Astragalus polysaccharide protects against neuron degeneration and mitochondrial dysfunction in Parkinson's disease by upregulating CEND1.

BACKGROUND: Parkinson's disease (PD) is a chronic neurodegenerative disorder featuring dopaminergic neuron loss, which is associated with mitochondrial dysfunction. Astragalus polysaccharide (APS) extracted from Astragalus membranaceus possesses antioxidant, anti-inflammatory, and neuroprotective properties. APS was previously revealed to exert neuroprotective effects in experimental PD models. However, the underlying mechanism remains poorly understood. Therefore, our study was designed to reveal the molecular mechanism through which APS exerts neuroprotective effects in PD. METHODS: SH-SY5Y

Reduced DJ-1/Nrf2/HO-1 antioxidant signalling is associated with type 2 diabetes-related aggravation of neurodegeneration in a mouse model of Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) and type 2 diabetes mellitus (T2DM) are increasingly recognized as comorbid conditions. Epidemiological evidence suggests that T2DM is associated with faster PD progression, but the biological processes that may connect these disorders remain incompletely understood. Oxidative stress is a shared pathological feature of both diseases, and the DJ-1/Nrf2/HO-1 pathway is an important antioxidant defence system. OBJECTIVE: To determine whether T2DM is associated with more severe PD-like neurodegeneration and whether these changes are accompanied by reduced DJ-1

Neuroprotective and neurorestorative properties of Mesembryanthemum tortuosum in a Parkinson's disease zebrafish larvae model.

ETHNOPHARMACOLOGICAL RELEVANCE: Parkinson's disease (PD) is characterised by the loss of dopaminergic neurons, resulting in reduced dopamine levels in the brain. The exact cause of dopaminergic neuron loss remains unknown but factors such as increased oxidative stress (OS), neuroinflammation and mitochondrial dysfunction have been implicated in the progression of the disease. The behavioural and molecular repertoire of PD can be modelled in zebrafish larvae using a neurotoxin, 6-hydroxydopamine (6-OHDA). Currently, there is no cure for PD, however, medicinal plants such as Mesembryanthemum tor

Sleep deprivation drives hepatic steatosis via sympathetic sprouting-induced ER stress in young male mice.

BACKGROUND & AIMS: Accumulating evidence indicates that sleep loss is significantly associated with an increased risk of metabolic dysfunction-associated steatotic liver disease (MASLD). However, the underlying mechanisms remain poorly understood. This study aimed to investigate the role of hepatic sympathetic nerves in sleep loss-associated MASLD. METHODS: Chronic sleep restriction (CSR) was induced using an automated cylindrical sleep-deprivation apparatus with a rotating bar. The 3D architecture of hepatic sympathetic nerves was visualized using the iDISCO+ (immunolabeling-enabled 3D imagi

Simultaneous 5-HT1BR agonist/5-HT6R antagonist action as a potential treatment of Parkinson's disease and its comorbidities.

Parkinson's disease (PD) treatment focuses mainly on the augmentation of dopamine transmission, but to alleviate adverse motor effects accompanying L-DOPA use, additional treatments with serotonergic (5-HT) medications may be considered. We propose a novel concept based on the simultaneous activation of 5-HT1BR and 5-HT6R blockade as a putative therapeutic option for PD treatment. We have developed PZKKN-94, a dual human 5-HT1BR agonist (EC50 = 39 nM) and human 5-HT6R antagonist (KB = 7.7 nM), with selectivity over 43 targets, favorable drug-like properties, and brain penetration. Importantly,

Coherent Changes in Neural Motor Network Activity during Levodopa-Induced Dyskinesia in a Rat Model of Parkinson's Disease.

BACKGROUND: Long-term use of levodopa, a metabolic precursor of dopamine (DA) for alleviation of motor symptoms in Parkinson's disease (PD), can cause a serious side effect known as levodopa-induced dyskinesia (LID). With the development of LID, high-frequency gamma oscillations (~100 Hz) are registered in the motor cortex (MCx) in patients with PD and rats with experimental PD. Studying alterations in the activity within major components of motor networks during transition from levodopa-off state to dyskinesia can provide useful information about their contribution to the development of abnor

Preclinical evaluation of [18F]FP-CIT, the radiotracer targeting dopamine transporter for diagnosing Parkinson's disease: pharmacokinetic and efficacy analysis.

