Molecular epidemiology of norovirus infection among children with acute gastroenteritis in Shanghai, China, 2001-2005.

Norovirus is one of the major causes of outbreaks and sporadic cases of acute gastroenteritis in young children worldwide. Obtaining local baseline information regarding this virus is important for developing and evaluating prevention strategies of norovirus transmission in children. The age, seasonal distribution and circulating genotypes of norovirus in Shanghai, China, between 2001 and 2005 were determined. Of 5411 stool specimens collected from children under 5 years of age who were hospitalized with acute gastroenteritis 3,975 were rotavirus-negative, indicating the presence of another ca

Antigenic variant of swine influenza virus causing proliferative and necrotizing pneumonia in pigs.

A new antigenic variant of swine influenza virus was isolated from the lungs of pigs experiencing respiratory problems in 7 different swine herds in Quebec. Pigs of different ages were affected, and the main clinical signs were fever, dyspnea, and abdominal respiration. Coughing was not a constant finding of the syndrome. At necropsy, macroscopic lesions included the overall appearance of pale animals, general lymphadenopathy, hepatic congestion, and consolidation of the lungs. Histopathologic findings were mainly proliferative pneumonia with a significant macrophage invasion, necrotic inflamm

Octyl gallate targeting the 3C-like protease exhibits highly efficient antiviral activity against swine enteric coronavirus PEDV.

Infection with porcine epidemic diarrhea virus (PEDV) causes severe watery diarrhea in newborn piglets, leading to substantial financial losses for the swine industry. In this study, we screened small molecule drugs targeting 3 C-like protease (3CLpro) by molecular docking, and further evaluated the antiviral activity of the screened drugs against PEDV. Results showed that octyl gallate (OG), a widely used food additive, exhibited strong binding affinity with the 3CLpro active sites of PEDV. Bio-layer interferometry and fluorescence resonance energy transfer revealed that OG directly interacts

Fe-Co bimetallic nanozyme-amplified lateral-flow immunoassay for the sensitive detection of porcine rotavirus.

Porcine rotavirus (PoRV) causes severe viral diarrhea in piglets. Owing to the lack of specific antiviral therapies, rapid on-site detection is urgently required for swine disease control. Herein, a lateral-flow immunoassay (LFIA) amplified by Fe-Co bimetallic nanozyme was established for PoRV detection using two screened high-affinity anti-PoRV monoclonal antibodies. The peroxidase-like catalytic activity of Fe-Co nanozyme was employed for signal amplification on the immunochromatographic strip. The limit of detection of this LFIA was 100 TCID50/mL, which was two-fold more sensitive than conv

Survey for selected pathogens in wild pigs (Sus scrofa) from Guam, Marianna Islands, USA.

Pigs (Sus scrofa) were introduced to Guam in the 1600's and are now present in high densities throughout the island. Wild pigs are reservoirs for pathogens of concern to domestic animals and humans. Exposure to porcine parvovirus, transmissible gastroenteritis, and Leptospira interrogans has been documented in domestic swine but data from wild pigs are lacking. The close proximity of humans, domestic animals, and wild pigs, combined with the liberal hunting of wild pigs, results in frequent opportunities for pathogen transmission. From February-March 2015, blood, tissue and ectoparasite sample

A trivalent inactivated PDCoV-PEDV-TGEV vaccine confers protective efficacy in piglets coinfected with porcine rotavirus.

Porcine epidemic diarrhea virus (PEDV), porcine deltacoronavirus (PDCoV), and transmissible gastroenteritis virus (TGEV) are the primary enteric coronaviruses responsible for viral diarrhea in piglets. These pathogens frequently co-circulate with porcine rotavirus (PoRV) under field conditions, leading to increased disease severity and complicating prevention efforts. Although multivalent vaccines targeting these three coronaviruses have been developed, their protective efficacy in the presence of PoRV coinfection remains largely unknown. Based on our previously developed trivalent inactivated

CdSe/ZnS quantum dot-labeled antibody fluorescent immunoassay strip for swine acute diarrhea syndrome coronavirus S1 protein detection.

Swine acute diarrhea syndrome coronavirus (SADS-CoV), a porcine enteropathogenic coronavirus, causes acute diarrhea, vomiting, and death in newborn piglets, leading to economic losses in the pig industry. Previous studies have shown that SADS-CoV can infect a variety of mammalian cell lines, including human-derived cells, indicating its potential public health risk of cross-species transmission. In this study, we successfully expressed the SADS-CoV S1 protein using a eukaryotic expression system and screened monoclonal antibodies 1B8 and 3C9. Antibody 1B8 was used as a detection antibody coupl

Comparative in vivo analysis of the nsp15 endoribonuclease of murine, porcine and severe acute respiratory syndrome coronaviruses.

