Comparative anatomy of cranial nerve motor nuclei: with a comment on evolution of the craniofacial region.

Changes of cranial nerve nuclei are investigated at three crucial points of phylogenesis in frogs, lizards and rats. Profound changes can be observed in the trigeminal and facial motor nuclei parallel to transformation of the primary mandibular joint into the secondary mandibular joint. New muscles appear for mastication and facial expression and they are innervated by new types of neurons. The muscular tongue appears first in tetrapods coinciding with terrestrial life. It can be shown that an entirely new type of neuron develops at the medullospinal junction for the innervation of the intrins

A dynamic regulation of GDNF-family receptors correlates with a specific trophic dependency of cranial motor neuron subpopulations during development.

Glial cell line-derived neurotrophic factor (GDNF) family ligands promote the survival of developing motor neurons in vivo and in vitro. However, not all neurons survive with any single ligand in culture and GDNF null mutant mice display only a partial motor neuron loss. An interesting possibility is that subpopulations of motor neurons based on their function and/or their myotopic organization require distinct members of GDNF family ligands. Because responsiveness to the different ligands depends on the expression of their cognate ligand-binding receptor we have herein addressed this issue by

Role of inhibitory mechanisms in trigeminal neuralgia.

Segmental inhibition was elicited in the spinal trigeminal nucleus of cats by delivering a conditioning stimulus to the maxillary nerve 100 msec before the test stimulus. Carbamazepine, baclofen, and phenytoin markedly facilitated this segmental inhibition, as well as depressing the response to an unconditioned maxillary nerve stimulus. Phenobarbital, on the other hand, usually depressed the segmental inhibition. These results suggest that drugs that relieve trigeminal neuralgia both facilitate inhibitory mechanisms and depress excitatory mechanisms in the spinal trigeminal nucleus. The facili

Baclofen in trigeminal neuralgia: its effect on the spinal trigeminal nucleus: a pilot study.

Experiments with cats showed that baclofen resembles carbamazepine and phenytoin sodium in its ability to depress excitatory synaptic transmission in the spinal trigeminal nucleus. Baclofen was, therefore, given to 14 patients with refractory trigeminal neuralgia. Ten patients were relieved of the paroxysms of tic douloureux while taking 60 to 80 mg/day of baclofen. A reduction in the dosage of baclofen in six of these patients resulted in a recurrence of painful paroxysms in five patients. Seven patients have been pain-free or almost pain-free on a regimen of baclofen for four to 12 months. O

Neurotoxic effects of p-chloroamphetamine on the serotoninergic innervation of the trigeminal motor nucleus: a retrograde transport study.

The rat forebrain receives projections from both dorsal and median raphe nuclei. It has recently been shown that serotoninergic axons arising from the dorsal raphe nucleus, but not those from the median raphe nucleus, degenerate following systemic administration of p-chloroamphetamine (PCA). The present study was conducted to determine (i) whether the motor nucleus of the trigeminal nerve is innervated by overlapping projections from multiple serotonin cell groups and (ii) whether a particular subset of serotoninergic axon terminals in the trigeminal motor nucleus are sensitive to the neurotox

Evaluation of Masseteric Vestibular Evoked Myogenic Potentials in Patients With Migraine.

PURPOSE: Masseter vestibular evoked myogenic potentials (mVEMP) involve the connection between the vestibular complex and trigeminal nerve nuclei. Given the theory that migraine is caused by increased activation of the trigeminal nerve, it is believed that mVEMP responses may have influenced in migraine patients. METHOD: The study included 20 individuals with migraine and 20 healthy controls. Latency, amplitude, and interaural amplitude asymmetry ratio of mVEMP responses recorded in migraine patients were compared with control group. RESULTS: Considering the mVEMP normalization study conduct

Effects of lesions to the trigeminal motor nucleus on temporomandibular disc morphology.

