Nucleic acid and protein metabolism in undernutrition and protein deficiency.

This review discusses the metabolism of nucleic acids and proteins in various models of undernutrition in female rats and their neonatal and 21-day-old progeny. Based on the observations noted in our laboratories and those of other investigators, it is concluded that body and organ weights as well as various parameters of cellular growth (DNA, RNA, proteins, amino acids and total nucleotides) fail to increase normally in dietary-insulted animals. Protein and RNA synthesis demonstrate variable responses, leading to the speculation that modulation of mRNA metabolism and of protein synthesis occu

Advancing a Pre-Clinical Model of Full-Term Prenatal Exposure to Vaporized Δ9-Tetrahydrocannabinol.

INTRODUCTION: Use of cannabinoid vape products during pregnancy is a major public health concern. Δ9-tetrahydrocannabinol (THC), the main psychoactive constituent in cannabis, is rapidly absorbed after inhalation and transferred to the fetal brain. We developed a novel model of prenatal cannabinoid exposure (PCE) comprising THC vapor exposure across the human-equivalent full-term prenatal period in rats. This study examined gestational/parturitional outcomes and the distribution and metabolism of THC in maternal and filial blood and brains using this model. MATERIALS AND METHODS: Pregnant fem

Field evaluation of visual endoscope-assisted transcervical artificial insemination in goats: Effects on insemination time and pregnancy outcomes under tropical conditions.

BACKGROUND AND AIM: Artificial insemination (AI) in goats is constrained by the complex cervical anatomy, which limits the efficiency of conventional transcervical AI (C-TCAI), particularly under field conditions. Although laparoscopic AI (LAI) achieves higher fertility rates, its invasive nature, need for anesthesia, and high operational costs limit its routine application. Visual endoscope-assisted transcervical AI (VE-TCAI) offers a minimally invasive alternative that enables real-time cervical visualization and potentially improves procedural efficiency. This study evaluated the field perf

Dietary fiber supplementation during late gestation affects steroidogenesis, antioxidant capacity, gut microbiota and placental metabolomics of Rongchang sows.

Oxidative stress can impair placental function and sow health. This experiment aimed to explore the impacts of inulin and cellulose as sources of dietary fiber (DF) on reproductive performance, steroidogenesis, antioxidant capacity, placental metabolomics, and fecal microbiota of Rongchang sows from d 85 of gestation to parturition. Sixty sows (parity: 5.07 ± 0.24, backfat thickness: 43.06 ± 0.78 mm) were randomly assigned to 4 groups (n = 15): the control group fed a basal diet (CON), and the other 3 groups receiving diets supplemented with 21.8%, 43.6% or 65.5% more DF than the CON group, na

Neonatal overfeeding attenuates microgliosis and hippocampal damage in an infant rat model of pneumococcal meningitis.

BACKGROUND: Pneumococcal meningitis (PM) triggers apoptotic neuronal and progenitor cell death in the hippocampal dentate gyrus (DG), resulting in subsequent cognitive impairment. Microglia play a crucial role in PM-induced hippocampal damage. While the lasting effects of neonatal nutrition on health are well documented, the influence of early-life overfeeding on the host response to neuroinfections remains uncertain. This study aimed to examine whether neonatal overfeeding affects the outcome of PM in the hippocampus (HC). MATERIAL AND METHODS: Overfeeding was induced by adjusting litter siz

Preserved oxytocin-induced myometrium contraction and sensitivity to progesterone inhibition following rat uterus thermal insult. Impact on fertility.

Women seeking improved fertility often undergo diagnostic hysteroscopy that could cause uterine thermal injury with unclear impact on uterine contraction, embryo implantation and fertility. We tested whether uterine thermal insult adversely affects myometrium function and contraction related receptors, channels, junctional proteins and remodeling enzymes. Female Sprague-Dawley rats were anesthetized, the left uterine horn was infused with 85 ℃ hot saline (thermal Insult) and the right horn was infused with 25℃ warm saline (control) for 3 min. After 7-days recovery, uterine strips were prepared

The selective progesterone receptor modulator-promegestone-delays term parturition and prevents systemic inflammation-mediated preterm birth in mice.

BACKGROUND: Progesterone, acting via its nuclear receptors called progesterone receptors, promotes myometrial relaxation during pregnancy, and suspension of this activity triggers labor. We previously found that 20α-hydroxysteroid dehydrogenase causes a local withdrawal of progesterone in the term and preterm myometrium by converting the progesterone into an inactive form before it accesses the progesterone receptors. OBJECTIVE: We hypothesized that a selective progesterone receptor modulator called promegestone, which is not metabolized by 20α-hydroxysteroid dehydrogenase, would sustain prog

Prenatal exposure to aflatoxin B1: developmental, behavioral, and reproductive alterations in male rats.

Previous studies have shown that aflatoxin B1 (AfB1) inhibits androgen biosynthesis as a result of its ability to form a high-affinity complex with the steroidogenic acute regulatory protein. The results of the present study demonstrate the postnatal effects of in utero exposure to AfB1 in the rat. Pregnant Wistar rats were given 10, 20, or 50 μg AfB1/kg body weight daily from gestation day (GD) 12 to GD 19. At parturition, newborns were observed for clinical signs and survival. All animals were born alive and initially appeared to be active. Male pups from control and AfB1-exposed animals wer

Metabolic maturity at birth and neonate lamb survival and growth: the effects of maternal low-dose dexamethasone treatment.

