Inhibitory and excitatory amino acids in cerebrospinal fluid of neurolathyrism patients, a highly prevalent motorneurone disease.

Neurolathyrism is caused by overconsumption of seeds containing 3-N-oxalyl-L-2,3-diaminopropanoic acid (beta-ODAP). Amino acids levels of cerebrospinal fluid (CSF) were studied in 50 patients with neurolathyrism and 12 healthy volunteers. The levels of excitatory amino acids glutamate and aspartate were 281% and 71% respectively of control values. The concentration of inhibitory amino acids glycine and taurine were 277% and 198% respectively of the levels in CSF from control individuals. There was a significant correlation between the level of glycine and the duration of the disease. We also f

Leukaemia inhibitory factor (LIF) production in a mouse model of spinal trauma.

A model of spinal trauma was developed where spinal neurones of adult mice were exposed to the excitotoxic glutamate analogue beta-N-oxylamino-L-alanine (L-BOAA). After 24 h, the injured neurones received a single dose of [125I]-LIF at the same site of the spinal cord, and 2 h later, tissues were removed to assess the distribution of leukaemia inhibitory factor (LIF). There was a significant increase in LIF binding to the injured region of the spinal cord over saline controls, and this corresponded with a significant increase in LIF mRNA expression in the same region of the cord. There was a c

Insulin secretion in lipodystrophic HIV-infected patients is associated with high levels of nonglucose secretagogues and insulin resistance of beta-cells.

We examined whether plasma concentrations of nonglucose insulin secretagogues are associated with prehepatic insulin secretion rates (ISR) in nondiabetic, insulin-resistant, human immunodeficiency virus (HIV)-infected, lipodystrophic patients (LIPO). Additionally, the negative feedback of insulin on ISR was evaluated. ISR were estimated by deconvolution of plasma C-peptide concentrations during fasting (basal) and during the last 30 min of a 120-min euglycemic insulin clamp (40 mU.m(-2).min(-1)). Eighteen normoglycemic LIPO were compared with 25 normoglycemic HIV-infected patients without lipo

Effect of citrus pulp silage feeding on concentration of beta-cryptoxanthin in plasma and milk of dairy cows.

Citrus pulp is known to contain a functional molecule of beta-cryptoxanthin which is one of the carotenoids showing anti-oxidative capacity. Influences of citrus pulp silage feeding to dairy cows on beta-cryptoxanthin concentration in plasma, other blood properties and milking performances were investigated. Four Holstein cows were fed total mixed ration (TMR) containing citrus pulp silage 20% dry matter (DM) for 2 weeks with free access to the TMR. Dry mater intake, milk production and milk components 2 weeks later were not altered compared with those of the control group without citrus pulp

Identification of a novel integrin alpha 6 beta 1 binding site in the angiogenic inducer CCN1 (CYR61).

The angiogenic inducer CCN1 (cysteine-rich 61, CYR61), a secreted matricellular protein of the CCN family, is a ligand of multiple integrins, including alpha 6 beta 1. Previous studies have shown that CCN1 interaction with integrin alpha 6 beta 1 mediates adhesion of fibroblasts, endothelial cells, and smooth muscle cells, as well as migration of smooth muscle cells. Recently, we have reported that CCN1-induced tubule formation of unactivated endothelial cells is also mediated through integrin alpha 6 beta 1. In this study, we demonstrate that human skin fibroblasts adhere specifically to the

Multi-target action of β-alanine protects cerebellar tissue from ischemic damage.

Brain ischemic stroke is among the leading causes of death and long-term disability. New treatments that alleviate brain cell damage until blood supply is restored are urgently required. The emerging focus of anti-stroke strategies has been on blood-brain-barrier permeable drugs that exhibit multiple sites of action. Here, we combine single-cell electrophysiology with live-cell imaging to find that β-Alanine (β-Ala) protects key physiological functions of brain cells that are exposed to acute stroke-mimicking conditions in ex vivo brain preparations. β-Ala exerts its neuroprotective action thr

Chemometric-assisted spectrophotometric approach for stability assessment of safinamide and its synthetic precursor in antiparkinsonian formulation with sustainability profiling.

Parkinson's disease (PD) is a significant health issue that affects older individuals. Safinamide mesylate (SAF) is a recently developed adjunct therapy increasingly employed in the management of PD. Ensuring the stability of new drug formulations and establishing suitable stability-indicating methodologies is crucial for pharmaceutical analysis. The current study has developed three new, user-friendly, and robust mathematical methods using multivariate spectrophotometric analysis to quantify SAF, its synthetic precursor impurity (4-HBD), and stress-induced degradation products. The multivaria

Evaluation of protein-energy malnutrition in surgical patients from plasma valine and other amino acids, proteins, and anthropometric measurements.