BACKGROUND: N-(3-fluoropropyl)-2β-carboxymethoxy-3β-(4-iodophenyl) nortropane (FP-CIT), the representative cocaine derivative used in dopamine transporter imaging, is a promising biomarker, as it reflects the severity of Parkinson's disease (PD). 123I- and 18F-labeled FP-CIT has been used for PD diagnosis. However, preclinical studies evaluating [18F]FP-CIT as a potential diagnostic biomarker are scarce. Among translational research advancements from bench to bedside, translating preclinical findings into clinical practice is one-directional. The aim of this study is to employ a circular appro

Autoradiographic labelling of metabotropic glutamate type 2/3 receptors in the hemi-parkinsonian rat brain.

L-3,4-dihydroxyphenylalanine (L-DOPA) is the treatment of choice for Parkinson's disease (PD) motor symptoms, but its chronic use is hindered by complications such as dyskinesia. Pre-clinical studies discovered that activation of metabotropic glutamate type 2 and 3 (mGlu2/3) receptors alleviates L-DOPA-induced dyskinesia. To gain mechanistic insight into the anti-dyskinetic activity of mGlu2/3 activation, we performed autoradiographic binding with [3H]-LY-341,495 in brain sections from L-DOPA-treated 6-hydroxydopamine (6-OHDA)-lesioned rats that developed mild or severe dyskinesia, as well as

Abnormal Cortico-Basal Ganglia Neurotransmission in a Mouse Model of l-DOPA-Induced Dyskinesia.

l-3,4-dihydroxyphenylalanine (l-DOPA) is an effective treatment for Parkinson's disease (PD); however, long-term treatment induces l-DOPA-induced dyskinesia (LID). To elucidate its pathophysiology, we developed a mouse model of LID by daily administration of l-DOPA to PD male ICR mice treated with 6-hydroxydopamine (6-OHDA), and recorded the spontaneous and cortically evoked neuronal activity in the external segment of the globus pallidus (GPe) and substantia nigra pars reticulata (SNr), the connecting and output nuclei of the basal ganglia, respectively, in awake conditions. Restoration of complex sensorimotor behavior and skilled forelimb use by a modified nigral cell suspension transplantation approach in the rat Parkinson model.

While intrastriatal transplants of dopamine-rich ventral mesencephalic tissue are effective in reversing a variety of drug-induced behaviors in the rat Parkinson model, previous studies have failed to obtain significant graft-induced effects on deficits in certain aspects of complex sensorimotor behaviors. In the present study we have applied a modified cell suspension transplantation procedure, which allows more reproducible and consistent ventral mesencephalic transplants of large size, as well as more wide-spread distribution of the ventral mesencephalic tissue over multiple graft sites wit

Cocaine: cardiovascular effects in relation to inhibition of peripheral neuronal monoamine uptake and central stimulation of the sympathoadrenal system.

Cocaine (0.03-5.6 mg/kg i.v.) produced a dose-dependent and prolonged increase in mean arterial blood pressure and heart rate in conscious rats. The 0.3 and 1 mg/kg doses of cocaine potentiated the pressor response to exogenous norepinephrine (0.2 microgram/kg), whereas lower (0.03 and 0.1 mg/kg) and higher (3 and 5.6 mg/kg) doses were ineffective. Desipramine (0.03-1 mg/kg), a prototype norepinephrine uptake blocker, did not alter blood pressure or heart rate. Nisoxetine (0.01-1 mg/kg), another norepinephrine selective uptake blocker, produced a small and brief (< 5 min) pressor response, but

Medial Prefrontal Cortex Modulation of the Peripheral Immune Response in Sepsis Via the Autonomic Nervous System.

BACKGROUND: Sepsis is a life-threatening organ dysfunction syndrome caused by a dysregulated host response to infection, in which neuroimmune dysfunction plays a central role. The medial prefrontal cortex (mPFC) is involved in autonomic and immune regulation. However, its role in modulating peripheral immune responses during sepsis remains unclear. This study investigated whether glutamatergic neurons in the mPFC regulate systemic inflammation and organ injury during sepsis via autonomic pathways and adrenergic signaling. METHODS: Glutamatergic neurons in the mPFC were selectively manipulated

Mechanisms underlying the efficacy of a rodent model of vertical sleeve gastrectomy - A focus on energy expenditure.

OBJECTIVE: Bariatric surgery remains the only effective and durable treatment option for morbid obesity. Vertical Sleeve Gastrectomy (VSG) is currently the most widely performed of these surgeries primarily because of its proven efficacy in generating rapid onset weight loss, improved glucose regulation and reduced mortality compared with other invasive procedures. VSG is associated with reduced appetite, however, the relative importance of energy expenditure to VSG-induced weight loss and changes in glucose regulation, particularly that in brown adipose tissue (BAT), remains unclear. The aim

Increased pulmonary vascular permeability and oedema induced by intrathecally injected endothelins in rat.

