[Zinc homeostasis in plants: the role of ZIP, MTP and HMA transporters and chelating agents].

Zinc (Zn) is an essential micronutrient involved in numerous physiological processes in plants, serving catalytic, structural, and regulatory functions. Maintaining its appropriate concentration requires precise control of uptake, transport, distribution, and storage, as both Zn deficiency and excess lead to disruptions in plant growth and development. This paper presents the mechanisms regulating zinc homeostasis in plants, with a particular focus on the role of membrane transporters and chelating compounds. The ZIP, MTP, and HMA protein families play a key role, being responsible for cellula

Maximum water stress is decoupled from climate, traits, and growth in a xeric oak.

Our ability to explain and predict the spatial patterns of forest mortality remains limited. We tested the predictability of water stress across populations and explored how water availability and plant allocation mediate spatial variation in tree growth in a widespread oak species (Quercus douglasii Hook & Arn.). We conducted a range-wide survey of drought stress, xylem water isotopes, growth, and allocation traits across 15 sites spanning almost 1000 m difference in annual precipitation. We found little relationship between end-of-season water availability (predawn leaf water potentials) or

Retention of CdS/ZnS Quantum Dots (QDs) on the Root Epidermis of Woody Plant and Its Implications by Benzo[a]pyrene: Evidence from the in Situ Synchronous Nanosecond Time-Resolved Fluorescence Spectra Method.

The retention of CdS/ZnS QDs on the epidermis has been confirmed to be one of the core procedures during the root uptake process. However, the retention mechanisms of QDs on the epidermis of woody plant were poorly understood for lacking of an appropriate QD quantitative method. In this study, a novel method for in situ determination of CdS/ZnS QDs retained on the root epidermis was established using synchronous nanosecond time-resolved fluorescence spectroscopy. No correlations between Kf values of oleylamine-CdS/ZnS QDs retained on the epidermal tissues and the surface/bulk composition of ma

A microautoradiographic method for fresh-frozen sections to reveal the distribution of radionuclides at the cellular level in plants.

Microautoradiography (MAR) is a conventional imaging method based on the daguerreotype. The technique is used to visualize the distribution of radionuclide-labeled compounds within a tissue section. However, application of the classical MAR method to plant tissue sections is associated with several difficulties. In this study, we report an MAR method applicable to fresh-frozen plant sections. Our method had two features: (i) the sample was kept frozen from plant tissue collection to radioisotope detection, making it possible to fix solutes without solvent exchange; and (ii) 1.2 µm thick polyph

Drought-induced delays in stem hydraulic development shape gas exchange and growth recovery in Douglas fir.

The limiting factors of tree recovery from drought, particularly the coordination between carbon sources and sinks, remain poorly understood. In this study, juvenile Douglas fir (Pseudotsuga menziesii) were exposed to 28 d of mild or severe drought, followed by 35 d of recovery. We continuously monitored CO₂ and H₂O fluxes in shoots and roots to derive gas exchange and carbon accumulation, while measuring basal area to estimate stem growth and sapwood development. To identify underlying mechanisms of drought recovery, we periodically measured nonstructural carbohydrates (NSC), midday water pot

Accumulation and localization of cadmium in Echinochloa polystachya grown within a hydroponic system.

Phytoremediation is a technology for extracting or inactivating pollutants. Echinochloa polystachya [(H.B.K.) Hitchcock] (Poaceae) is a fast-growing perennial grass that is common in tropical areas and is often found in oil-polluted soils that contain high concentrations of heavy metals. However, its tolerance to heavy metals, and its ability to accumulate them, has yet to be investigated. Here we test the hypothesis that E. polystachya is able to accumulate high concentrations of cadmium (Cd). Plants were grown hydroponically with different levels of Cd(2+) (0, 0.25, 1, 2, 10, 50, and 100mgL(

Adaptive Structural and Transcriptional Responses Contribute to Cold Tolerance Variation in Xinluzhong 61 and Tahe 2 Cotton Cultivars.

