Nonsyndromic hearing impairment is associated with a mutation in DFNA5.

Nonsyndromic hearing impairment is one of the most heterogeneous hereditary conditions, with more than 40 loci mapped on the human genome, however, only a limited number of genes implicated in hearing loss have been identified. We previously reported linkage to chromosome 7p15 for autosomal dominant hearing impairment segregating in an extended Dutch family (DFNA5). Here, we report a further refinement of the DFNA5 candidate region and the isolation of a gene from this region that is expressed in the cochlea. In intron 7 of this gene, we identified an insertion/deletion mutation that does not

Audiological and haematological studies on the Todas of Nilgiris.

A detailed audiological study combined with certain haematological investigations which were never previously done was undertaken on Todas, a small vanishing tribe of Nilgiris, South India. Ten per cent of the Todas studied exhibited otosclerosis which, though lower than that of a previous study, is still a very high percentage when compared to the general population. The incidence of chronic suppurative otitis media in Todas is markedly low (1-6 per cent) and 13-3 per cent of Todas had sensorineural hearing loss. Evaluation of hearing thresholds at various frequencies in the different age gro

Onset aging conditions of adults with an intellectual disability associated with primary caregiver depression.

Caregivers of adults with an intellectual disability experience depressive symptoms, but the aging factors of the care recipients associated with the depressive symptoms are unknown. The objective of this study was to analyze the onset aging conditions of adults with an intellectual disability that associated with the depression scores of their primary caregivers. A cross-sectional survey was administered to gather information from 455 caregivers of adults with an intellectual disability about their symptoms of depression which assessed by a 9-item Patient Health Questionnaire (PHQ-9). The 12

Effect of metabolic presbyacusis on cochlear responses: a simulation approach using a physiologically-based model.

In the presented model, electrical, acoustical, and mechanical elements of the cochlea are explicitly integrated into a signal transmission line where these elements convey physiological interpretations of the human cochlear structures. As a result, this physiologically-motivated model enables simulation of specific cochlear lesions such as presbyacusis. The hypothesis is that high-frequency hearing loss in older adults may be due to metabolic presbyacusis whereby age-related cellular/chemical degenerations in the lateral wall of the cochlea cause a reduction in the endocochlear potential. The

Hormone replacement therapy diminishes hearing in peri-menopausal mice.

We recently discovered that progestin in hormone replacement therapy (HRT) for post-menopausal women has detrimental effects on the ear and central auditory system [Guimaraes, P., Frisina, S.T., Mapes, F., Tadros, S.F., Frisina, D.R., Frisina, R.D., 2006. Progestin negatively affects hearing in aged women. Proc. Natl. Acad. Sci. - PNAS 103, 14246-14249]. To start determining the generality and neural bases of these human findings, the present study examined the effects of combination HRT (estrogen+progestin) and estrogen alone on hearing in peri-menopausal mice. Specifically, auditory brainste

Auditory processing in aging listeners.

Many aspects of auditory processing in aging listeners are poorly understood. Methodological problems encountered with the aging population, ranging from ear canal collapse to a conservative criterion, have rarely been considered in past research. The relation between pure-tone thresholds and other aspects of auditory functions is unclear. There have been few studies of frequency and temporal analysis in aging listeners. There is disagreement on the effect of age on speech discrimination abilities in various listening conditions, and the cause of speech perception problems in aging listeners h

A machine-learning-based approach to predict early hallmarks of progressive hearing loss.

Machine learning (ML) techniques are increasingly being used to improve disease diagnosis and treatment. However, the application of these computational approaches to the early diagnosis of age-related hearing loss (ARHL), the most common sensory deficit in adults, remains underexplored. Here, we demonstrate the potential of ML for identifying early signs of ARHL in adult mice. We used auditory brainstem responses (ABRs), which are non-invasive electrophysiological recordings that can be performed in both mice and humans, as a readout of hearing function. We recorded ABRs from C57BL/6N mice (6

Expression of G9a in Auditory Cortex Is Downregulated in a Rat Model of Age-Related Hearing Loss.

G9a is essential for dendritic plasticity and is associated with neurological disorders. The possible relationship between age-related hearing loss and G9a expression in the auditory cortex has not been fully explored. This study aimed to understand the expression patterns of G9a-mediated histone methylations in the auditory cortex during aging. Using immunofluorescence and western blotting, we demonstrated that a significant reduction in G9a expression observed in the auditory cortex of 24-month-old rats compared to 3-month-old rats, was associated with remarkable hearing threshold elevation

A DPOAE assessment of outer hair cell integrity in ears with age-related hearing loss.

Distortion product otoacoustic emissions (DPOAEs) were used to assess outer hair cell (OHC) integrity in human ears with age-related hearing loss. Sound pressure measurements were made in the ear canal over the stimulus range 40-90 dB SPL (L2), with L1 = 0.45*L2 + 44 with F2 = 2 and 3 or 4 kHz. Model-generated DPOAE I/O functions were fit to DPOAE data to quantify the contribution of loss of nonlinearity (OHC loss) to the hearing loss. Results suggest OHC loss as a contributing cause of age-related hearing, regardless of audiogram configuration. It seems likely that OHC and strial pathology co

Factors associated with third-party disability in spouses of older people with hearing impairment.

