Results of follow-up of human contacts of bovine tuberculosis in cattle during 1993-7 in North Staffordshire.

The purpose of the study was to describe the results of follow up of human contacts of bovine tuberculosis. The bovine tuberculosis cases occurred on farms in North Staffordshire between 1993 and 1997. A total of 162 people were identified as having close contact with cattle diagnosed as having bovine tuberculosis, or who had drunk unpasteurized milk from a herd with bovine tuberculosis. A retrospective review of chest clinic notes was performed. One hundred and thirty-eight people attended for follow up, and Heaf test results, necessity for chest X-ray and further clinical follow-up are descr

An outbreak of bovine tuberculosis in two herds in south west Scotland--veterinary and human public health response.

BACKGROUND: Although the incidence of bovine tuberculosis in UK cattle has declined markedly, outbreaks still occur. There is often confusion as to how veterinary and human public health agencies should interact in such circumstances. We report an outbreak of bovine tuberculosis which occurred in South West Scotland in 1992. METHODS: The outbreak, which affected two cattle herds, and potentially involved 17 humans, was discovered when visible tuberculous lesions were found at routine post mortem examination of a calf. Investigation of animals and humans, exchange of information between human

Bovine tuberculosis resulting from infection with Mycobacterium orygis in a closed herd of Indian water buffalo (Bubalus bubalis).

Mycobacterium orygis (M. orygis), a member of the Mycobacterium tuberculosis complex, has emerged as an important cause of animal and human tuberculosis in South Asia. Despite the widespread prevalence of bovine tuberculosis (bTB) in India and the major role of over 100 million Indian water buffaloes in dairy production, the pathological and epidemiological features of M. orygis infection in buffaloes are largely unexplored. A closed herd of 279 female water buffaloes in Haryana, India, was screened for bTB by single intradermal cervical tuberculin skin (SCT) test, the comparative cervical tub

Deep Learning-Enabled Engineering of a Hyper-Stable and Soluble MPB70-83 Antigen for Sensitive Bovine Tuberculosis Surveillance.

Bovine tuberculosis (bTB) diagnostics are frequently hindered by the structural instability and insolubility of recombinant multi-epitope proteins in prokaryotic expression systems. To address this, we developed an integrated strategy combining immunoinformatics with the ProteinMPNN deep learning framework. We constructed a fusion protein utilizing B-cell epitopes from MPB70 and MPB83, where ProteinMPNN was employed to redesign non-epitope scaffolds for optimized thermodynamic stability. The structural integrity was verified via 200-ns molecular dynamics (MD) simulations. MD analysis revealed

KLK12 Regulates MMP-1 and MMP-9 via Bradykinin Receptors: Biomarkers for Differentiating Latent and Active Bovine Tuberculosis.

It has been established that kallikrein12 (KLK12) expression is closely related to bovine tuberculosis (bTB) development. Herein, we sought to clarify the regulatory mechanism of KLK12 and its application in tuberculosis diagnosis. KLK12 knockdown macrophages were produced by siRNA transfection. Bradykinin receptors (BR, including B1R and B2R) were blocked with specific inhibitors. Mannose-capped lipoarabinomannan (ManLAM) was extracted from Mycobacterium bovis (M. bovis) and used to study the mechanism of KLK12 activation. In addition, we constructed different mouse models representing the la

Replication and transmission features of two experimental vaccine candidates against bovine tuberculosis subcutaneously administrated in a murine model.

Cattle vaccination is an attractive approach in compliance with control and eradication programs against Bovine Tuberculosis (bTB). Today, there is no anti bTB vaccine licensed. Two vaccine candidates, MbΔmce2 and MbΔmce2-phoP previously designed were evaluated in BALB/c mice, including the parental M. bovis NCTC10772 and a M. bovis hypervirulent Mb04-303 strains as controls. Sentinel mice (non-inoculated) cohoused with subcutaneous inoculated mice. Persistence, visible tuberculosis lesions (VTL) in lungs and spleens and bacillary load were investigated subcutaneously delivered at 60 and 90 da

Polyfunctionality of CD4+ T lymphocytes in buffaloes and cattle: comparative antigen-specific cytokine responses in bovine tuberculosis infection.

INTRODUCTION: Bovine tuberculosis (BTB), caused by Mycobacterium bovis, is a chronic infectious disease of major veterinary and public health concern. It affects a broad range of domestic and wild animals, including water buffalo, and poses a risk to humans due to its zoonotic nature. The economic consequences of BTB, arising from production losses and trade restrictions, further underline its global importance. While cattle immune responses to BTB are well characterized, the immune mechanisms in buffalo remain poorly understood, despite their increasing role as livestock in endemic regions. G

Development of an improved ESAT-6 and CFP-10 peptide-based cytokine flow cytometric assay for bovine tuberculosis.

Control of bovine tuberculosis (bTB) continues to be a problem world-wide because of difficulties in identifying infected animals at all stages of infection. The use of the IFN-γ release assays (IGRA) as an ancillary test with the tuberculin skin tests has improved the ability to identify infected animals. However, infected animals may still be missed. The objective of the present study was to evaluate a rapid flow-cytometric assay based on intracellular cytokine staining as an alternative to the in vitro IFN-γ release assay (IGRA). Antigen-specific cells producing IFN-γ were identified after

Comparative performance of tuberculin and defined-antigen cocktails for detecting bovine tuberculosis in BCG-vaccinated cattle in natural settings.

