The First Record of Echinococcus ortleppi (G5) Tapeworms in Grey Wolf (Canis lupus).

The aim of this study is to confirm the presence and molecular identification of Echinococcus tapeworms in wolves from south-eastern Poland. An investigation was carried out on the intestines of 13 wolves from south-eastern Poland. The small intestines were divided into three equal segments. Each segment was separately examined using the sedimentation and counting technique (SCT). The detected Echinococcus tapeworms were isolated and identified by PCRs and sequencing (nad1 and cox1 genes). Additionally, DNA isolated from the feces of wolves positive for Echinococcus tapeworms was examined with

Echinococcus in wild canids in Québec (Canada) and Maine (USA).

Zoonotic Echinococcus spp. cestodes (E. canadensis and E. multilocularis) infect domestic animals, wildlife, and people in regions of Canada and the USA. We recovered and quantified Echinococcus spp. cestodes from 22 of 307 intestinal tracts of wild canids (23 wolves, 100 coyotes, 184 red and arctic foxes) in the state of Maine and the province of Québec. We identified the species and genotypes of three Echinococcus spp. cestodes per infected animal by sequencing mitochondrial DNA at two loci. We further confirmed the absence of E. multilocularis by extracting DNA from pools of all cestodes fr

Dispelling dog dogma: an investigation of heterochrony in dogs using 3D geometric morphometric analysis of skull shape.

Heterochrony is an evolutionary mechanism that generates diversity via perturbations of the rate or timing of development that requires very little genetic innovation. As such, heterochrony is thought to be a common evolutionary mechanism in the generation of diversity. Previous research has suggested that dogs evolved via heterochrony and are paedomorphic wolves. This study uses three-dimensional landmark-based coordinate data to investigate heterochronic patterns within the skull morphology of the domestic dog. A total of 677 adult dogs representing 106 different breeds were measured and com

Prevalence and molecular characterization of Giardia duodenalis in companion dogs, domestic livestock and wildlife in the Jordan Basin, Israel.

Giardiasis is a small intestinal disease caused by the zoonotic parasite, Giardia duodenalis. This study presents the molecular findings of G. duodenalis infection in companion dogs, domestic livestock and wildlife in the Northern Jordan Basin, Israel. Identification of G. duodenalis was accomplished by nested PCR (nPCR) targeting the 18S rRNA gene. Samples were collected from water (five samples from four sources of which one was recycled water), as well as feces from wolves (Canis lupus) (n = 34), jackals (Canis aureus) (n = 24), wild boars (Sus scrofa) (n = 40), cattle (Bos taurus) (n = 40)

Isolation and RFLP genotyping of Toxoplasma gondii from the gray wolf (Canis lupus).

Little is known of the genetic diversity of Toxoplasma gondii circulating in wildlife. In the present study feral gray wolves (Canis lupus) from Minnesota were examined for T. gondii infection. Antibodies to T. gondii were detected in 130 (52.4%) of 248 wolves tested by the modified agglutination test (cut-off titer of 25). Tissues (hearts, brains or both) of 109 wolves were bioassayed in mice for protozoal isolation. Viable T. gondii was isolated from 25 and the isolates were further propagated in cell culture. T. gondii DNA from these isolates was characterized using 10 PCR-RFLP markers (SAG

"Old hosts die hard": the return of the grey wolf (Canis lupus) and the re-emerging threat of cardiopulmonary dirofilariosis in northern Italy.

Over the past decades, the grey wolf (Canis lupus) has expanded its range across Italy, reclaiming historical habitats, including anthropized landscapes. This resurgence raises concerns regarding its potential role in maintaining and spreading pathogens. In this study, we focused on Dirofilaria immitis, the causative agent of canine heartworm disease, investigating its prevalence in wolves and exploring the environmental factors influencing infection risk. A total of 488 wolf carcasses collected from northern and central Italy between 2021 and 2024 were analysed. Morphological identification o

The wolf (Canis lupus) as an indicator species for the sylvatic Trichinella cycle in the Central Balkans.

