Electron microscopy of vesicular stomatitis virus replicative ribonucleoproteins.

The objective of this investigation was to examine by electron microscopy the replicative ribonucleoprotein (RNP) structures synthesized in vesicular stomatitis virus-infected HeLa cells. Pulse-labeled in vivo products of vesicular stomatitis virus replication and transcription can be separated by centrifugation in Renografin gradients. Transcription complexes are dissociated, allowing nascent messenger RNPs to remain at the top of the gradient, whereas RNPs biochemically consistent with replication complexes sediment to the middle of the gradient. Examination of these structures by electron m

On the mechanism of inhibition of influenza virus replication by amantadine hydrochloride.

The results of analyses of fowl plague virus-specific RNA and protein synthesis in infected chick embryo fibroblasts incubated in amantadine hydrochloride are reported. They indicate that provided amantadine is present from the time of virus addition no expression of the virus genome occurs and that the synthesis of even the first detectable transcripts catalysed by the polymerase of the infecting virus particles is prevented. In agreement with previous reports it is concluded that amantadine prevents an unknown event which occurs immediately following virus infection.

Effect of cordycepin on the replication of western equine encephalitis virus.

Cordycepin (3'-deoxyadenosine) inhibited viral RNA synthesis in the replication of western equine encephalitis virus, thereby causing a reduction of virus production. The rate of inhibition of viral RNA synthesis was dependent on drug concentration and the period of treatment with the drug. These results suggest that the virus RNA synthesizing system is sensitive to the drug.

The ER-Golgi transport of influenza virus through NS1-Sec13 association during virus replication.

Influenza A virus is a respiratory virus that can cause complications such as acute bronchitis and secondary bacterial pneumonia. Drug therapies and vaccines are available against influenza, albeit limited by drug resistance and the non-universal vaccine administration. Hence there is a need for host-targeted therapies against influenza to provide an effective alternative therapeutic target. Sec13 was identified as a novel host interactor of influenza. Endoplasmic reticulum-to-Golgi transport is an important pathway of influenza virus replication and viral export. Specifically, Sec13 has a fun

MCPIP1 suppresses hepatitis C virus replication and negatively regulates virus-induced proinflammatory cytokine responses.

Human MCP-1-induced protein 1 (MCPIP1, also known as ZC3H12A and Regnase-1) plays important roles in negatively regulating the cellular inflammatory response. Recently, we found that as an RNase, MCPIP1 has broad-spectrum antiviral effects by targeting viral RNA. In this study, we demonstrated that MCPIP1 expression was induced by hepatitis C virus (HCV) infection in Huh7.5 hepatoma cells. MCPIP1 expression was higher in liver tissue from patients with chronic HCV infection compared with those without chronic HCV infection. Knockdown of MCPIP1 increased HCV replication and HCV-mediated express

Hepatitis C virus polymerase-polymerase contact interface: significance for virus replication and antiviral design.

The hepatitis C virus (HCV) replicates its genome in replication complexes located in micro-vesicles derived from endoplasmic reticulum. The composition of these replication complexes indicates that proteins, both viral and cellular in origin, are at high concentrations. Under these conditions, protein-protein interactions must occur although their role in the replication pathways is unknown. HCV RNA-dependent RNA-polymerase (NS5B) initiates RNA synthesis in these vesicles by a de novo (DN) mechanism. After initiation, newly synthesized dsRNA could induce conformational changes that direct the

Interactions between rubella virus capsid and host protein p32 are important for virus replication.

The distribution and morphology of mitochondria are dramatically affected during infection with rubella virus (RV). Expression of the capsid, in the absence of other viral proteins, was found to induce both perinuclear clustering of mitochondria and the formation of electron-dense intermitochondrial plaques, both hallmarks of RV-infected cells. We previously identified p32, a host cell mitochondrial matrix protein, as a capsid-binding protein. Here, we show that two clusters of arginine residues within capsid are required for stable binding to p32. Mutagenic ablation of the p32-binding site in

Enhancement of hepatitis C virus replication by Epstein-Barr virus-encoded nuclear antigen 1.

Based on our recent observation that Epstein-Barr virus (EBV) is detected in 37% of the tissues of hepatocellular carcinoma, and especially frequently in cases with hepatitis C virus (HCV), the effect of EBV infection on the replication of HCV was investigated. EBV-infected cell clones and their EBV-uninfected counterparts in cell lines MT-2 (a human T-lymphotropic virus type I-infected T-cell line), HepG2 (a hepatoblastoma cell line) and Akata (a Burkitt's lymphoma cell line) were compared in terms of their permissiveness for HCV replication following inoculation of HCV derived from patients

Homologous interference of lymphocytic choriomeningitis virus involves a ribavirin-susceptible block in virus replication.

Depending on the multiplicity of infection (MOI), infection of L929 cells results in either productive lymphocytic choriomeningitis virus replication or homologous interference M. Bruns, A. Gessner, H. Lother, and F. Lehmann-Grube, Virology 166:133-139, 1988). As shown in this communication, productive lymphocytic choriomeningitis virus replication as observed at a low MOI was effectively inhibited by ribavirin. In contrast, virus yields increased if cells were infected with a high MOI and in the presence of 5 microM of the antiviral compound. This drug-dependent release of infectious virus wa

Clinical-Stage Nanatinostat Triggers Productive Epstein-Barr Virus Lytic Replication.

A phase 1b/2 study of the all-oral "kick-and-kill" combo nanatinostat + valganciclovir (nana-val) showed a ~40% overall response rate in patients with relapsed/refractory Epstein-Barr virus (EBV)-associated lymphomas. Despite the promising results in these difficult-to-treat cancers, there is little known about the effects of nanatinostat on EBV and cell death. In this work we demonstrate that nanatinostat, an HDAC1/3 inhibitor, robustly induces the full EBV lytic cycle in a panel of EBV-positive lymphoma and transformed cell lines (Burkitt, DLBCL, lymphoblastoid), spanning viral latency types

Lactate facilitates largemouth bass virus replication by promoting cholesterol biosynthesis.

