A multi-epitope vaccine CTB-UE relieves Helicobacter pylori-induced gastric inflammatory reaction via up-regulating microRNA-155 to inhibit Th17 response in C57/BL6 mice model.

Vaccination is an effective mean of preventing infectious diseases, including those caused by Helicobacter pylori. Th17 cell responses are critical for the pathogenesis of Helicobacter pylori infection. In view of Th17 responses to multi-epitope vaccine CTB-UE, the IL-17 production in antiserum was examined. CTB-UE immunization decreased IL-17 production, implying that Th17 responses may be inhibited. Furthermore, IL-17 aggravated GES-1 cell injury induced by H. pylori SS1; In contrast, CTB-UE antiserum could alleviate this cell injury, which suggesting that CTB-UE can protect GES-1 cell infec

Decreased IgA antibody production in the stomach of gastric adenocarcinoma patients.

Gastric adenocarcinoma is closely associated with Helicobacter pylori infection. It is also much more frequent in patients with common variable immunodeficiency or selective IgA-deficiency than in the general population. To investigate a possible link between local antibody production and gastric tumors, we studied gastric B cell infiltration and local IgA production in patients with H. pylori induced gastric adenocarcinomas. These studies showed that total and H. pylori-specific IgA antibody levels were substantially lower in gastric tissue from the cancer patients compared to those from asym

Evaluation of nested PCR in detection of Helicobacter pylori targeting a highly conserved gene: HSP60.

OBJECTIVE: To comparatively evaluate a new nested set of primers designed for the detection of Helicobacter pylori targeting a highly conserved heat shock protein gene (Hsp60). METHODS: A total of 60 subjects having peptic ulcer diseases were tested for the detection of H. pylori using rapid urease test (RUT), histology, culture, and polymerase chain reaction (PCR) in their antral biopsy specimens. A newly designed Hsp60 gene-based primer set was evaluated against commonly used PCR primers for detection of H. pylori. RESULTS: Forty-six of the 60 study subjects were found positive for culture

Proteus mirabilis isolates of different origins do not show correlation with virulence attributes and can colonize the urinary tract of mice.

Proteus mirabilis has been described as an aetiological agent in a wide range of infections, playing an important role in urinary tract infections (UTIs). In this study, a collection of P. mirabilis isolates obtained from clinical and non-clinical sources was analysed in order to determine a possible correlation between origin, virulence factors and in vivo infectivity. Isolates were characterized in vitro, assessing several virulence properties that had been previously associated with P. mirabilis uropathogenicity. Swarming motility, urease production, growth in urine, outer-membrane protein

Defensin pattern in chronic gastritis: HBD-2 is differentially expressed with respect to Helicobacter pylori status.

BACKGROUND/AIMS: Recent reports have suggested that Helicobacter pylori infection induces the mucosal antibiotic peptide human beta defensin 2 (HBD-2). Therefore, the present study investigated mRNA and peptide expression of four different defensins in the upper gastrointestinal tract in patients with H pylori positive and negative chronic gastritis. MATERIALS/METHODS: Biopsies from the oesophagus to the duodenum were taken during routine gastroscopy in 71 individuals. Total RNA was extracted and the reverse transcription polymerase chain reaction was performed with primers for human defensin

Management of Helicobacter pylori infection in ulcer disease.

Eradication of the bacterium not only heals the ulcer but essentially cures the underlying peptic ulcer disease. Although there is still no simple, all-purpose regimen, a decade of experience has identified safe, well- tolerated drug combinations that yield eradication rates exceeding 90% without generating a high frequency of bacterial resistance.

Significance and regulation of gastric secretion of platelet-activating factor (PAF-acether) in man.

Platelet-activating factor (PAF) has been implicated in the pathogenesis of acute inflammatory and ulcerative diseases of the upper gastrointestinal tract. In the present study, we compared the gastric output of PAF and its precursors with gastric acid output, in patients with various upper gastrointestinal tract diseases and healthy controls. PAF and precursors were also extracted from gastric biopsies from subjects with chronic gastritis and/or gastric colonization by Helicobacter pylori. Under basal conditions, hourly gastric PAF output increased in esophagitis and erosive gastritis, but no

Conditional deficiencies of ornithine or arginine.