This study elucidated the molecular, morphological and anatomical mechanisms underlying differential cold tolerance between two cotton cultivars, cold-tolerant Xinluzhong 61 (C61) and cold-sensitive Tahe 2 (C2). Seedlings were subjected to 0 °C cold stress for 12 and 24 h, followed by comparative transcriptomic (RNA-Seq) and comprehensive anatomical analyses of cotyledons, true leaves and stems. Transcriptomic profiling identified 8834 and 14,664 differentially expressed genes (DEGs) in C2 and C61 at 12 h, and 14,399 and 16,791 DEGs at 24 h, respectively. KEGG enrichment revealed prominent inv

Overexpression of Silicon Uptake Transporter OsLsi1 and OsLsi2 Decreased Cadmium Accumulation in Rice.

Silicon (Si) fertilizer reduces cadmium (Cd) accumulation in rice; whether the rice Si uptake genes mediate this process remains unclear. This study investigated how overexpressing OsLsi1 and OsLsi2 affects Cd accumulation in rice. Compared to wild-type ZH11, OsLsi1 and OsLsi2 expression increased by 5- and 150-fold in roots, respectively. The overexpression of OsLsi1 reduced root Cd, while that of OsLsi2 decreased shoot Cd in the seedlings. Both lines showed lower Cd in xylem sap, dependent on Si, and alleviated Cd-induced root stress. At maturity, grain Cd decreased by 44-66% and straw Cd by

Novel loop-mediated isothermal amplification (LAMP) assay with a universal QProbe can detect SNPs determining races in plant pathogenic fungi.

Tomato wilt pathogen Fusarium oxysporum f. sp. lycopersici (Fol) is grouped into three races based on their pathogenicity to different host cultivars. Rapid detection and discrimination of Fol races in field soils is important to prevent tomato wilt disease. Although five types of point mutations in secreted in xylem 3 (SIX3) gene, which are characteristic of race 3, have been reported as a molecular marker for the race, detection of these point mutations is laborious. The aim of this study is to develop a rapid and accurate method for the detection of point mutations in SIX3 of Fol. Loop-medi

SbLOX3 modulates jasmonate-associated flavonoid accumulation, root growth, and drought-induced root hollow formation in Scutellaria baicalensis.

The hollowed root formation and 4'-deoxyflavone accumulation in Scutellaria baicalensis are associated with jasmonate (JAs) signaling, but the upstream regulators integrating these processes with drought tolerance remain unknown. This study investigated whether the 13-lipoxygenase gene SbLOX3 may act as a modulator. Transgenic plants overexpressing SbLOX3 or with RNAi suppression were exposed to polyethylene glycol-simulated and long-term soil drought stress. Metabolite profiling, gene expression analysis, root morphological measurements, and histological observations were conducted. Overexpre

Transcriptomic Differences Between Two Fusarium oxysporum Formae Speciales During Cucumber Infection.

Fusarium oxysporum f. sp. radicis-cucumerinum (Forc) V03-2g and Fusarium oxysporum f. sp. radicis-lycopersici (Forl) ZUM2407 both cause foot and root rot in cucumber, but differ in host range. Forc V03-2g possesses Secreted in Xylem (SIX) effector genes, whereas Forl ZUM2407 does not, raising questions about their distinct infection strategies on this host. Using comparative transcriptomic analysis (in cucumber at 7 and 14 days post-inoculation (dpi) and in tomato at 2 dpi) we show that Forl ZUM2407 induces a delayed defense response in cucumber compared to Forc V03-2g. In turn, Forc V03-2g ra

Retrospective study of methylmercury and other metal(loid)s in Madagascar unpolished rice (Oryza sativa L.).

The rice ingestion rate in Madagascar is among the highest globally; however studies concerning metal(loid) concentrations in Madagascar rice are lacking. For Madagascar unpolished rice (n = 51 landraces), levels of toxic elements (e.g., total mercury, methylmercury, arsenic and cadmium) as well as essential micronutrients (e.g., zinc and selenium) were uniformly low, indicating potentially both positive and negative health effects. Aside from manganese (Wilcoxon rank sum, p < 0.01), no significant differences in concentrations for all trace elements were observed between rice with red bran (n

Arsenic, cadmium, and chromium concentrations in contrasting phosphate fertilizers and their bioaccumulation by crops: Towards a green label?

