Manganese (Mn) constitutes an essential co-factor in the oxygen-evolving complex of photosystem II (PSII). Consequently, Mn deficiency reduces photosynthetic efficiency and leads to changes in PSII composition. In order to study these changes, multiplexed protein assays are advantageous. Here, we developed a multiplexed antibody-based assay and analysed selected PSII subunits in barley (Hordeum vulgare L.). A selection of antibodies were labelled with specific lanthanides and immunoreacted with thylakoids exposed to Mn deficiency after western blotting. Subsequently, western blot membranes wer

Optimization of photosynthesis requires precise coordination between electron transport and CO2 assimilation, but how plants maintain this coordination under suboptimal CO2 and light conditions remains poorly understood. Here, we identify two tomato (Solanum lycopersicum) β-carbonic anhydrases (βCAs) responsive to deficient CO2. Although both are required for biomass accumulation and efficient photosynthesis, the chloroplast βCA1 does not directly participate in carbon assimilation but instead specifically coordinates photosystem performance and leaf internal structure. This role is distinct f

We have previously shown that heat-shock in the dark evokes photomorphogenesis-like effects and circadian rhythmicity at the level of mRNAs when applied to emerging pea plantlets during several consecutive days [15]. Here we extend these findings by showing that a temperature shift to 10 degrees C above average and a single heat-shock are sufficient for induction of circadian rhythmicity and changes in morphogenesis. The maximum response to a single heat-shock occurs at days 2 to 3 after sowing indicating additional developmental control of the response. An increasing number of heat-shock trea

Ilex × altaclerensis 'Belgica Aurea' is an attractive ornamental plant bearing yellow-green variegated leaves. However, the mechanisms underlying the formation of leaf variegation in this species are still unclear. Here, the juvenile yellow leaves and mature variegated leaves of I. altaclerensis 'Belgica Aurea' were compared in terms of leaf structure, pigment content and transcriptomics. The results showed that no obvious differences in histology were noticed between yellow and variegated leaves, however, ruptured thylakoid membranes and altered ultrastructure of chloroplasts were found in ye

To examine the role of sulfoquinovosyl diacylglycerol (SQDG) in thylakoid membranes, we compared the structural and functional properties of photosystem II (PSII) between a mutant of Chlamydomonas reinhardtii defective in SQDG ( hf-2) and the wild type. The PSII core complex of hf-2, as compared with that of the wild type, showed structural fragility when solubilized with a detergent, dodecyl beta- d-maltoside, suggesting that the physical properties of the PSII complex were altered by the loss of SQDG. On the other hand, exposure of the cells to 41 degrees C for 120 min in the dark decreased

Chilling stress induces photosystem I (PSI) photoinhibition in various plants, severely impairing their growth. However, the mechanisms suppressing chilling-induced PSI photoinhibition remain unclear. To identify factors preventing PSI photoinhibition, we compared two cucumber cultivars with different susceptibilities to PSI photoinhibition and chilling stress tolerance. Chilling stress caused uncoupling of thylakoid membrane in both cultivars, as previously reported. However, in the chilling-sensitive cultivar, electron efflux from ferredoxin (Fd) was more restricted under chilling stress, re

Chromic oxide nanoparticles (Cr2O3 NPs) are widely used in commercial factories and can cause serious environmental problems. However, the mechanism behind Cr2O3 NP-induced phytotoxicity remains unknown. In this study, the effects of Cr2O3 NPs on the growth, chlorophyll fluorescence, SEM-EDS analysis, and chloroplast ultrastructure of soybean (Glycine max) were investigated to evaluate its phytotoxicity. The growth of soybean treated with various Cr2O3 NP suspensions (0.01, 0.05, 0.1, and 0.5 g L-1) was significantly inhibited. Specially, shoot and root biomass decreased by 9.9 and 46.3%, resp

Isoprene synthase (ISPS) catalyzes the elimination of pyrophosphate from dimethylallyl diphosphate (DMADP) forming isoprene, a volatile hydrocarbon emitted from many plant species to the atmosphere. In the present work, immunological techniques were applied to study and localize ISPS in poplar leaves (Populus x canescens). Immunogold labeling using polyclonal antibodies generated against His-tagged recombinant ISPS protein detected ca. 44% of ISPS in the stroma of the chloroplasts and ca. 56% of gold particles attached to the stromal-facing side of the thylakoid membranes. ISPS isolated from l

The effect of daily heat-shock treatments on gene expression and morphogenesis of etiolated barley (Hordeum vulgare) was investigated. Heat-shock treatments in the dark induced shortening of the primary leaves and the coleoptiles to the length of those in light-grown plantlets. In addition, the mRNA levels of the light-induced genes that were investigated were raised under these conditions and showed distinct oscillations over a period of at least 3 d. While the mRNA levels for chlorophyll a/b binding protein (LHC II), plastocyanin, and the small subunit of ribulose-1,5-bisphosphate carboxylas

A newly observed general chlorophyll fluorescence induction effect in plants is described. Fluorescence yield can rise through as many as four different phases (alpha, beta, gamma, ) in the dark, when intact cells or leaves are rapidly heated (within approx. 2.5 s) from 20 to 40-50 degrees C. An analysis of this temperature-jump fluorescence induction in Scenedesmus obliquus leads to the following: 1. Phase alpha is due to removal of S-quenching and appears to be related to heat deactivation of the water-splitting enzyme system. With prolonged heating, irreversibility of alpha upon recooling r

