A study of the health assessment questionnaire to evaluate functional status in polymyalgia rheumatica.

OBJECTIVE: To evaluate the Health Assessment Questionnaire (HAQ) in the assessment of functional status, its responsiveness to change with treatment and its correlation with conventional disease activity indices in polymyalgia rheumatica (PMR). METHODS: Newly diagnosed patients with PMR, satisfying modified Jones and Hazleman criteria, were recruited to the study. The clinical assessments, including early morning stiffness (EMS), pain measured on a horizontal 10 cm visual analogue scale (VAS), C-reactive protein (CRP) and the HAQ, were carried out 0, 6, 12 and 24 weeks after treatment had bee

Contracture of the deltoid muscle. Results of distal release.

We prospectively studied the results of distal release, performed between February 1989 and December 1994 for the treatment of a contracture of the deltoid muscle, in forty patients (forty-nine shoulders). Forty-seven contractures (in thirty-eight patients) developed after multiple intramuscular injections of various medications. The two remaining contractures (in two patients) were congenital. The average age at the onset of the symptoms was thirty-two years (range, birth to fifty-eight years). The primary symptoms included pain around the neck and the shoulder girdle, dimpling of the skin, a

Central nervous system effects of the neurohypophyseal hormones and related peptides.

This review of the CNS effects of the neurohypophyseal hormones and related neuropeptides discusses recent data illustrating the significance of these principles in brain function, synthesis, distribution, in particular in extrahypothalamic brain structures, binding sites, and signal transduction. Binding sites for vasopressin of the vascular V1a type have been found in the CNS and there is evidence for the existence of a subtype of the antidiuretic V2 receptor in the brain. Also two types of oxytocin binding sites have been detected. One widely distributed throughout the CNS is comparable to

Perinatal malnutrition, suprachiasmatic nuclear lesioning, and circadian-ultradian aspects of spontaneous behavior of albino rats.

The effect of reducing the dietary protein content at different stages of development was investigated. A group of control rats was fed a diet with 25% protein throughout the entire study. Four other groups were fed only 6% protein in a diet isocaloric with the control diet. In an "intrauterine"-deprivation (prenatal) group, the feeding of the low protein diet was limited to the span from mating to delivery. In a "postnatal"-deprivation (lactation) group, the reduced protein diet was limited to the 25 days allowed for lactation. In a "perinatal"-deprivation group, the low-protein diet was fed

PX-RICS-deficient mice mimic autism spectrum disorder in Jacobsen syndrome through impaired GABAA receptor trafficking.

Jacobsen syndrome (JBS) is a rare congenital disorder caused by a terminal deletion of the long arm of chromosome 11. A subset of patients exhibit social behavioural problems that meet the diagnostic criteria for autism spectrum disorder (ASD); however, the underlying molecular pathogenesis remains poorly understood. PX-RICS is located in the chromosomal region commonly deleted in JBS patients with autistic-like behaviour. Here we report that PX-RICS-deficient mice exhibit ASD-like social behaviours and ASD-related comorbidities. PX-RICS-deficient neurons show reduced surface γ-aminobutyric ac

Methylphenidate treatment during pre- and periadolescence alters behavioral responses to emotional stimuli at adulthood.

BACKGROUND: Methylphenidate (MPH) is a psychomotor stimulant medication widely used for the treatment of attention-deficit/hyperactivity disorder (ADHD). Given the extent of prescribed use of MPH, and because MPH interacts with the same brain pathways activated by drugs of abuse, most research has focused on assessing MPH's potential to alter an individual's risk for adult drug addiction. Data examining other potential long-term behavioral consequences of early MPH administration are lacking, however. METHODS: We investigated the long-term behavioral consequences of chronic administration of

Ferula ammoniacum gum aqueous extract exerts anti-inflammatory and antioxidant mechanisms to combat aluminum chloride-induced Alzheimer's disease in rats: possible involvement of opioid pathways.

BACKGROUND: Alzheimer's disease (AD), the most common form of dementia, significantly affects memory and behavior due to dysregulated pathways involving oxidative stress, inflammation, and opioidergic systems. Currently, no effective treatments are available, underscoring the need for novel alternatives. Ferula ammoniacum (D.Don) Spalik, M. Panahi, Piwczyński, and Puchałka [Apiaceae] (FA), an Iranian medicinal plant, is known for its anti-seizure, anti-inflammatory, and analgesic properties, with its gum utilized as a nerve tonic. PURPOSE: This study investigates the anti-AD effects of F. amm

Sex-specific behavioral and monoaminergic network alterations following adolescent binge-like ethanol and WIN55,212-2 exposure under chronic nicotine in rats.