Potentially toxic elements such as arsenic (As), cadmium (Cd), and chromium (Cr) are severely regulated in fertilizers and deserve continuous investigation. Phosphate-derived Cd has been a stepping-stone toward achieving sustainable and safe worldwide food production, especially after a new regulation aiming for reduced limits of Cd in P fertilizers (EU, 2019/1009). Three pot experiments were conducted to assess the variability of As, Cd, and Cr concentrations - with a particular focus on Cd - from monoammonium phosphates (MAP 1, MAP 2, and MAP 3 from different geographic origins) and their ac

Vascular Bundles Mediate Systemic Reactive Oxygen Signaling during Light Stress.

Systemic signaling and systemic acquired acclimation (SAA) are essential for plant survival during episodes of environmental stress. Recent studies highlighted a key role for reactive oxygen species (ROS) signaling in mediating systemic responses and SAA during light stress in Arabidopsis (Arabidopsis thaliana). These studies further identified the RESPIRATORY BURST OXIDASE HOMOLOG D (RBOHD) protein as a key player in mediating rapid systemic ROS responses. Here, we report that tissue-specific expression of RBOHD in phloem or xylem parenchyma cells of the rbohD mutant restores systemic ROS sig

Reveal the lignification of ginkgo after click chemistry by novel monolignol analogs.

Lignin is one of the abundant natural resources in the world, but most of it is discarded as waste. So, understanding the deposition and structure of lignin plays an important role lignin valorization. In order to avoid changing the side chain structure of lignin monomers to make negligible influence on lignin deposition process, propargyl group was introduced at 2/6 position of benzene ring to explore deposition mechanism of lignin in plants by polymerization of alkynyl-labeled lignin precursors (HALK, GALK) in vivo and in vitro. The results showed that HALK and GALK were biocompatibility, co

FvNST1b NAC Protein Induces Secondary Cell Wall Formation in Strawberry.

Secondary cell wall thickening plays a crucial role in plant growth and development. Diploid woodland strawberry (Fragaria vesca) is an excellent model for studying fruit development, but its molecular control of secondary wall thickening is largely unknown. Previous studies have shown that Arabidopsis NAC secondary wall thickening promoting factor1 (AtNST1) and related proteins are master regulators of xylem fiber cell differentiation in multiple plant species. In this study, a NST1-like gene, FvNST1b, was isolated and characterized from strawberry. Sequence alignment and phylogenetic analysi

Cadmium distribution in ratoon rice: OsNramp5 mutant reduces accumulation while physiological factors modulate node-specific partitioning.

Cadmium (Cd) accumulation in ratoon rice poses significant food-safety risks and complicates the sustainable expansion of ratooning systems. This study elucidates the physiological mechanisms governing Cd accumulation patterns in ratoon rice by comparing two genotypes contrasting in Cd accumulation under controlled Cd exposure. OsNramp5 loss-of-function reduced grain Cd by 39.5-51.2% across the main and ratoon seasons, accompanied by downregulation of OsNramp1 (root uptake) and OsHMA2 (xylem loading) and enhanced leaf Cd chelation. Across the ratoon crop, upper-node tillers accumulated 30.8-59

PtoeIF5A1: A Pleiotropic Regulator of Development, PCD, and Salt Tolerance in Populus tomentosa.

Eukaryotic translation initiation factor 5A (eIF5A) is a highly conserved protein family unique to eukaryotes, yet its functional characterization in woody plants remains limited. In this study, we identified four eIF5A genes (PtoeIF5A1-PtoeIF5A4) from the genome of Populus tomentosa, a fast-growing tree species indigenous to China, and characterized their expression patterns and functional roles through bioinformatics analysis, quantitative real-time PCR, stable overexpression in Arabidopsis thaliana, and transient expression in Nicotiana benthamiana leaves. Excess nitric oxide alters cellular pH to restrict salicylic acid movement and systemic immunity.

Plants carrying a mutation in S-nitrosoglutathione reductase 1 (GSNOR1) accumulate high nitric oxide (NO) and exhibit impaired immunity characterized by reduced pathogen-responsive salicylic acid (SA) accumulation and failure to activate normal SA signaling. We show that this reduced SA responsiveness in gsnor1 plants arises from impaired vascular-associated movement of SA, thereby compromising systemic acquired resistance (SAR). Elevated NO perturbs cellular pH homeostasis, acidifying the apoplast and alkalinizing the cytosol, which likely interferes with phloem-associated SA movement and ren
