Urolithin A prevents age-related hearing loss in C57BL/6J mice likely by inducing mitophagy.

Mitochondrial dysfunction with aging is associated with the development of age-related hearing loss. Mitophagy is a cardinal mechanism to maintain a healthy mitochondrial population through the turnover of damaged mitochondria. Declining mitophagy with age causes a buildup of damaged mitochondria, leading to sensory organ dysfunction. The effect of Urolithin A (UA), a mitophagy inducer, was investigated on age-related hearing loss in a mouse model. C57BL/6J mice were treated with UA from 6 to 10 months of age. UA attenuated an auditory brainstem responses (ABR) threshold shift at 8, 16, and 32

A Novel Method for Classifying Hearing Impairment in Epidemiological Studies of Aging: The Wisconsin Age-Related Hearing Impairment Classification Scale.

Purpose Longitudinal population-based cohort data were used to develop a standardized classification system for age-related hearing impairment using thresholds for frequencies (0.5-8 kHz) typically measured in cohort studies. Method Audiometric testing data collected in the Epidemiology of Hearing Loss Study from participants (n = 1,369) with four visits (1993-1995, 1998-2000, 2003-2005, and 2009-2010) were included (10,952 audiograms). Cluster analyses (Wald's method) were used to identify audiometric patterns. Maximum allowable threshold values were defined for each cluster to create an orde

Afferent-efferent connectivity between auditory brainstem and cortex accounts for poorer speech-in-noise comprehension in older adults.

Speech-in-noise (SIN) comprehension deficits in older adults have been linked to changes in both subcortical and cortical auditory evoked responses. However, older adults' difficulty understanding SIN may also be related to an imbalance in signal transmission (i.e., functional connectivity) between brainstem and auditory cortices. By modeling high-density scalp recordings of speech-evoked responses with sources in brainstem (BS) and bilateral primary auditory cortices (PAC), we show that beyond attenuating neural activity, hearing loss in older adults compromises the transmission of speech inf

Effects of Cdh23 single nucleotide substitutions on age-related hearing loss in C57BL/6 and 129S1/Sv mice and comparisons with congenic strains.

A single nucleotide variant (SNV) of the cadherin 23 gene (Cdh23c.753A), common to many inbred mouse strains, accelerates age-related hearing loss (AHL) and can worsen auditory phenotypes of other mutations. We used homologous recombination in C57BL/6 NJ (B6N) and 129S1/SvImJ (129S1) embryonic stem cells to engineer mouse strains with reciprocal single base pair substitutions (B6-Cdh23c.753A>G and 129S1-Cdh23c.753G>A). We compared ABR thresholds and cochlear pathologies of these SNV mice with those of congenic (B6.129S1-Cdh23Ahl+ and 129S1.B6-Cdh23ahl) and parental (B6N and 129S1) strain mice.

Age-related changes in the auditory brainstem response.

PURPOSE: This cross-sectional study had two goals: (1) Identify and quantify the effects of aging on the auditory brainstem response (ABR); (2) Describe how click rate and hearing impairment modify effects of aging. RESEARCH DESIGN AND ANALYSIS: ABR measures were obtained from 131 predominately male Veteran participants aged 26 to 71 yr. Metrics analyzed include amplitude and latency for waves I, III, and V, and the I-V interpeak latency interval (IPI) at three repetition rates (11, 51, and 71 clicks/sec) using both polarities. In order to avoid confounding from missing data due to hearing imp

Presbyacusis and calcium-binding protein immunoreactivity in the cochlear nucleus of BALB/c mice.

The BALB/c mouse is an established model for the early development of sensorineural hearing loss, and is homozygous for the Ahl allele (age-related hearing loss). The present study was designed to determine how auditory peripheral pathology influences calcium-binding protein immunoreactivity in the cochlear nucleus in aged BALB/c mice. To address this issue the loss of hair cells, spiral ganglion neurons (SGN), and neurons in the dorsal (DCN) and posteroventral (PVCN) cochlear nucleus of BALB/c mice at 1 and 24 months of age were quantified using CAST stereological methods. These values were t

A novel Z-score-based method to analyze candidate genes for age-related hearing impairment.