Wildlife is the most important reservoir of Trichinella spp. worldwide. Although the Balkans are a recognized European endemic region for Trichinella infections, data on wildlife are scarce. To monitor the circulation of these zoonotic parasites in the Central Balkan region, the wolf (Canis lupus) was selected because of its abundance (>2,000 individuals) and because it is at the top of the food chain. A total of 116 carcasses of wolves were collected in Serbia and in the neighboring areas of Macedonia and Bosnia and Herzegovina (B&H) between 2006 and 2013. Trichinella spp. larvae were found i

Detection of Trichinella spp. in free-ranging carnivores and wild boars in Switzerland.

Wolves (Canis lupus lupus) and lynx (Lynx lynx) are officially monitored species in Switzerland. Deceased individuals are subjected to post-mortem examination and collection of baseline health data. The procedure includes an assessment of different infectious agents, including Trichinella spp., the cause of a notifiable zoonotic infection. Between May 2009 and May 2023, a total of 100 wolves and 250 lynx were tested at the National Reference Laboratory for Trichinellosis by the artificial digestion method. Additionally, muscle samples from 8838 wild boars (Sus scrofa), 27 red foxes (Vulpes vul

Trichinellosis dissemination among wild carnivores in the Republic of Kazakhstan: A 10-year study.

BACKGROUND AND AIM: Trichinellosis is caused by a species of roundworm called Trichinella and is an invasive disease causing severe medical, veterinary, and socioeconomic problems worldwide. More than 100 mammalian species are Trichinella hosts. Among domestic animals, pigs and dogs are prone to trichinellosis. An essential aspect of controlling the spread of infection is to identify the number and level of infections in wild carnivores in the country. However, the number, habitats, and movements of wild animal Trichinella hosts in Kazakhstan have not been reported yet. This study aimed to mon

Giardia duodenalis genotypes in domestic and wild animals from Romania identified by PCR-RFLP targeting the gdh gene.

Sixty Giardia duodenalis isolates from domestic (n=49) and wild (n=11) animals (dogs, cats, deers, wolves, raccoon dog and muskrat) were analysed by PCR-RFLP at glutamate dehydrogenase locus (gdh). The isolates were obtained from positive feces samples for Giardia cysts analysed by flotation technique with saturated sodium chloride solution (specific gravity 1.28). Three G. duodenalis genotypes were identified: C (10/60; 16.7%); D (42/60; 70.0%); and E (7/60; 11.7%). In dogs all three genotypes were found, with the following prevalences: 76.9% genotype D (30/39); 23.1% C (9/39); 2.6% genotype

Parasite survey on a captive wolf population using classical techniques and ELISA coproantigen detection, USA.

As laws change around the United States, wildlife that were once kept as companion animals are now often confiscated by local authorities. They are then euthanized unless a home is found for them at a sanctuary. Wolf sanctuaries are, therefore, becoming increasingly important for their conservation and management. However, little data is available on best practices for the health management of captive wolves, including data on parasitic diseases. Our objective was to assess the prevalence of parasites of captive wolves combining classical coprological techniques and immunoassays based on the d

High richness of ungulate Sarcocystis species in intestines of the grey wolf (Canis lupus) from Lithuania.

There is a lack of studies on the richness of Sarcocystis species in naturally infected grey wolves that serve as definitive hosts of these parasites. In this work, we used a nested PCR approach to examine the intestines of 13 grey wolves from Lithuania for the presence of Sarcocystis species that use canids as definitive host and ungulates as intermediate host. Under a light microscope, Sarcocystis spp. sporocysts/oocysts were found in 12 animals (92.3%). Based on comparisons of newly acquired cytochrome c oxidase subunit I (cox1) gene nucleotide sequences, we identified a total of 12 differe

Molecular survey of Ehrlichia canis and Coxiella burnetii infections in wild mammals of southern Italy.