Copper metallochaperones supply copper to cupro-proteins through copper-mediated protein-protein-interactions and it has been hypothesized that metallochaperones thereby inhibit copper from causing damage en route. Evidence is presented in support of this latter role for cyanobacterial metallochaperone, Atx1. In cyanobacteria Atx1 contributes towards the supply of copper to plastocyanin inside thylakoids but it is shown here that in copper-replete medium, copper can reach plastocyanin without Atx1. Unlike metallochaperone-independent copper-supply to superoxide dismutase in eukaryotes, glutath

INTRODUCTION: Drought stress adversely affects photosynthesis, redox homeostasis and metabolic signalling in plants. Exogenous application of hormonal and nonhormonal elicitors, have emerged as promising strategies to improve plant resilience to changing climatic conditions.
METHODS: In the current study, the effect of foliar application of gibberellic acid (GA3) and 6-benzylaminopurine (6-BAP) individually and in combination was assessed at different doses (0.2 and 0.4 mM) in pigeonpea (Cajanus cajan) under drought stress through integrated physiological, metabolomic and transcriptomic appro

Citrus sinensis seedlings were irrigated with nutrient solution at a concentration of 0 (Mg-deficiency) or 2 (Mg-sufficiency) mM Mg (NO3)2 for 16 weeks. Mg-deficiency-induced interveinal chlorosis, vein enlargement and corkiness, and alterations of gas exchange, pigments, chlorophyll a fluorescence (OJIP) transients and related parameters were observed in middle and lower leaves, especially in the latter, but not in upper leaves. Mg-deficiency might impair the whole photosynthetic electron transport, including structural damage to thylakoids, ungrouping of photosystem II (PSII), inactivation o

The effects of ongoing anthropogenic climate change are not well known in marine diatoms, a key group of primary producers. Specifically, detailed characterizations of their carbon-concentrating mechanisms (CCMs) are lacking, which limits the understanding of how changing ocean carbonate chemistry will impact global primary production. While the model diatom Thalassiosira pseudonana has been widely studied, contrasting results have prevented the clear elucidation of its CCM. A quantitative proteomic analysis was therefore performed across three experimental treatments (low pCO2/high pH, high p

Antibiotic cocontamination poses a severe but poorly understood threat to crop photosynthesis. This study employed an integrated multiscale framework to uncover the phytotoxicity of tetracycline (TC) and ciprofloxacin (CIP) on rice seedlings and evaluate the mitigation potential of nanobiochar (nBC). Coexposure to TC and CIP amplified antibiotic residues in both root and shoot, exacerbating growth inhibition and photosynthetic pigment depletion, with the net photosynthetic rate plummeting to 1.44 μmol CO2 m-2 s-1. Mechanically, chlorophyll fluorescence and transmission electron microscopy reve

Ferredoxin5 (FDX5), a minor ferredoxin protein in the alga Chlamydomonas (Chlamydomonas reinhardtii), helps maintain thylakoid membrane integrity in the dark. Sulfur (S) deprivation has been used to achieve prolonged hydrogen production in green algae. Here, we propose that FDX5 is involved in algal responses to S-deprivation as well as to the dark. Specifically, we tested the role of FDX5 in both the initial aerobic and subsequent anaerobic phases of S-deprivation. Under S-deprived conditions, absence of FDX5 causes a distinct delay in achieving anoxia by affecting photosynthetic O2 evolution

A novel kind of lutein-protein complex (LPC) was extracted from heterotrophic Chlorella vulgaris through aqueous extraction. The purification procedure contained solubilization of thylakoid proteins by a zwitterionic detergent CHAPS, anion exchange chromatography and gel filtration chromatography. Both wavelength scanning and HPLC analysis confirmed that lutein was the major pigment of the protein-based complex, and the mass ratio of lutein and protein was determined to be 9.72 : 100. Besides showing lipid peroxidation inhibition activity in vitro, LPC exerted significant antioxidant effects a

We report for the first time significant changes in the P680*+ reduction kinetics of Photosystem II (PS II) in which the 17 and 23 kDa extrinsic polypeptides are intact, in the presence of Ca(2+) or ethylene glycol bis (beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) which were added to vary the Ca(2+) concentration from 5 microM to 30 mM. The decrease in the extent of normal P680*+ reduction decay with lifetimes of 40-370 ns and a corresponding increase in the extent of kinetics with lifetimes of 20-220 micros was interpreted as being due to electron transfer from Y(Z) to P680*+ bein

Microcystis aeruginosa (M. aeruginosa) blooms threaten aquatic ecosystems and drinking water safety. Algicidal bacteria offer an eco-friendly approach to control Microcystis blooms, yet few strains can simultaneously inhibit algae growth and reduce microcystin production. Herein, we isolated an efficient algicidal bacterium Pseudomonas fulva EH5 and explored its inhibitory effects and underlying mechanisms against M. aeruginosa. Strain EH5 exerted prominent algicidal activity mainly via secreted active substances, with a 97.33% inhibition rate at a 4% inoculation ratio on day 5. EH5 displayed
