Erythroid induction of chronic myelogenous leukemia K562 cells following treatment with a photoproduct derived from the UV-A irradiation of 5-methoxypsoralen.

Induction of terminal erythroid differentiation can be an efficient strategy to inhibit proliferation of chronic myelogenous leukemia cells. Psoralens, well-known photo-chemotherapeutic agents, were found to be efficient at inducing erythroid differentiation of K562 cells, an in vitro cell line isolated from the pleural effusion of a patient with chronic myelogenous leukemia in blast crisis. The effects of crude pre-irradiated solutions of 5-methoxypsoralen on erythroid differentiation of human leukemic K-562 cells were evaluated. The major photoproduct was characterized and analyzed, and it w

Dissection of three Ca2+-dependent steps leading to secretion in chromaffin cells from mouse adrenal slices.

In neurosecretory cells, intracellular Ca2+ ([Ca2+]i) not only acts as the trigger for secretion but also regulates earlier steps in the secretory pathway. Here, a novel approach was developed to control [Ca2+]i over a broad concentration range, which allowed the quantification of three distinct actions of [Ca2+]i on large dense-core vesicle (LDCV) fusion in chromaffin cells from mouse adrenal slices. Basal [Ca2+]i regulated the transfer of vesicles toward a slowly releasable state, whereas further maturation to the readily releasable state was Ca2+ independent. [Ca2+]i levels above 3 microM t

Photochemical initial reaction mechanism of the ε-CL-20 explosive: a molecular-level study based on TDDFT.

Hexanitrohexaazaisowurtzitane (CL-20) is a pure explosive with the highest energy density, and its environmental release and photochemical transformation have attracted considerable attention. However, the initial reaction mechanism under photoexcitation remains unclear. Time-dependent density functional theory (TDDFT) was employed to systematically investigate the photochemical reaction pathways, bond cleavage sequence, and reaction mechanisms of ε-CL-20 in this study. Electron density difference analysis reveals that the ground-state to first-excited-state transition for all five polymorphs

Nanotitania crystals induced efficient photocatalytic color degradation, antimicrobial and larvicidal activity.

Textile industries release tonnes of harmful toxic dyes into the environment, causing severe effects on living organisms, including humans. Mosquitoes vectors spread important diseases which cause millions of human deaths worldwide. To control mosquitoes a number of synthetic mosquitocidal agents have been employed but all these pesticides pose harmful effects to human health and non-target species and also led to resistance development in treated vectors. Microbial strains are also developing resistance to the available antibiotics, this currently represents a major public health challenge. T

Ah receptor agonists in UV-exposed toluene solutions of decabromodiphenyl ether (decaBDE) and in soils contaminated with polybrominated diphenyl ethers (PBDEs).

GOAL, SCOPE AND BACKGROUND: The use of polybrominated diphenyl ethers (PBDEs) as flame retardants increases the risk for emissions of other brominated compounds, such as polybrominated dibenzodioxins (PBDDs) and dibenzofurans (PBDFs). The large homology in structure of PBDD/Fs and mechanism of toxic action, i.e. the capacity to activate the Ah receptor (AhR) pathway, compared to their well-studied chlorinated analogues, justifies a raised concern to study the environmental levels and fate of these compounds. Decabromodiphenyl ether (decaBDE) is the most widely used PBDE today. Studies on photo

Photolytic degradation of benorylate: effects of the photoproducts on cultured hepatocytes.

The photodegradation of benorylate [4'-(acetamido)phenyl-2-acetoxybenzoate], a drug frequently used in rheumatoid arthritis therapy, has been examined under different sets of experimental conditions. Several photoproducts have been isolated and identified on the basis of their IR, NMR, and MS spectra. The most significant photochemical process is the photo-Fries rearrangement of benorylate, leading to 5-acetamido-2'-acetoxy-2-hydroxybenzophenone (1). This compound undergoes a rapid transacylation to the isomeric 5'-acetamido-2'-acetoxy-2-hydroxybenzophenone (2). A primary culture of rat hepato

Effect of Zinc Precursors on the Antibacterial and Photocatalytic Applications of Green Synthesized ZnO Nanostructures Using Calluna vulgaris.

