Chronic inhalation toxicity and carcinogenicity study of methyl bromide in Wistar rats.

The toxicity and carcinogenicity of methyl bromide (MeBr) were studied in male and female Wistar rats exposed by inhalation to 0, 3, 30 or 90 ppm MeBr 6hr/day, 5 days/wk for 29 months. After 13, 52 and 104 wk ten rats/sex/group were killed to provide interim information. Body weights, clinical signs, haematology, biochemistry and gross and microscopic pathology were studied. Mortality was increased by wk 114 in the 90-ppm group. Body weights in males and females of the 90-ppm group were lower than those of the controls throughout the study. Increased incidences of degenerative and hyperplastic

Dual-Solvent SPE-UPLC-MS/MS Method for Quantifying 17 Organophosphate Flame Retardants in Environmental Waters.

RATIONALE: Organophosphate esters (OPEs) have emerged as dominant flame retardants following the phase-out of brominated alternatives, highlighting the need for accurate quantification due to their associated toxicity. Comprehensive analysis of OPEs, however, is challenging due to the wide polarity range of congeners and issues with co-elution. To address these challenges, we developed an ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for the simultaneous quantification of 17 OPEs. METHODS: Using an ACQUITY UPLC BEH C18 column with sub-2-μm particles, we

Long-Term Adverse Effects of Chlorinated Saline Primary Sewage Effluent and Halo-Nitrophenolic Disinfection Byproducts on a Marine Polychaete.

The practice of flushing toilets with seawater in coastal cities can result in bromide- and iodide-rich saline sewage effluent, which generates brominated and iodinated aromatic disinfection byproducts (DBPs) with relatively high developmental toxicity during chlorine disinfection. While the acute developmental effects of such DBPs are increasingly documented, their long-term impacts remain understudied. In this study, we investigated the developmental effects of saline primary sewage effluent on the marine polychaete Platynereis dumerilii (located at the bottom of the marine food chain) durin

An in Vitro Comparison of the Effects of Tetrabromobisphenol A and Tetrabromobisphenol S on Threshold-Like Hemolysis in Human Erythrocytes.

BACKGROUND/AIMS: The use of tetrabromobisphenol A (TBBPA) as a brominated flame retardant has raised growing concerns of its potential toxicity due to its widespread use and persistence. The present study compares the threshold-like mechanism of hemolysis induced by TBBPA with that of its sulfone analog, tetrabromobisphenol S (TBBPS), using an in vitro model of human erythrocytes. METHODS: Isolated red blood cells (5% hematocrit) obtained from healthy donors were exposed to TBBPA (10-50 µg/mL) or TBBPS (10-100 µg/mL) for 24 hours at 37°C. Hemolysis, methemoglobin formation, lipid peroxidation

Associations of plasma persistent organic pollutants with brain atrophy, cognitive decline and risk of dementia in older adults.

BACKGROUND: Persistent Organic Pollutants (POP), including polychlorinated biphenyls (PCBs) and organochlorine pesticides, are established neurotoxicants in experimental models; yet it remains uncertain whether exposures in the general population increase the risk to develop brain aging pathologies. We assessed the prospective associations of plasma POP concentrations with three dementia-related outcomes in a population-based cohort of older adults. METHODS: Analyses included 515 participants from the Three-City Study, free of dementia at baseline at the time of blood measurements (1999-2000,

High-resolution mass spectrometry recognized Tetrabromobisphenol A bis (2,3-dibromo-2-methylpropyl ether) (TBBPA-DBMPE) as a contaminant in sediment from a flame retardant manufacturing factory.

Information on contamination status of tetrabromobisphenol, and triazine-based novel brominated flame retardants (NBFRs) in sediment environment is very rare. Here, by use of high performance liquid chromatography coupled with quadrupole orbitrap mass spectrometry (HPLC-Q-Orbitrap/MS), we developed an analytical method for determination of three tetrabromobisphenol, and two triazine-based FRs in sediment samples. By applying this method for analysis of n = 6 sediment samples from a flame retardant manufacturing factory, we observed that total concentrations of 6 NBFRs (∑6NBFRs) ranged from 7.4

Activation of persulfate by a layered double oxide supported sulfidated nano zero-valent iron for efficient degradation of 2,2',4,4'-tetrabromodiphenyl ether in soil.

The nano zero-valent iron (nZVI) activated persulfate (PS) is recognized as a promising approach to degrade 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), which is ubiquitous in the soil at electronic waste sites. However, all the reported studies were performed in liquids, gaps in the real behaviour and microbial contribution to the degradation of BDE-47 in soil media need to be urgently filled. The removal efficiency of BDE-47 is low using traditional nZVI as activator because of its aggregation and corrosion. Herein, we designed a novel layered double oxide supported sulfidated nano zero-vale

Chemical uptake into silicone wristbands over a five day period.

Silicone wristbands are a noninvasive personal exposure assessment tool. However, despite their utility, questions remain about the rate at which chemicals accumulate on wristbands when worn, as validation studies utilizing wristbands worn by human participants are limited. This study evaluated the chemical uptake rates of 113 organic pollutants from several chemical classes (i.e., polychlorinated biphenyls (PCB), organophosphate esters (OPEs), alkyl OPEs, polybrominated diphenyl ethers (PBDEs), brominated flame retardants (BFR), phthalates, pesticides, and polycyclic aromatic hydrocarbons (PA

Algae impacted drinking water: Does switching to chloramination produce safer drinking water?