Perinatal mortality is a major contributing factor to reproductive wastage in grazing sheep industries. Enhanced metabolic and endocrine maturity at birth may improve the behavioral competency and thermoregulatory ability of neonates, potentially improving lamb survival over the first 72 h of life. Maternal glucocorticoid treatment in late gestation was investigated as a mechanism for manipulating metabolic and endocrine maturity in the ovine neonate. Multiparous, fine-wool Merino ewes (n = 150) were divided into 3 groups to lamb on pasture. Within each group, 5 single-lamb and 5 twin-lamb bea

Excess Maternal Fructose Consumption Increases Fetal Loss and Impairs Endometrial Decidualization in Mice.

The most significant increase in metabolic syndrome over the previous decade occurred in women of reproductive age, which is alarming given that metabolic syndrome is associated with reproductive problems including subfertility and early pregnancy loss. Individuals with metabolic syndrome often consume excess fructose, and several studies have concluded that excess fructose intake contributes to metabolic syndrome development. Here, we examined the effects of increased fructose consumption on pregnancy outcomes in mice. Female mice fed a high-fructose diet (HFrD) for 6 weeks developed glucose

The effects of strain and prenatal nicotine exposure on ethanol consumption by adolescent male and female rats.

Two studies of variables affecting voluntary ethanol consumption by adolescent male and female rats are reported. Sprague-Dawley (SD) and spontaneously hypertensive rats (SHRs) were compared in Experiment 1. Starting on postnatal day 30 all had 24-h access to 2%, then 4%, and then 6% ethanol, followed by 1-h access to the 6% until intake stabilized. During the 1-h access SHR females consumed more ethanol than all other groups. In Experiment 2, the same procedure was used to compare SD groups prenatally exposed to nicotine, with controls. Nicotine-exposed females consumed more ethanol during 1-

Reduced fertility in an adenomyosis mouse model is associated with an altered immune profile in the uterus during the implantation period.

STUDY QUESTION: Does a reduction in fertility and/or systemic immune cell change occur during the early implantation period in a mouse model of adenomyosis? SUMMARY ANSWER: A reduction in fertility was observed in mice with adenomyosis, coinciding with local and systemic immune changes observed during the implantation period. WHAT IS KNOWN ALREADY: Adenomyosis is a pathology responsible for impaired fertility in humans, with a still unclear pathophysiology. One hypothesis is that changes in immune cells observed in adenomyosis-affected uteri may alter fertility, notably the physiological imm

Optimal dietary energy and protein levels for breeding pigeons in the winter "2 + 3" lactation pattern.

The nutritional requirements of breeding pigeons depend on their physiological period, breeding pattern, and environmental conditions. Despite works on reduced litter size in winter production to combat high mortality and the poor welfare of squabs, there are few studies on the related nutritional requirements of these pigeons. A total of 432 pairs of European Mimas pigeons were randomly divided into 9 groups in which 3 crude protein (CP) levels (15, 16.5, and 18%) and 3 metabolizable energy (ME) levels (12.2 MJ/kg, 12.4 MJ/kg, and 12.6 MJ/kg) were tested to determine the optimal energy and pr

The Sex Chromosome Trisomy mouse model of XXY and XYY: metabolism and motor performance.

BACKGROUND: Klinefelter syndrome (KS), caused by XXY karyotype, is characterized by low testosterone, infertility, cognitive deficits, and increased prevalence of health problems including obesity and diabetes. It has been difficult to separate direct genetic effects from hormonal effects in human studies or in mouse models of KS because low testosterone levels are confounded with sex chromosome complement. METHODS: In this study, we present the Sex Chromosome Trisomy (SCT) mouse model that produces XXY, XYY, XY, and XX mice in the same litters, each genotype with either testes or ovaries. Th

Induction of enrichment of the stages of the seminiferous epithelial cycle in prepubertal rats.

The aim of the present study was to examine the feasibility of inducing enrichment of the stages of the seminiferous epithelium in prepubertal male rats produced by vitamin A-deficient (VAD) females that had received a supplement of a marginal level of retinol. Seventy-day-old retinoic acid-supplemented VAD female rats were given daily doses of 5, 10, or 20 micrograms retinol for 2 wk. Breeding of these females resulted in the production of viable offspring in litters of normal size. Subsequently, male rats so produced (VAD males) were given a single injection of 10 or 20 micrograms retinol at

Ultrastructure of Met5-enkephalin terminals in the caudate-putamen nuclei of adult rats receiving neonatal intranigral 6-hydroxydopamine.

Destruction of dopamine neurons of the nigrostriatal pathway in the early postnatal rat enhances the levels of Met5-enkephalin in the adult dorsal striatum (caudate-putamen nuclei) and may contribute to the abnormal self-injurious behavior seen in humans with Lesch-Nyhan disease. We examined whether there were ultrastructural changes in Met5-enkephalin immunoreactive terminals in the rat model that might reflect cellular sites for enhanced activity of these opioid neurons. At 3 days postnatal, 10-20 nl injections of a 1% solution of the dopamine neurotoxin, 6-hydroxydopamine (6-OHDA), or vehic

An extended evaluation of an in vivo teratology screen utilizing postnatal growth and viability in the mouse.

A teratology test system proposed by Chernoff and Kavlock utilizes growth and viability for 3 days after birth to prioritize chemicals for standard teratology testing. The present study is an extended observation of the growth and viability of Chernoff and Kavlock's animals from 41 treatments for 250 days to determine if neonatal weight reductions persisted throughout life, if mortality occurred later in life or, if other serious abnormalities developed that could not be anticipated from the neonatal data. One chemical which was positive in the present study would be a false negative in the st
