogenesis, and virion/subviral particle (SVP) production. Here, we identify UFL1, the E3 ligase of UFMylation, as a host factor that modifies and stabilizes SHBs by antagonizing its ubiquitination to potentiate HBV output and SHBs-driven pro-oncogenic activities. Using affinity purification-mass spectrometry, we discovered UFL1

BACKGROUND: Colorectal cancer (CRC) is the third most common gastrointestinal cancer in terms of morbidity and mortality worldwide. Reprogramming of glucose metabolism will directly affect the energy supply pattern of tumor cells and determine their malignant biological behavior. Mitofusin-2 (MFN2) has been shown to be associated with glycolysis. Therefore, targeting MFN2 may be a potential therapeutic approach for CRC.
METHODS: Firstly, GSE143939, GSE81558 dataset and GSE184093 dataset were used to screen differentially expressed mRNAs in CRC. The expression of MFN2 in colorectal cancer was

Ebola virus disease (EVD) caused by Zaire Ebolavirus (EBOV) infection is a major threat to public health in Africa and even worldwide, due to its extremely high mortality rate. However, there are still no effective antiviral therapies that can completely cure EVD. A comprehensive understanding of virus-host interactions would be beneficial for developing new antiviral agents. Here, we showed that CXCR4-induced macroautophagy/autophagy and was internalized to endosomes by interacting with glycoprotein (GP) on viral particles during EBOV infection; this promoted the EBOV attachment and entry, wh

BACKGROUND: Post-resuscitation brain injury is the main contributor to the death and disability of cardiac arrest (CA) victims. Neuronal ferroptosis is involved in the pathogenesis of brain injury after resuscitation, which becomes an effective therapeutic target. Recent studies demonstrated that mesenchymal stem cell-derived exosomes (MSC-Exo) were a promising approach for the alleviation of regional cerebral ischemia reperfusion injury; however, its effectiveness in post-resuscitation brain injury remains to be investigated. In this study, we investigated whether MSC-Exo could alleviate post

AIMS: Dermatomyositis (DM) is a progressive, idiopathic inflammatory myopathy with poorly understood pathogenesis. A hallmark of DM is an increased risk for developing breast, ovarian, and lung cancer. Since autoantibodies against anti-TIF-1-γ, a member of the tripartite motif (TRIM) proteins, has a strong association with malignancy, we examined expression of the TRIM gene family to identify pathways that may be contributing to DM pathogenesis.
MATERIALS AND METHODS: We employed the Search Tag Analyze Resource for GEO platform to search the NCBI Gene Expression Omnibus to elucidate TRIM fami

OBJECTIVES: Sjögren's disease (SD) is an autoimmune disease affecting the exocrine glands that is associated with autoantibodies against Ro60/SS-A, anti-Ro52/TRIM21, La/SS-B and others. We examined the role of acute Epstein-Barr virus (EBV) infection in the pathogenesis of these autoantibodies in a previously healthy patient (patient 1) with primary EBV infection who developed SD with anti-Ro/La and anti-Smith/U1 ribonucleoprotein (Sm/U1RNP) autoantibodies and had lymphoplasmacytic foci on labial salivary gland biopsy.
METHODS: Immune responses to Epstein-Barr nuclear antigen-1 (EBNA1) and th

BACKGROUND: Chronic Hepatitis C Virus (HCV) infection has been commonly linked to the development of autoimmunity, in part through activation of B cells. B cells are also postulated to play a pathogenic role in the autoimmune liver disease Primary Biliary Cholangitis (PBC). Patients with concurrent PBC and HCV infection carry an increased risk of more progressive disease, although the mechanism underlying this effect is poorly understood. Utilizing a case series of patients with concurrent PBC and HCV, the aim of this study was to evaluate for the potential impact of HCV eradication upon autoi

Crimean-Congo hemorrhagic fever virus (CCHFV) is the cause of a sometimes-severe viral hemorrhagic fever, CCHF. Severe cases of CCHF often have little-to-no CCHFV-specific antibody responses prior to death. However, it is unclear if the failure to mount an antibody response is the cause of a poor outcome or a correlate of a failed host response. Here, we used a mouse-adapted strain of CCHFV (MA-CCHFV) to investigate the requirement of humoral immunity in control of acute CCHFV infection in immunocompetent mice. We found that mice infected with MA-CCHFV develop a rapid non-neutralizing antibody

Melanoma-associated antigen D4 (MAGED4) belongs to the melanoma-associated antigen family and is upregulated in various cancer types. However, the functional role and molecular mechanisms of MAGED4 in hepatocellular carcinoma (HCC) remain largely unknown. In this study, we observed that MAGED4 expression levels were significantly higher in HCC tissues than in non-cancerous tissues and elevated expression was associated with poor patient outcomes. Functional assays demonstrated that MAGED4 promoted proliferation and migration of HCC. We found that MAGED4 can activate the Janus kinase 2/signal t