Stereotaxic surgery was performed on the trigeminal motor nuclei (TMNu) of 47 actively growing rats in order to test the effects of neuromuscular alterations on TMJ disc morphology. Lesioned animals received a small electrolytic lesion to their left-side TMNu, while shams had their TMNu stimulated without an actual lesion being produced. Both lesioned and sham-lesioned animals demonstrated significant (P less than 0.05; P less than 0.01) morphological alterations of their discs 28, 56, and 84 days postoperatively. Data indicated that (i) TMNu lesions do indeed affect disc morphology, and (ii)

Deafferentation pain after posterior rhizotomy, trauma to a limb, and herpes zoster.

After incisional or alcoholic destruction of trigeminal posterior rootlets, constant dysesthesias of major degree referred to some part of the markedly denervated zone develop in 5 to 15% of the patients. The full severity may not appear for weeks or months. There is no allodynia or hyperpathia of the denervated zone. Bulbar trigeminal tractotomy with sparing of touch sensation produces severe dysesthesias in a tiny percentage of the patients, as does selective destruction of pain fibers by radiofrequency heating or glycerol. Spinal posterior rhizotomy elicits in less than 4% a lasting dysesth

Trigeminal neuralgia as a presentation of high cervical stenosis: a systematic review of the literature.

PURPOSE: An atypical presentation of cervical spondylopathy (CS), trigeminal neuralgia (TN) is attributable to the extension of trigeminal nuclei into the spinal cord and is frequently overlooked, leading to limited discussion with patients regarding potential anterior cervical surgery. Our systematic review assesses the effectiveness of cervical surgery for concurrent trigeminal neuralgia in cases of cervical spondylopathy. METHODS: A systematic review exploring cases of trigeminal neuralgia related to cervical spondylopathy was conducted searching on PubMed, Scopus and Embase databases for

Functional anatomy of the vagus system: How does the polyvagal theory comply?

Due to its pivotal role in autonomic networks and interoception, the vagus attracts continued interest from both basic scientists and therapists of various clinical disciplines. In particular, the widespread use of heart rate variability as an index of autonomic cardiac control and a proposed central role of the vagus in biopsychological concepts, e.g., the polyvagal theory, provide a good opportunity to recall basic features of vagal anatomy. In addition to the "classical" vagal brainstem nuclei, i.e., dorsal motor nucleus, nucleus ambiguus and nucleus tractus solitarii, the spinal trigeminal

Functional anatomy of the vagus system - Emphasis on the somato-visceral interface.

Due to its pivotal role in autonomic networks, the vagus attracts continuous interest from both basic scientists and clinicians. In particular, recent advances in vagus nerve stimulation strategies and their application to pathological conditions beyond epilepsy provide a good opportunity to recall basic features of vagal peripheral and central anatomy. In addition to the "classical" vagal brainstem nuclei, i.e., dorsal motor nucleus, nucleus ambiguus and nucleus tractus solitarii, the spinal trigeminal and paratrigeminal nuclei come into play as targets of vagal afferents. Brain-Derived Neurotrophic Factor Systemic Response in the Periodontium, Trigeminal Nucleus Caudalis, and Hippocampus Induced by Occlusal Trauma.

Occlusal trauma (OT), by causing periodontal tissue damage, can activate and enhance the activity of the peripheral and central nervous system (CNS) neuropeptides. The brain-derived neurotrophic factor (BDNF) gene is activity-dependent and exhibits marked alterations, characterized by protection against injury and repair. Our results show the possible molecular mechanism through which noxious environmental stimuli induce alterations in BDNF activity in the local periodontal tissue, the primary sensory neurons-Vc, and the hippocampus, suggesting systemic impairment. BDNF serves a more positive

Glycine receptor distribution in mouse CNS: autoradiographic localization of [3H]strychnine binding sites.