The purpose of this study was to compare the biochemical and biological properties of nonstructural protein (nsp) 15 among mouse hepatitis virus (MHV), severe acute respiratory syndrome coronavirus (SARS-CoV) and transmissible gastroenteritis virus (TGEV) in virus-infected and ectopically expressed cells. In virus-infected cells, MHV nsp15 distributed unevenly throughout the cytoplasm but predominantly in the perinuclear region. When expressed as N-terminal enhanced green fluorescence protein (EGFP) fusion, it predominantly formed speckles in the cytoplasm. In contrast, SARS-CoV and TGEV EGFP-

Pathological and immunohistochemical assessment of the impact of three different strains of swine enteric coronaviruses in the intestinal barrier.

Swine enteric coronaviruses, such as porcine epidemic diarrhea virus (PEDV) or transmissible gastroenteritis virus (TGEV), have risen concern for the porcine industry and research community due to the increase in their virulence, their potential recombination capacity and the emergence of new variants. This in vivo study aims to compare the impact of three different strains of swine enteric coronaviruses [(two G1b (S-INDEL) PEDV strains and a recombinant TGEV-PEDV or Swine enteric coronavirus (SeCoV)] in the intestine of 3-weeks-old infected piglets, focusing on the pathology and main componen

L-Leucine Promotes STAT1 and ISGs Expression in TGEV-Infected IPEC-J2 Cells via mTOR Activation.

L-leucine (Leu), as one of the effective amino acids to activate the mTOR signaling pathway, can alleviate transmissible gastroenteritis virus (TGEV) infection. However, the underlying mechanism by which Leu alleviates the virus infection has not been fully characterized. In particular, how Leu impacts TGEV replication through mTOR signaling has yet to be elucidated. In the present study, we found that TGEV proliferated efficiently in intestinal porcine epithelial cells (IPEC-J2 cells) as evidenced by the increase in viral contents by flow cytometry, the inhibition of cell proliferation by CCK

Epidemiology of calicivirus infections in Sweden, 1994-1998.

Outbreaks of acute gastroenteritis are frequently caused by caliciviruses. Electron microscopy was used to search for these viruses in fecal samples from patients with acute gastroenteritis. Of 5800 samples collected and analyzed from November 1994 to June 1998, 3700 were associated with outbreaks. A total of 676 outbreaks were analyzed, and viruses were found in 67%. Caliciviruses, usually Norwalk-like viruses (NLVs), were found in 407 (89%) of 455 outbreaks, while Sapporo-like viruses were identified in nine outbreaks, including six that were suspected to include foodborne transmission. Development and Application of a Reverse-Transcription Recombinase-Aided Amplification Assay for Porcine Epidemic Diarrhea Virus.

Porcine epidemic diarrhea virus (PEDV) is a coronavirus currently widespread worldwide in the swine industry. Since PEDV was discovered in China in 1984, it has caused huge economic losses in the swine industry. PEDV can infect pigs of all ages, but piglets have the highest infection with a death rate as high as 100%, and the clinical symptoms are watery diarrhea, vomiting, and dehydration. At present, there is not any report on PEDV detection by RT-RAA. In this study, we developed an isothermal amplification technology by using reverse-transcription recombinase-aided amplification assay (RT-R

FUT8-mediated core fucosylation of receptor APN drives entry of multiple alphacoronaviruses.

Understanding the interaction mechanisms between coronaviruses (CoVs) and their hosts is crucial for understanding the viral replication cycle and identifying novel antiviral targets. In this study, we found alpha-(1,6)-fucosyltransferase (FUT8), via its fucosyltransferase activity, is involved in the several alphacoronaviruses (α-CoVs) spike-receptor aminopeptidase N (APN) interaction to regulate viral entry. Mechanistically, pAPN lacking FUT8-mediated modification showed no binding to the transmissible gastroenteritis virus (TGEV) RBD. The viral entry depends on core fucosylation at pAPN N73

The essential role of heparan sulfate in the entry of PDCoV and other porcine coronaviruses.

Porcine enteric coronaviruses, including porcine deltacoronavirus (PDCoV), porcine epidemic diarrhea virus (PEDV), swine acute diarrhea syndrome coronavirus (SADS-CoV), and transmissible gastroenteritis coronavirus (TGEV), can cause acute diarrhea, vomiting, dehydration, and high mortality in suckling piglets. Recent studies revealing human PDCoV infections and the potential of SADS-CoV to penetrate human cell lines have heightened apprehensions about the zoonotic transmission risks of these viruses. While heparan sulfate (HS) serves as a receptor in PDCoV binding, the key host genes involved

Porcine Respiratory Coronavirus as a Model for Acute Respiratory Coronavirus Disease.

In the light of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, we have developed a porcine respiratory coronavirus (PRCV) model for in depth mechanistic evaluation of the pathogenesis, virology and immune responses of this important family of viruses. Pigs are a large animal with similar physiology and immunology to humans and are a natural host for PRCV. Four PRCV strains were investigated and shown to induce different degrees of lung pathology. Importantly, although all four strains replicated equally well in porcine cell lines in vitro and in the upper respirator
