Microcomputed Tomography Mineral Density Profile as Reference Standard for Early Carious Lesion Activity Assessment.

Early caries diagnosis is crucial to treatment decisions in dentistry and requires identification of lesion activity: whether a carious lesion is active (progressively demineralizing) or arrested (progressively remineralizing). This study aimed to identify microtomographic (micro-CT) differences between active and arrested smooth surface enamel lesions, to quantify those micro-CT differences by creating thresholds for ex vivo caries activity assessment to serve as a future reference standard, and to validate those thresholds against the remaining sample. Extracted human permanent teeth (n = 59

Influence of two dentin bonding systems on the demineralization of the root surface.

It has been assumed that dentin adhesives can prevent root surface caries. The aim of this study was to determine the caries-protective effect of two different dentin bonding systems on the demineralization of root surfaces in vitro. The root surfaces of 60 freshly extracted caries-free human molars were thoroughly cleaned and polished, thereby removing the cementum. The teeth were then coated with acid-resistant nail varnish, exposing two rectangular windows of 6 mm2 each. One window served as an untreated control, while the other window was treated with a dentin bonding system. The specimens

An in-vitro model for tooth eruption utilizing periodontal ligament fibroblasts and collagen lattices.

An in-vitro model that simulates tooth eruption was devised to determine the possible role of periodontal ligament fibroblasts (PLF) in providing a major force for eruption. The model consisted of agarose-coated wells in multiwell tissue-culture plates to which wire mesh was attached at the rim and a slice of demineralized root was attached to the floor. When the wells were filled with a collagen lattice containing PLF, the lattice contracted and elevated the root slice. The root slice in models that were unattached at the rim was not elevated. Cultures without cells did not contract and root

Formulation of Tooth Mousse Containing Grape Seed Extract and Comparative Evaluation of Its Remineralizing Effect with GC Tooth Mousse: An In Vitro Study.

BACKGROUND: Dental caries is one of the most prevalent chronic infectious diseases, with its incidence increasing significantly over time. The global prevalence rate of dental caries in primary teeth stands at approximately 46.2% (Furiga et al.). To combat this widespread issue, remineralizing agents play a crucial role in reducing the prevalence rate, helping to restore and strengthen the demineralized tooth surface. OBJECTIVES: The aim of this in vitro study is to formulate the grape seed extract (GSE)-containing tooth mousse and to compare the remineralizing property with commercially avai

The Influence of Different Preservation Protocols on the Teeth's Osteoinductive Characteristics: An In Vitro Study.

The purpose of this study was to evaluate in vitro whether the type of tooth preservation before treatment with the Tooth Transformer® (TT) device affects the osteoinductive characteristics of the extracted tooth. Forty extracted teeth from healthy non-smoking patients were selected. All teeth were cleaned of caries, tartar, and filling material and then roughly sectioned and divided into four experimental groups according to storage type: room-temperature (RT) tooth samples, frozen tooth samples, RIPA tooth samples, and fresh tooth samples. Each sample was minced, demineralized, and disinfect

A Remineralizing and Antibacterial Coating for Arresting Caries.

Dental caries is a dynamic disease induced by the unbalance between demineralization of dental hard tissues caused by biofilm and remineralization of them; however, although various effective remineralization methods have been well documented, it is a challenge to reestablish the balance by enhancing remineralization alone while ignoring the antibacterial therapy. Therefore, the integration of remineralizing and antibacterial technologies offers a promising strategy to halt natural caries progression in clinical practice. Here, the conception of interrupting dental caries (IDC) was proposed ba

Biomimetic remineralization of acid etched enamel using agarose hydrogel model.

Background: Minimally invasive dentistry aims to prevent progression of caries and treats non-cavitated lesions through non-invasive approaches to preserve the integrity of tooth structure. The aim of this research was to investigate the possible biomimetic effect of agarose hydrogel in remineralizing a human demineralized enamel model. Methods: Mandibular third molars were distributed into three groups (G1, G2 and G3) according to the follow up time (2, 4 and 6 days respectively). Caries like lesion was prepared by applying 37% phosphoric acid gel for 1 minute and then remineralization was pe

Chapter 5.1: Physicochemical Interactions between Enamel and Oral Fluids.

Despite all the current knowledge in cariology, research is still being carried out nowadays trying to make dental enamel resistant to dental caries. Since enamel is mainly composed by a mineral, efforts have been put together to make it more resistant to acids produced by dental biofilm when exposed to dietary sugars. Fluoride was once thought to be a micronutrient that impacted caries resistance when incorporated in the tooth mineral, but we now know that the complex interactions at the mineral surface are most important. Every slightly soluble mineral, and enamel is no exception, has a beha

Failure of a glass ionomer to remineralize apatite-depleted dentin.