The intrathecal (i.t.) injection of endothelins to conscious rats was found to cause respiratory arrest. To gain some insights into this central phenomenon, peripheral vascular permeability and lung oedema were measured after i.t. and i.v. injections of these peptides. When injected at T-8 spinal cord level, endothelin-1 (65 and 650 pmol) and endothelin-3 (650 pmol) enhanced vascular permeability in the lungs by 22-fold and 7-fold, respectively, and caused sudden death at the highest dose. Less prominent increases (between 1.4- and 2.2-fold) of vascular permeability were observed in other tiss

Brain-death-induced cardiac contractile dysfunction: studies of possible neurohormonal and blood-borne mediators.

The aim of these studies was to investigate the mechanism underlying the haemodynamic changes associated with brain death. The initial series of studies were to assess whether these changes involved some blood-borne factor. When control rats (n = 6) were exsanguinated whilst being simultaneously transfused with blood from rats that had been brain-dead for 60 min, their haemodynamic function did not deteriorate. Likewise, when brain-dead rats (n = 6) were exsanguinated and transfused with blood from control rats there was no improvement of haemodynamic function. The absence of any blood-borne f

[Effects of Compound Dihuang Granules on PERK/ATF4/CHOP pathway in substantia nigra of rat model of Parkinson's disease with syndrome of Yin deficiency and stirring wind].

This study aims to investigate the therapeutic mechanism of Compound Dihuang Granules in a rat model of Parkinson's disease(PD) with the syndrome of Yin deficiency and stirring wind, by examining their effects on the expression of activating transcription factor 4(ATF4), CCAAT/enhancer-binding protein homologous protein(CHOP), eukaryotic initiation factor 2 alpha(eIF2α), and protein kinase RNA-like endoplasmic reticulum kinase(PERK) in the substantia nigra. A rat model of PD with the syndrome of Yin deficiency and stirring wind was established via unilateral injection of 6-hydroxydopamine into

Genetically encoded nAChR upregulation is neuroprotective in female parkinsonian mice.

Parkinson's disease is projected to rise to pandemic proportions by 2050, which has resulted in an urgent need for disease-modifying treatments. In this regard, we previously showed that in a mouse model of parkinsonism with unilateral 6-hydroxydopamine (6-OHDA) injection into the dorsolateral striatum (DLS), low doses of the neuronal nicotinic acetylcholine receptor (nAChR) partial agonist and smoking cessation drug, cytisine exerts sex-specific neuroprotection in substantia nigra pars compacta (SNc) dopaminergic (DA) neurons of only female mice by reducing apoptotic endoplasmic reticulum (ER

Role of the iNOS isoform in the cardiovascular dysfunctions of male rats with 6-OHDA-induced Parkinsonism.

INTRODUCTION: Available studies have shown the involvement of nitric oxide (NO) in the processes that lead to neurodegeneration in Parkinson's disease (PD). Also, the use of inhibitors of the inducible isoform of NO-synthase (iNOS) promotes neuroprotection and attenuates dopamine (DA) loss in experimental models of Parkinsonism. In addition, NO also appears to be involved in cardiovascular changes in 6-hydroxydopamine (6-OHDA)-induced Parkinsonism. The current study aimed to evaluate the effects of iNOS inhibition on cardiovascular and autonomic function in animals that were subjected to Parki

[History of deprenyl--the first selective inhibitor of monoamine oxidase type B].

(-)Deprenyl (Selegiline, Jumex, Eldepryl, Movergan), a close structural relative of phenylethylamine (PEA), is a drug with a unique pharmacological spectrum. Whereas PEA and its long-lasting variants, the amphetamines, are mixed-acting stimulants of the sympathetic system in the brain, they primarily enhance the impulse propagation generated release of catecholamines (catecholamine activity enhancer, CAE, effect) and displace catecholamines in higher concentration (catecholamine releasing effect). (-)Deprenyl is the first CAE substance in clinical use devoid of catecholamine releasing activity

Chronic alterations in dopaminergic neurotransmission produce a persistent elevation of deltaFosB-like protein(s) in both the rodent and primate striatum.

Using an antibody that recognizes the products of all known members of the fos family of immediate early genes, it was demonstrated that destruction of the nigrostriatal pathway by 6-hydroxydopamine (6-OHDA) lesions of the medial forebrain bundle produces a prolonged (>3 months) elevation of Fos-like immunoreactivity in the striatum. Using retrograde tract tracing techniques, we have previously shown that this increase in Fos-like immunoreactivity is located predominantly in striatal neurons that project to the globus pallidus. In the present study, Western blots were performed on nuclear extr