OBJECTIVES: This study had two aims: (1) to describe the extent of third-party disability in a sample of spouses of older people with hearing impairment, and (2) to investigate factors associated with third-party hearing disability. Third-party disability is defined as the disability and functioning of family members as a result of the health condition of their significant other. DESIGN: One hundred older spouses who had partners with hearing impairment participated. All assessments were self-reported, and included a set of measures (the 36-item Significant Other Scale for Hearing Disability,

Recent advances in the study of age-related hearing loss: a mini-review.

Hearing loss is a common age-associated affliction that can result from the loss of hair cells and spiral ganglion neurons (SGNs) in the cochlea. Although hair cells and SGNs are typically lost in the same cochlea, recent analysis suggests that they can occur independently, via unique mechanisms. Research has identified both environmental and genetic factors that contribute to degeneration of cochlear cells. Additionally, molecular analysis has identified multiple cell-signaling mechanisms that likely contribute to pathological changes that result in hearing deficiencies. These analyses should

Normal adult aging and the contextual influences affecting speech and meaningful sound perception.

It is widely accepted that hearing loss increases markedly with age, beginning in the fourth decade ISO 7029 (2000). Age-related hearing loss is typified by high-frequency threshold elevation and associated reductions in speech perception because speech sounds, especially consonants, become inaudible. Nevertheless, older adults often report additional and progressive difficulties in the perception and comprehension of speech, often highlighted in adverse listening conditions that exceed those reported by younger adults with a similar degree of high-frequency hearing loss (Dubno, Dirks, & Morga

Separate and combined effects of Sod1 and Cdh23 mutations on age-related hearing loss and cochlear pathology in C57BL/6J mice.

Both the ahl allele of Cdh23 and the null mutation of Sod1 have been shown to contribute to age-related hearing loss (AHL) in mice, but mixed strain backgrounds have confounded analyses of their individual and combined effects. To test for the effects of Sod1 deficiency independently from those of Cdh23(ahl), we produced mice with four digenic genotypes: Sod1(+/+)Cdh23(ahl)(/ahl), Sod1(+/+)Cdh23(+/+), Sod1(-/-)Cdh23(ahl)(/ahl), and Sod1(-/-)Cdh23(+/+), all on a uniform C57BL(/)6J strain background. We assessed hearing loss by ABR threshold measurements and evaluated cochlear pathologies in age

Association between age-related hearing loss and stroke in an older population.

BACKGROUND AND PURPOSE: Very few studies have investigated the association between hearing loss and stroke. A recent article in Stroke reported an increased incidence of stroke among patients with sudden hearing loss over a 5-year follow-up period. Our study aimed to explore this association among subjects with age-related hearing loss from a representative population. Further, we looked at the association between severity of hearing loss and risk of stroke in older persons, acknowledged as a limitation by the authors of the Stroke report. METHODS: The Blue Mountains Hearing Study is a popula

The burdens of age-related and occupational noise-induced hearing loss in the United States.

OBJECTIVES: Aging and noise are generally considered the most common causes of adult hearing loss in developed countries. This study estimates the contributions of aging and occupational noise in the United States. DESIGN: A model of hearing loss burden in American adults was constructed using data from the Census Bureau, from an international standard that predicts age-related and noise-induced hearing loss (ISO-1999), from the American Medical Association method of determining hearing impairment, and from sources estimating the distribution of occupational noise exposure in different age an

Prepulse inhibition of the startle response in mice: relationship to hearing loss and auditory system plasticity.

Prepulse inhibition was used with C57BL/6J (C57) mice to assess behavioral-perceptual correlates of previously demonstrated physiological changes in the central auditory system associated with age-related hearing loss. Normal-hearing CBA/CaJ (CBA) mice and DBA/2J (DBA) mice, which exhibit extremely rapid hearing loss, were also tested. Tone prepulse stimuli (S1s) were presented 100 ms prior to a startle-evoking noise stimulus (S2), and a decrease in startle amplitude served as the measure of startle modification. As high-frequency hearing declined in C57 mice between 1 and 12 months of age, th

Physiology of the aged Fischer 344 rat inferior colliculus: responses to contralateral monaural stimuli.

1. Presbycusis, age-related hearing loss, is an ever increasing problem in our aging society. It involves changes in both the peripheral and central portions of the auditory system. The inferior colliculus (IC) has been shown to display age-related changes including decreased gamma-aminobutryic acid (GABA) levels, decreased glutamate decarboxylase levels, and decreased binding by GABAB receptors, as well as rearrangement of axon terminals in aging Fischer 344 (F344) rats. Age-related physiological changes have also been noted in the ICs of C57 and CBA mice. 2. Given the age-related alterations