In this study, we report the environmentally friendly green synthesis of zinc oxide (ZnO) nanostructures using Calluna vulgaris (C. vulgaris) leaves extract. The extract was used as a reductant and stabilizer in aqueous medium instead of chemicals and two different precursors, zinc acetate dihydrate and zinc nitrate hexahydrate, were used to synthesize ZnO-Acetate and ZnO-Nitrate nanostructures, respectively. The optical, structural, and morphological characteristics of the green synthesized ZnO nanostructures were investigated by UV-Visible Spectroscopy (UV-Vis), X-ray diffraction (XRD), and

Development of a mechanistic SAR model for the detection of phototoxic chemicals and use in an integrated testing strategy.

Phototoxicity is of increasing concern in dermatology, since modern lifestyle is often associated with exposure to sunlight. The most commonly reported process is via oxidative reactions. Therefore characterizing the "photo-pro-oxidant" potential of a compound early in its industrial development is of utmost interest, especially for compounds likely to undergo sunlight exposure in skin. Today there is a need for filtering compounds to be tested in the 3T3 neutral red uptake in vitro test for phototoxicity since testing requires resources. A computational model aiming at predicting the mechanis

An evaluation of chemical photoreactivity and the relationship to photogenotoxicity.

In the accompanying paper (Kleinman et al., submitted for publication), the link between phototoxicity and photoreactivity (i.e. production of singlet oxygen, superoxide and chemical photostability) has been established. It is proposed this may be used to refine existing triggers for photosafety testing. Using a series of compounds we have determined whether these photochemical reactivity measurements may be used to mechanistically predict phototoxic and/or photogenotoxic liability. Therefore, a subset of compounds tested in the in vitro 3T3 NRU assay from the accompanying paper were tested in

A close association of RyRs with highly dense clusters of Ca2+-activated Cl- channels underlies the activation of STICs by Ca2+ sparks in mouse airway smooth muscle.

Ca(2+) sparks are highly localized, transient releases of Ca(2+) from sarcoplasmic reticulum through ryanodine receptors (RyRs). In smooth muscle, Ca(2+) sparks trigger spontaneous transient outward currents (STOCs) by opening nearby clusters of large-conductance Ca(2+)-activated K(+) channels, and also gate Ca(2+)-activated Cl(-) (Cl((Ca))) channels to induce spontaneous transient inward currents (STICs). While the molecular mechanisms underlying the activation of STOCs by Ca(2+) sparks is well understood, little information is available on how Ca(2+) sparks activate STICs. In the present stu

Six-coordinate and five-coordinate Fe(II)(CN)(2)(CO)(x) thiolate complexes (x = 1, 2): synthetic advances for iron sites of [NiFe] hydrogenases.

The dicyanodicarbonyliron(II) thiolate complexes trans,cis-[(CN)(2)(CO)(2)Fe(S,S-C-R)](-) (R = OEt (2), N(Et)(2) (3)) were prepared by the reaction of [Na][S-C(S)-R] and [Fe(CN)(2)(CO)(3)(Br)](-) (1). Complex 1 was obtained from oxidative addition of cyanogen bromide to [Fe(CN)(CO)(4)](-). In a similar fashion, reaction of complex 1 with [Na][S,O-C(5)H(4)N], and [Na][S,N-C(5)H(4)] produced the six-coordinate trans,cis-[(CN)(2)(CO)(2)Fe(S,O-C(5)H(4)N)](-) (6) and trans,cis-[(CN)(2)(CO)(2)Fe(S,N-C(5)H(4))](-) (7) individually. EPR studies of amine radical cations. Part 2. Thermal and photo-induced rearrangements of propargylamine and allylamine radical cations in low-temperature freon matrices.