Eight plasma proteins, four anthropometric measurements, and 21 amino acids were measured in 24 fasting patients before surgery. A matrix of partial correlation coefficients, correcting for age and height, showed many correlations including several between biochemical and anthropometric data such as between valine or prealbumin and arm muscle circumference. Valine was the most highly correlated variable and was used to rank and group the patients. Ten patients with low valine had the greatest weight loss and also had low values for 18 variables which could be subdivided. 1) Thirteen of the low

Protective mechanism of beta-SQAG9 liposome, a sulfonoglycolipid extracted from sea urchin intestines, against hepatic ischemia reperfusion injury.

We previously reported that beta-SQAG9 liposome, a sulfonoglycolipid extracted from sea urchin intestines, had a protective effect against hepatic ischemia reperfusion (I/R) injury. In this study, we made a detailed investigation of this protective effect and its mechanism. Rats were pretreated either with beta-SQAG9 liposome (treated group) or with phosphate-buffered saline solution (control group). Thereafter, they were subjected to partial hepatic I/R. The serum levels of aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase were measured, and histological damage w

Multi-omic analysis reveals that the CARNS1-mediated carnosine synthesis pathway modulates muscle metabolic responses to nutrient restriction in goats.

Seasonal feed shortages pose critical challenges to ruminant productivity, yet the mechanisms underlying muscle metabolic responses to varying degrees of restriction remain unclear. This study simulated feed shortage in goats using three dietary treatments: normal feeding (RF0), 25% restricted feeding (RF25), and 50% restricted feeding (RF50). A total of 30 male Xiangdong black goats with similar initial body weight (19.14 ± 2.75 kg) and age (4.0 ± 0.3 months) were randomly assigned into 3 groups (n = 10 per group) for an 87-d trial (7-d preliminary period followed by an 80-d experimental peri

West Nile virus NS2A protein facilitates virus-induced apoptosis independently of interferon response.

The flavivirus NS2A protein is a small, multifunctional protein, involved in replication, virion formation and regulation of the innate immune response. Using the Kunjin strain of West Nile virus (WNV(KUN)) we previously demonstrated that a single amino acid change from alanine to proline at position 30 of the NS2A protein (A30P) reduced viral cytopathicity in cells and virulence in mice. To further investigate functions of the NS2A protein we have substituted alanine at position 30 with different amino acids (A30 mutants) in a WNV(KUN) infectious clone. The virulence of mutant viruses in wild

Amino acid metabolism in the brain with convulsive disorders. Part 2: The effects of anticonvulsants on convulsions and free amino acid patterns in the brain of El mouse.

To elucidate the changes of free amino acid patterns in brains of El mice induced by several anticonvulsants, the free amino acid levels in brains were measured by an amino acid autoanalyzer 24 hours after intraperitoneal injection of PB, PHT or vitamin B6 or subcutaneous injection of ACTH. Compared to the preconvulsion group, the total free amino acid level increased in the ACTH group clearly. In the PB group, the levels of threonine, glutamine, lysine, histidine and homocarnosine increased and that of ornithine decreased; in the PHT group, an increase of glutamine and histidine and a decreas

The neurotransmitters glycine and GABA stimulate glucagon-like peptide-1 release from the GLUTag cell line.

The incretin hormone, glucagon-like peptide-1 (GLP-1) is released from intestinal L-cells following food ingestion. Its secretion is triggered by a range of nutrients, including fats, carbohydrates and proteins. We reported previously that Na(+)-dependent glutamine uptake triggered electrical activity and GLP-1 release from the L-cell model line GLUTag. However, whereas alanine also triggered membrane depolarization and GLP-1 secretion, the response was Na+ independent. A range of alanine analogues, including d-alanine, beta-alanine, glycine and l-serine, but not d-serine, triggered similar de

Enhancement of GABA-mediated postsynaptic inhibition in cultured mammalian spinal cord neurons: a common mode of anticonvulsant action.

Murine spinal cord neurons grown in dissociated cell culture were used to study the effects of barbiturate (phenobarbital, mephobarbital) and benzodiazepine (diazepam, chlordiazepoxide( anticonvulsants on amino acid responses. Both types of anticonvulsant augmented GABA-mediated postsynaptic inhibition without augmenting beta-alanine or glycine-mediated postsynaptic inhibition. Barbiturates, but not benzodiazepines, antagonized glutamate-mediated postsynaptic excitation. Augmentation of GABA-mediated inhibition by the anticonvulsants should contribute to their anticonvulsant action; antagonism