Bovine tuberculosis (bTB) is a threat to cattle health and public safety. The current control programs are hampered by wildlife reservoirs and socioeconomic barriers. Vaccinating cattle with Bacillus Calmette-Guérin (BCG) effectively reduces transmission, offering a potential solution for controlling bTB. A key requirement for vaccination strategies using BCG is the validation of defined antigens to differentiate infections among vaccinated animals (DIVA). We compared tuberculin with DIVA peptide cocktails (ESAT-6, CFP-10, and Rv3615c) in 67 unvaccinated and 67 BCG-vaccinated cattle exposed to

Re-Evaluation of ELISA for the Detection of Bovine Tuberculosis and a New Proposal for Its Use in Eradication Efforts on Outbreak Farms.

Bovine tuberculosis (bTB) is a zoonotic infectious disease and a chronic wasting illness. Accordingly, detecting and eradicating bTB remains a significant challenge in South Korea. This study evaluated the efficacy of a modified enzyme-linked immunosorbent assay (ELISA) protocol for detecting bTB in cattle. The protocol included two ELISA tests: one performed on the day of purified protein derivative (PPD) inoculation and another seven days post-inoculation. Results show a significant increase in ELISA detection rates, from 11% to 76%, particularly in cattle that tested positive for the tuberc

Shaken, not stirred: magnetic bead DNA extraction as a rapid and effective method for the scaling up of bovine tuberculosis diagnosis.

BACKGROUND: The growing use of real-time PCR (qPCR) as a diagnostic method for bovine TB (bTB) requires rapid and effective DNA extraction methods, which are crucial for its success. Automated DNA extraction methods based on magnetic beads are a promising alternative to conventional silica column-based protocols (COL protocol) due to their high throughput capacity and reduced hands-on time. This study aimed to assess the performance of the MagMax CORE Nucleic Acid Purification kit and the KingFisher Flex instrument (KF protocol) as an alternative for scaling up the use of qPCR in bTB diagnosis

Estimation of sensitivity and specificity of bacteriology, histopathology and PCR for the confirmatory diagnosis of bovine tuberculosis using latent class analysis.

Bacteriology and histopathology are the most commonly used tests used for official confirmatory diagnosis of bovine tuberculosis (bTB) in cattle in most countries. PCR is also being used increasingly because it allows a fast diagnosis. This test could be applied as a supplement to or replacement for current bTB confirmatory diagnostic tests but its characteristics have first to be evaluated. The aim of this study was to estimate and compare sensitivities and specificities of bacteriology, histopathology and PCR under French field conditions, in the absence of a gold standard using latent class

Identification of New Mycobacterium bovis antigens and development of a multiplexed serological bead-immunoassay for the diagnosis of bovine tuberculosis in cattle.

Serological assays for bovine tuberculosis diagnosis require the use of multiple Mycobacterium bovis specific antigens to ensure the detection of infected animals. In the present study, identification and selection process of antigens, based on data from published proteomic studies and involving the use of bioinformatics tools and an immuno-screening step, was firstly performed for identifying novel antigens that elicit an antibody response in M. bovis infection. Based on this approach, a panel of 10 M. bovis antigens [with known relevance (MPB70, MPB83, MPB70/83, and ESAT6/CFP10) and novel (M

Development and evaluation of a Mycobacterium bovis interferon-γ enzyme-linked immunospot (ELISpot) assay for detection of bovine tuberculosis.

Bovine tuberculosis (bTB) caused by Mycobacterium bovis is an important zoonotic disease. This infection is difficult to control because of the limited ability of the tuberculin skin test (TST) and ancillary IFN-γ release assay to detect all infected animals. In this study, we aimed to develop an efficient assay based on the enzyme-linked immunospot (ELISpot) technique for the diagnosis of bTB, with IFN-γ monoclonal antibodies 3E9 and Bio-labeled 6F8 used as capture and detection antibodies, respectively. As expected, there were significantly more M. bovis-specific spot-forming units (SFU) in

Risk, knowledge and preventive measures of smallholder dairy farmers in northern Malawi with regard to zoonotic brucellosis and bovine tuberculosis.

Milk production using local cattle breed-types is an age-old practice in Malawi. Although dairy farming is becoming more common as a result of the increasing population and demand for milk and milk products, there is limited knowledge of the farmers' awareness of zoonotic disease risks, their preventative practices and the disease burden in animals. This study determined dairy farmers' general knowledge of zoonoses, assessed their risks for infection with zoonotic bovine tuberculosis (bTB) and brucellosis, and evaluated farm practices to prevent disease transmission. A questionnaire was drawn

Field-oriented assessment of bovine tuberculosis in Tunisian cattle: IDR, PCRMpb70 and serological test prediction based on AI approaches.

Bovine tuberculosis (bTB), caused by Mycobacterium bovis (M. bovis), remains a major zoonotic and economically burdensome disease worldwide. In Tunisia, where bTB has remained present for many years, Efforts to eliminate the disease have been slowed down by limited diagnostic tools and a lack of resources. The tuberculin Intra Dermal Reaction test (IDR), also known as Tuberculin Skin Test (TST), is still the main tool for surveillance, but its low sensitivity and specificity show the need for better diagnostic methods. This study evaluated the diagnostic performance of IDR, PCRMpb70 and three
