Putative type IV secretion genes in Bacillus anthracis.

Although the physiology of Bacillus anthracis, the causative agent of anthrax, has been studied extensively, we still do not know how toxins are dispatched from the bacterial cell. Here, by means of distant homology and genome context analyses, we identify genes encoding putative type IV secretion system-related elements on the B. anthracis plasmids pXO1 and pXO2 and in the chromosome. We argue that this type IV secretion system-like system could be responsible for anthrax toxin secretion, although we also discuss the possibilities of its involvement in the processes of sporulation, germinatio

The intracellular agent of Q fever, Coxiella burnetii, alters human alveolar macrophage metabolism and mitochondrial physiology.

Coxiella burnetii, the etiologic agent of Q fever, is a gram-negative intracellular bacterium that infects humans via contaminated aerosols typically while working with livestock. C. burnetii initially targets alveolar macrophages (AMs) to establish a growth niche within a phagolysosome-like compartment termed the Coxiella-containing vacuole (CCV). C. burnetii deploys a type IV secretion system (T4SS) to secrete effector proteins that control host cell functions to benefit the bacterium, orchestrating an immunosuppressive, pro-bacterial environment for replication to high numbers. Although mul

A Brucella Type IV Effector Targets the COG Tethering Complex to Remodel Host Secretory Traffic and Promote Intracellular Replication.

Many intracellular pathogens exploit host secretory trafficking to support their intracellular cycle, but knowledge of these pathogenic processes is limited. The bacterium Brucella abortus uses a type IV secretion system (VirB T4SS) to generate a replication-permissive Brucella-containing vacuole (rBCV) derived from the host ER, a process that requires host early secretory trafficking. Here we show that the VirB T4SS effector BspB contributes to rBCV biogenesis and Brucella replication by interacting with the conserved oligomeric Golgi (COG) tethering complex, a major coordinator of Golgi vesi

Coxiella burnetii as a useful tool to investigate bacteria-friendly host cell compartments.

Coxiella burnetii is an obligate intracellular and airborne pathogen which can cause the zoonotic disease Q fever. After inhalation of contaminated aerosols alveolar macrophages are taking up C. burnetii into a phagosome. This phagosome matures to a very large vacuole called the C. burnetii-containing vacuole (CCV). Host endogenous and bacterial driven processes lead to the biogenesis of this unusual compartment, which resembles partially a phagolysosome. However, there are several important differences to the classical phagolysosome, which are crucial for the ability of C. burnetii to replica

Regulation of virulence in Brucella: an eclectic repertoire of transcription factors defines the complex architecture of the virB promoter.

Many intracellular bacterial pathogens use type IV secretion systems to deliver effector molecules and subvert the eukaryotic host cell defenses. The genus Brucella comprises facultative intracellular bacteria that cause brucellosis, a disease affecting a wide range of mammals including humans. The virB operon codes for a type IV secretion system that plays a central role in intracellular survival and replication of Brucella within the host. Expression of the virB genes is under the control of various transcription factors that allow this system to respond to different types of environmental s

Description of Bartonella bennetti sp. nov., a novel rodent-associated species, with comparative genomics of the Bartonella genus.

The genus Bartonella comprises over 40 species, most of which are haemoparasites of mammals. Herein, we describe Bartonella bennetti sp. nov., a novel member of the genus, isolated from field voles (Microtus agrestis) in Kielder Forest, UK. Polyphasic characterization of three strains (C271T, D105 and J117) of the proposed species indicated that they were closely related to members of phylogenetic lineage 3 (L3) of the genus. The average nucleotide identity (ANI) between C271T and other L3 species ranged between 88.8 and 90.6%, supporting the proposal of a new species. C271T shared ANIs approa

IcmF and DotU are required for optimal effector translocation and trafficking of the Legionella pneumophila vacuole.

The gram-negative bacterium Legionella pneumophila causes a severe form of pneumonia called Legionnaires' disease, characterized by bacterial replication within alveolar macrophages. Prior to intracellular replication, the vacuole harboring the bacterium must first escape trafficking to the host lysosome, a process that is dependent on the Dot/Icm type IV secretion system. To identify genes required for intracellular growth, bacterial mutants were isolated that were delayed in escape from the macrophage but which retain a minimally functional Dot/Icm machinery. The mutations were found in eigh

Fatal attraction of mammalian cells to Legionella pneumophila.

Legionella pneumophila is a protozoan parasite that causes Legionnaires' disease. Its ability to do so is dependent on its capacity to replicate intracellularly within a phagosome that is not trafficked through the endosomal-lysosomal pathway and is surrounded by the rough endoplasmic reticulum. Within this unique niche, the bacterium undergoes alterations in gene expression. In addition, many virulence-related phenotypes that are induced in vitro by starvation are expressed intracellularly as the bacteria exit the logarithmic growth phase. (p)ppGpp appears to signal expression of the virulenc

[Molecular bases for brucella virulence].

The authors review published reports on the molecular bases of Brucella virulence, including type IV secretion proteins, S-lipopolysaccharide biosynthesis enzymes (O-antigen), regulatory proteins of various systems, and cellular metabolism proteins. High efficiency of modified transposon mutagenesis technique (selective labeled transposon mutagenesis) in search for virulence genes is shown. Analysis of DNA sequences of Brucella genome promotes identification of new virulence factors.

A comprehensive two-hybrid analysis to explore the Legionella pneumophila effector-effector interactome.

