Diversity and seasonal dynamics of bacterial community in indoor environment.

BACKGROUND: We spend most of our lives in indoor environments and are exposed to microbes present in these environments. Hence, knowledge about this exposure is important for understanding how it impacts on human health. However, the bacterial flora in indoor environments has been only fragmentarily explored and mostly using culture methods. The application of molecular methods previously utilised in other environments has resulted in a substantial increase in our awareness of microbial diversity. RESULTS: The composition and dynamics of indoor dust bacterial flora were investigated in two bu

Anti-acne cinnamic acid bigel: a hybrid topical system for concurrent modulation of inflammation and microbial load.

BACKGROUND: Acne vulgaris is a chronic inflammatory skin disease aggravated by Cutibacterium acnes (P. acnes). Conventional drug delivery treatments researchers reported irritation, resistance and poor efficacy. Poor skin permeability, along with oxidative degradation hindering its efficacy on Dermal drug delivery application. PURPOSE: In this study, the novel work was to incorporate and optimize cinnamic acid phytoconstituent within a bigel system to improve solubility, stability, permeation and achieve prolonged release. STUDY DESIGN: Box-Behnken design was used to optimize formulation, in

Multifunctional mesoporous superparamagnetic Iron oxide-based nanocomposites for synergistic Photothermal/Fenton therapy in the treatment of acne vulgaris.

Acne vulgaris is a common chronic inflammatory dermatosis primarily mediated by the dysregulated proliferation of Cutibacterium acnes (C. acnes). The current therapeutic efficacy is severely compromised by the widespread antibiotic resistance of C. acnes. To overcome this limitation, we designed mesoporous superparamagnetic iron oxide (MSPIONs) nanoparticles through an innovative one-step bubble-templated approach, and then the PMS (phase change material modified MSPIONs) loaded with CuO and Quercetin (Que) nanocomposites (PMS/CuO/Que) were synthesized. PMS/CuO/Que. exhibited remarkable biocom

Novel-designed antimicrobial peptides with dual antimicrobial and anti-inflammatory actions against Cutibacterium acnes for acne vulgaris therapy.

Acne vulgaris is a prevalent skin condition among adolescents, primarily instigated by over-colonization and subsequent inflammation triggered by Cutibacterium acnes. Although topical and oral antibiotics are standard treatments, they often lead to the proliferation of antibiotic-resistant bacteria and are associated with undesirable side effects. Antimicrobial peptides (AMPs) are considered a promising solution to these challenges. In this study, we aimed to develop novel short AMPs to combat C. acnes. By comparing sequences and abstracting the distribution of residue types of established AMP

Propionic acid produced by Cutibacterium acnes fermentation ameliorates ultraviolet B-induced melanin synthesis.

Ultraviolet irradiation induces melanin accumulation, which can be reduced by the use of chemical whitening products. However, the associated safety concerns of such products have prompted the search for natural and harmless alternatives. This study aimed to identify a natural acidic formulation to reduce skin pigmentation. The metabolite propionic acid (CH3CH2COOH, PA) was the most abundant fatty acid in the filtrate from Pluronic F68 (PF68) fermentation of Cutibacterium acnes (C. acnes) and reduced the DOPA-positive melanocytes by significantly inhibiting cellular tyrosinase activity via bin

PEG-8 Laurate Fermentation of Staphylococcus epidermidis Reduces the Required Dose of Clindamycin Against Cutibacterium acnes.

The probiotic activity of skin Staphylococcus epidermidis (S. epidermidis) bacteria can elicit diverse biological functions via the fermentation of various carbon sources. Here, we found that polyethylene glycol (PEG)-8 Laurate, a carbon-rich molecule, can selectively induce the fermentation of S. epidermidis, not Cutibacterium acnes (C. acnes), a bacterium associated with acne vulgaris. The PEG-8 Laurate fermentation of S. epidermidis remarkably diminished the growth of C. acnes and the C. acnes-induced production of pro-inflammatory macrophage-inflammatory protein 2 (MIP-2) cytokines in mice

Genome-wide meta-analysis implicates mediators of hair follicle development and morphogenesis in risk for severe acne.

Acne vulgaris is a highly heritable common, chronic inflammatory disease of the skin for which five genetic risk loci have so far been identified. Here, we perform a genome-wide association study of 3823 cases and 16,144 controls followed by meta-analysis with summary statistics from a previous study, with a total sample size of 26,722. We identify 20 independent association signals at 15 risk loci, 12 of which have not been previously implicated in the disease. Likely causal variants disrupt the coding region of WNT10A and a P63 transcription factor binding site in SEMA4B. Risk alleles at the

Resistin gene polymorphisms are associated with acne and serum lipid levels, providing a potential nexus between lipid metabolism and inflammation.

