Epidemiology of neuropathic pain: an analysis of prevalence and associated factors in UK Biobank.

INTRODUCTION:: Previous epidemiological studies of neuropathic pain have reported a range of prevalences and factors associated with the disorder. OBJECTIVES:: This study aimed to verify these characteristics in a large UK cohort. METHODS:: A cross-sectional analysis was conducted of 148,828 UK Biobank participants who completed a detailed questionnaire on chronic pain. The Douleur Neuropathique en Quatre Questions (DN4) was used to distinguish between neuropathic pain (NeuP) and non-neuropathic pain (non-NeuP) in participants with pain of at least 3 months' duration. Participants were also

Incident Clinical and Mortality Associations of Myocardial Native T1 in the UK Biobank.

BACKGROUND: Cardiac magnetic resonance native T1-mapping provides noninvasive, quantitative, and contrast-free myocardial characterization. However, its predictive value in population cohorts has not been studied. OBJECTIVES: The associations of native T1 with incident events were evaluated in 42,308 UK Biobank participants over 3.17 ± 1.53 years of prospective follow-up. METHODS: Native T1-mapping was performed in 1 midventricular short-axis slice using the Shortened Modified Look-Locker Inversion recovery technique (WIP780B) in 1.5-T scanners (Siemens Healthcare). Global myocardial T1 was

Bilirubin as an indicator of cardiometabolic health: a cross-sectional analysis in the UK Biobank.

BACKGROUND: Mildly elevated bilirubin, a by-product of hemoglobin breakdown, might mitigate cardiometabolic risk factors including adiposity, dyslipidemia, and high blood pressure (BP). We investigated the cross-sectional relationship between (total) bilirubin and baseline cardiometabolic risk factors in 467,519 UK Biobank study participants. METHODS: We used multivariable-adjusted linear regression to estimate associations between bilirubin levels and risk factors of cardiometabolic diseases including body mass index (BMI), waist and hip circumferences (WC, HC), waist-to-hip ratio (WHR), fat

SARS-CoV-2 is associated with changes in brain structure in UK Biobank.

There is strong evidence for brain-related abnormalities in COVID-19 1-13 . It remains unknown however whether the impact of SARS-CoV-2 infection can be detected in milder cases, and whether this can reveal possible mechanisms contributing to brain pathology. Here, we investigated brain changes in 785 UK Biobank participants (aged 51-81) imaged twice, including 401 cases who tested positive for infection with SARS-CoV-2 between their two scans, with 141 days on average separating their diagnosis and second scan, and 384 controls. The availability of pre-infection imaging data reduces the likel

Association of Diligence and Sociability with Stroke: A UK Biobank Study on Personality Proxies.

BACKGROUND: There is a growing interest in how personality may be related to the risk of developing disease. Associations between personality and stroke have so far only been studied in relation to stroke mortality. However, many stroke survivors suffer severe impairment of quality of life due to sequelae such as aphasia, hemiparesis, depression and anxiety. In this study we assess the association between personality and risk of stroke, regardless of mortality. METHODS: Using self-reported data on psychological factors, mental health and social support, proxies for the Big Five personality tr

Detection and characterization of male sex chromosome abnormalities in the UK Biobank study.

PURPOSE: The study aimed to systematically ascertain male sex chromosome abnormalities, 47,XXY (Klinefelter syndrome [KS]) and 47,XYY, and characterize their risks of adverse health outcomes. METHODS: We analyzed genotyping array or exome sequence data in 207,067 men of European ancestry aged 40 to 70 years from the UK Biobank and related these to extensive routine health record data. RESULTS: Only 49 of 213 (23%) of men whom we identified with KS and only 1 of 143 (0.7%) with 47,XYY had a diagnosis of abnormal karyotype on their medical records or self-report. We observed expected associati

Migraine and gastrointestinal disorders in middle and old age: A UK Biobank study.

