X-linked hyper IgM syndrome with a novel CD40-ligand gene mutation suffered from infection of Talaromyces marneffei.

BACKGROUND: X-linked Hyper-IgM Syndrome (X-HIGM) is a primary immunodeficiency caused by pathogenic variants in CD40L. Loss of functional CD40 ligand interferes with T-cell-dependent B-cell responses and immunoglobulin class-switch recombination, resulting in susceptibility to recurrent and opportunistic infections. Because CD40L is located on the X chromosome, X-HIGM is usually diagnosed in males. Symptomatic females are rare, and the clinical and immunological features of these patients have not been well defined. METHODS: We report a 4-year-old Chinese girl who developed disseminated Talar

Manganese-Induced Production of Antioxidant Polyene Steroids in the Extremophile Fungus Talaromyces fuscoviridis Isolated From Copper-Mine Soil.

The fungus Talaromyces fuscoviridis was isolated from soil collected in a copper-mine tailings basin in Pará State, Brazil, an extreme environment that can induce the production of bioactive secondary metabolites. Extracts and fractions obtained after cultivation were analyzed by liquid chromatography coupled to high-resolution mass spectrometry (LC-HRMS) and compared using Global Natural Products Social molecular networking (GNPS), which revealed a family of seven putative polyunsaturated steroids. Isolation led to the characterization of two major steroids: the known Ergosta-3(4),5(6),7(8),1

Machine learning-based in-hospital mortality prediction of HIV/AIDS patients with Talaromyces marneffei infection in Guangxi, China.

OBJECTIVE: Talaromycosis is a serious regional disease endemic in Southeast Asia. In China, Talaromyces marneffei (T. marneffei) infections is mainly concentrated in the southern region, especially in Guangxi, and cause considerable in-hospital mortality in HIV-infected individuals. Currently, the factors that influence in-hospital death of HIV/AIDS patients with T. marneffei infection are not completely clear. Existing machine learning techniques can be used to develop a predictive model to identify relevant prognostic factors to predict death and appears to be essential to reducing in-hospit

Disseminated Mycobacterium kansasii disease complicating Talaromyces marneffei infection in patient with anti-IFN-γ autoantibodies: a case report.

BACKGROUND: Patients with immunodeficiency syndromes associated with anti-interferon-gamma (IFN-γ) autoantibodies are prone to a wide range of infections, particularly those caused by intracellular pathogens such as nontuberculous mycobacteria (NTM) or Talaromyces marneffei (T. marneffei). However, disseminated Mycobacterium kansasii (M. kansasii) infection co-occurring with T. marneffei infection is relatively rare. Here, we present a case of disseminated M. kansasii disease complicated by T. marneffei infection in a patient with anti-IFN-γ autoantibodies. CASE PRESENTATION: A 52-year-old ma

Inflammation-related indices as markers for mortality in people living with HIV co-infected with Talaromyces Marneffei: a retrospective analysis.

Talaromyces marneffei (T. marneffei) co-infection remains a significant cause of mortality in people living with HIV (PLWH). Although early detection of individuals at high risk is essential, there remains a lack of markers to predict outcomes. This multicenter retrospective study analyzed 282 PLWH (95 treatment-naïve T. marneffei co-infected, 187 without opportunistic infections) to evaluate six inflammatory indices: systemic immune-inflammation index (SII), neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), lymphocyte-to-monocyte ratio (LMR), neutrophil-to-platelet rat

Cerebral vasculitis caused by Talaromyces marneffei and Aspergillus niger in a HIV-positive patient: a case report and literature review.

Cerebral vasculitis is a long-standing but flourishing and fadeless research topic. Infections are a frequent cause of cerebral vasculitis, vital to diagnose due to involvement of specific anti-infection treatments. A 65-year-old man visited the hospital for his neurological symptoms without obvious inducements. After admission, radiological examination and comprehensive conventional microbiological tests (CMTs) revealed suspected intracranial infectious vasculitis. Metagenomic next-generation sequencing (mNGS) and reverse transcription-polymerase chain reaction further confirmed that his cere

Treg Cells Impair Talaromyces Marneffei Clearance by Promoting M2 Macrophage Polarization Via the IL-10/IL-10R/STAT3 Axis.

Talaromyces marneffei (TM) causes a deeply invasive fatal fungal disease in immunocompromised individuals that is widespread in Southeast Asia and southern China, and little is known about how this fungus can evade the human immune system. An imbalance between M1 and M2 macrophages plays a critical role in fungal clearance. However, whether regulatory T (Treg) cells mediate the immune evasion of TM by influencing the polarization direction of macrophages remains unexplored. In this study, we found that, compared with those in healthy volunteers, the number of circulating Treg cells in the peri