Relative deficiencies of ornithine or arginine occur in the presence of excessive ammonia, excessive lysine, growth, pregnancy, trauma, or protein deficiency and malnutrition. Ammonia excess may occur in the presence of a normal liver when amino acid mixtures lacking ornithine, arginine, or citrulline are infused; when specific amino acids such as glycine are injected; when ammonium salts, urea, or urease are injected; or when the gastrointestinal tract contains an excess of protein, urea, or NH4+, as occurs after a gastrointestinal hemorrhage. In these states, ornithine is often rate-limiting

Insights into Remediation of Cd Contamination in Vegetable Soils by Red Mud-Modified Biochar.

Recently, heavymetal contamination in soil has been widely concerned. Herein, three types of biochar-based materials, including biochar (BC), Fe-modified biochar (Fe-BC), and red mud-modified biochar (RM-BC), were prepared and characterized, and the effects of BC, Fe-BC, and RM-BC on the bioavailability of Cd in vegetable soils were studied by pot experiments. Results showed that the RM-BC treatment exhibited better immobilization effect on soil Cd than BC and Fe-BC, and reduced the bioavailability of soil Cd and effectively inhibited the migration of Cd from soil to the edible parts of onion

The gut-kidney axis in chronic kidney disease: a vicious cycle of microbial dysbiosis and uremic toxin accumulation.

The progression of Chronic Kidney Disease (CKD) is governed by a pathogenic interplay between the host and the gut microbiota, a relationship encapsulated in the gut-kidney axis. This review synthesizes current knowledge on gut microbial alterations, metabolic reprogramming, and immunopathological mechanisms in CKD. Patients with CKD exhibit reduced microbial diversity, depletion of beneficial butyrate-producing bacteria, and enrichment of pathogenic taxa harboring urease, tryptophanase, and tyrosine phenol-lyase. These compositional shifts drive overproduction of protein-bound uremic toxins (

Phytochemicals as Natural Therapeutics Against Helicobacter pylori: A Comprehensive Review.

Helicobacter pylori is considered as a high-priority pathogen by World Health Organization which causes chronic gastritis and peptic ulcer disease and gastric carcinoma. The organism uses several methods to establish its disease-causing ability through urease production CagA protein synthesis biofilm development and ongoing gastric tissue damage. The rising rates of antibiotic resistance have decreased the success rate of standard treatment methods that use proton pump inhibitors and other eradication protocols. The plant-based compounds provide immunomodulatory properties together with gastro

Beyond the stomach: the association between Helicobacter pylori and the spectrum of digestive cancers.

Helicobacter pylori (H. pylori) is a Group 1 gastric carcinogen increasingly implicated in extragastric digestive malignancies. This review synthesizes evidence on its role in liver, biliary, esophageal, colorectal, and pancreatic cancers. Based on its unique spiral morphology, flagellar motility bundle, and urease activity-mediated acidic niche adaptation, H. pylori disrupts host cellular homeostasis through multifactorial virulence mechanisms involving CagA/VacA synergy, and exploits antigenic variation and immunomodulatory strategies to achieve persistent gastric mucosal colonization and ch

Harnessing the biomolecular mechanisms of marine biomineralisation for carbon sequestration.

Anthropogenic activities, primarily fossil fuel combustion, have increased atmospheric carbon dioxide (CO2) levels and climate change effects. Carbon dioxide removal (CDR) is now widely accepted as essential in all pathways to limit global warming in line with the Paris Agreement. Biomineralisation offers compelling and promising natural pathways for durable carbon sequestration by converting CO₂ into stable carbonate minerals, a process driven by a suite of biomolecules within bio-calcifying organisms. These processes are orchestrated by biomolecules such as proteins (e.g., carbonic anhydrase

Simultaneous stabilization of high-concentration multi-heavy metal(loid)s co-contaminated soil using Fe/Mn (hydr)oxide-humic acid composites:A novel approach towards sustainable environmental remediation.

In this work, a green and highly efficient functional material (FMHs) was developed through the precise optimization of Fe2+, Fe3+, Mn2+, and humic acid (HA) ratios. The stabilization efficacy of FMHs on the high-concentration Cd-Pb-Cu-Zn-As-Sb co-contaminated soil and its potential ecological impacts were comprehensively evaluated from multiple perspectives. After 60 days of soil remediation with 5 wt% FMHs, the extractable Cd, Pb, Cu, Zn, As, and Sb were reduced by 59.06 %, 99.83 %, 92.60 %, 92.43 %, 98.06 %, and 77.04 %, respectively. These reductions were primarily attributed to the FMHs-d
