Molecular determinants of skeletal muscle force loss in response to 5 days of dry immersion in human.

BACKGROUND: Astronauts in Earth's orbit experience microgravity, resulting in a decline of skeletal muscle mass and function. On Earth, models simulating microgravity have shown that the extent of the loss in muscle force is greater than the loss in muscle mass. The reasons behind this disproportionate loss of muscle force are still poorly understood. In the present study, we hypothesize that alongside the loss in skeletal muscle mass, modifications in the expression profile of genes encoding critical determinants of resting membrane potential, excitation-contraction coupling and Ca2+ handling

Simulated microgravity enhances intracellular energy metabolism and electricity generation by electrochemically active bacteria for in situ space bioenergy production.

Electrochemically active bacteria (EAB) characterized by extracellular respiration is promising for bioregenerative life support systems and elemental cycling at extraterrestrial planetary bases. However, it remains unclear how microgravity modulates genes related to electricity generation and reshapes intracellular energy metabolism. In this study, the impact of simulated microgravity on EAB was systematically analyzed through electricity generation, gene enrichment, energy metabolism, and central carbon metabolic reprogramming. The results demonstrate that simulated microgravity increases th

A Platform for the Continuous Culture of Endothelial Cells Under Physiological Flow Conditions in Simulated Microgravity.

Microphysiological systems (MPS) are valuable in vitro models that recreate the structure and function of human tissues. Specifically, tissue- or organ-on-chips that incorporate microfluidic technology replicate the in vivo mechanical and biochemical environment while enabling real-time monitoring of cellular growth and development. These systems can be particularly useful for spaceflight studies, as they can be sent into space, such as to the International Space Station (ISS), to investigate the impact of space travel on the human body. A major impediment to the success of the MPS is the form

Plantar stimulation as a possible countermeasure to microgravity-induced neuromotor degradation.

Since the early days of human spaceflight it has been reported that extended exposure to gravitational unloading results in a myriad of neuromotor adaptations that, while appropriate for microgravity, are maladaptive upon return to Earth. If not countered, these adverse effects of microgravity can result in negative health consequences and place crewmembers at risk for injury. The most commonly used countermeasures in today's space programs are those requiring active participation in prescribed exercise regimes that are time intensive, not completely effective, and have led to relatively low c

[Ocular counterrolling as an indicator of vestibular otolith function].

Space Motion Sickness (SMS) occurred in the first few days of space flight correlated with the vestibular function. As the main influence of weightlessness in space focused on the otolith receptors. The test of otolith function is of great importance. Although there are a lot of methods to test otolith function, at present ocular counterrolling is the only relative pure indicator of otolith function. It provide a valid method for predicting SMS susceptibility, it has great application prospect in the research of the SMS mechanism and the readaptability of vestibular function after space flight

Spaceflight influences on ocular counterrolling and other neurovestibular reactions.

Exposure to extended periods of weightlessness in orbital flight has profound effects on the neurovestibular system and influences head and eye movements, postural control, and spatial orientation. The associated space motion sickness is among the earliest of the signs of adaptation to this new environment. This report both reviews the prominent neurovestibular phenomena associated with going into space and returning to earth and relates the issues to vestibular compensation and rehabilitation. New results from the Spacelab SLS-2 mission are included, showing significant reductions in postflig

Altered sensory-motor control of the head as an etiological factor in space-motion sickness.

Mechanical unloading during head movements in weightlessness may be an etiological factor in space-motion sickness. We simulated altered head loading on Earth without affecting vestibular stimulation by having subjects wear a weighted helmet. Eight subjects were exposed to constant velocity rotation about a vertical axis with direction reversals every 60 sec. for eight reversals with the head loaded and eight with the head unloaded. The severity of motion sickness elicited was significantly higher when the head was loaded. This suggests that altered sensory-motor control of the head is also an

Grip forces exerted against stationary held objects during gravity changes.

In the present study, grip forces exerted against a stationary held object were recorded during parabolic flights. Such flight maneuvers induce changes of gravity with two periods of hypergravity, associated with a doubling of normal terrestrial gravity, and a 20 s period of microgravity. Accordingly, the object's weight changed from being twice as heavy as normally experienced and weightless. Grip-force recordings demonstrated that force control was seriously disturbed only during the first experience of hyper- and microgravity, with the grip forces being exceedingly high and yielding irregul

The vestibular functions of the manganese-deficient rat.

Vestibular functions were examined in 25 animals of the second generation of rats raised on a manganese-deficient diet; 9 animals appeared to be normal and 16 showed behavioural abnormalities, characterized by inability to swim and lack of tonic eye deviations. The nystagmic responses elicited by rotatory stimulation appeared to be normal. Histological studies showed that the only defect was the absence of both utricular and saccular otoconia. These rats' swimming disability was similar to that of bilabyrinthectomized rats in normogravic condition and to that of normal rats immersed in water d

Spaceflight Respiratory System from a Nursing Perspective.

The pulmonary system is sensitive to aerospace exposures, including changes in atmospheric composition and pressure, alterations in gravitational forces, and exposure to radiation and toxins. Providing nursing care in the aerospace environment requires understanding of pulmonary conditions such as high-altitude pulmonary edema, decompression-related illness and injury, arterial gas embolism, acceleration atelectasis, hypoxia, hypercapnia, and exposure syndromes. In addition to these conditions, we discuss changes in pulmonary physiology in microgravity, in-flight nursing medical capabilities,

Effects of simulated microgravity on biological features and virulence of the fungal pathogen Cryptococcus neoformans.