UNLABELLED: Legionella pneumophila uses over 300 translocated effector proteins to rewire host cells during infection and create a replicative niche for intracellular growth. To date, several studies have identified L. pneumophila effectors that indirectly and directly regulate the activity of other effectors, providing an additional layer of regulatory complexity. Among these are "metaeffectors," a special class of effectors that regulate the activity of other effectors once inside the host. A defining feature of metaeffectors is direct, physical interaction with a target effector. Metaeffect

Genomic analysis of Neisseria gonorrhoeae strains from disseminated, urogenital, and rectal infections reveals differences in genes for iron acquisition and type IV secretion.

Neisseria gonorrhoeae, an obligate human pathogen, causes the sexually transmitted infection gonorrhea. Delayed treatment or immunodeficiencies can result in a higher risk of disseminated gonococcal infection (DGI), where the infection spreads to normally sterile anatomic sites, including the bloodstream. The gonococcus produces TonB-dependent transporters (TdTs) to sequester metals from host nutritional immunity proteins. These transporters are critical for survival and are important virulence factors. We characterized genes encoding iron-regulated TdTs in the genomes of strains from dissemin

Despite nearly a century of therapy for gonococcal infection with a variety of antimicrobials, more than 80 million cases of the disease are reported annually worldwide. The gonorrhea pathogen, Neisseria gonorrhoeae, exhibits an exceptional capability of developing antimicrobial resistance due to its high genetic flexibility. As an obligate pathogen, the gonococcus has evolved mechanisms to evade host defenses by engaging with the innate and adaptive immune responses in both men and women. N. gonorrhoeae can establish residence within epithelial cells, macrophages, and neutrophils. Strains res

Innate immune responses and monocyte-derived phagocyte recruitment in protective immunity to pathogenic bacteria: insights from Legionella pneumophila.

Legionella species are Gram-negative intracellular bacteria that evolved in soil and freshwater environments, where they infect and replicate within various unicellular protozoa. The primary virulence factor of Legionella is the expression of a type IV secretion system (T4SS), which contributes to the translocation of effector proteins that subvert biological processes of the host cells. Because of its evolution in unicellular organisms, T4SS effector proteins are not adapted to subvert specific mammalian signaling pathways and immunity. Consequently, Legionella pneumophila has emerged as an i

Comparative proteome analysis of laboratory grown Brucella abortus 2308 and Brucella melitensis 16M.

Brucella species are pathogenic agents that cause brucellosis, a debilitating zoonotic disease that affects a large variety of domesticated animals and humans. Brucella melitensis and Brucella abortus are considered major health threats because of their highly infectious nature and worldwide occurrence. The availability of the annotated genomes for these two species has allowed a comparative proteomics study of laboratory grown B. melitensis 16M and B. abortus 2308 by two-dimensional (2-D) gel electrophoresis and peptide mass fingerprinting. [Molecular basis of Brucella persistence].

This review focuses on Brucella persistence. Data on Brucella--macrophage interaction and the role of molecular-genetic systems including homologues of Mos operon Rhizobium, a type IV secretion system, a flagellum apparatus and a "quorum sensing"--virulence factors using signal or effector molecules are updated. Brucella enters macrophages through lipid raft microdomains, avoids its bactericidal attacks, phagolysosome fusion, expressing a set of virulence genes and inhibits TNF-alpha secretion and apoptosis for persistence in macroorganisms. Comparative whole-genome microarray analyses reveal

Comparative analyses of Legionella species identifies genetic features of strains causing Legionnaires' disease.

BACKGROUND: The genus Legionella comprises over 60 species. However, L. pneumophila and L. longbeachae alone cause over 95% of Legionnaires’ disease. To identify the genetic bases underlying the different capacities to cause disease we sequenced and compared the genomes of L. micdadei, L. hackeliae and L. fallonii (LLAP10), which are all rarely isolated from humans. RESULTS: We show that these Legionella species possess different virulence capacities in amoeba and macrophages, correlating with their occurrence in humans. Our comparative analysis of 11 Legionella genomes belonging to five spec

Identification of a type IV secretion substrate of Brucella abortus that participates in the early stages of intracellular survival.

Brucella abortus, the aetiological agent of bovine brucellosis, is an intracellular pathogen whose virulence is completely dependent on a type IV secretion system. This secretion system translocates effector proteins into the host cell to modulate the intracellular fate of the bacterium in order to establish a secure niche were it actively replicates. Although much has been done in understanding how this secretion system participates in the virulence process, few effector proteins have been identified to date. We describe here the identification of a type IV secretion substrate (SepA) that is

Brucellosis and type IV secretion.

Brucellosis is a global disease of domestic and wild mammals that is caused by intracellular bacteria of the genus Brucella. Although humans are not a natural reservoir for Brucella, infection in the human population is common in many countries, and brucellosis is one of the most common zoonotic infections. Brucella species have evolved to avoid the host's immune system and infection is usually characterized by long-term persistence of the bacteria. One important Brucella virulence factor for intracellular survival and persistence in the host is the type IV secretion system. This review will d

Coxiella burnetii manipulates the lysosomal protease cathepsin B to facilitate intracellular success.

The obligate intracellular bacterium Coxiella burnetii establishes an intracellular replicative niche termed the Coxiella-containing vacuole (CCV), which has been characterised as a bacterially modified phagolysosome. How C. burnetii withstands the acidic and degradative properties of this compartment is not well understood. We demonstrate that the key lysosomal protease cathepsin B is actively and selectively removed from C. burnetii-infected cells through a mechanism involving the Dot/Icm type IV-B secretion system effector CvpB. Overexpression of cathepsin B leads to defects in CCV biogenes
