Neurophysiology of CSWS-associated cognitive dysfunction.

The phenomenon of continuous spikes and waves during slow-wave sleep (CSWS) is associated with a number of epileptic syndromes, which share a behavioral phenotype characterized by deterioration of cognitive, behavioral, or sensorimotor functions. Available evidence seems to suggest that spike-wave activity is a result of a complex interaction between cortical and subcortical inhibitory networks and can "per se" produce a transient loss of underlying cortical functions. Syndromes like Landau-Kleffner syndrome, CSWS, and phenomena such as negative myoclonus could share in common--at least at the

Two successive phases in the threat-related attentional response of anxious subjects: neural correlates.

BACKGROUND: It is well known that highly anxious individuals are characterized by the allocation of a greater amount of attentional resources to threatening stimulation. However, neural data in relation to the time course of attentional biases in anxiety are still surprisingly scarce and preliminary. The present research explored attentional biases in anxious subjects (grouped according to their scores in trait and state forms of the State-Trait Anxiety Inventory) over time by using event-related potentials (ERPs) and through the application of source localization methodologies. METHODS: Part

Phonological but not auditory discrimination is impaired in dyslexia.

Deficient phonological skills are considered to be a core problem in developmental dyslexia. Children with dyslexia often demonstrate poorer performance than non-impaired readers when categorizing speech-sounds. Using the automatic mismatch response, we show that in contrast to this deficit at the behavioural level, neurophysiological responding in dyslexic children indicates their ability to automatically discriminate syllables. Therefore, the phonological deficit is unlikely to be caused by a temporal deficit or by a noisy functional organization in the respective representational cortex. We

Brain mechanisms for reading in children with and without dyslexia: a review of studies of normal development and plasticity.

In this article we review our experience with the application of magnetic source imaging (MSI), the newest of the functional imaging methods, to the study of brain mechanisms for reading among children who read normally and among those with dyslexia. After giving a general description of MSI, we present evidence for reliable and valid maps of the brain mechanism for aural language comprehension as well as for reading. Next, we present data from 39 normal readers, 40 children with dyslexia, and 30 younger children at risk for developing a reading disability. These data show different brain acti

Optimal configuration of on-scalp OPMs with fixed channel counts.

Recent technological developments have brought optically pumped magnetometers (OPMs) within reach of the larger neuroscientific community. The current state-of-the-art consists of whole-head systems that measure the magnetic field at >100 locations. OPM sensors can be constructed to measure the field in either 1, 2, or 3 orientations. Consequently, the number of channels can differ from the number of sensors. This allows for magnetoencephalography (MEG) system designs with multiple measurement orientations at fewer locations, many locations with fewer orientations, or, ideally, many locations

Clinical and experimental insight into pathophysiology, comorbidity and therapy of absence seizures.

Absence seizures in children and teenagers are generally considered relatively benign because of their non-convulsive nature and the large incidence of remittance in early adulthood. Recent studies, however, show that 30% of children with absence seizures are pharmaco-resistant and 60% are affected by severe neuropsychiatric comorbid conditions, including impairments in attention, cognition, memory and mood. In particular, attention deficits can be detected before the epilepsy diagnosis, may persist even when seizures are pharmacologically controlled and are aggravated by valproic acid monothe

Slowing of oscillatory brain activity is a stable characteristic of Parkinson's disease without dementia.

Extensive changes in resting-state oscillatory brain activity have recently been demonstrated using magnetoencephalography (MEG) in moderately advanced, non-demented Parkinson's disease patients relative to age-matched controls. The aim of the present study was to determine the onset and evolution of these changes over the disease course and their relationship with clinical parameters. In addition, we evaluated the effects of dopaminomimetics on resting-state oscillatory brain activity in levodopa-treated patients. MEG background oscillatory activity was studied in a group of 70 Parkinson's di

[Neurobiological correlates of cognitive flexibility in ADHD - A systematic review of the literature].

INTRODUCTION: Attention deficit hyperactivity disorder (ADHD) is one of the most commonly diagnosed childhood psychiatric disorders, which persists to adulthood in 30-50% of the cases. Previous studies have shown that cognitive flexibility, which means to switch between two different rules, that can be tested with task switching paradigms, is affected. Although poor performance in cognitive flexibility tests has been demostrated, the neurobiological background is only partly known. OBJECTIVE: The aim of our review was to examine the neurobiological background of impairment of cognitive flexib

Orbitofrontal dysfunction related to depressive symptomatology in subjects with borderline personality disorder.

OBJECTIVE: In order to explore the relationships of fronto-limbic dysfunction with the clinical features of borderline personality disorder (BPD), the authors investigated brain electrophysiological activity in BPD patients following stimulation with emotionally arousing images. METHODS: Seventeen non-medicated patients with borderline personality disorder were studied with magneto-encephalography. Regional cortical activities were obtained by minimum norm estimate (MNE) of steady-state visual evoked fields (ssVEFs). Linear regression models were conducted to explore clinical correlates of br

Which physiological components are more suitable for visual ERP based brain-computer interface? A preliminary MEG/EEG study.