Biochemical and electrophysiological studies of mammalian CNS indicate that the amino-acid, glycine, is a major inhibitory neurotransmitter whose location is, for the large part, confined to the spinal cord and brain stem. In this study, autoradiographs of C57BL/6J mouse brain sections labeled with [3H] strychnine, a potent antagonist of glycine, were used to map the distribution of glycine receptors in the CNS. Autoradiographs showed highly localized areas of grain density, which confirmed the gross distributions described in homogenate binding studies and gave a more precise regional localiz

The reticular premotor neurons of the jaw muscle motor nuclei in the mallard (Anas platyrhynchos L.).

The trigeminal and facial motor nuclei in the mallard comprise several subnuclei which innervate tongue, jaw and other head muscles. The premotor cells of the subnuclei innervating jaw muscles are distributed in two longitudinal cell columns within the parvocellular reticular formation (RPc). The ventromedial part of RPc contains cells projecting to subnuclei innervating either jaw-closer muscles or jaw-opener muscles. In the dorsolateral part of RPc three subdivisions may be recognized: a rostral part which projects to two trigeminal subnuclei innervating jaw-closer muscles, an intermediate p

Projections from the dorsal peduncular cortex to the trigeminal subnucleus caudalis (medullary dorsal horn) and other lower brainstem areas in rats.

This study has revealed direct projections from the dorsal peduncular cortex (DP) in the medial prefrontal cortex (mPfC) to the trigeminal brainstem sensory nuclear complex and other lower brainstem areas in rats. We first examined the distribution of mPfC neurons projecting directly to the medullary dorsal horn (trigeminal subnucleus caudalis [Vc]) and trigeminal subnucleus oralis (Vo) which are known to receive direct projections from the lateral prefrontal cortex (insular cortex). After injections of the retrograde tracer Fluorogold (FG) into the rostro-dorsomedial part of laminae I/II of V

Trigeminal sensory symptoms due to midbrain lesions.

Three patients with midbrain lesion are described who showed trigeminal sensory symptoms in the contralateral side. Two with ischemic stroke showed partial oculomotor paresis while the third with probable vascular malformation had trochlear palsy. Considering their clinical manifestations and magnetic resonance findings, their sensory changes are believed to be caused by the involvement of the crossed trigeminothalamic fibers near the ocular motor nuclei.

The somatostatin systems of the guinea-pig brainstem.

The tetradecapeptide somatostatin has been shown to have a widespread distribution in the rat brain. Except for its role in the inhibition of growth hormone secretion, the function of this molecule in the remainder of the central nervous system is unknown. To address this problem, the distribution of somatostatin-like immunoreactivity in the guinea-pig brainstem was examined systematically. Of 116 nuclei and/or areas, 34 nuclei had somatostatin neurons, 32 did not have any immunoreactivity and the remainder had immunoreactive fibers and/or terminals. Cranial nerve motor nuclei--somatic, branch

Nasal trigeminal inputs release the A5 inhibition received by the respiratory rhythm generator of the mouse neonate.

Experiments were performed on neonatal mice to analyze why, in vitro, the respiratory rhythm generator (RRG) was silent and how it could be activated. We demonstrated that in vitro the RRG in intact brain stems is silenced by a powerful inhibition arising from the pontine A5 neurons through medullary alpha(2) adrenoceptors and that in vivo nasal trigeminal inputs facilitate the RRG as nasal continuous positive airway pressure increases the breathing frequency, whereas nasal occlusion and nasal afferent anesthesia depress it. Because nasal trigeminal afferents project to the A5 nuclei, we appli

Induction of facial muscle neurotization by temporalis muscle transposition: literature review and animal model evaluation using horseradish peroxidase uptake.

OBJECTIVE: To study the concept of facial muscle reinnervation from the trigeminal pathway following facial nerve paralysis. DESIGN AND METHODS: We studied this phenomenon in an animal model using the neuronal marker, horseradish peroxidase (HRP). The temporalis transposition procedure was performed at varying intervals post facial nerve transection. To evaluate the trigeminal-facial reinnervation process and its timing, the zygomaticus major muscle was injected with HRP at varied periods after temporalis transposition, and histologic sections of the brainstem nuclei were examined for the fin