INTRODUCTION: Recreational drug use often begins during adolescence, a critical period of brain maturation characterized by heightened vulnerability to drug-induced neuroplastic changes. Although weekend consumption of alcohol and cannabis and nicotine is commonly perceived as relatively harmless, the long-term neurobiological consequences of such patterns remain poorly understood. METHODS: In this study, we investigated the behavioral and neurochemical consequences of combined adolescent exposure to moderate doses of alcohol, cannabinoid receptor agonist WIN55,212-2, and nicotine in male and

Behavioral decline in Shank3Δex4-22 mice during early adulthood parallels cerebellar granule cell glutamatergic synaptic changes.

BACKGROUND: SHANK3, a gene encoding a synaptic scaffolding protein, is implicated in autism spectrum disorder (ASD) and is disrupted in Phelan-McDermid syndrome (PMS). Despite evidence of regression or worsening of ASD-like symptoms in individuals with PMS, the underlying mechanisms remain unclear. Although Shank3 is highly expressed in the cerebellar cortical granule cells, its role in cerebellar function and contribution to behavioral deficits in ASD models are unknown. This study investigates behavioral changes and cerebellar synaptic alterations in Shank3Δex4-22 mice at two developmental s

Characterization of the biochemical and behavioral effects of cannabidiol: implications for migraine.

Cannabidiol (CBD) is the main pharmacologically active phytocannabinoid. CBD exerts an analgesic effect in several pain models, does not have side effects and has low toxicity. The data about CBD mechanisms of action in pain and its therapeutic potential in this area are limited. Here, we tested CBD effects in animal models specific for migraine. We assayed CBD distribution in plasma and in cranial areas related to migraine pain in male Sprague Dawley rats treated chronically (5 days). Successively, we tested CBD activity on the behavioral and biochemical effects induced in the acute and the c

GENDER RELATED DIFFERENCES IN SEX HORMONE-MEDIATED ANXIOLYTIC EFFECTS OF ELECTROMAGNETIC STIMULATION DURING IMMOBILIZATION STRESS.

Physical and psychological stressors activate the hypothalamic-pituitary-adrenal axis (HPA). Elevated glucocorticoid levels over a long time harm neurocognitive and neurobiological development and cause deleterious effects. The effects of glucocorticoids depend not only on the concentration and duration of action, but also on the period of life when the effect occurs and gender. Electromagnetic stimulation (EMS) is a non-invasive treatment for some neurodegenerative diseases. The main objective of the study was to investigate the therapeutic effects of EMS on immobilization-induced depressive

Neurocognitive sequelae of antenatal corticosteroids in a late preterm rabbit model.

BACKGROUND: Late preterm birth is associated with short-term respiratory and adaptive problems. Although antenatal corticosteroids seem to reduce the respiratory burden, this may come at the cost of adverse neuropsychological outcomes later in life. This impact has not been investigated. OBJECTIVE: Herein, we investigate what the short- and long-term neurodevelopmental effects of a single course of betamethasone in simulated late preterm birth. STUDY DESIGN: Time-mated pregnant does received 0.1 mg/kg betamethasone (n=8) or 1 mL saline intramuscular (n=6) at the postconceptional ages of 28 a

Behavior and inflammation of the rumen and cecum in Holstein bulls fed high-concentrate diets with different concentrate presentation forms with or without straw supplementation.

Twenty-four individually housed Holstein bulls (395 ± 7.3 kg BW and 252 ± 3.1 d age) were exposed to a 2 × 2 factorial design (meal vs. pellets; with vs. without straw) to evaluate the effect of concentrate form and provision of straw in finishing diets on behavior and expression of rumen and cecum epithelium genes related to inflammation and behavior. Concentrate and straw consumption were recorded monthly and behavior (self-grooming, social, oral nonnutritive, tongue rolling, eating, drinking, ruminating, and lying) was recorded every two weeks. Bulls were slaughtered after 64 d of exposure

Older driver distraction: a naturalistic study of behaviour at intersections.