Cryptococcus neoformans is a fungal pathogen that causes cryptococcal meningitis, mainly in immunocompromised individuals, such as those with HIV. Its recent detection on the International Space Station raises concerns about potential health risks in space, where immune systems may be compromised. However, its behavior in space-like conditions remains unclear. In this study, we examined the effects of simulated microgravity on C. neoformans. We found that the condition enhanced the fungus's resistance to membrane and osmotic stress and increased key virulence factors, including capsule formati

The article considers the questions of development of new technologies for evaluation of health of apparently healthy people based on experience of long-term researches of cosmonauts' performed at Russian Academy of Sciences Institute of Biomedical Problems under the direction of academician A. I. Grigoriev. In 2007 in monograph "Concept of Health and Space Medicine" the principle of health estimation in apparently healthy people working in conditions of chronic stress was described. These approaches were realized the same year to produce a new hard and software "Ecosan-2007". In the following

[Mechanism of development of space motion sickness].

Astronauts and scientists are scheduled to stay aboard the International Space Station (ISS) for three-month periods, and various problems relating to the human ability to live in space for such long periods of time, as well as medical problems, are being studied. For the crews that have operated and worked on the US space shuttle, the longest time spent in the gravity-free (accurately speaking, microgravity) environment of space has been about two weeks. Even such short periods have generated reports of various physical changes and symptoms caused by the absence of gravity. Particularly strik

Subtle shake-up in bone-loss research.

Recent research in the prevention of bone loss during weightlessness is described. Scientists are studying the effects of vibration on bone loss in laboratory animals and humans. Research is focused on determining how bone formation is triggered, the effects of stimulation of the stress response in bones, and the mechanisms behind the effects. Clinton Rubin at the State University of New York at Stony Brook is experimenting with 90 Hz vibration frequencies in rats. Other researchers are studying the effects of vibration plates in the treatment of osteoporosis in postmenopausal women and in chi

Simulation of Space-Adaptation Syndrome on Earth.

The three Spacelab D-1 Scientist Astronauts were exposed to a 1.5 h centrifuge run in the supine position, resulting in a linear 3 g acceleration. They used their space experience to evaluate their readaptation to normal gravity and compared their observations with 'Space Adaptation Syndrome'. After the centrifuge runs, the vestibular visual system appeared to be modified in a very specific and reproducible manner. Readaptation to the normal 1 g environment took at least 6 h. During this period there was a striking similarity to the astronauts' experience during adaptation to weightlessness in

Twenty four hours head-down tilt decreases arterial baroreflex function in conscious rat.

In human zero gravity induces a cardiovascular deconditioning. The transfer of blood from the periphery to the thoracic compartment, occuring immediately at the beginning of the exposure to microgravity seems to be involved in this phenomenon. We have previously shown that LBNP (-40 mm Hg) produced an exaggerated heart rate response at the end of 24 hours of bed rest without modification of the cardiopulmonary receptors in human. Some other studies have shown that microgravity induces an impairment of the baroreflex sensitivity in human, but its involvement in the cardiovascular deconditioning

Hepcidin knockout exacerbates hindlimb unloading-induced bone loss in mice through inhibiting osteoblastic differentiation.

BACKGROUND: An oligopeptide hepcidin is encoded by the human HAMP gene (Hamp in mice). Its deficiency can result in iron overload, while excess may lead to iron deficiency. Hepcidin knockout mice exhibited iron accumulation in multiple tissues, accompanied by degeneration of bone microarchitecture and reduced biomechanical properties. Astronauts who are exposed to weightlessness during prolonged spaceflight experience bone loss. After space missions, an interrelation exists between iron stores and bone mineral density (BMD). Bone loss in mice due to unloading is linked to iron excess and invol

Comparison of Streptococcus mutans growth-phase-dependent transcriptome changes in two different simulated microgravity models.

Streptococcus mutans is a cariogenic member of human dental plaque biofilm. Our previous study of S. mutans early stationary-phase rotary cell culture system (RCCS) cultures revealed significant transcriptome changes under low-shear modelled microgravity (LSMMG) compared to normal gravity (NG), as well as an altered cell clumping phenotype. In this current study, S. mutans mid-exponential phase and late-exponential phase cultures were compared using the RCCS (LSMMG and NG orientations) as well as simulated microgravity ('0 g') using a random positioning machine (0 g RPM). Multiple simulated spaceflight stressors impact cardiac fibrosis, calcium dynamics, immune function, cytokines and gene variants in rat, Rattus norvegicus.

Space radiation and microgravity pose significant health risks for astronauts conducting deep space exploration. The impact of galactic cosmic radiation (GCR) and weightlessness alone, and their combined effects on cardiac disease, the immune system, cytokines and calcium dynamics should be understood before undertaking such missions. Male WAG/RijCmcr rats, Rattus norvegicus, 8-9 months old were hind limb unloaded (HLU) to simulate exposure to microgravity for 5 days before, during, and 25 days after whole-body exposure to a simplified GCR simulation (simGCRsim) dose of 1.5 Gy that was adminis
