Multidrug Intrinsic Resistance Factors in Staphylococcus aureus Identified by Profiling Fitness within High-Diversity Transposon Libraries.

UNLABELLED: Staphylococcus aureus is a leading cause of life-threatening infections worldwide. The MIC of an antibiotic against S. aureus, as well as other microbes, is determined by the affinity of the antibiotic for its target in addition to a complex interplay of many other cellular factors. Identifying nontarget factors impacting resistance to multiple antibiotics could inform the design of new compounds and lead to more-effective antimicrobial strategies. We examined large collections of transposon insertion mutants in S. aureus using transposon sequencing (Tn-Seq) to detect transposon mu

Antibiotics in neonatal infections: a review.

The bacteria most commonly responsible for early-onset (materno-fetal) infections in neonates are group B streptococci, enterococci, Enterobacteriaceae and Listeria monocytogenes. Coagulase-negative staphylococci, particularly Staphylococcus epidermidis, are the main pathogens in late-onset (nosocomial) infections, especially in high-risk patients such as those with very low birthweight, umbilical or central venous catheters or undergoing prolonged ventilation. The primary objective of the paediatrician is to identity all potential cases of bacterial disease quickly and begin antibacterial tre

Meningitis caused by Streptococcus in adults.

Meningitis caused by Streptococcus is uncommon in adults. Otitis media, mastoiditis, sinusitis, or trauma has antedated most cases reported in the past. Data on 10 recent cases suggest changes in the current pathogenesis of streptococcal meningitis in adults and emphasize the importance of accurate streptococcal speciation. Endocarditis was present in five cases; trauma played a role in two others. Brain abscess, corticosteroids, alcoholic cirrhosis, and peritonitis secondary to chronic peritoneal dialysis were etiologic factors in five patients. Otitis media, mastoiditis, and sinusitis were c

Risk Factors Associated With Multidrug Resistant Organism (MDRO) Isolates in Infected Diabetic Foot Ulcers (DFU): An Analytical Cross-sectional Study.

BACKGROUND: Increasing use and abuse of antibiotics has led to a rise in multidrug resistant organisms (MDRO) isolates among infected diabetic foot ulcers (DFUs) and is seen in more than half of all diabetic foot infection cases. This study aimed to identify risk factors for MDRO infection since there are no Filipino studies currently available to establish an association. METHODOLOGY: A cross-sectional, analytical study was conducted to evaluate risk factors for MDRO in DFUs. Adult patients admitted in a tertiary hospital from January 2019 - December 2023 for DFU were included in the study.

Molecular characteristics and pathogenicity-associated phenotypes of methicillin-resistant Staphylococcus aureus sequence type 398 clinical isolates from a women and children's hospital in Southwest China.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) sequence type 398 (ST398), initially recognized as a livestock-associated lineage, has been increasingly reported in human infections. However, the molecular characteristics, antimicrobial resistance profiles, phylogenetic context, and pathogenicity-associated phenotypes of clinical MRSA ST398 isolates in China remain insufficiently defined. Here, we characterized such isolates from Southwest China using genomic, antimicrobial susceptibility, and phenotypic analyses. METHODS: From January 2020 to June 2023, 23 nonduplicate MRSA ST

Distinct genetic programs drive antibiotic resistance and intracellular invasion in emerging MRSA strains.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major global health threat because of its ability to adapt. In North America, the USA300 lineage ST8 has become the predominant MRSA clone, whereas the ST72 lineage has emerged as an important MRSA in East Asia. Here, we compare USA300 and SAWL001 from the ST72 at the phenotypic, genomic, and transcriptomic levels. Phenotypic assays assessed antibiotic susceptibility, intracellular invasion, oxidative stress survival, biofilm formation under β-lactam exposure, blood-induced cell clumping, persister formation, and virulence in a mouse seps

Genomic and phenotypic characterization of multidrug-resistant Staphylococcus haemolyticus isolated from burn patients in Chongqing, southwestern China.

Staphylococcus hemolyticus is one of the frequently isolated nosocomial pathogens in immunocompromised patients; however, its genotypic and phenotypic characteristics in burn patients remain largely unknown. In this study, 146 S. haemolyticus strains were collected from the burn unit of a tertiary hospital in Chongqing, southwestern China. Whole genome sequencing was applied to determine the molecular characteristics of these S. haemolyticus isolates. Results showed that sequence type 1 (ST1; 26.7%, 39/146) was the most prevalent lineage, followed by clones ST42 (17.8%, 25/146), ST3 (15.8%, 23

Triterpenoid saponin from Panax ginseng increases the sensitivity of methicillin-resistant Staphylococcus aureus to β-lactam and aminoglycoside antibiotics.

The triterpenoid saponins, ginsenosides, are the major bioactive compound of red ginseng and can exert various physiological activities. In the present study, we examined whether red ginseng extract (RGE) exerts antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA). RGE had no bactericidal activity, at least in the range of dissolvable concentration. However, RGE reduced 0.03-0.25-fold the minimum inhibitory concentration (MIC) values of β-lactam antibiotics (oxacillin, ampicillin, carbenicillin, and cefazolin) and aminoglycoside antibiotics (kanamycin and gentamici

Canine Staphylococcaceae circulating in a Kenyan animal shelter.

Animal shelters, especially in resource-poor countries, bring together pets from different regions and with different backgrounds. The crowding of such animals often results in infectious diseases, such as respiratory infections. This study characterized Staphylococcaceae from diseased and apparently healthy dogs housed in an animal shelter in Kenya, to determine their antibiotic resistance profiles, their genetic relatedness, and the presence of dominant clones. Therefore, bacteria were collected from all 167 dogs present in the shelter in June 2015 and screened for Staphylococcaceae using st