OBJECTIVE: Approximately half of the variance of Age-Related Hearing Impairment (ARHI) is attributable to environmental risk factors, and the other half to genetic factors. None of these genes has ever been identified, but the genes involved in monogenic nonsyndromic hearing impairment are good candidates. Here we define and validate a quantitative trait value for ARHI, correcting for age and gender, to allow the genetic study of ARHI as a quantitative trait. DESIGN: Based on the ISO 7029 standard, we convert audiometric data into a Z-score, an age- and gender-independent value expressing to

Low-frequency specificity of the auditory brainstem response threshold elicited by clicks masked with 1590-Hz high-pass noise in subjects with sloping cochlear hearing losses.

In this study, the frequency specificity of the ABR threshold to stimulation with a click masked with 1590-Hz high-pass noise was determined in subjects with sloping cochlear hearing losses both high- and low-frequency in character. The results show that the ABR threshold elicited by this stimulus is low-frequency specific. The standard error in estimating the 1,000-Hz pure-tone threshold from the high-pass-noise-masked click-evoked ABR threshold is 10.2 dB which equals that for estimating the 3,000-Hz pure-tone threshold from the routinely used unmasked click ABR threshold. The ABR threshold

Binaural interaction and localization with various hearing impairments.

Binaural interaction and localization abilities of hearing-impaired listeners are summarized separately for each of several types of hearing and neurological disorders. The dependence of binaural abilities on the nature of the impairment is emphasized. Unfortunately, available data are not sufficient to characterize the specific aspects of the stimulus information available in each type of impairment.

Chromatin and genomic instability in the cochlea contributing to age-related hearing loss: Insights from in vitro and in vivo models.

Age-related hearing loss (ARHL) is one of the most common causes of hearing impairment in older adults. However, the cellular and molecular mechanisms underlying ARHL remain unclear. Chromatin and genomic instability, including DNA damage, heterochromatin loss, and telomere attrition, are hallmarks of aging. In the present study, we investigated DNA damage levels, chromatin accessibility, telomere length, and telomerase changes in vitro and in vivo mouse models of ARHL. To further explore changes in outer hair cells (OHCs), one of the prominent auditory sensory cells in the cochlea, we constru

Speech based transmission index for all: An intelligibility metric for variable hearing ability.

A method to measure the speech intelligibility in public address systems for normal hearing and hearing impaired persons is presented. The proposed metric is an extension of the speech based Speech Transmission Index to account for accurate perceptual masking and variable hearing ability: The sound excitation pattern generated at the ear is accurately computed using an auditory filter model, and its shapes depend on frequency, sound level, and hearing impairment. This extension yields a better prediction of the intensity of auditory masking which is used to rectify the modulation transfer func

Age dependence of otoacoustic emissions: the loss of amplitude is primarily caused by age-related hearing loss and not by aging alone.

The amplitude of otoacoustic emissions (OAE) is known to decrease with increasing age, but it is still unclear whether this is due to aging alone or to age-related hearing loss. This study describes the exploration of a large database (5,142 patients from 0.4 to 89.8 years) collected in a routine clinical testing. Reliable pure tone audiograms, transitory evoked otoacoustic emissions (TEOAE) and distortion product otoacoustic emissions (DPOAE) recordings were available from 5,424 ears without conductive loss, acute sudden deafness or retrocochlear disorder. From this database, group 1 with beh

Influence of lecithin on mitochondrial DNA and age-related hearing loss.

OBJECTIVES: Lecithin is a polyunsaturated phosphatidylcholine (PPC), which are high energy functional and structural elements of all biologic membranes. PPC play a rate-limiting role in the activation of numerous membrane-located enzymes, including superoxide dismutase and glutathione, which are important antioxidants protecting cell membranes from damage by reactive oxygen species (ROS). ROS-induced damage to mitochondrial DNA may lead to reduced mitochondrial function in the cochlea and resultant hearing loss. STUDY DESIGN AND SETTING: The effects of lecithin on aging and age-associated hea