INTRODUCTION: Migraine is a prevalent condition causing a substantial level of disability worldwide. Despite this, the pathophysiological mechanisms are not fully understood. Migraine often co-occurs with gastrointestinal disorders, but the direction of a potential causal link is unclear. The aim of this project was to investigate the associations between migraine and several gastrointestinal disorders in the same cohort in order to determine the relative strengths of these associations. METHODS: This cross-sectional study examined whether migraine is associated with irritable bowel syndrome

Insights into the aetiology of snoring from observational and genetic investigations in the UK Biobank.

Although snoring is common in the general population, its aetiology has been largely understudied. Here we report a genetic study on snoring (n ~ 408,000; snorers ~ 152,000) using data from the UK Biobank. We identify 42 genome-wide significant loci, with an SNP-based heritability estimate of ~10% on the liability scale. Genetic correlations with body mass index, alcohol intake, smoking, schizophrenia, anorexia nervosa and neuroticism are observed. Gene-based associations identify 173 genes, including DLEU7, MSRB3 and POC5, highlighting genes expressed in the brain, cerebellum, lungs, blood an

Polygenic risk for schizophrenia and subcortical brain anatomy in the UK Biobank cohort.

Research has shown differences in subcortical brain volumes between participants with schizophrenia and healthy controls. However, none of these differences have been found to associate with schizophrenia polygenic risk. Here, in a large sample (n = 14,701) of unaffected participants from the UK Biobank, we test whether schizophrenia polygenic risk scores (PRS) limited to specific gene-sets predict subcortical brain volumes. We compare associations with schizophrenia PRS at the whole genome level ('genomic', including all SNPs associated with the disorder at a p-value threshold < 0.05) with 'g

Predictors of mosaic chromosome Y loss and associations with mortality in the UK Biobank.

Mosaic loss of the Y chromosome (mLOY) is the most commonly reported large structural somatic event. Previous studies have indicated age and cigarette smoking increase the risk of mLOY, but the relationship of other exposures with mLOY and mLOY with disease has not been adequately investigated. We characterized mLOY in a large cohort of 223,338 men from the UK Biobank by scanning for deviations in genotyping array median log2 intensity ratios (mLRR) of the Y chromosome using a standard algorithm. A total of 3,789 (1.7%) men showed evidence for mLOY (mLRR < -0.15). In multivariable-adjusted log

Защитная роль аполипопротеина А и В в болезни Паркинсона: перспективное исследование из UK Biobank.

Periodontitis and adverse outcomes in atrial fibrillation: prospective evidence from the UK Biobank.

BACKGROUND: Periodontitis (PD), a prevalent chronic inflammatory disease, has been implicated in the pathogenesis of various cardiovascular conditions. Although prior evidence suggests an association between PD and incident atrial fibrillation (AF), its influence on the clinical outcomes in patients with established AF remains poorly understood. OBJECTIVE: This prospective cohort study aimed to explore the associations of PD with all-cause mortality, cardiovascular mortality, incident heart failure (HF) and incident stroke among individuals with AF based on the data from the UK Biobank. Detecting scoliosis at scale using automated Cobb angle analysis in the UK Biobank.

PURPOSE: Adult degenerative scoliosis arises after skeletal maturity in an initially normal spine, primarily driven by age-related degeneration. The Cobb angle, the angle between the most tilted vertebrae typically derived from radiographs, remains the clinical standard for assessing curvature severity, yet large-scale evaluation using MRI has not been feasible. This study developed an automated method for Cobb angle estimation from chemical-shift-based water-fat separation (Dixon) MRI and applied it to the UK Biobank to characterise the prevalence and curvature within the general population.

Factors Associated With Fibromyalgia Diagnosis Among People Meeting Criteria: Results From UK Biobank.

OBJECTIVE: The diagnosis of fibromyalgia (FM) is challenging due to the absence of definitive biomarkers, numerous overlapping comorbidities, and its reliance on patient-reported symptoms. Discrepancies between diagnostic criteria and clinical practice imply the possibility of diagnostic biases, complicating timely and accurate identification. This study aimed to identify factors associated with receiving an FM diagnosis among individuals who met 2016 criteria and explore potential barriers to diagnosis. METHODS: Using data from the UK Biobank, we analyzed a total of 27 potential factors acro
