Alpha-lipoic acid promotes the growth of rat hepatic pre-neoplastic lesions in the choline-deficient model.

alpha-lipoic acid (alpha-LA) is an antioxidant used in a number of conditions related to liver diseases. Herein, we investigated the effect of alpha-LA on the development of rat pre-neoplastic lesions generated by a model of hepatocarcinogenesis, which has similarities in its histopathological sequence to human hepatocellular carcinoma development with cirrhosis. Initiation of hepatocytes was achieved by treatment with a single dose of diethylnitrosamine and promotion by feeding a choline-methionine-deficient diet (CMD), with or without alpha-LA. Pre-neoplastic lesions were identified by their

Enhanced HGF with increased receptor affinity and nitration-dysfunction resistance through interaction with lipoic acid trisulfide.

Myogenic stem cell activator HGF (hepatocyte growth factor) undergoes nitration of tyrosine residues (Y198, Y250) predominantly on fast-twitch fibers to lose its binding affinity to the signaling receptor c-met, in response to peroxynitrite (ONOO-) generation during aging. Here we show that HGF adopts an enhanced form with dynamically increased receptor-binding affinity and nitration-dysfunction resistance through interaction with lipoic acid trisulfide (LASSS) under physiological conditions. When evaluated after exposure to LASSS at a 1:8000 molar ratio to HGF and subsequent ultra-filtration

Lipoic Acid as a Possible Pharmacological Source of Hydrogen Sulfide/Sulfane Sulfur.

The aim of the present study was to verify whether lipoic acid (LA) itself is a source of H₂S and sulfane sulfur. It was investigated in vitro non-enzymatically and enzymatically (in the presence of rat tissue homogenate). The results indicate that both H₂S and sulfane sulfur are formed from LA non-enzymatically in the presence of environmental light. These results suggest that H₂S is the first product of non-enzymatic light-dependent decomposition of LA that is, probably, next oxidized to sulfane sulfur-containing compound(s). The study performed in the presence of rat liver and kidney homoge

[Effect of lipoic acid on tyrosine metabolism in patients with open-angle glaucoma].

Lipoic acid effects on tyrosine metabolism and ocular hydrodynamics were examined in patients with primary open-angle glaucoma. Glaucoma patients not administered the drug and patients with ocular hypertension made up two reference groups. In the majority of glaucoma patients administration of lipoic acid did not result in improvement of tyrosine metabolism, whereas hydrodynamic parameters somewhat increased. Tyrosine metabolism normalized in the patients administered lipoic acid in complex with tyrosine catabolism cofactors (pyridoxal phosphate, ascorbic acid, iron ions). The studied paramete

Natural Antioxidant Lipoic Acid Constructed Nanodrug for Oral Therapy of Inflammatory Bowel Disease.

Although antioxidant therapy is promising for inflammatory bowel disease (IBD), the current drugs, such as natural antioxidants and metal nanozymes, cannot meet the needs of efficacy and/or long-term or even lifelong safe administration. Herein, we developed a safe and efficient nanodrug for IBD through self-polymerization of the natural antioxidant lipoic acid (LA) into cross-linked poly(lipoic acid) particles (cLAPs). Owing to the structural homology with the LA monomer, the cLAPs exhibited exceptional biosafety. Orally administered cLAPs selectively adhered to positively charged inflammator

Peptide-Directed Aqueous-Phase Fabrication of Lipoic Acid Hydrogels for Covalent Protein Loading and On-Demand Release in Wound Healing.

Despite their biomedical potential, α-lipoic acid (LA)-based hydrogels face challenges regarding polymerization, stability, and functionalization. Here, we propose a "polymerization-functionalization" strategy for fabricating stable LA-based hydrogels using a bioderived peptide (SpyTag-Cys, ST-Cys) as an initiator. Through multiscale interactions (peptide-monomer, -polymer, and -peptide), ST-Cys drives efficient polymerization of LA monomers under mild aqueous conditions and fortifies the network against hydrolysis while simultaneously providing anchoring sites for covalent protein loading. Alpha-Lipoic Acid and Benfotiamine in Diabetic Peripheral Neuropathy: A Critical Review of Mechanistic Rationale and Clinical Evidence Within a Nutritional Therapeutic Framework.