We investigated which evoked response component occurring in the first 800 ms after stimulus presentation was most suitable to be used in a classical P300-based brain-computer interface speller protocol. Data was acquired from 275 Magnetoencephalographic sensors in two subjects and from 61 Electroencephalographic sensors in four. To better characterize the evoked physiological responses and minimize the effect of response overlap, a 1000 ms Inter Stimulus Interval was preferred to the short (<400 ms) trial length traditionally used in this class of BCIs. To investigate which scalp regions conv

Binding 3-D object perception in the human visual cortex.

How do visual luminance, shape, motion, and depth bind together in the brain to represent the coherent percept of a 3-D object within hundreds of milliseconds (msec)? We provide evidence from simultaneous magnetoencephalographic (MEG) and electroencephalographic (EEG) data that perception of 3-D objects defined by luminance or motion elicits sequential activity in human visual cortices within 500 msec. Following activation of the primary visual cortex around 100 msec, 3-D objects elicited sequential activity with only little overlap (dynamic 3-D shapes: MT-LO-Temp; stationary 3-D shapes: LO-Te

Cortical reorganization during recovery from complex regional pain syndrome.

OBJECTIVE: To characterize reorganization of the primary somatosensory cortex (S1) during healing process in complex regional pain syndrome (CRPS). BACKGROUND: Recently, the authors showed extensive reorganization of the S1 cortex contralateral to the CRPS affected side. Predictors for these plastic changes were CRPS pain and the extent of mechanical hyperalgesia. It is unclear how these S1 changes develop following successful therapy. METHODS: The authors used magnetic source imaging to explore changes in the cortical representation of digits (D) 1 and 5 in relation to the lower lip on the

Electrophysiological characterization of adult-onset Niemann-Pick type C disease.

In infantile and juvenile Niemann-Pick type C (NPC) disease electrophysiological studies have shown central (CNS) and peripheral (PNS) nervous system abnormalities. However, an extensive electrophysiological evaluation of CNS and PNS in adult form of NPC is still lacking. The aim of the study is to assess in adult-onset NPC disease the involvement of CNS and PNS by a multimodal electrophysiological approach. Three patients affected by adult form of NPC disease underwent electrophysiological evaluation including nerve conduction study (NCS), magnetic motor (MEPs), visual (VEPs), somatosensory (

Stimulus-contrast-induced biases in activation order reveal interaction between V1/V2 and human MT+.

The luminance contrast of a visual stimulus is known to modulate the response properties of areas V1 and the human MT complex (hMT+), but has not been shown to modulate interactions between these two areas. We examined the direction of information transfer between V1/V2 and hMT+ at different stimulus contrasts by measuring magnetoencephalographic (MEG) responses to moving and stationary stimuli presented centrally or peripherally. To determine the direction of information flow, the different response latencies among stimuli and hemispheres in V1/V2 was compared with those of hMT+. At high cont

Brain electrical source differences between depressed subjects and healthy controls.

Many brain regions show metabolic and perfusion abnormalities in major depressive disorder (MDD), including anterior cingulate and prefrontal cortices. Some of these same areas also show abnormal function with low resolution electromagnetic tomography (LORETA). However, LORETA results are not always consistent across studies, nor with findings from other imaging modalities. These discrepancies may be due, among other factors, to the sensitivity of EEG source localization to different electrode montages. Thirty-six channel EEG was collected from healthy controls and age- and gender-matched unme

Combined MEG and EEG methodology for non-invasive recording of infraslow activity in the human cortex.

OBJECTIVE: Periinfarct depolarisation and spreading depression represent key mechanisms of neuronal injury after stroke. Changes in cortical electrical potentials and magnetic fields in the very low frequency range are relevant parameters to characterize these events, which up to now have only been recorded invasively. In this study, we proved whether a non-invasive combined MEG/EEG recording technique is able to quantitatively monitor cortical infraslow activity in humans. METHODS: We used repetitive very slow and slow right finger movements as a physiological motor activation paradigm to in

Cognitive dimensions of orthographic stimuli affect occipitotemporal dynamics.

Previous research documented letter-string specific cortices in the ventral visual stream near the left occipitotemporal junction (i.e., anterior fusiform gyrus). These neural areas potentially code the perceptual elements comprising orthographic stimuli, and thus function as feature detectors in high-level vision. While abundant evidence supports this region's role in detecting isomorphic perceptual features, any influence cognitive dimensions (e.g., the lexicality of letter-strings) may play in modulating this area's processing remains an open question. To investigate this, we examined the s

Assessment of corticodiaphragmatic pathway and pulmonary function in acute ischemic stroke patients.

The aims of this study were to investigate the effect of stroke on the corticodiaphragmatic pathway and to clarify the relationships between neurophysiological data and degree of motor disability, site of infarction in CT scan, diaphragmatic excursion, blood gases and pulmonary function in stroke patients. The corticodiaphragmatic pathway was assessed using magnetic stimulation of the scalp sites and cervical roots. The study included 34 sequentially selected patients out of 250 patients with acute ischemic stroke. Twenty-five (age and sex matched) volunteers served as controls. Sixteen patien