Ehrlichiosis and Q fever caused by the intracellular bacteria Ehrlichia canis and Coxiella burnetii, respectively, are tick-borne diseases with zoonotic potential and widespread geographical distribution. This study investigated the prevalence of both infections in wild mammals in southern Italy. Tissue samples obtained from the red fox (Vulpes vulpes), European badger (Meles meles), gray wolf (Canis lupus), beech marten (Martes foina), and crested porcupine (Hystrix cristata) were processed for molecular detection of both pathogens. E. canis was detected in 55 out of 105 (52 %) red foxes and

The domestication of the wolf larynx-testing the neural crest connection.

The neural crest domestication syndrome (NCDS) hypothesis suggests that selection pressures for tameness during animal domestication resulted in a down-regulation of neural crest cell migration and proliferation, providing a mechanistic explanation for traits commonly observed across domesticates. We test the viability of this hypothesis via a derived prediction. According to the NCDS hypothesis, neural crest-derived structures should be reduced throughout the entire organism in domesticated variants compared to their wild ancestors. Here, we test this prediction by comparing the larynges of w

Playing with fire: Wolf response to active wildfires, fire fronts and smoke.

Wildfires are a key natural disturbance in fire-prone ecosystems, but their impacts on wildlife are increasingly significant as fire regimes intensify globally. Although considerable research has examined animal responses to post-wildfire landscapes, little is known about how animals respond to burning fires. While fires often trigger avoidance behaviours, predators may benefit from fire-related hunting opportunities. We present one of the first studies, to our knowledge, explicitly examining fine-scale behavioural responses of a large carnivore to actively burning wildfires, testing whether p

A simple repeat polymorphism in the MITF-M promoter is a key regulator of white spotting in dogs.

The white spotting locus (S) in dogs is colocalized with the MITF (microphtalmia-associated transcription factor) gene. The phenotypic effects of the four S alleles range from solid colour (S) to extreme white spotting (s(w)). We have investigated four candidate mutations associated with the s(w) allele, a SINE insertion, a SNP at a conserved site and a simple repeat polymorphism all associated with the MITF-M promoter as well as a 12 base pair deletion in exon 1B. The variants associated with white spotting at all four loci were also found among wolves and we conclude that none of these could

Evaluating noninvasive methods for estimating cestode prevalence in a wild carnivore population.

Helminth infections are cryptic and can be difficult to study in wildlife species. Helminth research in wildlife hosts has historically required invasive animal handling and necropsy, while results from noninvasive parasite research, like scat analysis, may not be possible at the helminth species or individual host levels. To increase the utility of noninvasive sampling, individual hosts can be identified by applying molecular methods. This allows for longitudinal sampling of known hosts and can be paired with individual-level covariates. Here we evaluate a combination of methods and existing

Structural variants in genes associated with human Williams-Beuren syndrome underlie stereotypical hypersociability in domestic dogs.

Although considerable progress has been made in understanding the genetic basis of morphologic traits (for example, body size and coat color) in dogs and wolves, the genetic basis of their behavioral divergence is poorly understood. An integrative approach using both behavioral and genetic data is required to understand the molecular underpinnings of the various behavioral characteristics associated with domestication. We analyze a 5-Mb genomic region on chromosome 6 previously found to be under positive selection in domestic dog breeds. Deletion of this region in humans is linked to Williams-

High genetic diversity of Echinococcus canadensis G10 in northeastern Asia: is it the region of origin?

Echinococcus canadensis consists of 4 genotypes: G6, G7, G8 and G10. While the first 2 predominantly infect domestic animals, the latter are sylvatic in nature involving mainly wolves and cervids as hosts and can be found in the northern temperate to Arctic latitudes. This circumstance makes the acquisition of sample material difficult, and little information is known about their genetic structure. The majority of specimens analysed to date have been from the European region, comparatively few from northeast Asia and Alaska. In the current study, Echinococcus spp. from wolves and intermediate