This study examined older driver engagement in distracting behaviours (secondary activities) at intersections using naturalistic driving data from a larger study based in Melbourne, Australia. Of interest was whether engagement in secondary activities at intersections was influenced by factors such as driver gender and situational variables, in particular, those relating to the complexity of the driving environment. Specifically we expected that when making left/right turns, older drivers would reduce the proportion of time engaged in secondary behaviours at intersections which required gap ju

Unstable maternal environment, separation anxiety, and heightened CO2 sensitivity induced by gene-by-environment interplay.

BACKGROUND: In man, many different events implying childhood separation from caregivers/unstable parental environment are associated with heightened risk for panic disorder in adulthood. Twin data show that the occurrence of such events in childhood contributes to explaining the covariation between separation anxiety disorder, panic, and the related psychobiological trait of CO(2) hypersensitivity. We hypothesized that early interference with infant-mother interaction could moderate the interspecific trait of response to CO(2) through genetic control of sensitivity to the environment. METHODO

Chronic adrenocorticotrophic hormone treatment alters tricyclic antidepressant efficacy and prefrontal monoamine tissue levels.

Several animal models are currently utilised in the investigation of major depressive disorder; however, each is validated by its response to antidepressant pharmacotherapy. Few animal models consider the notion of antidepressant treatment resistance. Chronic daily administration of adrenocorticotropic hormone (ACTH) or corticosterone can alter behavioural responses to antidepressants, effectively blocking antidepressant efficacy. Herein, we demonstrate that ACTH-(1-24) (100μg/day; 14 days) blocks the immobility-reducing 'antidepressant' effects of a single dose of imipramine (10mg/kg) in the

A mouse model of fragile X syndrome exhibits heightened arousal and/or emotion following errors or reversal of contingencies.

This study was designed to further assess cognitive and affective functioning in a mouse model of Fragile X syndrome (FXS), the Fmr1(tm1Cgr) or Fmr1 "knockout" (KO) mouse. Male KO mice and wild-type littermate controls were tested on learning set and reversal learning tasks. The KO mice were not impaired in associative learning, transfer of learning, or reversal learning, based on measures of learning rate. Analyses of videotapes of the reversal learning task revealed that both groups of mice exhibited higher levels of activity and wall-climbing during the initial sessions of the task than dur

Bortezomib-induced gastrointestinal dysfunction, peripheral neuropathy and neuroimmune signalling are influenced by the gut microbiota in mice.

BACKGROUND: Peripheral neuropathy and gastrointestinal dysfunction are frequent, debilitating side effects of the neurotoxic myeloma drug bortezomib that reduce quality of life and adherence to optimal therapy. Therapeutic strategies to manage these complications are limited. Here, we have developed a mouse model of bortezomib side effects to investigate the role of the gut microbiota in their development. METHODS: C57BL/6 specific pathogen-free (SPF), germ-free (GF) and ex-GF mice (colonised with healthy gut microbiota via faecal microbiota transplantation [FMT]) were treated with bortezomib

NOP agonist AT-403 promoted sleep in lactic acid-induced acute pain model.

The majority of patients with acute pain experience sleep disturbances that persist despite analgesic treatments such as mu opioid receptor (MOP) agonists and non-steroidal anti-inflammatory drugs (NSAIDs). Further, sleep disturbances increase pain sensitivity, demonstrating a bi-directional relationship between pain and sleep. Given that commonly prescribed MOP agonists disrupt sleep in pain-naïve subjects, it is possible that analgesics exacerbate sleep disturbances associated with pain states. Thus, pain-induced sleep disturbances remain an understudied and undertreated symptom impacting ov

Group housing increases alertness and social interaction compared to traditional single stabling in two-year-old sport horse stallions during pretraining.

Horses in training are often stabled individually, but this is increasingly questioned. We have investigated the effects of different stabling systems in Warmblood stallions during a 12-week pretraining programme. Stallions were 24 months old and housed either in a group stable (Group 24, n = 9) or in individual boxes (Box 24, n = 10), or they were 30 months of age and housed in individual boxes (Box 30, n = 10). Cortisol, heart rate, heart rate variability (HRV), body development, injuries and selected behaviour patterns were analysed (1) at transfer from pasture to stable and (2) thereafter,