Acne vu lgaris is a multifactorial inflammatory skin disease causing social stigma and psychological effect on patients. We hypothesized that the genes that can affect both lipid metabolism and inflammation may be central for acne formation and present targets for treatment. Pro-inflammatory adipokine resistin, one such likely target, activates NFkB and JNK pathways inducing TLR-2, IL-1, IL-6, and TNFα genes. The polymorphisms in promoter and intron region of the resistin gene affect its expression levels. Therefore, we explored the association of resistin polymorphisms (RETN +299G > A and -42

Streptococcal cell wall induced arthritis: leukocyte activation in extra-articular lymphoid tissue.

Rheumatoid arthritis (RA) is a common systemic inflammatory disease thought to be T-helper-1 cell driven, though current controversy involves the relative role of T cells versus other leukocytes. Thus, there is a need for better understanding of the role of various leukocytes and their subsets in RA. Using the streptococcal cell wall (SCW) induced arthritis model, we examined leukocytes isolated from peripheral blood, spleen, and lymph nodes using monoclonal antibodies directed against lineage specific cell surface markers. Activation status of these cells was assessed using CD44 and CD71 as m

Mediastinitis after cardiac surgery: improvement of bacteriological diagnosis by use of multiple tissue samples and strain typing.

The diagnosis of postsurgical mediastinitis (PSM) among patients with sternal wound complication (SWC) after cardiac surgery is sometimes difficult, as fever, elevated C-reactive protein levels, and chest pain can be caused by a general inflammatory reaction to the operative trauma and/or sternal dehiscence without infection. The definitions of PSM usually used emphasize clinical signs and symptoms easily observed by the surgeon. The aim of the study was to investigate whether the use of standardized multiple tissue sampling, optimal culturing methods, and strain typing, together with a microb

Natural cell-mediated cytotoxicity in rats. II. In vivo augmentation of NK-cell activity.

Natural cell-mediated cytotoxicity in rats as well as in mice has been shown to vary consistently with age, with peak levels detectable at 5-10 weeks. The levels of cell-mediated cytotoxicity against tumor cells could be augmented in strains of inbred rats with either high or low levels of natural reactivity, by IP injection of a variety of agents, including C. parvum, LCMV, KRV, and poly I:C. The specificity of the augmented cytotoxicity appeared to be the same as the specificity of natural killer cells which are found in normal rat spleen cells. Similarly, the cells mediating the augmented c

Experimental cutaneous leishmaniasis. II. Effects of immunosuppression and antigenic competition on the course of infection with Leishmania enriettii in the guinea-pig.

Intradermal inoculation of the guinea-pig with Leishmania enriettii results in a self-healing cutaneous lesion which provides a laboratory model of human cutaneous leishmaniasis and which is dominated by cell-mediated immunological responses (Bryceson et al., 1970). In this study we sought to design experimental situations resembling non-healing forms of cutaneous leishmaniasis in man and to determine whether these experimental situations were accompanied by abnormalities in the immunological response to infection. This paper describes three procedures which impair the resistance of guinea-pig

Comparison of Blood Culture Contamination Rates Between Initial Central Venous Puncture and Catheter Hub Sampling.

OBJECTIVES: Blood cultures are essential for guiding antimicrobial therapy, but contamination remains a major challenge, particularly when samples are drawn from central venous catheters (CVCs). Blood cultures obtained from CVCs, even when newly inserted, show higher contamination rates than those from peripheral venipuncture, likely due to translocation of skin bacteria during skin dilation and catheter insertion. In critically ill patients with suspected infection or difficult peripheral venous access, CVC insertion often coincides with the indication for blood culture sampling. We hypothesi

Cutibacterium acnes periprosthetic joint infections.

AIMS: Cutibacterium acnes (C. acnes; previously known as Propionibacterium acnes or P. acnes) periprosthetic hip and knee infections are under-reported. While culture contamination with C. acnes occurs, true infections are important to recognize and treat. We sought to describe the demographics and treatment outcomes of patients with C. acnes periprosthetic joint infections (PJIs) of the hip and knee. METHODS: Patients with C. acnes PJI between January 2005 and December 2018 were retrospectively reviewed utilizing the institutional total joint registry. Patients with monomicrobial PJI and two
