Chitosan coating of zein-carboxymethylated short-chain amylose nanocomposites improves oral bioavailability of insulin in vitro and in vivo.

Non-invasive means of insulin administration circumvent some of the inconveniences of injections. Oral administration in particular is convenient, pain-free, and allows favorable glucose homeostasis, but is subject to chemical instability, enzymatic degradation, and poor gastrointestinal absorption. Natural polymeric nanoparticles have emerged as a promising oral delivery system for peptide therapeutics due their safety, biocompatibility, and stability. In this study, self-assembled nanocomposites from chitosan (CS) and insulin-loaded, zein-carboxymethylated short-chain amylose (IN-Z-CSA) nano

Design and optimization of a biodegradable porous zein conduit using microtubes as a guide for rat sciatic nerve defect repair.

Various degradable biomaterials have been used to bridge injured peripheral nerve defect; however, drawbacks such as poor mechanical properties, inappropriate degradation rate, and toxic degradation products continue to limit the application of them in nerve repair. Considering the unique properties of zein, such as its biocompatibility, biodegradability and ease of fabrication, we report the use of zein conduits to repair injured rat sciatic nerves with a 10-mm defect. Three-dimensional zein conduits were designed with/without pores, and with/without microtubes including in the lumen of condu

Zein-calcium alginate dual-shell liquid-core beads for tailored starch digestibility: Structural design, functional performance, and mechanistic analysis.

A single calcium alginate shell has poor barrier against digestive enzymes, restricting starch protection and necessitating a more robust shell. To address this, 1% sodium alginate were immersed in 0.25-1% zein solutions to form a hydrophobic protective layer. Chromaticity analysis showed that the b⁎ value increased from 7.45 to 24.08 with the increase of zein concentration. SEM images and laser confocal microscopy observations the formation of a zein/calcium alginate double-layer shell on the starch granule surface. Additionally, as the zein concentration in the immersion solution increased f

Thermally induced self-assembly of zein into amyloid fibrils under alkaline-ethanol conditions: Kinetics, structural evolution, and molecular interactions.

The formation behavior of zein into amyloid fibrils (ZAFs) under alkaline-ethanol conditions remains insufficiently understood. This study adopts orthogonal experiments combined with multidimensional structural and physicochemical characterization to elucidate the formation mechanism of ZAFs under heat treatment in high-ethanol alkaline environments. Ethanol concentration was identified as the dominant factor governing ZAFs formation, with optimal conditions determined as 60% ethanol, pH 10, and 90 °C. Under these conditions, zein self-assembly followed a typical sigmoidal kinetic profile, exh

Zein-EGCG‑sodium alginate conjugates with tunable interfacial properties for improved stability and antioxidant performance of walnut oil emulsions.

Designing food-grade emulsifiers with enhanced interfacial properties and functional activities is key to improving walnut oil-in-water emulsion stability and oxidation resistance. This study used zein, epigallocatechin gallate (EGCG), and sodium alginate (SA) to construct binary/ternary emulsifiers with different compositions and bonding modes. An oil-to-water ratio of 2:8 was optimized for minimal droplet size. Emulsions stabilized by zein-EGCG-SA conjugates exhibited the greatest interfacial thickness (732 nm) and highest protein adsorption rate, resulting in superior stability against ther

Lipidomic fingerprinting of plasmalogen-loaded zein nanoparticles during in vitro multiple-stage digestion using rapid evaporative ionization mass spectrometry.

Plasmalogens (Pls) as the hydrophobic bioactive compound have shown potential in enhancing neurological disorders. However, the bioavailability of Pls is limited because of their poor water solubility during digestion. Herein, the hollow dextran sulfate/chitosan - coated zein nanoparticles (NPs) loaded with Pls was prepared. Subsequently, a novel in situ monitoring method utilizing rapid evaporative ionization mass spectrometry (REIMS) coupled with electric soldering iron ionization (ESII) was proposed to assess the lipidomic fingerprint alteration of Pls-loaded zein NPs during in vitro multip

Disrupting the inflammation-oxidative stress feedback loop via transdermal nanodelivery of Shikonin using a zein/chitosan core-shell platform.

Psoriasis is a chronic inflammatory skin disorder for which effective topical therapy remains challenging due to limited skin penetration and insufficient modulation of the pathological microenvironment. Disease progression is sustained by a self-amplifying loop between inflammation and oxidative stress, largely mediated by hyperactivation of the IL-17A/JAK2/STAT3 signaling pathway. Herein, we report the development of a core-shell nano-delivery system (SKN-ZCNPs) to enhance the transdermal delivery and therapeutic efficacy of Shikonin (SKN), a multi-target anti-inflammatory and antioxidant ag

Controlled release and enhanced bioavailability of curcumin using multilayered LPSG/PVA and zein/gluten nanofibers fabricated by emulsion electrospinning.

The low stability and bioavailability, and poor aqueous solubility of curcumin (Cur) make it difficult to fully utilize its biological benefits. To overcome these limitations, emulsion electrospinning technique was used to encapsulate Cur in multilayered nanofibers made of Lepidium perfoliatum seed gum (LPSG)/polyvinyl alcohol (PVA) as the middle layer, and zein or zein/gluten nanofibers as the outer layers. Cur was first dissolved in a corn oil/Tween 80 mixture to prepare the oil phase. For the middle layer, emulsions with LPSG/PVA (30:70 v/v) and corn oil (2, 5, 10 % v/v) were prepared. The