Matrix EPR studies and quantum chemical calculations have been used to characterize the consecutive H-atom shifts undergone by the nitrogen-centered parent radical cations of propargylamine (1b*+) and allylamine (5*+) on thermal or photoinduced activation. The radical cation rearrangements of these unsaturated parent amines occur initially by a 1,2 H-atom shift from C1 to C2 with pi-bond formation at the positively charged nitrogen; this is followed by a consecutive reaction involving a second H-atom shift from C2 to C3. Thus, exposure to red light (lambda > 650 nm) converts 1b*+ to the vinyl-

Biochemical reactivity of melatonin with reactive oxygen and nitrogen species: a review of the evidence.

Melatonin (N-acetyl-5-methoxytryptamine), an endogenously produced indole found throughout the animal kingdom, was recently reported, using a variety of techniques, to be a scavenger of a number of reactive oxygen and reactive nitrogen species both in vitro and in vivo. Initially, melatonin was discovered to directly scavenge the high toxic hydroxyl radical (*OH). The methods used to prove the interaction of melatonin with the *OH included the generation of the radical using Fenton reagents or the ultraviolet photolysis of hydrogen peroxide (H202) with the use of spin-trapping agents, followed

Degradation of the antibiotic oxolinic acid by photocatalysis with TiO2 in suspension.

In the work presented here, a photocatalytic system using titanium Degussa P-25 in suspension was used to evaluate the degradation of 20mg L(-1) of antibiotic oxolinic acid (OA). The effects of catalyst load (0.2-1.5 g L(-1)) and pH (7.5-11) were evaluated and optimized using the surface response methodology and the Pareto diagram. In the range of variables studied, low pH values and 1.0 g L(-1) of TiO(2) favoured the efficiency of the process. Under optimal conditions the evolution of the substrate, chemical oxygen demand, dissolved organic carbon, toxicity and antimicrobial activity on Esche

Photoreduction of methemoglobin by irradiation in the near-ultraviolet region.

Ferric metHb can be photoreduced to the ferrous state by direct photoexcitation in the near-ultraviolet region. In this research, we studied the mechanism and facilitating conditions for the photoreduction and the resulting restoration of O(2) binding. MetHb in phosphate-buffered saline or pure water in a CO atmosphere was photoreduced to form HbCO by illuminating the N band (365 nm), one of the porphyrin pi --> pi transitions, whereas the photoreduction did not occur in Ar, N(2), or O(2). The transient absorption spectrum exhibited the generation of deoxyHb within 30 ns in both the CO and Ar

Nabumetone induced photogenotoxicity mechanism mediated by ROS generation under environmental UV radiation in human keratinocytes (HaCaT) cell line.

Nabumetone (NB) is a non-steroidal anti-inflammatory drug (NSAID), prescribed for managing pain associated with acute/chronic rheumatoid arthritis, osteoarthritis and other musculoskeletal disorders. Though some incidences of photosensitivity have been reported, there is limited information available on its phototoxicity potential. In this study, NB photodegraded in a time-dependant manner (0-4 h) under UVA (1.5 mW/cm2), UVB (0.6 mW/cm2) and natural sunlight as observed through UV-vis spectrophotometer and the results were further confirmed with Ultra High-Performance Liquid Chromatography (UH

Exploiting natural chemical photosensitivity of anhydrotetracycline and tetracycline for dynamic and setpoint chemo-optogenetic control.

The transcriptional inducer anhydrotetracycline (aTc) and the bacteriostatic antibiotic tetracycline (Tc) are commonly used in all fields of biology for control of transcription or translation. A drawback of these and other small molecule inducers is the difficulty of their removal from cell cultures, limiting their application for dynamic control. Here, we describe a simple method to overcome this limitation, and show that the natural photosensitivity of aTc/Tc can be exploited to turn them into highly predictable optogenetic transcriptional- and growth-regulators. This new optogenetic class

Midazolam is a short-acting benzodiazepine central nervous system depressant available as an injection, tablet, or oral syrup. The need for alternative dosage form options for patients unable to take tablets and shortages of other forms of the drug have led compounding pharmacies to seek alternatives, mainly solutions and suspensions. Additionally, some patients are unable to use suspending agents containing alcohol or sorbitol. The objective of this study was to determine the stability of midazolam in sorbitol-free, alcohol-free SyrSpend SF and SyrSpend SF Cherry suspending agents. The studie