Remineralization of demineralized dentin lesions adjacent to glass-ionomer cements (GICs) has been reported in the literature. This study tested the hypothesis that a strontium-based GIC can remineralize completely demineralized dentin by nucleation of new apatite crystallites within an apatite-free dentin matrix. Human dentin specimens were acid-etched, bonded with Fuji IX(GP), and immersed in a calcium-and-phosphate-containing 1.5X simulated body fluid (SBF) for 1-4 months. Polyacrylic acid and polyvinylphosphonic acid biomimetic analogs were added to the SBFs to create 2 additional reminera

Characterization of the effects of 3DSS peptide on remineralized enamel in artificial saliva.

Aspartate-serine-serine (DSS) repeats are abundant in naturally occurring proteins that are critical for tooth formation. In this study, we reported a possible role of triplet repeats of aspartate-serine-serine (3DSS) peptides in promoting mineral deposition onto human enamel from free ions. Human enamel specimens were acid demineralized, exposed briefly to 3DSS peptide solution, and then immersed in artificial saliva. At various stages of treatments, nanomechanical behaviors, surface morphology, surface roughness and the sorts of deposited minerals were characterized by nanoindentation, scann

[The effect of gastric juice on the development of erosive changes in hard dental tissue].

INTRODUCTION: Gastroesophageal reflux disease (GERD) is an esophageal disorder where the refluxed gastric contents enters first into the esophagus followed by the pharynx, oral cavity, larynx, airway and middle ear, causing a range of disorders and symptoms. Hydrochloric acid from the gastric contents is responsible for the demineralization of dental hard tissues and release of matrix metalloproteinase from the dentin. OBJECTIVE: The aim of this study was to verify the SEM (scanning electron microscopy) analysis of the surface enamel, the enamel-dentin border and dentine after the exposure of

Nursing-bottle caries: the importance of a development perspective.

Early diagnosis of nursing-bottle caries is so difficult because its first stage is difficult to detect, and occurs at an age the child does not visit a dentist regularly. Also, the first stage does not appear alarming. If for the parent serious complaints from the child arise, irreversible defects already have developed. The developmental stages of nursing-bottle caries are clearly linked to the eruption pattern of the primary teeth. The maxillary primary incisors are affected first, followed by the maxillary first molars. In all stages, a stabilized condition, arrested caries, can occur, if

Effects of free and nano-encapsulated bovine lactoferrin on the viability and acid production by Streptococcus mutans biofilms.

Tooth decay is one of the most common diseases that humans face during their lifetime. Streptococcus mutans is one of the most critical factors of early tooth decay. These bacteria may produce biofilms in the mouth, which are more resistant to antimicrobial agents. Streptococcus mutans may also demineralize tooth enamel by producing lactic acid. In this study, the effect of nanoliposomes containing lactoferrin at three different concentrations (1·5, 3 and 6 mg ml-1 ) on the biofilm formed by S. mutans was investigated. Nanoliposomes were prepared using the thin-layer hydration method. An activ

Structural changes in the dental and periodontal tissues of the rat incisor following application of orthodontic loads.

The present study deals with the morphometric changes in the loaded rat incisor and its supporting structures 3 months after load removal. The material examined was limited to that part of the tooth that is located between 8 and 20 mm from the tooth apex. Of the ten albino rats in whom laterointrusive loads (19 +/- 0.6 g) were applied to the lower left incisor for a period of 2 weeks, the rate of tooth eruption returned to normal in four animals (group A); in six rats, eruption remained grossly impaired (group B). These two groups were compared with a control group of four rats in whom the inc

Comparison of Biofilm Accumulation on Conventional and CAD/CAM Orthodontic Band Alloys (In Vivo) and Subsequent Enamel Demineralization (Ex Vivo).

Biofilm accumulation can lead to enamel decalcification, gingivitis, and periodontal disease. The objective of this study was to compare the accumulation of biofilm under in vivo conditions and consequent ex vivo acid production and enamel demineralization around the material used for "off-the-shelf" conventional and CAD/CAM orthodontics bands. The study design required both in vivo and in vitro approaches. An experimental model was utilized to combine the exposure of an in vivo formed biofilm to in vitro cariogenic conditions to achieve the objective. Twenty-one consenting participants took p