Cellular metabolism has been hijacked by viruses for efficient replication due to their regulatory roles on both innate and adaptive immunity. Largemouth bass virus (LMBV), a member of Ranavirus, causes high mortality and severe economic losses in several fish species, seriously threatening the sustainable development of aquaculture industry. However, the roles of the metabolic events in LMBV replication remained largely uncertain. Here, our results showed that LMBV boosted aerobic glycolysis both in vitro and in vivo. Supplement with glucose significantly enhanced LMBV replication in vitro. C

MARK1 suppresses infectious bursal disease virus replication via phosphorylating VP3.

Infectious bursal disease virus (IBDV) of the Birnaviridae family is a non-envelope, double-stranded RNA virus that encodes a VP3 protein with multiple functions, which controls viral genome replication, IFN-β production, and virus traffic in infected cells. Posttranslational modifications (PTMs), such as ubiquitination, of VP3 have been demonstrated for affecting its function and stability. To clarify the mechanism by which VP3 is regulated in IBDV infected cells, we focused on the phosphorylation of VP3. Mass spectrometry analysis identified that microtubule-affinity regulating kinases 1 (MA

Sulfatide Binds to Influenza B Virus and Enhances Viral Replication.

Seasonal influenza epidemics caused by influenza A viruses (IAV) and influenza B viruses (IBV) pose a substantial public health burden. Despite the significant impact of IBV, its restricted host range and the absence of documented pandemics have resulted in limited research attention relative to IAV. Understanding the viral infection mechanisms of both IAV and IBV is crucial for controlling seasonal epidemics. Previously, we demonstrated that 3'-O-sulfated galactosylceramide sulfatide binds to IAV and enhances viral replication, a finding with potential therapeutic implications. However, the r

IRF8 facilitates bovine ephemeral fever virus replication by downregulating IRF9.

Interferon regulatory factor 8 (IRF8), an essential member of the IRFs protein family, serves as a critical transcriptional regulator in cytokine signaling, gene transcription, and the differentiation and proliferation of immune cells. However, its function on the bovine ephemeral fever virus (BEFV) infection has not been described. In this study, we demonstrate that BEFV infection upregulates the expression of IRF8, and IRF8 promotes the replication of BEFV. Subsequent investigations reveal that IRF8 suppresses the type I IFN signaling pathway via the degradation of IRF9 in the context of BEF

Bisbenzylisoquinoline alkaloids inhibit influenza virus replication by disrupting endosomal acidification.

Influenza virus, known for causing recurrent epidemics and pandemics, pose a significant public health challenge due to their rapid mutation rates and the emergence of drug resistance. This emphasizes the urgent need for the development of novel antiviral drugs. In this study, we identified five bisbenzylisoquinoline alkaloids (BBAs)-cepharanthine (CEP), tetrandrine (TET), fangchinoline (FCN), berbamine (BBM) and iso-tetrandrine (Iso-TET)-that exhibit antiviral activity against influenza virus, as determined through cytopathic effect inhibition screening. These compounds showed dose-dependent

Temporal and spatial characterization of keratinocytes supporting orf virus replication.

Reflecting their tropism for keratinocytes, most poxviruses that infect vertebrates replicate to high titers and cause pathology in the skin. Keratinocytes, the main cells of the epidermis, are found in different stages of a differentiation program that produces the critical barrier against environmental damage. While systemic poxviruses (e.g. smallpox virus, sheeppox virus) also infect other cell types, the parapoxvirus orf virus (ORFV), which causes localized infections in sheep and goats, has not been shown to replicate in cells other than keratinocytes. Notably, ORFV infection only occurs

eIF4A3 inhibits pseudorabies virus replication by facilitating antiviral immune response.

Eukaryotic translation initiation factor 4A3 (eIF4A3)-mediated RNA metabolism is essential for cellular homeostasis and viral replication. However, its role in regulating antiviral innate immunity during pseudorabies virus (PRV) infection remains unknown. Here, we demonstrate that eIF4A3 protein expression was significantly downregulated both in vitro and in vivo during PRV infection. Functional assays showed that eIF4A3 overexpression suppressed PRV replication, whereas its knockdown enhanced viral replication. Mechanistically, eIF4A3 inhibited m6A modification of STING mRNA, thereby increasi

Oxysterol binding protein (OSBP) contributes to hepatitis E virus replication.

Hepatitis E virus (HEV) is a positive-sense, single-stranded RNA virus and causes primarily acute self-limiting infections. The ORF1 of the HEV genome encodes a polyprotein around 190 kDa, which contains several putative domains, including helicase and RNA-dependent RNA polymerase. The HEV-encoded helicase is a member of the superfamily 1 helicase family and possesses multiple enzymatic functions, such as RNA 5'-triphosphatase, RNA unwinding, and NTPase, which are thought to contribute to viral RNA synthesis. However, the helicase interaction with cellular proteins remains less known. Oxystero

Novel Potent Autophagy Inhibitor Ka-003 Inhibits Dengue Virus Replication.

Every year, dengue virus (DENV) affects millions of people. Currently, there are no approved drugs for the treatment of DENV infection. Autophagy is a conserved degradation process that was shown to be induced by DENV infection and required for optimal DENV replication. The modulation of autophagy is, therefore, considered an attractive target to treat DENV infection. This study carried out a high-content image screen analysis using Crispr-Cas9 GFP-LC3 knocked-in HeLa cells of a compound library synthesized from or inspired by natural products and their biocongener precursors to discover novel
