Hematological, biochemical, and serological parameters of experimentally infected rabbits with Trichinella nativa and Trichinella spiralis for early identification of trichinellosis.

BACKGROUND AND AIM: Trichinellosis remains a dangerous disease for humans and animals, which can lead to a lethal outcome. The study of specific body reactions in response to invasion by different types of Trichinella can help in the early diagnosis of the disease. This study aimed to investigate the hematological, biochemical, and serological characteristics of rabbits experimentally infected with trichinellosis, as well as the possibility of using changes in these parameters at various disease stages for early hematological, biochemical, and serological diagnosis of trichinellosis. The immune protection induced by a serine protease from the Trichinella spiralis adult against Trichinella spiralis infection in pigs.

Trichinellosis is a major foodborne parasitosis caused by Trichinella spiralis. In the present study, a serine protease gene from an adult T. spiralis (Ts-Adsp) cDNA library was cloned, expressed in Escherichia coli and purified by Ni-affinity chromatography. Previous studies of our laboratory have found that mice vaccinated with recombinant Ts-Adsp protein (rTs-Adsp) exhibited partial protection against T. spiralis infection. In this study, the protective effect of rTs-Adsp against T. spiralis infection in pigs was further explored. The cell-mediated and humoral immune responses induced by rT

Matrix metalloproteinase (MMP)-2 and MMP-9 as inflammation markers of Trichinella spiralis and Trichinella pseudospiralis infections in mice.

Trichinella spiralis and Trichinella pseudospiralis exhibit differences in the host-parasite relationship such as the inflammatory response in parasitized muscles. Several studies indicate that matrix metalloproteinases (MMPs) represent a marker of inflammation since they regulate inflammation and immunity. The aim of this study was to evaluate the serum levels of gelatinases (MMP-9 and MMP-2) in mice experimentally infected with T. spiralis or T. pseudospiralis, to elucidate the involvement of these molecules during the inflammatory response to these parasites. Gelatin zymography on SDS polya

Trichinella nelsoni and Trichinella T8 mixed infection in a lion (Panthera leo) of the Kruger National Park (South Africa).

In South Africa, Trichinella sp. was first discovered in 1966 in the wildlife of the Kruger National Park (KNP). Since then, both Trichinella T8 and Trichinella nelsoni have been detected in the KNP, leading to a debate on the existence of a gene flow between the two taxa. In 2006-2008, four lions were killed in the Manyeleti Game Reserve, the Mthethomusha Nature Reserve, Numbi Gate, and Skukuza, which border the KNP. Larvae were isolated from muscles by artificial digestion. The molecular identification of single larva by multiplex PCR, followed by a specific PCR to distinguish between Trichi

Trichinella infection in a hunting population of Papua New Guinea suggests an ancient relationship between Trichinella and human beings.

A new Trichinella species, Trichinella papuae, was discovered in 11.5% of wild pigs in a remote region of Papua New Guinea. A survey was conducted to determine whether the inhabitants of this region are infected with Trichinella, as wild-pig meat represents their main source of protein. The prevalence of anti-Trichinella antibodies and its determinants were assessed among the inhabitants in 51 of the villages in the Morehead District of Papua New Guinea. We tested and interviewed 1536 people (28.8% of the adult population). Anti-Trichinella IgG were detected in 10.0% (95% Cl 8.5-11.6%) of them

Mitochondrial ATP-ASE as a measure of uncoupling of rat muscle mitochondria in experimental infection with Trichinella spiralis and Trichinella pseudospiralis.

Changes in a bioenergetic state of Trichinella spiralis and Trichinella pseudospiralis infected rat and mouse muscle mitochondria were evaluated enzymatically, and in both infections 3-4-fold increase of mitochondrial, Mg++-stimulated ATP-ase (EC 3.6.1.3) was observed. Looking for the dynamics of those bioenergetic changes in T. pseudospiralis infected rat and mouse muscle mitochondria 1-2 weeks, pi, the 5-6-fold stimulation of mATP-ase activity, followed by a significant drop between the week 3-4th was found. In in vitro experiments cytoplasmic fractions isolated from T. spiralis and T. pseud

New records of Trichinella patagoniensis from Argentina.

Trichinella spiralis was long considered the sole Trichinella species in Argentina. However, since 2004, various Trichinella species, including the encapsulated Trichinella patagoniensis and Trichinella britovi, as well as the unencapsulated Trichinella pseudospiralis, have been detected in the country. The present study aimed to identify Trichinella ML at the species level from cougars naturally infected from Argentina. To this end, muscle tissue samples from one cougar each from Córdoba, Neuquén, and Santa Cruz Provinces were individually analysed using the artificial digestion technique. DN

Trichinella spiralis immunomodulation: an interactive multifactorial process.

Many epidemiological data support the postulate that infection with helminths might provide some protection against allergic and autoimmune diseases. Hence arises the concept that helminths strongly influence the immune system and enable protective pathways against these hyperimmune-associated disorders. This review discusses how Trichinella spiralis can make the immune system smarter in dealing with hyperimmune-associated disorders. T. spiralis possesses the capacity to direct the immune system towards a mixed Th1/Th2 phenotype with predominance of Th2 response, or it may interfere with dendr

Nematode infections: soil-transmitted helminths and trichinella.

Infection with soil-transmitted helminths occurs via ingestion of nematode eggs with contaminated food and water, via hands, or inhalation of dust, or by penetration of larvae through the skin. Trichinella infections are caused by the ingestion of larvae contained in undercooked meat. In highly endemic areas, preventive chemotherapy (ie, regular administration of anthelmintic drugs to at-risk populations) is the key strategy against soil-transmitted helminthiasis. Integrated control approaches, including improved hygiene, sanitation, and water, are required for lasting effects. Because of grow

Trichinella spiralis shares epitopes with human autoantigens.

Like other helminths, Trichinella spiralis has evolved strategies to allow it to survive in the host organism, including the expression of epitopes similar to those present in either expressed or hidden host antigens. To identify T. spiralis-derived antigens that are evolutionarily conserved in the parasite and its host and that could be responsible for its evasion of the host immune response, we examined the reactivity of six different types of autoantibodies to T. spiralis larvae from muscle. T. spiralis antigens that share epitopes with human autoantigens were identified by assessing the cr

Trichinella spiralis--a potential anti-tumor agent.

Murine forestomach carcinoma (cell line MFC), ascitic hepatoma (cell line H22) and sarcoma (cell line S180) solid tumor models were used to test the anti-tumor effect of Trichinella spiralis in vivo. Mice previously infected by oral administration of 400 viable T. spiralis larvae per mouse for 7 days were grafted with various solid tumor cell lines. Other groups of tumor-bearing mice were given caudal vein injection of crude extracts of adult and newborn larvae at 17.5, 35.0 or 70.0 mg kg(-1). These treatments to inhibit tumor growth were dose-dependent (p<0.05). The anti-proliferative activit

Trichinella sp. in wolves from interior Alaska.

Tongue samples were collected from 148 wolf (Canis lupus) carcasses during 1993 and 1994 near Fairbanks (Alaska, USA). A standard peptic digestion procedure was used to detect Trichinella sp. larvae. Larvae were found in 54 of 148 (36%) samples. There was no significant difference in sex-specific prevalence. Prevalence was significantly related to age. There was no relationship between the number of larvae/g of host tissue and the age or sex of the host. Trichinella spp. infection may cause illness in individual wolves. However, there was no indication the parasite had any impact on the popula