Harmful algal (cyanobacterial) blooms (HABs) are increasing throughout the world. HABs can be a direct source of toxins in freshwater sources, and associated algal organic matter (AOM) can act as precursors for the formation of disinfection by-products (DBPs) in drinking water. This study investigated the impacts of algae on DBP formation using treatment with chloramine, which has become a popular disinfectant in the U.S. and in several other countries because it can significantly lower the levels of regulated DBPs formed. Controlled laboratory chloraminations were conducted using live field-c

Combined chemical exposure using exposure loads on human biomonitoring data of the 4th Flemish Environment and Health Study (FLEHS-4).

To improve our understanding of internal exposure to multiple chemicals, the concept exposure load (EL) was used on human biomonitoring (HBM) data of the 4th FLEHS (Flemish Environment and Health Study; 2016-2020). The investigated chemicals were per- and polyfluoroalkyl substances (PFASs), bisphenols, phthalates and alternative plasticizers, flame retardants, pesticides, toxic metals, organochlorine compounds and polycyclic aromatic hydrocarbons (PAHs). The EL calculates "the number of chemicals to which individuals are internally exposed above a predefined threshold". In this study, the 50th

Increased Flame Retardancy of Enzymatic Functionalized PET and Nylon Fabrics via DNA Immobilization.

Poly(ethylene terephthalate) (PET) and nylon find their main applications in working clothes, domestic furniture and as indoor decoration (curtains and carpets). The increasing attention on healthy lifestyle, together with protection and safety, gained a strong interest in today's society. In this context, reducing the flammability of textiles has been tackled by designing flame retardants (FRs) able to suppress or delay the flame propagation. Commercially available FRs for textiles often consist of brominated, chlorinated and organo-phosphorus compounds, which are considered a great concern f

Impact of wastewater infrastructure upgrades on the urban water cycle: Reduction in halogenated reaction byproducts following conversion from chlorine gas to ultraviolet light disinfection.

The municipal wastewater treatment facility (WWTF) infrastructure of the United States is being upgraded to expand capacity and improve treatment, which provides opportunities to assess the impact of full-scale operational changes on water quality. Many WWTFs disinfect their effluent prior to discharge using chlorine gas, which reacts with natural and synthetic organic matter to form halogenated disinfection byproducts (HDBPs). Because HDBPs are ubiquitous in chlorine-disinfected drinking water and have adverse human health implications, their concentrations are regulated in potable water supp

Matrix solid-phase dispersion combined with gas chromatography-mass spectrometry for the determination of fifteen halogenated flame retardants in mollusks.

This study presents the development and validation of a new analytical method for the simultaneous determination of fifteen analytes classified as halogenated flame retardants (HFRs) - nine brominated diphenyl ethers (BDEs) and six novel HFRs - in different kinds of mollusks using matrix solid-phase dispersion (MSPD) followed by gas chromatography coupled to negative chemical ionization-mass spectrometry (GC-NCI-MS). The proposed method is the first one developed for such a broad range of HFRs in aquatic biota, featuring several advantages, including low solvent and sample intake, simplicity o

Bioaccumulation and metabolism of polybrominated diphenyl ethers in carp (Cyprinus carpio) in a water/sediment microcosm: important role of particulate matter exposure.

Microcosms were built up to simulate a pond system with polybrominated diphenyl ether (PBDE) contaminated sediment and bioorganisms. The microcosms were divided into groups A and B. In group A, both benthic invertebrates (tubificid worms) and carp (Cyprinu carpio) were added, while in group B, only fish were added. After exposure for 20 d, the fish were sampled (exposure I). A net was fixed in the microcosms, and new fish were added (exposure II). These fish were prohibited from contacting the sediment by the net, and the accumulation and depuration of PBDEs in the fish were investigated. Amon

Inhibition of human placental aromatase activity by hydroxylated polybrominated diphenyl ethers (OH-PBDEs).

Polybrominated diphenyl ethers (PBDEs) are widely used as flame retardants in many different polymers, resins and substrates. Due to their widespread production and use, their high binding affinity to particles, and their lipophilic properties, several PBDE congeners can bioaccumulate in the environment. As a result, PBDEs and their hydroxylated metabolites (OH-PBDEs) have been detected in humans and various wildlife samples, such as birds, seals, and whales. Furthermore, certain OH-PBDEs and their methoxylated derivatives (MeO-PBDEs) are natural products in the marine environment. Recently, o

CYP2E1-catalyzed oxidation contributes to the sperm toxicity of 1-bromopropane in mice.

1-bromopropane (1-BrP) induces dose- and time-dependent reproductive organ toxicity and reduced sperm motility in rodents. The contribution of cytochrome P4502E1 (CYP2E1) to both 1-BrP metabolism and the induction of male reproductive toxicity was investigated using wild-type (WT) and Cyp2e1-/- mice. In gas uptake inhalation studies, the elimination half-life of [1,2,3-(13)C]-1-BrP was longer in Cyp2e1-/- mice relative to WT (3.2 vs. 1.3 h). Urinary metabolites were identified by 13C nuclear magnetic resonance. The mercapturic acid of 1-bromo-2-hydroxypropane (2OHBrP) was the major urinary met