BACKGROUND: Diabetic peripheral neuropathy (DPN) affects up to 50% of diabetes patients and is driven by hyperglycemia-induced oxidative stress, mitochondrial dysfunction, polyol pathway activation, advanced glycation end-product formation, and inflammation. Current management is largely symptomatic, prompting interest in metabolic/nutritional therapies. This review critically evaluates the mechanistic rationale and clinical evidence for alpha-lipoic acid (ALA) and benfotiamine as adjunctive treatments for DPN. METHODS: A structured narrative review of PubMed/MEDLINE was conducted using prede

Protective Effect of Lipoic Acid on Oxidative Stress and Tissue Damage Induced by Aflatoxin B1 in Young Laying Hens.

The aim of this study is to investigate the alleviating effect of lipoic acid on oxidative stress and tissue damage induced by aflatoxin B1 (AFB1) in young laying hens. The experiment was divided into a control group, an AFB1 group, and three lipoic acid treatment groups. The AFB1 group and three lipoic acid treatment groups were given diets supplemented with 90 μg/kg of AFB1. The additional amounts of lipoic acid were 20, 100, and 500 mg/kg, respectively, with a feeding period of 4 weeks. The experimental results showed that AFB1 significantly increased the levels of alanine aminotransferase

Strain-Promoted mRNA Transdermal Delivery by Lipoic Lipid Nanoparticles for Therapeutic Skin Genome Editing.

Lipid nanoparticle (LNP)-mRNA formulations have revolutionized the field of nucleic acid therapeutics, yet their broader clinical application is constrained by inflammatory side effects and oxidative stress, particularly in the context of inflammatory diseases. Herein, we report the rational design and synthesis of a lipoic acid-based ionizable lipid library to address these limitations. By leveraging the antioxidant properties and thiol-mediated uptake potential of lipoic acid, we identified LA-A2B2CD3 as an optimal candidate through a structure-activity relationship study and design of exper

Evaluation of the Protective Effects of Alpha Lipoic Acid on Bleomycin-Induced Ovarian Toxicity.

Chemotherapeutic drugs administered during cancer therapy, may lead to the depletion of ovarian follicles, and subsequent infertility in fertile patients. We aimed to determine toxic effects of bleomycin (BLM) on rat ovary, and to evaluate protective effects of alpha lipoic acid (ALA) on BLM toxicity. The total of 30 adult female rats were split into 4 groups. First, an intramuscular injection (i.m) of BLM (30 mg/m2) was administered to BLM and BLM + ALA groups except the control and ALA groups on the 1st, 8th and 15th days. The control group received 0.1 mL (i.m) saline on those days. BLM + A

Preclinical Evaluation of a Pilocarpine-(R)-Lipoic Acid Eye Drop for Presbyopia.

PURPOSE: Presbyopia is a progressive, age-related loss of near vision. Although current therapies offer symptomatic relief, they fail to target the underlying pathology. These studies investigated a novel dual-mechanism eye drop, CLX-162 (pilocarpine lipoate salt), focusing on three key characteristics: (1) tolerability, (2) pharmacokinetics and ocular tissue penetration, and (3) chemical stability within a dual-chamber delivery system. METHODS: Tolerability and pharmacokinetic studies involved administering CLX-162 and lipoic acid choline ester (LACE) ophthalmic formulations to New Zealand W

R-α-Lipoic Acid Conjugated to d-α-Tocopherol Polyethylene Glycol 1000 Succinate: Synthesis, Characterization, and Effect on Antiseizure Activity.

α-Lipoic acid (LA), a dithiol micronutrient, acts as a vital cofactor in various cellular catabolic reactions and is also known as a universal antioxidant. The therapeutic efficacy of LA is compromised by a poor aqueous solubility as well as a short half-life. In the present study, LA was conjugated to d-α-tocopherol polyethylene glycol succinate (TPGS) using carbodiimideacid-alcohol coupling reaction. The synthesized conjugate (TPGS-LA) was characterized using 1H and 13C nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FT-IR), UV-vis spectroscopy, and matrix-assisted

Alpha-Lipoic Acid and Glucose Metabolism: A Comprehensive Update on Biochemical and Therapeutic Features.

Alpha-lipoic acid (ALA) is a natural compound with antioxidant and pro-oxidant properties which has effects on the regulation of insulin sensitivity and insulin secretion. ALA is widely prescribed in patients with diabetic polyneuropathy due to its positive effects on nerve conduction and alleviation of symptoms. It is, moreover, also prescribed in other insulin resistance conditions such as metabolic syndrome (SM), polycystic ovary syndrome (PCOS) and obesity. However, several cases of Insulin Autoimmune Syndrome (IAS) have been reported in subjects taking ALA. The aim of the present review i