Immune checkpoint blockade (ICB) targeting PD-1/PD-L1 improves outcomes across multiple malignancies, yet resistance to immune checkpoint blockade remains common. Here, we identify HILPDA as a tumor-intrinsic regulator of immune evasion in breast cancer. HILPDA overexpression increases the infiltration and suppressive activity of regulatory T cells while decreasing the infiltration, activation, and cytotoxicity of CD8+ T cells and natural killer cells, thereby establishing an immunosuppressive tumor microenvironment. Mechanistically, HILPDA binds to HSP90 and protects the transcription factor

BACKGROUND: Alzheimer's disease (AD) is characterized by progressive neurodegeneration driven by amyloid-β (Aβ) plaques and tau neurofibrillary tangles. Stress granules (SGs), dynamic ribonucleoprotein condensates formed under cellular stress have been implicated in several neurodegenerative disorders, but their role in AD pathogenesis remains incompletely understood.
METHODS: Following PRISMA-ScR guidelines, we systematically searched PubMed, Embase, Scopus, Web of Science, and Cochrane Library (through February 2026) for peer-reviewed studies investigating SGs in AD models or human tissue.

OBJECTIVE: To synthesize current knowledge of molecular mechanisms underlying human lumbar disc degeneration (LDD) and identify knowledge gaps to be addressed by future research.
SUMMARY OF BACKGROUND DATA: Chronic low back pain (CLBP) is a leading cause of disability worldwide. Epidemiological studies based on radiologic findings suggest that patients with LDD are more likely to develop CLBP. Despite many reviews on disc degeneration, no systematic synthesis has focused on the molecular mechanisms of LDD using human tissue models.
METHODS: A systematic search

Myocardial infarction (MI) and ischemia-reperfusion injury (IRI) are critical challenges in clinical practice. The function of macrophage in this process has gained increasing attention. Macrophage polarization from pro-inflammatory (M1) to reparative (M2) phenotypes is crucial for cardiac repair. However, the regulatory mechanisms behind macrophage polarization, particularly involving m6A modification are not well understood. We utilized a mouse model of myocardial IRI to study m6A modifications and related enzymes in a mouse model of myocardial IRI. We co-cultured bone marrow-derived macroph

Small cell lung cancer (SCLC) is an aggressive malignancy that responds poorly to immune checkpoint inhibitor (ICI) therapy, due to its immunosuppressive tumor microenvironment. Here, we analyse preclinical mouse tumor models and patient-derived SCLC samples and identify Exportin 1 (XPO1) as a regulator of immune suppression in SCLC. Mechanistically, XPO1 drives the nuclear export of TRIM21, enabling TRIM21-dependent proteasomal degradation of IRF3 and repressing the immunostimulatory cytokine TNFSF15. Loss of TNFSF15 promotes TREM2+ macrophage polarization, which subsequently attenuates macro

BACKGROUND & AIMS: Liver regeneration is a tightly regulated process requiring coordinated interactions between hepatocytes and non-parenchymal cells; however, its molecular mechanisms remain incompletely defined. Here, we aimed to investigate the role of interferon-related developmental regulator 1 (IFRD1) in regulating metabolic-immune crosstalk during liver regeneration.
METHODS: We integrated public transcriptomic datasets, human liver disease samples, and multiple in-house-generated experimental models to characterize the dynamic expression of IFRD1 during liver regeneration. Genetic los

Frontotemporal lobar degeneration with neuronal inclusions of the TAR DNA-binding protein 43 (FTLD-TDP) represents the most common pathological subtype of FTLD. We established the international FTLD-TDP whole-genome sequencing consortium to thoroughly characterize the known genetic causes of FTLD-TDP and identify novel genetic risk factors. Through the study of 1131 unrelated Caucasian patients, we estimated that C9orf72 repeat expansions and GRN loss-of-function mutations account for 25.5% and 13.9% of FTLD-TDP patients, respectively. Mutations in TBK1 (1.5%) and other known FTLD genes (1.4%)

Immune checkpoint inhibitors (ICIs) can re-active the immune response and induce a complete response in mismatch repair-deficient and microsatellite instability-high (dMMR/MSI-H) colorectal cancer (CRC). However, most CRCs exhibit proficient mismatch repair and microsatellite stable (pMMR/MSS) phenotypes with limited immunotherapy response because of sparse intratumoral CD8+ T-lymphocyte infiltration. Cellular senescence has been reported to involve immune cell infiltration through a senescence-associated secretory phenotype (SASP). However, the relationship between CRC cellular senescence and

Abnormal accumulation of Poly(ADP-ribose) polymerase 1 (PARP1) promotes cancer progression, yet its stabilization mechanisms remain unclear. Here, we identify E3 ubiquitin ligase tripartite motif-containing 21 (TRIM21) as a PARP1-binding partner. PARP1 interacts directly with TRIM21 via its 662-908 domain, while the PRY-SPRY domain of TRIM21 is essential for this binding. TRIM21 facilitates PARP1 polyubiquitination at residue K654, leading to its degradation. In small cell lung cancer (SCLC), TRIM21 is significantly downregulated, and its tumor-suppressive function is partly mediated through t
