Profound increase of Helicobacter pylori urease activity in gastric antral mucosa at low pH.

The effect of pH on H. pylori urease activity in its ecological niche was studied in gastric antral biopsy specimens. Specimens were incubated in 10 mmol/liter urea solutions at pH range 3.3-8.2. Activity of urease was studied by measuring production of ammonia and change in pH of the solutions. Urease activity was reduced at pH 8.2 (1424 +/- 218 mumol/liter) but decreasing initial pH to neutral and acidic values resulted in significant maximal 6.5-fold increase in ammonia production (9491 +/- 1073 mumol/liter, P < 0.0005), which considerably raised the pH of the test solutions. Peak urease ac

Urease catalyzed high-density sodium alginate microspheres enable high oral bioavailability of macromolecular drugs.

Destruction of insulin caused by the gastric microenvironment and rapid deactivation pose inevitable barriers to oral macromolecular absorption, especially for most peptide and protein drugs. In this study, we developed high-density sodium alginate microspheres composed of magnesium oxide and urease to address these challenges. These microspheres aim to anchor the gastric mucus layer and induce microenvironmental liquefaction, thereby enhancing gastric retention and the protection of insulin. The sedimentation test confirmed the capability of the Ins/Ur/MgO@SA microsphere to rapidly traverse t

Co-encapsulated urease and glucose oxidase in polyelectrolyte microcapsules for simultaneous glucose and urea determination.

BACKGROUND: Clinical monitoring of metabolic disorders requires stable tools for simultaneous analyte detection. However, conventional free-enzyme assays suffer from rapid degradation, poor operational stability, and single-use limitations. Herein, we report a novel bi-enzymatic diagnostic system based on the rational co-encapsulation of glucose oxidase (GOx) and urease within polyelectrolyte microcapsules (PMCs) for the simultaneous determination of glucose and urea. RESULTS: Fabricated via layer-by-layer assembly, the PMCs achieved high enzyme loading (73 ± 2% for GOx and 93 ± 5% for urease

Structural insights into how metallochaperones UreE and UreG interact to deliver a toxic metal to urease.

Maturation of urease, a virulence factor for Helicobacter pylori infection, requires the delivery of nickel ions to the metalloenzyme. To avoid cytotoxicity, Ni(II) ions are transferred within protein complexes of metallochaperones to ensure that the toxic metal does not escape into the cytoplasm. In the urease maturation pathway, metallochaperone UreG receives its Ni(II) ions by forming a 2:2 complex with another metallochaperone UreE. Using C-terminal truncation variants of UreE [UreE(1-158) and UreE(1-148)], we determined the crystal structures of the UreE2G2 complex bound with Ni(II) ions

Synthesis and Evaluation of Isatin Analogs as Potential Urease and Tyrosinase Inhibitors: An Approach of Molecular Docking.

A series of isatin-based Schiff base derivatives (1-12) was synthesized via a two-step reaction and characterized using spectroscopic techniques such as 1H-NMR and mass spectrometry. The urease and tyrosinase inhibitory activities of the synthesized compounds were evaluated using thiourea (IC50 = 21.25 ± 0.15 µM) and kojic acid (IC50 = 121 ± 0.5 µM) as standard inhibitors. Among the synthesized analogs, only three compounds-1 (IC50 = 38.9 ± 0.06 µM), 3 (IC50 = 56.7 ± 0.02 µM), and 10 (IC50 = 71 ± 0.09 µM) showed moderate urease inhibition, while the remaining compounds were inactive. In Silico and In Vitro Elucidation of the Inhibitory Mechanism of Chlorogenic Acid Against Helicobacter pylori Urease.

Chlorogenic acid (CGA), a phenolic compound widely distributed in plants, has exhibited inhibitory activity against Helicobacter pylori (H. pylori) by effectively suppressing urease, a key virulence factor of the pathogen. This study aimed to investigate the binding affinity between CGA and urease and explore the underlying molecular mechanism. Inhibition kinetics were characterized using Lineweaver-Burk plots, fluorescence quenching, isothermal titration calorimetry (ITC), and surface plasmon resonance (SPR). The binding mode was elucidated using molecular docking and molecular dynamics simul

Staphylococcus aureus urease is controlled by a complex regulatory network to promote dissemination during CAUTI.

UNLABELLED: Catheter-associated urinary tract infections (CAUTIs) are one of the most common hospital-associated infections in the United States, accounting for >1 million cases annually. One CAUTI pathogen, Staphylococcus aureus, is commonly found persisting asymptomatically in the bladder of catheterized individuals, increasing these individuals' risk of developing infection. Importantly, S. aureus is not only associated with severe symptoms during CAUTI, including bacteremia and septic shock, but it also produces a common uropathogen-associated virulence factor, urease. Despite its importan

Synthesis and evaluation of novel ebselen derivatives as urease inhibitors for combating Helicobacter pylori infections.

The development of novel inhibitors targeting urease that affects the colonization of Helicobacter pylori (H. pylori) is a promising strategy to address the growing challenge of antibiotic resistance. In this study, a novel urease covalent inhibitor, XBP2 (IC50 = 0.14 ± 0.01 μM and MIC = 8 μg/mL), was identified through structure-based design using Ebselen as a template. XBP2 binds to a newly identified site, forming stable dipole interaction with residues ASP223 and HIS322, which enhances both stability and inhibitory activity against urease. It exhibits potent in vitro antibacterial activity

Rational design and in vitro testing of new urease inhibitors to prevent urinary catheter blockage.

Catheter associated urinary tract infections (CAUTI) caused by urease-positive organisms can lead to catheter blockage: urease metabolizes urea in urine to ammonia causing an increase in pH and hence precipitation of struvite and apatite salts into the catheter lumen and bladder leading to blockage. Acetohydroxamic acid (AHA) is the only urease inhibitor currently approved for patient use, however, it is rarely used owing to its side effects. Here, we report the identification and development of new urease inhibitors discovered using a rational in silico drug design approach. A series of compo

Therapeutic potential of N4-substituted thiosemicarbazones as new urease inhibitors: Biochemical and in silico approach.

Urease enzyme plays a key role in pathogenesis of gastritis and peptic ulcers. Its inhibition averts our bodies from many disorders including formation of urinary calculi. In agriculture, the high urease content causes severe environmental and hence economic problems. Due to deficiency of effective and safer drugs to tackle the aforementioned disorders, the quest for new scaffolds becomes mandatory in the field of medicinal chemistry. In this regard, we herein report a new series of N4-substituted thiosemicarbazones 3a-v as potential candidates for urease inhibition. These new N4-substituted t

Utility of anthranilic acid and diethylacetylenedicarboxylate for the synthesis of nitrogenous organo/organometallic compounds as urease inhibitors.

Fumarate diester 3 was synthesized upon reacting anthranilic acid with diethylacetylenedicarboxylate. Compound 3 was reacted with different nucleophiles in mild reaction conditions. Selected reaction routes that afforded products 6, 9, 10, 11, and 12 were explained. The estimated mechanism for the reaction of 3 with ethylenediamine to afford 9 was proved by X-ray single-crystal and retro-synthetic reaction. Acetyl anthranilic acid was utilized with zinc and copper to afford the organometallic compounds 14a and 14b, respectively. Three single crystals were afforded for 3, 9 and the organocopper
